생장 및 부상성이 향상된 남세균 돌연변이 균주 Arthrospira platensis M20CJK3 (A Mutant Arthrospira platensis M20CJK3 Showing Enhanced Growth Rate and Floatation Activity)

  • 건강보조식품 등으로 이용되는 Arthrospira platensis는 세계적으로 대량생산되고 있으나 생산공정 중 수확단계에서 많은 비용이 소요된다. 본 연구에서는 부상을 이용한 효과적인 수확을 위해 균주의 개량을 시도하였으며, 개량균주의 생리적 물리적 특성을 파악하고자 하였다. Ethyl methanesulfonate (EMS)를 모균주 A. platensis KCTC AG20590에 0.24%의 농도로 10, 20, 30분씩 처리하여, 형태 및 부상성이 우수한 균주 A. platensis M20CJK3 균주를 분리하였다. A. platensis M20CJK3은 느슨한 형태에서 촘촘한 형태로 세포사(trichome)의 길이 및 코일간 간격이 감소하였으며, 생장 및 $CO_2$ 고정능이 각각 15%, 17% 향상되었다. 또한, 개량균주의 부상성은 모균주에 비해 2배 이상 향상되었다. 이차원 전기영동 분석을 통해 모균주와 개량균주의 단백질 발현양상을 비교분석한 결과 광합성 관련 색소의 구조와 광전자전달계에 관련된 단백질의 발현 양상이 차이를 보였다. 본 연구에서 개발된 A. platensis M20CJK3은 고밀도 대량배양 및 수확에 유리하며, A. platensis 유전자 연구의 유용한 모델 균주로 활용될 수 있을 것으로 사료된다.

항에스트로겐이 성체 수컷 마우스 수출소관의 형태적 변화에 미치는 효과 (Effects of the Antiestrogen on Morphologic Changes of efferent ductules of the Adult Male Mouse)

  • 본 연구는 수컷 생식기관내 에스트로겐 작용을 구명하기 위한 과정의 일환으로 항에스트로겐 물질인 ICI 182,780을 투여하고, 이로 인한 수출소관의 형태적 변화를 관찰하였다. 대조군과 처리군 간에 수출소관의 관강 직경은 처리후 8일째에 팽창하기 시작하여 59일째까지 지속되었고, 처리군 마우스의 수출소관내의 상피세포 높이는 대조군과 비교해 볼 때, 처리 후 경과기간 내내 감소되었다. 사이세포 높이가 최대로 감소된 곳은 처리된 마우스 수출소관의 근위부로 38% 감소하였으며, 수출소관 상피세포내의 핵상 세포질의 높이는 처리군 마우스의 근위부, 원추부, 원위부에서 각각 46%, 39%, 33% 감소된 것으로 나타났다. 항에스트로겐, ICI 182,780을 수컷 마우스에 투여한 본 실험에서, ${\alpha}ERKO$ 유전자 변형 동물에서 관찰되었던 것과 유사한 생식기관의 형태적 변화가 나타났다. 즉 항에스트로겐이 에스트로겐 수용체와 결합하여 에스트로겐의 기능을 차단하고 이로 인해 수출관의 관강 분비물 재흡수가 억제되어 분비물이 축적됨으로써 다양한 형태적 변환가 초래된 것으로 사료된다. 따라서 수컷 생식기관내의 중요한 에스트로겐 기능중의 하나는 수출소관 부위의 분비물 재흡수 작용이라고 할 수 있다.

Quali-Quantitative Analysis of Flavonoids for Mulberry Leaf and Fruit of 'Suhyang'

  • BACKGROUND: Globally, mulberry (Morus sp.) is exploited for feeding leaf to silkworms in order to obtain silk fiber or for animal feedstock production. Also, mulberry fruit is known to a by-product that was produced from mulberry tree after harvesting leaves for silkworm rearing, as a yield and consumption of mulberry fruit was increased, it has been fixing to a newincome crop. Mulberry leaves and fruits are used for the health benefits of human beings. Mulberry contains various bioactive components, such as alkaloids and flavonoids. Mulberry flavonoids are an important part of the diet because of their effects on human nutrition. The flavonoids in mulberry leaf and fruit of 'Suhyang'(Morus alba L.) were determined. METHODS AND RESULTS: Flavonoids for mulberry leaf and fruit of 'Suhyang' were analysed using ultrahigh performance liquid chromatography coupled with diode array detection and quadrupole time-of-flight mass spectrometry (UPLC-DAD-QTOF/MS)technique. An UPLC-DAD-QTOF/MS system was used, and identification of mulberry leaves constituents was carried out on the basis of the complementary information obtained from LC spectra, MS ions, and MS/MS fragments. The mulberry leaf (16 flavonoids) and fruit (9 flavonoids) were isolated and analyzed from Suhyang using UPLC-DAD-QTOF/MS chromatogram. To the best of our knowledge, Quercetin 3-O-(6"-O-malonyl) glucoside and quercetin 3-O-rutinoside (rutin) was detected on the highest content in leaf and fruit, respectively and further research will be devoted to evaluate their biological activity. CONCLUSION: Obtaining information about the concentration of functional materials in mulberry leaves could contribute to the development and promotion of processed, functional products and offer possible industrial use of 'Suhyang', holding promises to enhance the overall profitability of sericulture.

담관 결찰에 의한 간섬유증 발생에서 비만세포 동원에 미치는 Stem Cell Factor의 역할 (Role of Stem Cell Factor on the Recruitment of Mast Cells in the Development of Liver Fibrosis Induced by Bile Duct Ligation in the Rat)

  • Mast cells (MCs) have been implicated in the pathogenesis of tissue fibrosis. However, the role of MC in the development of liver fibrosis has not been fully elucidated. Stem cell factor (SCF) is known to recruit MCs to the liver following injury as it induces mast cell proliferation, survival and differentiation from resident tissue precursors. This study examines the interaction between activated hepatic stellate cells (HSCs) and MCs in rat fibrotic liver, and SCF production by HSCs during culture in vitro. Rats were studied 4, 7, 14 and 21 days after bile duct ligation (BDL). Fibrogenesis was assessed by a measurement of collagen stained with sirius red F3B. Activated HSCs and MCs were identified by ${\alpha}$-smooth muscle actin (${\alpha}-SMA$) immunohistochemical and alcian blue staining and measured by a computerized image analysis system. SCF production was determined in rat HSC cultures using Western blotting. Mild fibrotic changes were noted in BDL rat livers as early as 4 days after induction of cholestasis. Significant expansion and organization of fibrous tissue has occurred in day 14 BDL rats which progressed to bridging fibrosis by day 21. In BDL rats, both a large number of activated HSCs and MCs were detected in portal tracts and fibrous septa. Both area of activated HSCs infiltration and density of MCs were significantly higher in all BDL group compared with Shams. In BDL rats, both areas of activated HSCs infiltration and density of MCs were no significant difference between day 4 and 7 and were significantly higher in day 14. However, the areas of activated HSCs infiltration were significantly lesser in day 21 and the densities of MCs were significantly higher in day 21 compared with day14 BDL. In BDL rats, both areas of activated HSCs infiltration and density of MCs were highly correlated with areas of fibrosis. Western blotting showed that SCF protein was consistently produced in activated HSCs by culture on plastic and freshly isolated HSCs expressed relatively little 30kD SCF compared to late primary culture activated HSCs (day 14) and passaged HSCs. These results suggest that HSCs activated in vitro produce SCF, and may play an important role in recruiting mast cells to the liver during injury and fibrosis.

VR을 이용한 균형능력 측정장비의 성능평가 (Performance Evaluation of Balance Ability Equipment Using VR)

  • Purpose : Conventional Balance Measurement can only measure the center of gravity and the shaking movement of the body. As a result, it has the disadvantages of not responding to visual changes and blocking functions of variables. This study was carried out to evaluate the performance of new equipment that measures the balance of the body using changes in body segment and pressure using the acceleration sensor to compensate for the disadvantages of the existing equipment. Methods : To this end, balance ability was measured in 43 healthy male/female adults without orthopedic injuries and nervous system damage in the last 6 months. in a situation where the visual information was restricted by Virtual Reality (VR) gear, all subjects measured and evaluated the balance ability utilizing the new equipment. Balance measurement (Prime Medilab, Korea) and Wii fit (Nintendo, Japan) were used to measure the balance ability of the subjects, and the balance ability test was performed in 4 postures using each device for data acquisition. The test duration for each posture was 30 seconds. For data acquisition, the average value of three experiments measured using each equipment was analyzed, and the statistical test was performed using the independent sample and the corresponding sample t-test, and the significance level was set to α=.05. Results : As a result of measuring the balance ability using individual equipment, blocking visual information using VR gear, the average speed, maximum speed, and moving area of the COP increased equally. It was found that the obtained absolute size of the result in Wii was somewhat larger than that of BM. Conclusion : It is considered that in the future research, it is necessary to measure changes in the body's center of gravity through image analysis, etc., to make clear comparison and evaluation of the usability.

교통사고에 의한 소아 족부 압궤손상에 대한 치료결과 (The Results of Treatment for Motor Vehicle-related Crushing Injuries of Foot in Children)

  • Purpose: The purpose of this work was to describe the results of treatment for motor vehiclerelated crushing injuries among children and adolescents under sixteen years in Korea. Materials and Methods: A retrospective analysis was conducted of data from children who were under sixteen year and injured foot by motor vehicles. Cases were documented 1) age at the time of injury, 2) injured site, 3) the area of accident, 4) the kind of vehicle, 5) associated injuries, 6) methods of treatment for soft tissue reconstruction and 7) complications. The relationships between the area of accident and associated injuries, and the kind of vehicle and associated injuries were analyzed using Chi-square test and Fisher exact test. Results: There were 97 children who were 15 year and younger. The mean age was 7.4 years, and 65% were boys. The left foot was more dominant side of injury (57%). Seasonal variation was seen with the number of injuries peaking during the summer (43%, p<0.05). Among the vehicles, 78.3% were the large vehicles (bus, truck or van). The where of accident was more frequent at an alley or less than two lanes of traffic. But, the relationships between the place of accident and associated injury or the kind of vehicles and associated injury were not statistically significant. The associated injury were fracture or dislocation (23 cases, 35.9%), injury of tendon (21 cases, 32.8%). There were amputation or disarticulation of foot in 8 cases (8.2%) and post-traumatic deformities such as flatfoot, hindfoot varus or valus deformities by tendon injury in 7 cases (7.2%). Conclusion: More than 50% of crushing or degloving injuries of child's foot by traffic accidents happened in boys between 5 to 9 years old. The associated injury was unrelated with size of vehicles or accident place at the time of accident. But, even though foot injury happened in an alley or one lane by small vehicles, child who hurt feet by car need thorough investigation about associated injury. If a surgeon keep in mind and treat child to associated injury necessarily, can minimize complication. Microsurgical reconstruction for soft tissue defect was prior to other methods.

미강 페놀산 농축물의 특성 및 항산화 활성 (Properties and Antioxidative Activities of Phenolic Acid Concentrates of Rice Bran)

  • 본 연구에서는 현미의 미강으로부터 생리활성이 있는 페놀산물질을 얻기 위해 다양한 추출법을 시도하였고, 얻어진 미강 페놀산 농축물의 성분 분석 및 항산화 활성에 대해 조사하였다. 미강 추출물 중 알칼리 가수분해를 시행한 다음 에틸아세테이트로 분획하여 얻은 RBE-II의 총 폴리페놀 함량과 항산화 활성이 가장 높은 것으로 조사되었고, 페놀산 성분의 농축 및 정제를 위해 Sep-pak $C_{18}$ Vac cartridge를 통과시켜 미강 페놀산 농축물(F1-RBE-II)을 얻었다. HPLC 분석을 통해 얻어진 페놀산 성분 중 ferulic acid가 가장 많았고, p-coumaric acid, sinapic acid, vanillic acid, 그리고 syringic acid가 존재하였으며, 그 구성성분이 주로 hydroxycinnamic acid 계열에 속한다는 것을 알 수 있었다. DPPH radical과 ABTS radical 소거능에서 미강 페놀산 농축물(F1-RBEII)는 강력한 항산화 활성을 나타냈으며, ${\beta}$-CLMS을 이용한 지질산화 유도반응에서도 우수한 peroxy radical 저해능을 발휘하여, 합성 항산화제인 BHA의 활성과 비교했을 때 높은 과산화 지질억제력을 보였다. 본 연구를 통해 미강 페놀산 농축물의 성분 및 강한 항산화 능력이 확인됨으로천연 항산화제 및 지방 산화 방지제로 개발하기에 부족함이 없는 것으로 사료된다.

파노라마촬영 시 조직등가물질을 이용한 갑상선보호대의 갑상선피폭선량 감소효과 (The Effects of a Thyroid Shield Made of a Tissue-Equivalent Material on the Reduction of the Thyroid Exposure Dose in Panoramic Radiography)

  • 파노라마방사선촬영 시 방사선감수성이 높은 갑상선의 방사선 피폭을 최소화하면서 진단에 유용한 영상을 얻기 위하여 조직등가물질인 보루스(bolus)를 차폐체로 사용하여 피폭선량 감소효과를 측정하였다. 실험은 2011. 6. 1일부터 6. 30일까지 두경부팬톰을 이용하여 갑상선 위치의 표면입사선량과 심부흡수선량을 측정하였다. 그 결과 갑상선 부위 입사표면선량은 평균 43.84 ${\mu}Gy$이었고, 10 mm 두께의 보루스로 만든 갑상선보호대에서는 평균 28.39 ${\mu}Gy$로 15.45 ${\mu}Gy$(35.24%)가 감소되었다. 20 mm 갑상선보호대를 착용했을 경우에는 평균 25.38 ${\mu}Gy$로 미착용 시 평균 43.84 ${\mu}Gy$보다 18.46 ${\mu}Gy$(42.10%)가 감소되었다. 그리고 표면에서 20 mm 깊이에서는 10 mm 두께의 갑상선보호대는 선량 감소효과가 없었고, 20 mm 갑상선보호대를 착용했을 경우에는 0.06 mSv (20%)의 선량감소효과가 있었다.

중등학교 조리실습에 대한 현황과 효율화방안 (The Present Situtation and Efficient Way of Cooking Practice in the Middle and High School)

  • Cooking practice is keenly related to not only the betterment of family health but to the improvement of dietary life. Cooking is a kind of practical skill and one is skill in it by repeated practice. The aim of cooking practice in school is to fix cooking method practically and scientifically, to acquire cooking skill. To achieve this aim there are many unrecognized or unsolved problems in the first-line schools. The problems and efficient ways of cooking practice in the middle and high school are as follows; 1. The aim of cooking practice: It should be first of all remembered that the practice is so practical that the knowledge and skill of cooking should be related to the future domestic life. Second, the practice should be able to serve the communal and national life by scientifying and improving life. 2. The content of the practice: First, it should be so arranged step by step that the content of each year should not be repeated. Second, the ratio of Korean cooking practice should be increased and the material of the practice should be chosen in consideration of the peculiarity of the community, seasons and economical problems so that it may be applied to the very day life. Third, to improve dietary life, the practice should be a way of the simplification of dietary life, of the nutritional efficiency. Forth, for the betterment of physical condition of the family the practice should involve balanced dietary plans and dietary therapy. 3. Teaching method: First, the practice should be scientific and comprehensive in consideration of not only cooking skills but also knowledge of nutrition. Second, micro-class system should be adopted, and practice labor should be allotted each student to develop cooperative attitude and the sense of responsibility. Third, in addition to the practice conducted in a body, comparative cooking, applicable cooking and experimental ability and the content of the text. Fourth, teachers should let the students examine and find problems by bringing them to a focus on the basis of theory. 4. Administration method: First, the practice schedule should be planned that a class has practices at least more than twice a semester. Second, two hours of class should be continued without cessation after beforehand survey and theory study. Third, facilities and utensils for practice should be enlarged. That is, cooking practice rooms, tools, utensils should be prepared. Fourth, enough cost of practice should be secured. The above mentioned points show the present situation and problems with which cooking practice teaching is confronted. In order to normalize cooking practice teaching, the first-line schools should give the above mentioned problems their careful consideration and improve the present situation so that efficient, creative, practical cooking practice will be possible.

녹차 함유 카테친 및 카페인 동시분석을 위한 최적 HPLC 분석 조건 (Optimal HPLC Condition for Simultaneous Determination of Catechins and Caffeine in Green Tea Extracts)

  • 고 기능성 유용 차나무 품종육성 및 녹차의 이용성 증진 연구의 기초 자료를 제공하기 위해 녹차 함유 catechin 화합물 및 caffeine 성분을 동시에 분석할 수 있고, 재현성 및 검출 감도가 증진된 최적의 HPLC 분석 조건을 검토한 결과를 요약하면 다음과 같다. 1. 녹차에 함유된 catechin 화합물 및 caffeine의 동시분석을 위한 최적 HPLC 분석 조건을 검토한 결과 컬럼은 YMC-pak ODS-AM 303(25 cm)컬럼을 이용하고, 검출파장은 280 nm, 컬럼온도를 $30^{\circ}C$, 분당유속은 1 mL, 이동상 용매로는 A용매로 0.1% 인산 - 증류수, B용매로 100% MeOH를 사용하여 농도구배 조건으로 분석하는 것이 가장 효율적 분석법으로 조사되었다. 2. 녹차에 함유된 catechin 화합물 및 caffeine을 동시에 분석할 수 있는 최적 HPLC분석조건의 재현성 검정을 실시한 결과 머무름 시간(Rt.)의 변이계수는 최대 1.23% 미만, peak area의 변이계수는 최대 5.13% 미만을 나타내어 확립된 녹차 함유 catechin 9종 화합물 및 caffeine의 HPLC 정량분석법은 재현성이 높은 분석법으로 확인되었고, 각 cafechin 화합물 및 caffeine 성분의 최소 검출한계는 최소 50ppb(0.050 ppm, 50 ng/mL)이하로 조사되어, 확립된 HPLC 분석법은 녹차의 작물학적 특성 평가용으로 활용하기에 전혀 무리가 없는 분석법으로 판단되었다.
