A Study on the Efficacy of Gryllus Bimaculatus in Korean Medicine - Focusing on Medical Documents - (실솔(蟋蟀)의 한의학적 효능에 대한 소고(小考) - 의학(醫學) 문헌(文獻)을 중심으로 -)

  • Objectives : In this paper, the efficacy of Gryllus Bimaculatus, a rarely used medicinal in Korean Medical material medica, was examined in medical documents. Its nature, flavour, effects, properties and mechanism were studied. Methods : Classical texts on the effects of Gryllus Bimaculatus was researched and analyzed. In addition, medical conditions to which the medicine is applied were compared with those in contemporary medicine which used this medicine for treatment. Analysis on the mechanism of the nature, flavour, and effects of Gryllus Bimaculatus were studied in terms of the Zhangfu correlation to the Five Flavours and the Qi and Flavour medicinal application theory from the Suwen·Zhangqifashilun, together with the pathology of each illness. Results & Conclusions : The nature and flavour of Gryllus Bimaculatus are pungent, salty, and warm, which has the effect of communication and smooth flowing, used as diuretic, and to eliminate blood stagnation and tumor. Illnesses to which the medicinal is applied are blocked urination, cyst, fluid in the abdomen, stagnated blood, etc. Contemporary research comparable to the effects of Gryllus Bimaculatus in the classical texts were mostly on its stimulation of urination, with others on its liver protection effects, anti-coagulation effects, prevention of blood stasis and cardiovascular diseases, anticonvulsant and antihypertensive effects. Newly researched areas were those on strengthening the immune system, anti-aging, and antioxidation. Areas in need of further research were dystocia, Kidney-deficient impotence, and dou zhen(痘疹).

Biological Activity of Fermented Gryllus bimaculatus extracts (발효 쌍별귀뚜라미 추출물의 생리활성 연구)

  • Gryllus bimaculatus extract (GbE) have reported that anti-inflammatory activity by suppression of pro-inflammatory cytokines. However, the effects of fermented Gryllus bimaculatus extract (FGbE) have not yet been investigated. In this study, we evaluated the anti-inflammatory and anti-wrinkle effect of the fermented Gryllus bimaculatus extracts using Bacillus subtilis (JB PMB-18) in RAW264.7 cells. Both GbE and FGbE exerted no cytotoxic effects until 1000 ㎍/mL concentration. FGbE decreased NO production and decreased iNOS and COX-2 mRNA levels in a concentration-dependent manner. In addition, the protein production of inflammatory cytokines TNF-α, IL-1β and IL-6 was effectively reduced compared to the GbE. Inhibitory activities of elastase and collagenase associated with skin wrinkle improvement were measured to be 45% and 69%, respectively, at a concentration of 500 ㎍/mL in FGbE. From these results, FGbE can be used as a health functional food and skin functional cosmetic materials for preventing inflammatory diseases because it has excellent anti-inflammatory and anti-wrinkle effects.

Changes in Nutritional Composition of Gryllus bimaculatus Fermented by Bacillus sp. and Mycelium of Basidiomycetes (고초균 및 버섯 균사체를 이용하여 발효한 쌍별귀뚜라미 발효물의 단백질 및 무기질 성분 변화)

  • This study examined the changes in the protein and mineral composition of Gryllus bimaculatus fermented with Bacillus substilis and the mycelia of Basidiomycetes. Normal Gryllus bimaculatus (S) and experimental group data obtained after an inoculation of Bacillus substilis (SC) (KACC 19623), Pleurotus eryngii (SP) and Cordyceps millitaris (SC) were compared. The crude protein content of the Gryllus bimaculatus (control) was 75.48%, but it decreased to 64.55, 54.32, and 63.53% after fermentation with SB, SP and SC, respectively (p<0.05). An analysis of the organic elements showed that the contents of the carbon and nitrogen sources were also reduced after fermentation, and the most significant decrease was observed after fermentation with SP. In SDS-PAGE, a 120 kDa and a 48 kDa protein of Gryllus bimaculatus were found. On the other hand, protein bands faded after fermentation with SP and SC, respectively. Moreover, no visible band was observed after fermentation with SB. According to amino acid analysis, the total free amino acid content increased 3.84 and 1.74 times after fermentation with SB and SP, respectively, compared to the corresponding baseline data. In contrast, it decreased by 0.52 times after fermentation with SC. Among the essential amino acids found in crickets fermented with SB, the valine and isoleucine content was 3.57 and 2.64 times higher, respectively, than the recommended daily amount of essential amino acids.

  • Platelets act a fundamental role in primary- and secondary-hemostasis, however, platelet activation may cause thrombosis simultaneously. Therefore, control of platelet aggregation is crucial in preventing thrombosis-mediated diseases. Recently, the development of insect materials is attracting attention. Among the highly nutritious functional food sources, insects such as two-spotted cricket (Gryllus bimaculatus). Gryllus bimaculatus (G. bimaculatus) contains high protein and unsaturated fatty acids and has been registered as a food material September 2015 by the Ministry of Food and Drug Safety of Korea. In this study, we examined whether G. bimaculatus extract (GBE) inhibits platelet aggregation, intracellular calcium mobilization, thromboxane A2 production and glycoprotein IIb/IIIa (integrin αIIb/β3) activation. We investigated whether GBE can regulate signaling molecules, such as 1, 4, 5-triphosphate receptor type I, extracellular signal-regulated kinase, cytosolic phospholipase A2, mitogen-activated protein kinases p38, vasodilator-stimulated phosphoprotein, phosphatidylinositol-3 kinase, Akt, glycogen synthase kinase-3α/β, and SYK. Taken together, GBE is a potential therapeutic drug candidate to prevent platelet-related thrombosis and cardiovascular disease.

Effects of Cricket Supplements of the Chicken Meats and Its Eggs (귀뚜라미 첨가 사료가 계육과 계란의 성분에 미치는 영향)

  • Chemical characteristics of the cricket, Gryllus bimaculatus, were investigated in total composition, amino acid composition, fatty acid composition and mineral components. After the treatment of 0.4% of G. bimaculatus in the chicken feed, the changes of fatty acid composition in the chicken meat and egg were also estimated. As the result, saturated fatty acid, especially palmitic acid, decreased 4% and unsaturated fatty acids, linoleic aced and arachidonic acid, increased 12 and 23%, respectively, compared with those of control. In addition, there was a small increase in Eicosapentaenoic acid(EPA). The sensory test of the chicken meat resulted in increased flavor, brightness and yellow color. The sensory scores of G. bimaculatus egg in the point of taste resulted in somewhat better estimate than control. These results are appear to be stemmed from fatty acids of cricket.

  • 생쥐 비장세포에 대한 아열대산 귀뚜라미(Gryllus bimaculatus)와 왕귀뚜라미 (Teleogryllus emma)로부터 3종 추출물(Al, A2, Bl)의 면역활성능을 조사하였다. 3종의 추출물 모두 normal spleen cell에 대하여 독성을 나타내지 않았다. Al과 A2는 세포의 proliferation을 증가시키는 경향을 나타내었으며, 1000ug/m1의 고농도에서는 약 10-20%의 증가를 보였다. Bl의 경우는 약간의 감소경향을 나타내었으며, T cell의 mitogen인 Con A에 대한 mitogenic response는 두드러진 활성을 나타내지 않았다. (중략)

  • A cDNA encoding the protein lethal (2) essential for life [l(2)efl] was cloned from Gryllus bimaculatus and named GBl(2)efl. This protein is composed of 189 amino acids, including an N-glycosylation site and 15 phosphorylation sites. Its predicted molecular mass is 21.19 kDa, with a theoretical isoelectric point of 6.2. The secondary structure of GBl(2)efl was predicted from the identification of random coils (56.08%), alpha helices (22.22%), extended strands (17.99%), and beta turns (3.7%) through sequence analyses. A homology analysis revealed that GBl(2)efl exhibited a high similarity with other species at the amino acid level, ranging from 52% to 69%. While GBl(2)efl mRNA expression was higher in the dorsal longitudinal flight muscle following a three-day starvation and in the Malpighian tubules following a one-day starvation, no changes in expression were detected in other tissues. Furthermore, tunicamycin-induced endoplasmic reticulum (ER) stress resulted in an approximately 1.8-fold higher expression in the fat body compared with the wild type.

The Antioxidant Activity and Protective Effects on Skin Cell Lines of Solvent Fractions from Gryllus bimaculatus (쌍별귀뚜라미의 용매분획별 항산화 활성과 피부세포주 보호 효과)

  • Oxidative stress caused by reactive oxygen species (ROS) is a key mechanism of skin aging, and the use of antioxidants is an effective strategy to prevent the symptoms associated with ROS-induced skin aging. The components rich in polyphenolic compounds with antioxidative activity were identified by fractionating an aqueous ethanolic extract of Gryllus bimaculatus (AE-GBE) using solvent using hexane, chloroform, ethyl acetate, butanol, and water. Their ability to migrate H2O2-induced oxidative stress in human dermal fibroblasts (HDFs) was then evaluated. The butanol fraction of AE-GBE had the highest polyphenol content and antioxidant effect, followed by the ethyl acetate and water fractions, suggesting that the likely antioxidant components are polar components. Furthermore, the butanol, ethyl acetate, and water fractions effectively reduced intracellular ROS production and DNA damage in HDF cells caused by H2O2. Overall, these findings suggest that the butanol fraction of AE-GBE shows promise as a natural insect-derived antioxidant material, capable of suppressing oxidative stress by showing a stronger antioxidant effect under H2O2 stimulation than the other fractions.

Effect of different diets on growth and development of the two-spotted cricket, Gryllus bimaculatus (Orthoptera: Gryllidae)

  • Many insects have gained increasing attention as an alternative protein for humans. Among those, the two-spotted cricket, Gryllus bimaculatus (Orthoptera: Gryllidae), was recently approved as a general food ingredient by the Korean Ministry of Food and Drug Safety. For industrial utilization of G. bimaculatus, mass rearing techniques and production system should be standardized first. In this study, we investigated the effects of five different feeds on the growth and development of G. bimaculatus. Feed is the one of the key factors that has considerable effects on rearing insects. With five different kinds of feed on $1^{st}$, $3^{rd}$, and $5^{th}$ instar nymphs, the change of survival rate, body weight were monitored up to eight wk after hatching. We concluded that 50% of soybean flour, 20% of corn powder, 10% of rice bran, 9% of milk serum, 10% of rice flour, 0.5% of microorganisms, and 0.5% of multivitamins and minerals (diet C) was the best mix for promoting growth and development of $3^{rd}$ instar nymphs compared to the control diet.
