Antifungal Activity of Chloroform Extract from Lepidolaena Taylorii on the Dermatophytic Fungus Trichophyton mentagrophytes

  • The effects of chloroform extract from Lepidolaena tayiorii (L. tayiorii) on antifungal activity were investigated. The crude chloroform extract of L. tayiorii inhibited the growth of the Gram positive bacteria Bacillus subtilis ATCC 19659, (5 mm inhibition zone at $150{\mu}g/disc$) and the dermatophytic fungus Trichophyton mentagrophytes ATCC 28185, (6 mm inhibition zone at $150{\mu}g/disc$), and cytotoxic to P388 murine leukaemia cells ATCC CCL 46 P388D1, ($IC_{50}\;405.0{\mu}g/mL\;at\;150{\mu}g$/disc) and cytotoxic to BSC monkey kidney cells (@ 5 mg/mL, $150{\mu}g/disc;$ +++: 100% activity). We suppose that this crude chloroform extract of L. tayiorii is the strong antimicrobial and cytotoxic activities.

  • This study divided hairpiece-related technologies into three categories: Materials, Structure, Hair Extensions. Then, it investigated the current patent application trends in the above technologies and attempted to determine what aspects should be focused on in terms of the growth potential of hairpiece-related technologies and future technologies. A hairpiece is used to cover a specific balding area resulting from innate factors such as heredity or acquired traits including disease or accident. It is also used to make short hair look longer. Recently, as the rates of hair loss caused by stress increase, there has been a rising demand for hairpieces that can quickly and conveniently cover a balding region as well as satisfy people's aesthetic needs through diverse hairstyling. Even though patents have been applied evenly across hairpiece technology fields, patent activity has declined since 2010 was confirmed. Therefore, this study targets the facilitation and expansion of the hairpiece technology market by suggesting desirable R&D directions for future hairpiece-related technologies after analyzing current hairpiece patent trends.

Study on Effect of Saengbal-eum-II(Shēngfà-yĭn-II)on Hair Regrowth Promotion in C57BL/6 Mice (생발음(生髮飮)-II 피부도포가 C57BL/6 마우스의 육모촉진에 미치는 효과)

  • Objectives : Saengbal-eum-II($Sh{\bar{e}}ngf{\grave{a}}-y{\breve{i}}n-ll$) is a hair care product which is composed of ten plant extracts used in oriental medicine. This study was carried out to investigate the effects of Saengbal-eum-II($Sh{\bar{e}}ngf{\grave{a}}-y{\breve{i}}n-ll$) on hair regrowth and cytokine changes in a shaving model of C57BL/6 mice. Materials and Methods : Five-week-old mice were acclimated for 1 week at a temperature between $21-23^{\circ}C$, 40-60% relative humidity, and 12h of a light/dark cycle before beginning of the experiment. There were three experimental groups including 50% ethanol (EtOH, control), a positive control of 3% Minoxidil, and 30% Saengbal-eum-II($Sh{\bar{e}}ngf{\grave{a}}-y{\breve{i}}n-ll$) in 50% ethanol in 18 female mice. The test compounds were topically treated once a day over 12 days. The hair regrowth was photographically and histologically determined during the experimental period of 12 days. Revelation of EGF, $TGF-{\beta}1$ and IL-6 in hair follicle were also determined using immunohistochemistry. In addition to that, IL-6, $TNF-{\alpha}$, and $IL-1{\beta}$ in skin tissue were determined using enzyme-linked immunosorbent assay(ELISA). Results : Hair regrowth in 3% Minoxidil and Saengbal-eum-II($Sh{\bar{e}}ngf{\grave{a}}-y{\breve{i}}n-ll$) groups was promoted earlier and faster than the control group. Concentrations of hairs and thick-hair ratio in 3% Minoxidil and Saengbal-eum-II($Sh{\bar{e}}ngf{\grave{a}}-y{\breve{i}}n-ll$) groups were promoted than the control group. EGF was moderately positive in hair follicle of 3% Minoxidil and Saengbal-eum-II($Sh{\bar{e}}ngf{\grave{a}}-y{\breve{i}}n-ll$) groups, but negative in the control group. $TGF-{\beta}1$ was not significantly difference between the groups. IL-6 in hair follicle of Saengbal-eum-II($Sh{\bar{e}}ngf{\grave{a}}-y{\breve{i}}n-ll$) group was negative, but weakly positive in 3% Minoxidil and control group. IL-6 and $IL-1{\beta}$ in skin tissue were significantly decreased in Saengbal-eum-II($Sh{\bar{e}}ngf{\grave{a}}-y{\breve{i}}n-ll$) group, but there was not significantly decreased in 3% Minoxidil and control group. $TNF-{\alpha}$ in skin tissue was significantly decreased in 3% Minoxidil and Saengbal-eum-II($Sh{\bar{e}}ngf{\grave{a}}-y{\breve{i}}n-ll$) groups. Conclusions : These results suggest that Saengbal-eum-II($Sh{\bar{e}}ngf{\grave{a}}-y{\breve{i}}n-ll$) has hair growth promoting activity and it can be used for treatment of alopecia. And these effects relate to EGF revelation of hair follicle and a decrease IL-6, $TNF-{\alpha}$, and $IL-1{\beta}$ in skin tissue.

A case of Menkes disease with unusual hepatomegaly (멘케스병에서 간비대를 보인 1례)

  • Menkes disease is an X-linked recessive copper transport disorder characterized by neurological deterioration, connective-tissue damage, and abnormal hair growth. It is caused by the mutation of the ATP7A gene. This report describes a four-month-old boy with neurological symptoms typical of Menkes disease plus unusual liver involvement. He developed seizures at three months of age and exhibited hypotonia, cephalhematoma, a sagging face, redundant and hypopigmented skin, and abnormal hair growth. In addition, he had unexplained hepatomegaly and high hepatic transaminase. We confirmed the diagnosis of Menkes disease by mutation analysis of the ATP7A gene. To exclude other possible causes for the hepatic abnormalities, a liver biopsy was performed, revealing intracytoplasmic cholestasis, focal spotty necrosis, and minimal lobular activity. The patient's liver involvement may be an underestimated complication of Menkes disease.

Effects of 1-tetradecanol and β-sitosterol Isolated from Dendropanax morbifera Lev. on Skin Whitening, Moisturizing and Preventing Hair Loss (미백, 보습 및 탈모방지에 대한 황칠나무(Dendropanax modifera Lev.)에서 분리한 1-tetradecanol, β-sitosterol의 효과)

  • Dendropanax morbifera Leveille (Araliaceae) is an endemic species growing in the south-western part of South Korea and has been used in folk medicine. However, the effects of Dendropanax morbifera Lev. on skin biology remain to be elucidated. In this study, we isolated 1-tetradecanol and ${\beta}$-sitosterol from the n-hexane fraction of Dendropanax mobifera Lev. and To investigate the whitening effect of the fraction, we tested the inhibition of tyrosinase activity of 1-tetradecanol. The results show that the inhibitory effect of the 1-tetradecanol was higher than water extract and n-hexane fraction. And 1-tetradecanol significantly reduced melanin contents of B16F10 cells compared to more than water extract and n-haxane fraction dose-dependantly without cell cytotoxicitiy (below $100{\mu}g/mL$). We also investigated the skin moisturizing effect using HR-1 hairless mice. The transepidermal water loss (TEWL) in the 1-tetradecanol treated group was significantly smaller than that in the other groups. To investigate the effect of the preventing hair loss by ${\beta}$-sitosterol, we observed HR-1 hairless mice through periodic growth feature. The results suggest that hair loss of mice by ${\beta}$-sitosterol was delayed and it's hair density showed the highest. These data provide evidence that Dendropanax morbifera Lev. may be a potent candidate for the improvement of both skin whitening, moisturizing and alopecia from the point of cosmetic industry view.

In Vitro Anti-aging and Hair Follicle Dermal Papilla Cells Activation Effects of Usnea diffracta Vain Extract (송라 추출물의 세포 수준에서 항노화 및 모유두세포 활성화 효과)

  • Songla (Usnea diffracta Vain.) is one of the lichens belonging to the genus Usnea, and pharmacological activities such as antioxidant, antimicrobial, anti-inflammatory, anti-tumor and cardiovascular protection have been reported in previous studies, but its efficacy in skin and hair is not well known. In this study, the effect of Usnea diffracta extract (UDE) on anti-aging and dermal papilla cell proliferation was verified in vitro. As a result of the experiment, it was confirmed that the UDE significantly reduced the expression of MMP-1 and the activity of MAPKs (ERK, p38, JNK) and AP-1 (c-Fos, c-Jun), which were increased by UVA in HDFn. In addition, the UDE significantly increased the proliferation of HFDPC and significantly increased the mRNA expression of VEGF and KGF, which are hair growth factors. Accordingly, the phosphorylation of ERK/CREB involved in hair proliferation and expression of growth factors was increased in a concentration-dependent manner. The main component represented by the main peak was separated and purified using Prep LC by concentrating the UDE, which was confirmed as diffractaic acid through NMR and Mess analysis. Isolated diffractaic acid significantly reduced the expression of MMP-1 increased by UVA in HDFn and increased the proliferation of HFDPC in a concentration-dependent manner. The result suggest that UDE proved its usability as a natural cosmetic material with anti-aging and dermal papilla cell activation effects.

Skin Deiodinase Profiles after Melatonin Manipulated in Chinese Inner Mongolia Cashmere Goats

  • The roles of melatonin in the control of deiodinase (MD) activity in cashmere goat skin and associated cashmere fibre growth were investigated. Eighteen half-sib Chinese Inner Mongolia cashmere wethers were allocated randomly to two groups (n = 9/group). One group was implanted subcutaneously with melatonin (2 mg/kg BW) at three 2-monthly intervals while the other group served as a control. All goats were maintained under natural photoperiodic conditions and were grazed on natural pasture. The plasma melatonin concentration showed a significant difference (p<0.01) between the implant group (M) and the control group (C) but plasma $T_4$ (or $T_3$) showed no significant difference (p>0.05). The monodeiodinase type II (MDII) activity in skin tended to increase gradually from the summer solstice to November. During July and August, the activity of MDII for the M group was higher (p<0.05) than that of the C group; also during this period, there was a significant positive correlation between MDII activity of skin and cashmere fibre growth rate. The monodeiodinase type III (MDIII) activity and the ratio of MDIII and MDII tended to decrease from the summer solstice to November. The ratio of MDIII and MDII for the M group was lower (p<0.05) than that of the C group in July and August. The cashmere fibre growth rate of the M group was significantly greater than that of the C group in July (p<0.01), August (p<0.001) and September (p<0.05). The cashmere fibre diameter and guard hair and body weight were not influenced (p>0.05) by melatonin implantation. The results demonstrate that melatonin plays an important role in the regulation of skin MD activity. Simultaneously, the cashmere fibre elongation stimulated by melatonin may result from enhanced MDII activity during a period of two months after melatonin treatment.

The Plant Growth-Promoting Fungus Aspergillus ustus Promotes Growth and Induces Resistance Against Different Lifestyle Pathogens in Arabidopsis thaliana

  • To deal with pathogens, plants have evolved sophisticated mechanisms including constitutive and induced defense mechanisms. Phytohormones play important roles in plant growth and development, as well as in the systemic response induced by beneficial and pathogen microorganisms. In this work, we identified an Aspergillus ustus isolate that promotes growth and induces developmental changes in Solanum tuberosum and Arabidopsis thaliana. A. ustus inoculation on A. thaliana and S. tuberosum roots induced an increase in shoot and root growth, and lateral root and root hair numbers. Assays performed on Arabidopsis lines to measure reporter gene expression of auxin-induced/ repressed or cell cycle controlled genes (DR5 and CycB1, respectively) showed enhanced GUS activity, when compared with mock-inoculated seedlings. To determine the contribution of phytohormone signaling pathways in the effect elicited by A. ustus, we evaluated the response of a collection of hormone mutants of Arabidopsis defective in auxin, ethylene, cytokinin, or abscisic acid signaling to the inoculation with this fungus. All mutant lines inoculated with A. ustus showed increased biomass production, suggesting that these genes are not required to respond to this fungus. Moreover, we demonstrated that A. ustus synthesizes auxins and gibberellins in liquid cultures. In addition, A. ustus induced systemic resistance against the necrotrophic fungus Botrytis cinerea and the hemibiotrophic bacterium Pseudomonas syringae DC3000, probably through the induction of the expression of salicylic acid, jasmonic acid/ethylene, and camalexin defense-related genes in Arabidopsis.

Sulforaphene Attenuates Cutibacterium acnes-Induced Inflammation

  • Acne is a chronic inflammatory disease of the sebaceous gland attached to the hair follicles. Cutibacterium acnes is a major cause of inflammation caused by acne. It is well known that C. acnes secretes a lipolytic enzyme to break down lipids in sebum, and free fatty acids produced at this time accelerate the inflammatory reaction. There are several drugs used to treat acne; however, each one has various side effects. According to previous studies, sulforaphene (SFEN) has several functions associated with lipid metabolism, brain function, and antibacterial and anti-inflammatory activities. In this study, we examined the effects of SFEN on bacterial growth and inflammatory cytokine production induced by C. acnes. The results revealed that SFEN reduced the growth of C. acnes and inhibited proinflammatory cytokines in C. acnes-treated HaCaT keratinocytes through inhibiting NF-κB-related pathways. In addition, SFEN regulated the expression level of IL-1α, a representative pro-inflammatory cytokine expressed in co-cultured HaCaT keratinocytes and THP-1 monocytes induced by C. acnes. In conclusion, SFEN showed antibacterial activity against C. acnes and controlled the inflammatory response on keratinocytes and monocytes. This finding means that SFEN has potential as both a cosmetic material for acne prevention and a pharmaceutical material for acne treatment.
