The author studied the hemolytic action of chlorpromazine on rabbit erythrocytes and obtained the following results: 1. Chlorpromazine caused hemolysis in vitro. The hemolytic action of chlorpromazine was milder than that of saponin(Colleman and Bell Co ) 2. Cholesterol had no marked effect on chlorpromazine hemolysis. 3. Dextrose, albumin and blood plasma protected erythrocytes against chlorpromazine hemolysis. 4. The mechanism of chlorpromazine hemolysis seemed to be different from that of saponin hemolysis. 5. Chlorpromazine increased tile osmotic fragility of rabbit erythrocytes. 6. The intravenous injection of chlorpromazine caused hemolysis in vivo.
Low hemolysis is one of the key factors in the production of successful rotary blood pumps. It is, however, difficult to identify the areas where hemolysis occurs. Computational fluid dynamics(CFD) analysis enables the engineer to predict hemolysis on a computer Fluid dynamics in five different axial flow pumps was analyzed 3-dimensionally using CFD software. The impeller was rotated at a speed which supplied a flow of 5L/min at a pressure difference of 100mmHg. Changes in the turbulent kinetic energy along streamlines through the pumps were computed. The measured NIH and the calculated hemolysis(dHb/Hb) shoed a good correlation; NIH=0.0003(dHb/Hb) (r=0.90, n=6) in the range of NIH between 0.003 and 1.1. Intravascular hemolysis occurs in the majority of patient with mechanical valve prosthesis. The primary cause is mechanical trauma to red cells from turbulent blood flow through the prosthesis. Degree of hemolysis is dependent upon the type, size and material of valve and aggravated by paravalvular leakage. Clinically important hemolytic anemia is required medical management or consideration of reoperation. In severe hemolysis, reoperation is recommended without delay when seems to be renal failure. In this case, postoperative severe mechanical hemolysis was developed immediately after aortic valve replacement with St. Jude medical valve in a 13 year-old male patient. Neither significant paravalvular leakage nor valvular dysfunction was found through redo, but the mechanical valve was strongly suspected the cause of severe hemolysis. The St. Jude Medical valve was changed with Ionescu-Shiley bioprosthesis and any significant clinical problems were not noted through the postoperative course.
The hemolytic characteristics of amphotericin B-containing liposomes have been investigated in vitro. From the hemolysis of human erythrocytes against free and liposomal amphotericin B, the marked reduction in the toxicity of amphotericin B was observed by incorporating the drug in egg PC liposomes. For 45 min, free amphotericin B at $9.6\;{\mu}g/ml$ could completely lyse 2 wt% human erythrocytes. However, liposomal amphotericin B had essentially no lytic effect even in the range over $9.6\;{\mu}g/ml$. In the 66 hr-hemolysis experiment, liposomal amphotericin B showed the slowly hemolysing chracteristics during the experimental period regardless of the concentration of amphotericin B but rapid hemolysis only for 12 hr was observed in the case of free amphotericin B and the degree of hemolysis for 12 hr was maintained after that time. Also the hemolysing ability of liposomal amphotericin B at $4\;{\mu}g/ml$ was lower than that of free amphotericin B at the same concentration for 66 hr. On the other hand, the dependence of hemolysis on amphotericin B contents in egg PC liposomes was significant between 1.64 mole% amphotericin B-containing liposomes and 15.79 or 27.27 mole% amphotericin B-containing liposomes. But no marked difference in hemolysis was observed between 15.79 and 27.27 mole% amphotericin B-containing liposomes. Especially, cholesterol as an excipient in amphotericin B-containing liposomes significantly reduced the hemolysis of human erythrocyte. The degree of hemolysis in 5 mole% amphotericin B-containing liposomes was reduced to approximately 50% of value in the cholesterol-free liposomes by adding 50% cholesterol.
In order to investigate the phototoxicity of five phenothiazine derivatives and one thioxanthen derivative were examined by using in vitro method based on growth inhibition of Candida albicans and red blood cell hemolysis. Effects of the test compounds on RBCs were monitored with a spectrophotometer and a drug PI in the Candida albicans was calculated on the basis of the lowest concentration giving a yeast-free zone. All phenothiazines phototoxic in the red blood cell hemolysis method were positive in the yeast test except promethazine. It was also observed that toxic photo-products were formed by perphenazine and chlorpromazine in the red blood cell hemolysis.
Streptozotocin (STZ) is a naturally occurring nitrosoamide used extensively to produce diabetes in experimental animals. Since streptozotocin reportedly decrease deformability of red blood cells (RBC), we sought to investigate its potential toxicity at RBC. In addition to elevation blood glucose, 100mg/kg iv streptozotocin caused significant RBC hemolysis in female Sprague-Dawley at 48 hrs post treatment. Streptozotocin induced hemolysis was found to be dose and time-dependent` complete hemolysis required a relatively high streptozotocin concentration (500 mM, 4 hrs incubation), which is much more than in vivo dose.
The first successful percutaneous transcatheter occlusion technique for patent ductus arteriosus achived by Rashkind in 1977.Transcatheter occlusion with the Rashikind double umbrella device is now widely accepted as treatment for patent ductus arterisus. The reported complications include embolization of the device, psudocoarctation left pulmonary artery stenosis, residual shunts, and mechanical hemolysis. We report two cases of severe hemolysis after occlusion of PDA with Rashkind occluder.
The present studies were carried out to develope a simple quantitative analysis for saponins in ginseng by hemolysis. Erythrocytes from pig, rabbit, human and cattle were useful for this purpose. The purpose of this study was to find the extent of lipid peroxidation of erythrocytes in cigarette smokers, and to determine the relative effectiveness of $\beta$-carotene, canthaxanthin , and $\alpha$-tocopherol as antioxidants. Thirty smokers and 30 nonsmokers participated in this study . No significant differences according to age, sex, and height were shown. Cigarette smokers in this study had higher hemoglobin concentrations and more oxidation of hemoglobin than non-smokers. In addition, the erythrocytes of cigarette smokers had significantly higher MDA concentrations than crythrocytes of nonsmokers, which suggests that smokers may have tocopherol were studied in vitro by measuring the concentration of malondialdehyde(MDA) and precent hemolysis of erythrocytes. The addition of any antioxidant to erythrocytes significantly decreased MDA concentrations(p<0.05) while antioxidants showed nonsignificant inhibition of hemolysis. Among the antioxidant used in this study, canthaxanthin showed the greatest inhibition of both lipid peroxidationand hemolysis. Meanwhile, $\alpha$-tocopherol showed potent inhibition of lipid peroxidation, but not of hemolysis.
