운동 및 고지방식이가 흰쥐의 Carnitine 농도와 carnitine Palmitoyltransferase-I mRNA 수준에 미치는 영향 (Effects of Exercise and/or High Fat Diet on Carnitine and Carnitine Palmitoyltransfersase-I mRNA Levels in Rats)

  • The effect of exercise and/or high fat diet on carnitine status and carnitine palmitoyltransferase I(CPT I) level were investigated in Weanling Sprague Dawley rats. The rats were fed an AIN 76 diet or a modified high fat AIN diet, supplemented with 35% corn oil, for 31 days. During the 31 day period half of the animals in each dietary group were exercised on a treadmill for 90 minutes per day. Carnitine concentrations were determined in plasma and liver and CPT I mRNA levels were measured by Northern blot analysis with CPT I cDNA probe in livers of rats. Exercise rats gained less weight than non exercised rats during the study for high fat diet group. Exercise rats had a higher plasma acid soluble acylcarnitine and acid insoluble acylcarnitine concnetrations than non exercised rats for normal diet group. Exercise or high fat diet increased liver carnitine concentration, but a mixed effect was not shown. In exercised rats, CPT I mRNA levels increased significantly relative to those of nonexercised rats. CPT I mRNA levels also increased when compared high fat fed rats with those of normal diet fed rats. These data suggest that there is a correlation between carnitine concen trations and CPT I mRNA levels and that CPT I can be regulated at the transcriptional level by exercise and/or high fat diet.

  • Polycrystalline silicon thin film transistors (poly-Si TFTs) are used in a wide variety of applications, and will figure prominently future high-resolution, high-performance flat panel display technology However, it was very difficult to fabricate high performance poly-Si TFTs at a temperature lower than 300$^{\circ}C$ for glass substrate. Conventional process on a glass substrate were limited temperature less than 600$^{\circ}C$ This paper proposes a high temperature process above 750$^{\circ}C$ using a flexible molybdenum substrate deposited hydrogenated amorphous silicon (a-Si:H) and than crystallized a rapid thermal processor (RTP) at the various temperatures from 750$^{\circ}C$ to 1050$^{\circ}C$. The high temperature annealed poly-Si film illustrated field effect mobility higher than 30 $\textrm{cm}^2$/Vs, achieved I$\sub$on//I$\sub$off/ current ratio of 10$^4$ and crystall volume fraction of 92%. In this paper, we introduce the new TFTs Process as flexible substrate very promising roll-to-roll process, and exhibit the properties of high temperature crystallized poly-Si Tn on molybdenum substrate.

다양한 산소분압에서 소결한 NdBCO 초전도체의 열적 안정성 (Thermal Stability of Superconductor NdBCO Sintered at Various Oxygen Partial Pressures)

  • The $Nd_{1+x}Ba_{2-x}Cu_3O_{7-{\delta}}$(Nd123) superconductor exhibits high performance in high magnetic field and high temperature. We have studied phase stability for Nd123 under reduced oxygen partial pressure and various heat-treatment conditions. The main phase is Nd123 and some samples contain small amounts of Nd422 depending on the temperature and oxygen partial pressure. The decomposition temperature decreases with decreasing oxygen partial pressure from $1052^{\circ}C(P(O_2)$=150 Torr) to about $845^{\circ}C(P(O_2)$=0.1 Torr). The liquidus line was steeper temperature with decreasing oxygen partial pressure. In same condition of oxygen partial pressure, the region of stable Nd123 phase was formed at slightly higher temperature than the region of stable YBCO phase.

The Signaling Pathways Involved in High Glucose-Induced Secretion of Insulin-Like Growth Factors (IGFs) and IGF Binding Proteins in Podocytes

  • It has been reported that the dysfunctions of podocytes are associated with the development of diabetic nephropathy. In addition, insulin-like growth factors (IGFs) are associated with the development of diabetic nephropathy. However, it is not yet known about the effect of high glucose on IGF-I, -II secretion, and IGF binding proteins (IGFBPs) expression in the podocytes. Thus, this study was conducted to examine the effect of high glucose on IGF system and its involvement of protein kinase C (PKC) and mitogen activated protein kinases (MAPKs) in podocytes. In this study, high glucose (25 mM) increased IGF-I and IGF-II secretion (P<0.05), which was blocked by SB 203580 (a p38 MAPK inhibitor) but not by PD 98059 (a p44/42 MAPK inhibitor). In addition, high glucose-induced stimulation of IGFs was blocked by bisindolylmaleimide I and staurosporine (protein kinase C inhibitors). High glucose also increased IGFBP-l expression, which was blocked by bisindolylmaleimide I and SB 203580. In conclusion, high glucose alters IGFs secretion and IGFBP expression via PKC and p38 MAPK pathways in podocytes.

Molecular Cloning of Insulin-like Growth Factor-I (IGF-I) and IGF-II Genes of Marine Medaka (Oryzias dancena) and Their Expression in Response to Abrupt Transfer from Freshwater to Seawater

  • Growth hormone (GH) is known as one of the main osmoregulators in euryhaline teleosts during seawater (SW) adaptation. Many of the physiological actions of GH are mediated through insulin-like growth factor-I (IGF-I), and the GH/IGF-I axis is associated with osmoregulation of fish during SW acclimation. However, little information is available on the response of fish IGF-II to hyperosmotic stress. Here we present the first cloned IGF-I and IGF-II cDNAs of marine medaka, Oryzias dancena, and an analysis of the molecular characteristics of the genes. The marine medaka IGF-I cDNA is 1,340 bp long with a 257-bp 5' untranslated region (UTR), a 528 bp 3' UTR, and a 555-bp open reading frame (ORF) encoding a propeptide of 184 amino acid (aa) residues. The full-length marine medaka IGF-II cDNA consists of a 639 bp ORF encoding 212 aa, a 109 bp 5' UTR, and a 416 bp 3' UTR. Homology comparison of the deduced aa sequences with other IGF-Is and IGF-IIs showed that these genes in marine medaka shared high structural homology with orthologs from other teleost as well as mammalian species, suggesting high conservation of IGFs throughout vertebrates. The IGF-I mRNA level increased following transfer of marine medaka from freshwater (FW) to SW, and the expression level was higher than that of the control group, which was maintained in FW. This significantly elevated IGF-I level was maintained throughout the experiment (14 days), suggesting that in marine medaka, IGF-I is deeply involved in the adaptation to abrupt salinity change. In contrast to IGF-I, the increased level of marine medaka IGF-II mRNA was only maintained for a short period, and quickly returned a level similar to that of the control group, suggesting that marine medaka IGF-II might be a gene that responds to acute stress or one that produces a supplemental protein to assist with the osmoregulatory function of IGF-I during an early phase of salinity change.

심근형 Troponin I의 증가로 급성 심근염의 진단을 받은 환아에서 정맥용 면역글로부린 치료 효과에 대한 연구 (A Study of the Level of Cardiac Troponin I in Patients with Clinically Suspected Acute Myocarditis Treated with Intravenous Gammaglobulin)

  • 목 적 : 급성 심근염이 의심되는 환아에서 심근 손상의 정도에 따라 나타나는 혈청 심근형 troponin I(cTnI)과 좌심실 구축률(LVEF)을 측정하여 심근염을 진단하고, 치료제인 정맥용 면역글로부린(IVGG)(2.0gm/kg)을 투여한 후, cTnI치와 LVEF의 변화를 추적 관찰하여 심근염에 대한 IVGG치료 효과 판정을 위해 본 연구를 시행하였다. 방 법 : 1995년 1월부터 1998년 6월까지 내원한 환아 중 심근염으로 생각되었던 환아의 혈청을 면역화학발광법을 이용하여 측정한 cTnI가 기준치 이상(정상<0.1ng/ml) 증가되고, 심초음파 검사상 LVEF(정상 64~83%)가 63% 이하로 저하된 한아 18명을 대상으로 IVGG 투여전 후 두 번 이상 혈청 cTnI와 LVEF를 측정하여 변화를 비교하였다. 대조군은 바이러스성 질환으로 생각된 환아중 심근염 소견이 없는 20명을 선정하여 cTnI를 측정하여 환아군과 비교하였다. 결 과 : 1) 환아군의 남녀 비는 1.3 : 1.0으로 남아에서 많았고, 환아의 평균 연령은 $27.2{\pm}12.9$개월이었다. 2) IVGG 투여 전 측정한 cTnI치는 환아군 18례에서 평균 $0.306{\pm}0.209ng/ml$로 증가되어 있었고, LVEF는 $60.1{\pm}1.6%$로 심기능이 저하되어 있었다. 3) IVGG투여 후 측정한 혈청 cTnI치는 증가되었던 18례 중 16례가 임상증상의 호전과 함께 정상으로 되었지만(p<0.05), 2례는 가와사끼병 환아로 임상 증상이 호전된 후에도 cTnI가 추적검사에서 지속적으로 증가되어 있었고, 좌심실 구축율(LVEF)은 모든 예에서 유의하게 회복되었다.(($71.4{\pm}3.7%$)(p<0.05). 4) 혈청 cTnI치는 대조군에 비하여 환아군에서 유의하게 증가되어 있었다(p<0.05). 5) 모든 환아에서 CRP, ESR, C3 등이 증가하는 급성기 염증반응을 볼 수 있었고 백혈구 증다증을 보인 경우는 56%(10/18)였다. 결 론 : 급성 심근염으로 생각된 환아의 혈청 cTnI치와 LVEF를 IVGG 투여 전 후 비교한 결과, IVGG가 급성 심근염에 유의하게 효과가 있고, cTnI와 LVEF가 심근염에 대한 진단과 치료 효과 판정에 유용한 방법이 될 수 있다고 생각되었다.

Influence of bending strain on $I_c$ Degradation Behavior in YBCO Coated Conductor Tapes processed using RABiTS/MOD

  • The critical current, $I_c$, degradation behaviors with bending strain were investigated in a commercially available YBCO coated conductor tape. In particular, the strain reversibility of $I_c$ and the influence of repeated bending on $I_c$ have been studied. Also, repeated bending at 77 K was done in order to understand the Ie behavior under cyclic bending strains. A reversible behavior of $I_c$ has been found up to a high bending strain of 1.60% for the RABiTS/MOD processed CC sample with copper reinforcement. Under repeated bending, the Ie showed a 95% $I_c$ retention up to 100 cycles for bending strains of 1.0% or less. The n-value behavior showed a good agreement with the $I_c$ degradation behavior, representing that any cracking did not occur on the YBCO film resulting from the reinforcement provided by the copper stabilizers.

Citrullinemia Type I 환자의 가족에서 발견된 새로운 Argininosuccinate Synthetase 유전자 돌연변이 (A Novel Argininosuccinate Synthetase Gene Mutation in a Korean Family with Type I Citrullinemia)

  • Citrullinemia는 요소 회로 이상으로 argininosuccinate synthetase의 결핍으로 기인한다. 저자들은 citrullinemia type I 환자와 그의 가족에서 새롭게 발견된 돌연변이를 경험하여 이를 보고하고자 한다. 환아는 광범위 신생아 선천성 대사이상 선별검사에서 citrulline이 고도의 증가 소견과 혈청 암모니아는 $982{\mu}mol/L$까지 증가 소견을 보였다. 혈청 아미노산 분석결과 citrulline 1,581 nmol/mL로 현저한 증가 소견을 보였으며 또한 소변 유기산 분석 검사결과 orotic acid가 3,566 mmol/mol Cr로 매우 증가된 소견을 보였다. Citrullinemia 확진을 위하여 환아와 가족에 대하여 ASS1 gene 검사를 시행하였다. 환아는 c.689G>C (p.G230A)와 c.892G>A (p.E298k)의 변이가 발견되어 ASS1 gene의 돌연변이에 의한 citrullinemia type I 으로 진단되었으며, 두 가지 돌연변이는 아직 국내에 보고된 적 없는 새로운 것으로 확인되었다. 국내에서 새롭게 발견된 citrullinemia type I 유전자를 보고하며 citrullinemia를 보인 경우 확진 및 유전상담을 위하여 가족 유전자 검사를 시행하는 것이 필요하다고 생각한다.

서울시 인근의 한강 본류 및 지류에 대한 하상 저질의 중금속오염 평가 (An Assessment of the Heavy Metal Contamination of Sediment in Main Stream and Tributaries of the Han River in the Vicinity of Seoul)

  • This study was performed to investigate and assess the heavy metal contamination of sediment in main stream and tributaries of the Han-river in the vicinity of Seoul. Sediment were sampled at eight locations of main stream and eighteen locations of tributaries in the spring, summer, and fall in 1993. Six heavy metals of Cd, Cu, Pb, Zn, Cr, and Hg, were assessed. For an assessment of heavy metal contamination in sediment, the assessment method of Hakanson was modified to estimate for Han-river, then classification method of Muller in sediment quality was applied. For an assessment of contamination in each heavy metal, Contamination factor C$_f^i$ which was defined the ratio of concentration of i heavy metal in investigated location to background concentration of i heavy metal in uncontaminated upper area was used. The sediment quality in each heavymetal was classified as follows if C$_f^i$<1, Class I for Non contamination, if 1 < C$_f^i$ < 3, Class II for Low contamination, if 3< C$_f^i$ <6, Class III for Moderate contamination, if 6< C$_f^i$ <12, Class IV for Considerable contamination, and if C$_f^i$ > 12, Class V for High contamination.(abbreviation)

Czochralski 법으로 성장시킨 단결정 Silicon Wafer에서의 표면 무결함층(Denuded Zone) 형성에 관한 연구(I) (The Study on the Denuded Zone Formation of Czochralski-grown Single Crystal Silicon Wafer (I))

  • This study is intended to make defect-free region, denuded zone at the silicon wafer surface for semiconductor device substrates. In this experiment, initial oxygen concentration of starting material CZ-grown silicon wafer, various heat treatment combinations, denuding ambient and the amounts of oxygen reduction were measured, and then denuded zone (DZ) formation and depth were investigated. In Low/High anneal (DZ formation could be achieved), the optimum temperature for Low anneal was 700$^{\circ}C$∼750$^{\circ}C$. In case of High anneal, with the time increased, DZ depth was increased at 1000$^{\circ}C$, 1150$^{\circ}C$ respectively, but on the contrary, DZ depth was decreased at low temperature 900$^{\circ}C$. As well, out-diffusion time below 2 hours was unsuitable for effective Gettering technique even though the temperature was high, and DZ formation could be achieved when initial oxygen concentration was only above 14 ppm in silicon wafer.
