Protective Effects of Seonpyejeongcheon-tang on Elastase-Induced Lung Injury in Mice (Elastase 매개성 폐조직 손상에 대한 선폐정천탕(宣肺定喘湯)의 보호효과)

  • Objectives : This study aimed to evaluate the protective effects of Seonpyejeongcheon-tang (SJT) on elastase-induced lung injury. Materials and Methods : The extract of SJT was treated to A549 cells and an elastase-induced lung injury mouse model. Then, various parameters such as cell-based cytoprotective activity and histopathological findings were analyzed. Results : SJT showed a protective effect on elastase-induced cytotoxicity in A549 cells. This effect was correlated with analysis for caspase 3 levels, collagen and elastin contents, protein level of cyclin B 1, Cdk1, and Erk1/2, and gene expression of TNF-$\alpha$ and IL-$1{\beta}$ in A549 cells. SJT treatment also revealed a protective effect on elastase-induced lung injury in mouse model. This effect was evidenced via histopathological findings, including immunofluoresence stains against elastin, collagen, and caspase 3, and protein levels of cyclin B1, Cdc2, and Erk1/2 in lung tissue. Conclusion : These data suggest that SJT has pharmaceutical properties on lung injury. This study thus provides scientific evidence for the efficacy of SJT for clinical application to patients with chronic obstructive pulmonary disease.

Histopathologic Features in Animal Model of Atopic Dermatitis Induced by Topical Application of Oxazolone

  • Animal models of atopic dermatitis (AD) are widely used to investigate therapeutic effects of candidates for AD. However, the characteristics of each model are not fully understood. This study was designed to compare the animal models of dermatitis induced by dinitrofluorobenzene (DNFB) and oxazolone (Ox). We investigated the effects of DNFB and Ox on skin thicknesses and weights as well as skin lesions associated with AD such as scale, crust and erythematous eruption, and histopathological changes such as hyperkeratosis, dermal and epidermal hyperplasia and immune cell infiltration in inflamed tissues. Multiple application of 0.5% Ox onto the skin increased skin thickness and weight compared to those of DNFB treated mice, as well as those of normal mice. In addition, topical application of DNFB induced marked scale, crust and erythematous eruption, while Ox induced erythematous eruption and mild scale and crust. Histopathological examination revealed that 0.5% Ox induced marked hyperplasia in the dermis and epidermis, large vesicles, spongiotic changes, mild hyperkeratosis and immune cell infiltration in balb/c mice. These data suggest that multiple applications of Ox can induce chronic AD like dermatitis in balb/c mice.

Effects of Aloe vera on Hematology and Blood Chemical Values of Cobalt-60 Gamma Irradiated Mice (Aloe vera가 Cobalt-60 감마선을 조사한 마우스의 혈액상 및 혈액화학치에 미치는 영향)

  • This study was performed to investigate the effects of Aloe vera administration(500mg/kg. B.W., s.i.d., P.O.) on hematology, blood chemical values and histopathological changes of spleen in cobalt-60 gamma Irradiated mice. 1. Total leucocyte counts of aloe administered groups were less decreased and more rapidly returned to normal level than those of control group. 2. Lymphocyte counts were rapidly decreased on the 1st day after irradiation in all experimental groups, but the counts were less decreased in aloe administered groups than those of control group. 3. Erythrocyte counts, hemoglobin values and hematocrit values of aloe administered groups were maintained within normal level, but these values showed lower values on the 14th day after irradiation in control group then returned to normal level on the 21th day after irradiation. 4. There were no differences between aloe administered groups and control group In LDH, SGOT and glucose values. 5. Histopathological changes of spleen in aloe administered groups showed less lymphocytic necrosis and depletion than those of control group.

Effects of Buckwheat on Super Oxide Dismutase Activity and Histopathological Change of Rats Fed with Hypercholesterolemic Diet (메밀이 흰주 SOD 활성도와 장기에 미치는 영향)

  • This study was carried out to investigate the effects of rutin from buckwheat vegetable on rats fed with hypercholesterolemic diets. Sprague-Dawley male rats were fed the diets providing various level of buckwheat vegetable for five weeks. Body weight increased during the experimental period. Body weight gain decreased by addition of cholesterol group (C-C). Rutin feeding group (T-l) and buckwheat vegetable feeding group (T-2, T-3, and T-4) showed the reduction of weight gain. Total cholesterol level decreased whereas HDL-cholesterol level increased with the increasing amount of buckwheat vegetable added. The value of thiobarbituric acid in serum decreased with increasing buckwheat vegetable added. The histopathological changes of the kidney were different among the experimental groups. Kidney of the rats fed with cholesterol added diet showed severe calcification.

Comparison of Single-Dose Toxicity by Intravenous Infusion or Bolus Injection with CKD-602, a Camptothecin Anticancer Agent in Rats (II): Hematological and Serum Biochemical, and Histopathological Changes

  • The toxicity of CKD-602 was investigated at doses of 3, 9, and 27 mg/kg in rats, when the same total dose of CKD-602 was administered over 24 hr-continuous infusion or bolus injection. At 3 and 9 mg/kg, the 24-hr infusion group showed a more decreased WBC count on day 3, compared with the bolus group. Administration of CKD-602 caused more toxic effects such as the significant decreases of RBC counts, hematocrit, hemoglobin, and platelet count on day 7 post-administarion in the 24-hr infusion group than in the bolus group. Administration of CKD-602 also caused histopathological changes such as extramedullary hemopoiesis of liver and spleen, hyperplasia of femoral bone marrow, and caecal dilation. These toxic effects were more severe in the 24-hr infusion group than in the bolus injection group, indicating that the toxicity of CKD-602 may be dependant upon the duration of administration.

  • Spaceflight induces a number of cardiovascular physiological alterations. To study adaptations to microgravity on Earth, the tail-suspended, hindlimb-unloaded rat model has been used to simulate the effects of microgravity. Despite the extensive use of this model to infer physiological adaptations of many organs to microgravity, little information has been obtained on the effect of tail suspension(TS) on cardiac adaptations in the rat. This study was aimed to investigate the effects of simulated microgravity on the rat myocardium using the TS model. Twenty-four male Sprague-Dawley rats were randomly assigned to 3 experimental groups(1, 7 and 14 days of TS) and a control group. A microscopic examination was performed to assess histopathological changes in the myocardial morphology. The hearts from the control group, the 1 day-TS rats and the 7 day-TS rats revealed no evident abnormalities in cardiomyocyte size and morphology. At day 14 of TS, in contrast, the ventricular cardiomyocytes appeared more separated from each other and were slightly smaller in size compared with those of the control group. Also seen were scattered areas exhibiting focal disorganization of muscle fibers and some degenerating cardiomyocytes, of which the nuclei had become pyknotic or disappeared. In this study, we demonstrated that the ventricular cardiomyocytes underwent degeneration and atrophy at the microscopic level during exposure to simulated microgravity in TS rats.

Effects of Portulacae Herba (PH) on Allergic Contact Dermatitis (ACD) Induced by DNCB in Mice (마치현(馬齒莧)이 DNCB로 유발된 생쥐의 알레르기성 접촉피부염에 미치는 영향)

  • Objective : Portulacae Herba (PH) can clear away heat, detoxicate, cool down blood heat. The present study was designed to investigate the effects of PH on ACD induced by 2,4-dinitrochlorobenzene (DNCB) in mice. Method : In this experiment, the effects of PH on changes in body weights, thicknesses and weights of ear, thicknesses of dorsum skin, degree of symptoms on the dorsum skin, histopathological changes of ear and dorsum skin, spleen weights, TNF-${\alpha}$ and IL-10 in serum were observed. And the effects on the proliferation rates of splenocytes were also investigated in vitro and in vivo. Results : 1. PH spread (PHS) group and PH spread plus administered (PHS+Adm) group didn't show any changes in weight and thickness of ear. 2. But in PHS and PHS+Adm group, thickness of dorsum skin decreased significantly. 3. And PHS and PHS+Adm group showed meaningful effectiveness on ACD symptoms like erythema, desquamation and keratinization. 4. In histopathological observation, hyperplasia and edema of spongy tissues were remarkably diminished in PHS and PHS+Adm group. 5. And PH reduced the proliferation rates of splenocytes in vivo and vitro study. Conclusions : PH effectively decreased symptoms of ACD in this study, therefore PH can be one of medicinal ingredients to treat ACD.

Effects of Ginseng Radix on the Rat Hypothyroidism Induced by PTU (6-n-propyl-2-thiouracil) (인삼이 PTU (6-n-propyl-2-thiouracil)로 유발된 Rat 갑상선 기능저하증에 미치는 영향)

  • Object : This study was to evaluate the effect of Ginseng Radix, aqueous extracts of the root part of Panax ginseng on the 6-n-propyl-2-thiouracil(PTU)-induced rat hypothyroidism. Methods : Aqueous extracts of Ginseng Radix(GR; yield = 11.70%) were administered, once day for 42 days from 2 weeks before start of PTU treatment as an oral dose of 300 and 150 mg/kg(body weight), and hypothyroidism was induced by daily subcutaneous treatment of PTU 10mg/kg for 28 days. The changes in the body weight, thyroid gland weights, serum levels of thyroid hormone-thyroid stimulating hormone(TSH), tri-iodothyronine(T3) and thyroxine(T4), total cholesterol, low density lipoprotein(LDL), high density lipoprotein(HDL) and triglyceride, liver antioxidant defense system-lipid peroxidation, $H_2O_2$, superoxide dismutase(SOD) and catalase(CAT) were observed with histopathology of thyroid glands. Results : Results were compared with LevoT4 0.5mg/kg treated rats. GR extracts suppressed the decreases in the body weight, thyroid gland weights, T3 and T4, TG, liver $H_2O_2$ and SOD activities as results of PTU treatment. And GR extracts suppressed the increases of HDL contents, liver CAT activities, thyroid gland weight as results of PTU treatment. In addition, marked hyperplasia of follicular cells with decreases of follicular colloid contentsand sizes were demonstrated at histopathological inspections. However, these PTU-induced histopathological changes related to hypothyroidism were dramatically decreased by treatment of both different dosages of GR extract, respectively Conclusions : This study suggest that GR extracts have favorable effects on the thyroid hormone productions with beneficial effects on the hypothyroidism mediated by the modulatory effects on the antioxidant defense system.

Effects of Samhwangsasim-tang Gamibang on Atopic Dermatitis in Mice (삼황사심탕가미방(三黃瀉心湯加味方)이 생쥐에 유발된 아토피 피부염에 미치는 영향)

  • Objectives : Atopic Dermatitis (AD) is chronic skin disease characterized by allergic hypersensitivity reactions. Samhwangsasim-tang Gamibang(SHSST) can treat skin disease by cooling down blood heat, clearing away congenital fever and detoxicating. This study was conducted to investigate the effects of SHSST on AD induced by oxazolone in mice. Methods : We investigated the effects of SHSST on skin tickness, skin lesions, erythema and melanin index, water content and histopathological changes in vivo. The effects on body weights and spleen body weight ratio were also investigated in mice with AD. Results : In our results, oral administration of SHSST ameliorated skin lesion such as flare, erythema, exudate and petechia and lowered skin thickness induced by oxazolone. In addition, SHSST lowered erythema index and elevated water content compared to AD control respectively. In the histopathological observation, oral administration of SHSST prevented epidermal hyperplasia and spongiotic changes which is a hallmark of skin inflammation respectively. Finally, SHSST did not affect spleen/body weight ratio. Conclusions : These data imply that SHSST can ameliorate skin lesion of AD such as flare, erythema, exudate and petechia, and be used to treat AD patients with relative safety.
