Numerical changes of coliform bacteria in a recirculating aquarium were measured as fouling indicator. The contamination of the filter bed was mainly due to the food given to fishes, the fecal materials and the additional supply of water polluted already. Abundance of the bacteria in the aquarium was controlled by the physiochemical factors of water quality. Of these factors, pH was the main factor. Under the water temperature $20^{\circ}C$, if pH was controlled at lower than $5\~6$, the bacteria did not flourish seriously. Under the water temperature over $30^{\circ}C$, the bacterial growth was markedly decreased regardless of the pH value, and the bacterial flora seemed to be active in growth under temperature condition lower than $30^{\circ}C$.
The changes of microorganisms, pH, color and flavor were examined in the chicken treated with O%, 1%, 2%, and 3% lactic acid solutions during the storage at 4$^{\circ}C$. Lactic acid treatment inhibited the growth of mesophiles, psychrotrophs, and enteric bacilli as the concentration increased, however, it was not effective on yeasts and molds. Extension of lag phase for mesophilic bacteria and suppression of log phase for psychrotrophs and enteric bacilli were observed in the growth inhibition patterns. The pH of the chicken increased during the storage and antimicrobial effect of lactic acid appears to be due to mainly the decrease of pH. No flavor change of the chicken was observed by any lactic acid treatment, however, 3% lactic acid caused discoloration of the chicken.
The effects of temperature. pH, carbon and nitrogen sources on the growth of Rhizoctonia solani were studied by using five hyphal anastomosis groups(four cultural types, 7 isolates) of the fungus. The ranges of optimum temperature were $20^{\circ}C$ in the AG 2-1, AG 2-2 and AG 4, and $25^{\circ}C$ in the AG 1-IA, AG 1-IB, AG 3, AG 5. The optimum pH for the mycelial growth was 6-7 in the fungus. Glucose in the AG 1-lA, AG 1-IB, AG 2-2, AG 3 and AG 5, sucrose in the AG 2-1 and fructose in the AG 4 were the most effective for the mycelial growth, but glycerine, cellulose and lactose were not effectively utilized as nutrients. $Ca(NO_3)_2$ in the AG 1-IA, AG 1-IB and AG 4, asparagine in the AG 2-1, $KNO_3$ in the AG 2-2 and $NaNO_3$ in the AG 5 were the best nitrogen sources for the mycelial growth, but $NH_4NO_3$ was not easily utilized by the fungus. Nitrate and organic nitrogens for the fungal growth were utilized better than ammonium nitrogen.
Forty eight $Duroc{\times}Yorkshire{\times}Landrace$ pigs ($15.2{\pm}0.3kg$ average initial BW) were used conducted to evaluate the effects dietary phytase (Phytezyme, WOOJIN, Co. Ltd) supplementation on the growth performance and nutrient digestibility in growing pigs. There were three pigs per pen and four pens per treatment. Treatments were 1) Con (corn-wheat-SBM), 2) PE0.1 (low-P iet+0.1% Phytezyme), 3) PE0.2 (Iow-P diet+0.2% Phytezyme), 4) PE0.3 (low-P diet+0.3% Phytezyme). During d 0 to 28, average daily gain was not significantly different among the treatments. Pigs fed PE0.3 diet significantly decreased (p<0.05) their average daily feed intake compared to pigs fed Con diet. Also, gain/feed in pigs fed PE0.1 and PE0.3 diet was improved (p<0.05) compared to pigs fed Con and PE0.2 diet (p<0.05). For d 28 to 56, pigs fed PE0.2 diet grew significantly faster (p<0.05) than pigs fed Con and PE0.1 diet. Gain/feed was greater (p<0.05) for PE0.2 and PE0.3 treatments than for Con. For overall period, average daily gain was increased (P<0.05) by the addition of 0.2% Phytezyme compared with Con. Gain/feed was significantly improved (p<0.05) by supplementation of PE0.3. Pigs fed PE0.2 showed increased DM and N digestibilities compared to pigs fed other treatments. Supplemented diets PE0.2 and PE0.3 improved (p<0.05) the apparent digestibility of Ca and P compared to other treatments. In conclusion, the results obtained from these feeding trials suggest that the Phytezyme supplementation of diets for growing-finishing pigs had improved growth performance and nutrient availability.
This study determined the effects of dietary restriction on growth and the expression of lipid metabolism and growth hormone signaling genes in the longissimus dorsi muscle (LM) of Korean cattle. Thirty-one Korean cattle steers (average age 10.5 months) were allocated to normal (N; n = 16) or dietary restriction (DR; n = 15) groups. The feeding trial consisted of two stages: for the 8-month growing period, the DR group was fed 80% of the food intake of the normal diet, and for the 6-month growth-finishing period, the DR group was fed a DR total mixed ration with 78.4% of the crude protein and 64% of the net energy for gain of the normal diet. The LM was biopsied 5 months (period 1 [P1] at 15.5 months of age) and 14 months (period 2 [P2] at 24.5 months of age) after the start of feeding. The mRNA levels were determined using real-time polymerase chain reaction. Body weight, daily feed intake, average daily gain, and feed efficiency were lower in the DR group compared with the normal group at both P1 and P2. At P1, the lipogenic fatty acid synthase (FASN) mRNA levels were lower (p<0.05) in the DR group compared with the normal group. The DR group tended (p = 0.06) to have higher of levels of growth hormone receptor (GHR) mRNA than the normal group. At P2, the DR group tended to have lower (p = 0.06) androgen receptor (AR) mRNA levels than the normal group. In conclusion, our results demonstrate that dietary restriction partially decreases the transcription of lipogenic FASN and growth hormone signaling AR genes, but increases transcription of the GHR gene. These changes in gene transcription might affect body fat accumulation and the growth of the animals.
Ameliorating effects of dolomite and sludge on the polluted soil sampled from Ulsan and yeocheon Industrial Complexes were investigated. Ameliorating effects were analysed by changes of soil properties and plant growth after treatment of dolomite and sludge. Soil properties were investigated by analysing organic matter, N, P, K, Ca and Mg contents and pH. Growth of sample plants was investigated by leaf area calculated from length and breadth of leaves and by biomass from diameter and height of sample plants. Quercus serrata and Celtis sinensis selected as tolerant plants in field survey were used as experimental plants. Treatment with dolomite showed ameliorating effects by increassing n, Ca, and Mg com\ntents, and pH of soil and by decreasing Al content. Treatment of sludge showed similar effects by increasing N, Ca, Mg and organic matter contents, and by decreasing A1 content. But treatment of sludge did not show any effect on pH. Both soil ameliorators showed accelerating effects on the growth of experimental plants in Ulsan soil. But those effects in Yeocheon soil were somewhat different. Treatment of sludge showed accelerating effects of the growth of both sample plants but dolomitic liming did not so. From those results, we confirmed availability of sludge, a kind of industrial waste, as one of ameliorators of the polluted soil. In addition, we recognized that soil properties had to be considered to select soil ameliorators suitable for restoration of degraded ecosystems.

Strain PUPC1 produces an antifungal protease as well as plant growth promoting enzymes such as 1-aminocyclopropane-1-carboxylate (ACC) deaminase and phosphatase. Morphological, cultural, and physiological characteristics as well as 16S rRNA gene-sequence-based phylogenetic analysis confirmed the taxonomic affiliation of PUPC1 as Chryseobacterium aquaticum. The optimum growth of PUPC1 was observed at pH 6.0 and $30^{\circ}C$, and maximum protease production was observed in medium B amended with 1% tryptone, 0.5% sucrose, and 0.005% $MnCl_2$. The protease was purified by ammonium sulfate precipitation, Sephadex G-75 gel filtration chromatography, and electroelution from preparative SDS-PAGE. The protease had a molecular mass of 18.5 kDa. The optimum pH and temperature stability of the protease were pH 5.0-10.0 and temperature $40-70^{\circ}C$. Chryseobacterium aquaticum PUPC1 and its protease showed a broad-spectrum antifungal activity against phytopathogenic fungi. Strain PUPC1 also exhibited plant growth promoting traits. The objective of the present investigation was to isolate a strain for agricultural application for plant growth promotion and biocontrol of fungal diseases.
Objective of this study was to select ectomycorrhizal fungi for black spruce(Picea mariana) inoculation to overcome the growth inhibitory effects of Kalmia angustifolia. Nineteen isolates representing 11 species of ectomycorrhizal fungi were tested for their abilities to grow and form mycorrhizae with black spruce seedlings in the presence of water leachate of leaves of Kalmia. Mycelium growth of 9 isolates were inhibited by the leaf leachate. Colony diameter and biomass of the other 10 isolates were either increased or unaffected under the same conditions. Acidic pH of the culture medium(pH 3 and 4) inhibited some of the fungi, but a combination of acidic pH and the leaf leachate was more inhibitory. Thirteen isolates were able to form ectomycorrhizae with black spruce in presence of 25% leaf leachate in pure culture. Four isolates, Paxillus involutus(NF4), Cenococcum geophilum(GB12), Laccaria laccata(GB23), and E-strain(GB45) formed mycorrhizae more successfully than the others in presence of up to 50% Kalmia leaf leachate. Black spruce seedlings pre-inoculated with these fungi were grown with Kalmia leaf leachate and live Kalmia plants during a four month greenhouse experiment. Abundant mycorrhizae(77-91% of root tips) were developed on seedlings pre-inoculated with P. involutus, L. laccata and E-strain but relatively poor mycorrhization(32% of root tips) resulted with C. geophidum. Over 90% of the short root mycorrhizae were attributed to the inoculated fungi although indigenous mycorrhizae also occurred on most seedlings. Persistence of the mycorrhizae was not affected by living Kalmia plants. Over 80% of the mycorrhizae on seedlings inoculated with P. involutus, L. laccata and E-strain and 53% of the mycorrhizae on seedlings inoculated with C. geophilum were attributable to the inoculant fungi. Control seedlings formed about 45% ectomycorrhizal short roots with indigenous fungi. The L. laccata and C. geophilum inoculated seedlings exhibited enhanced mycorrhizae formation in presence of Kalmia leaf leachate. Mycorrhizae formation with inoculant fungi was 4-15% lower at pH 4 than at pH 5, with the greatest inhibition occurring for L. laccata. Seedlings inoculated with P. involutus had the greatest shoot and root growth followed by L. laccata and E-strain inoculated seedlings. The P. involutus and L. laccata inoculated seedlings were significantly taller with more shoot dry biomass than the uninoculated(control) seedlings. E-strain inoculated seedlings had significantly higher shoot dry biomass and significantly lower number of first order lateral roots compared to the control but other growth parameters such as height, root dry weight and number of short root tips were not significantly different from the control. Seedlings inoculated with C. geophilum were not significantly different from the uninoculated seedlings in any of the growth parameters except for the number of first artier lateral roots which was significantly less than the control seedlings.
The mycelial dispersed growth of Cordyceps sinensis was optimized in submerged batch culture at initial pH of 5.0, 150 rpm, and $25^{\circ}C$. The morphological data showed much more dispersed growth of C. sinenesis at initial pH of 5.0. Also, projected area, main hyphal length and number of tips for the mycelial growth of initial pH 5.0 were higher than those of other initial pHs. The industrial medium for mycelial production of C. sinensis was determined to be molasses of 100 g and crushed brewery yeast of 10 g per liter as carbon and nitrogen sources, respectively. With these culture conditions, the maximum production of mycelia was approximately 30.0 g per liter by batch culture in 5-liter jar fermenter with no controlled pH. This result suggests that large-scale mycelia production of C. sinensis may be possible in submerged batch culture. The hot water extract of mycelia from C. sinensis was mainly composed of 83.0% carbohydrate, 11.8% protein, 1.9% lipid, and 2.4% ash and there were present glucose, mannose, galactose, and arabinose as molar ratio of 8.79 : 2.59 : 1.34 : 1.0 in the carbohydrate, respectively. In the experiment using spleen cell and macrophage, the extract showed potent mitogenic and immuno-stimulating activities and among various components, an important factor that contribute to the immunological activities was turned out to be carbohydrate moiety.
The water extract of Sinomeniiacuti Radix (Sinomenium acutum) was found out to have a strong inhibition activity on the growth of Clostridium perfringens. The anti-bacterial activity was stable at the range of pH 1 to pH 13 and kept in a thermal treatment at $121^{\circ}C$ for 15 minutes. The minmal inhibition concentration of the Sinomeniiacuti Radix extract on the growth of Cl. peifringens was 1000 ppm. The Sinomeniiacuti Radix extract also suppressed the growth of Cl. ramosum, Cl. paraputnficum, Cl. butyn'cum, Bifidobacterium blficum, Bacteriodes fragilis, Eubacterium limosum. The extract, however, did not inhibit the growth of Bif adolescentis, Blf. infantis, Bif longum, E. coli. Enterococcus faecalis and Staphylococcus aureus. On the other hand, the extract showed a promoting effect for the growth of Bif animalis and Lactobacillus acidophilus.
