Hair growth effects of Mori Cortex Radicis Mixture on the hair of rat (탈모증에 대한 상백피 복합물의 모발성장 효과)

  • This study was carried out to investigate the hair growth effect of Mori cortex radicis mixture on the hair of rat. Cytarabine(50mg/kg) was injected to eight-day-old rats for 7 days. When the mixture of Mori cortex radicis was administered to the rat by route of skin, it promoted the growth of hair. These data suggest that Mori cortex radicis mixture have an effect on the hair growth in cytarabine-induced alopecia rat.

A Comparative Study on Effects of Kyungohkgo and Kyungohkgo Ga Nokyong on Growth in Growth Deficiency Rat with Insufficient Nutrition Diet (성장장애 흰쥐에서 경옥고(瓊玉膏)와 경옥고가록용(瓊玉膏加鹿茸)의 성장효과 비교 연구)

  • Objectives This study was designed to evaluate the difference of between Kyungohkgo and Kyungohkgo Ga Nokyong effects of growth Methods We divided male Spraque-Dawley rats into 6 groups. They were Normal group, Growth deficiency rat with insufficient nutrition diet group, Growth deficiency rat with 0.1% Kyungohkgo group, Growth deficiency rat with 0.2% Kyungohkgo group, Growth deficiency rat with 0.1% Kyungohkgo Ga Nokyong group and Growth deficiency rat with 0.2% Kyungohkgo Ga Nokyong group. They were administered for 5 weeks. We measured body weight, serum growth hormone, insulin-like growth factor and thyroid stimulating hormone, RBC, concentration of Hb and PCV ratio, total WBC and its composition, the values of plasma glutamic oxaloacetic transaminase(GOT) and glutamic pyruvic transaminase(GPT) activities. Results 1. In body weight, Kyungohkgo 0.1%, Kyungohkgo Ga Nokyong 0.1%, 0.2% groups were showed sighnificantly different than control group. But That groups were not showed significantly different than each others. 2. In serum growth hormone, insulin-like growth factor and thyroid stimulating hormone, we obtained the results of tendency to increase in Kyungohkgo Ga Nokyong group, however these values showed no significantly different. 3. In the counts of RBC, Kyungohkgo group and Kyungohkgo Ga Nokyong group were showed significantly different than control group. But these values showed no significantly different. In the concentration of Hb, Kyungohkgo Ga Nokyong 0.2%group was showed significantly different than control group. Kyungohkgo Ga Nokyong 0.2% group and Kyungohkgo 0.2% group were showed significantly different than control group. 4. The counts of total WBC and its composition showed no significantly different in all treatment groups. 5. The values of plasma glutamic oxaloacetic transaminase(GOT) and glutamic pyruvic transaminase(GPT) activities showed no significantly different in all treatment groups. Conclusions So Kyungohkgo Ga Nokyong and Kyungohkgo have an effect of promoting growth of rats. And We know that the effect of Kyungohkgo Ga Nokyong is better than Kyungohkgo.

Effect of 3-Methylcholanthrene on Rat Uterus: Uterine Growth and Mechanism of Action of 3-Methylcholanthrene

  • This study has been undertaken to examine the effect of 3-methylcholanthrene (3MC) on rat uterine growth and to understand the mechanism of action of 3MC in rat uterus. After diethylstilbesterol(DES) or tamoxifen(TAM) or 3MC or DES plus TAM or DES plus 3MC was administered into immature female rats, uterine weight over corn oil-treated uteri. 3MC treatment had no effect on uterine weight but, DES stimulated uterine weight was inhibited by 3MC concomitant tratment. While TAM alone treatment showed slight increase in uterine wieght, inhibited uterine growth simulated by DES when it was adiministrated with DES condirect binding assay with $[^3H]$ estradiol and the relative binding affinities of 3MC and TAM were estimated by competetion assy. Estradiol tumed out to have high affinity for rat uterine estrogen receptor (kd = 0.4 nM). The relative binding affinities of TAM and 3MC were 1% and 4.7% that of DES for rat uterine estrogen receptor, respectively. 3MC was shown to have similar affinity for eat uterine estrogen receptor to that of TAM. Effects of DES 3MC and TAM administration in vivo on rat uterine estrogen recptor level were examined. It was confirmed that the estrogen, DES and antiestrogen, TAM decreased estrogen receptor levels from rat ulterus and also 3MC decreased rat uterine estrogen receptor level when rats were treated with DES, TAM and 3MC in vivo. Data indicates that 3MC acts as an antiestrogen mediated through estrogen receptor system.

  • The insulin-like growth factor-I(IGF-I) is an important metabolic factor involved in cell growth and metabolism. Although secretion of IGF-I in rat liver is regulated by growth hormone, the effects of forskolin, adenylate cyclase activator, on secretion of IGF-I have not been reported. Therefore, a modified perfused rat liver model was used to investigate the regulatory effects of forskolin on IGF-I secretion in this experiment. The results were summerized as follows : 1. Modified perfused rat liver model was not changed to aspartate aminotransferase(AST), alanine aminotransferase(ALT) and lactic dehydrogenase(LDH) secretion in time. 2. The IGF-I secretion in hepatic cell was increased by forskolin($10^{-5}$, $10^{-6}$ and $10^{-7}M$) in a dose-dependent manner as compared with those of the controls, and significantly increased by $10^{-5}$ and $10^{-6}$ forskolin(p < 0.05). 3. Secretion of glucose in hepatic cell significantly was decreased by $10^{-5}$ forskolin as compared with those of controls(p < 0.05). These results suggest that forskolin may be involved in the regulation of IGF-I secretion in the perfused rat liver.

Effects of Yongohkgo on Growth and Learning Ability in Growth Deficiency Rat With Linsufficient Nutrition Diet (영양소 결핍으로 유도한 성장장애 흰쥐에서 용옥고(龍玉膏)가 성장 및 학습효과에 미치는 영향)

  • Effects of Kyungohkgo Ga Nokyong(Yongohkgo) on growth development and learning ability were investigated growth and intellectual impairment rat with insufficient nutrition diet. We divided male Sprague-Dawley rats into 4 groups. They were Normal group, Growth deficiency rat with insufficient nutrition diet group, Growth deficiency rat with 0.1% Yongohkgo group and 0.2% Yongohkgo group. They were administered for 5 weeks. We measured body weight, and morris water maze test in escape distance, escape time and escape speed, serum growth hormone, insulin-like growth factor and thyroid stimulating hormone, RBC, concentration of Hb and PCV ratio, total WBC and its composition, the values of GOT and GPT activities. The results are as follows that Yongohkgo 0.1%, 0.2% groups were showed significantly different than control groups in body weight and the counts of RBC. In the morris water maze test, in escape distance and escape time, in concentration of Hb and PCV ratio, 0.2% Yongohkgo group were significantly different than control groups. Serum growth hormone, insulin- like growth factor and thyroid stimulating hormone showed a tendency to increase in Yongohkgo groups. The counts of total WBC and its composition, GOT, GPT activities showed no significantly different in all treatment groups. These results suggested that Yongohkgo have an effect of promoting growth and learning ability of rats and might be effect to treat various kinds of growth and learning ability delay in children.

The Effect of Achyranthis Radix Extract on Hard Tissure Regeneration (우슬 추출물의 경조직 재생촉진효과)

  • This study was performed to investigate therapeutic effects of Achyranthis Radix extract and chitosan on the growth and differentiation of rat calvarial cells. it was found that treatment of methanol extract of Achyranthis Radix for 2 days caused 2.4-fold increase in the growth of rat calvarial cells. However, chitosan treatment caused only 1.9-fold increase in the cell growth. Treatment of methanol extract of Achyranthis Radix for 14 days caused 2-fold increase in the growth of rat calvarial cells. Alkaline phosphatase activity, one of the markers for bone cell differentiation, was increased approximately by 1.7-fold and 2.9-fold by the treatment of methanol extract of Achyranthis Radix for 2 days and 14 days, respectively. These results suggest that Achyranthis Radix extract could be beneficial for bone regeneration.

  • 1. Cooked soybean was fermented for about a week by Bacillus megatherium 88-3, D-28 and D-28a; and compared with the cooked soybean which was not fermented, this fermented soybean showed much increase in vitamin $B_{12}$ and $B_2$, while vitamin $B_1$ decreased. 2. In the process of fermenting the cooked soybean, Bacillus megatherium, 88-3 is the bacteria which produces brown pigment; and Bacillus megatherium D-28 and D-28a is the bacteria which produces yellow pigment. 3. In weight, fermented soybean-fed rat showed more increase than the unfermented soybean-fed rat. But the growth of the rat fed with Bacillus megatherium 88-3 fermented soybean was not good. Probably, this phenomenon came from the trouble in rat's metabolism by brown pigment. 4. In food efficiency, Bacillus megatherium D-28a is 4.3% lower than Bacillus megatherium B-938, but it is 17.l% higher than the unfermented cooked soybean. Bacillus megatherium 88-3, which is 47.1% lower in food efficiency than the unfermented cooked soybean, has been found "not good" in the growth of rat.th of rat.

  • S-Adenosyl-L-methionine synthetase (ATP: methionine S-Adenosyltransferase, EC 2.5.1.6; AdoMet synthetase) catalyzes the biosynthesis of S-Adenosyl-L-methionine(AdoMet) from methionine in the presence of ATP. To elucidate the role of transmethylation reaction in the pancreatic tissues, we examined AdoMet synthetase and isozyme activities, and AdoMet contents in the various tissues. The activities of AdoMet synthetase marked the highest in the kidney, and the lowest in the testis among the various tissues of rat. Considerable amounts of AdoMet synthetase activities were detected in the pancreatic tissues of various animals except for those of frog. The level of ${\alpha}$ and ${\gamma}$ isozyme activities were present in the pancreatic tissues of various animals, while ${\beta}$ isozyme activities were detected as trace. AdoMet synthetase activities of rat brain, liver, testis were decreased with growth. In the rat pancreatic tissues, AdoMet synthetase activities were increased during 16 days after birth and then decreased between 16 and 47 days of age. Levels of AdoMet contents of rat brain and testis were decreased with growth. However, AdoMet contents of rat pancreas were decreased until 26 days of age, and then increased thereafter. AdoMet synthetase isozyme patterns did not vary with growth in the pancreas and testis. But, in the liver, ${\beta}$ form is strikingly increased with growth.

Induction of Growth Hormone by the Roots of Astragalus membranaceus in Pituitary Cell Culture

  • The traditional Asian medicinal herb, roots of Astragalus (A.) membranaceus (Leguminosae), is used for many purposes, some of which are purported to stimulate the release of growth hormone in vivo. Extracts of A. membranaceus were tested to determine whether they stimulate the release of growth hormone in rat pituitary cell culture. A. membranaceus was extracted sequentially with 80% ethanol (fraction A), n-hexane (fraction B); the test compound from the herbal extraction was isolated using silica gel column chromatography and was identified with spectral data. Test compound was also extracted by traditional boiling water methods. Induction of growth hormone in pituitary cell culture was conducted with isolated compounds and extracted fractions of A. Radix (dried roots of A. membranaceus). The fraction A was not active in the rat pituitary cell culture, but the fraction B derived from the ethanol fraction stimulated the release of growth hormone in culture. Six compounds from fraction B (1-6) were isolated and identified previously. The compounds 1,2-benzendicarboxylic acid diisononylester (1), $\beta$-sitosterol (2), and 3-Ο-$\beta$-D-galactopyranosyl-$\beta$-sitosterol (5) did not induce growth hormone release in the culture. Formononetin (3), 9Z, 12Z-octadecadienoic acid (4), stigmast-4-en-6$\beta$-o1-3-one (6) and 98-E, a mixture of 1'-9, 12-octadecadienoic acid (Z,Z)-2',3'-dihydroxy-propylester (7) and 1'-hexadecanoic acid-2',3'-dihydroxy-propylester (8) stimulated the release of growth hormone in the rat pituitary cell culture significantly compared to the control. In conclusions, four compounds isolated from extracts of A. Radix induced growth hormone release in the rat pituitary cell culture. The 98-E isolate was the most active inducer of growth hormone release.

THE GROWTH OF RAT MANDIBLES FOLLOWING MANDIBULAR RETRACTIVE FORCE (백서 하악골 후방견인후의 하악골 성장에 관한 실험적 연구)

  • The purpose of this study was to investigate changes of the mandible of a growing rat when they are subjected to a retractive force and those after removal of the retractive force. The experimental animals were Sprague-Dawley male rats of four weeks of age. A mandible was retracted with 50 grams of force on each side in the posterior and superior direction for 8 hours per day. The animals were sacrificed after 1 week, 2-week and 4-week force application, and after 4-week force application-4-week force removal period. The changes of rat mandibular growth following retractive force on the growing rat mandible were observed histologically and biometrically. The findings were as follows ; 1. Histologically, the thickness of the condylar cartilage was slightly reduced in the anterosuperior region with the retractive force. However, in the group of 4-week force application-4-week force removal, there was no significant difference in the thickness of the condylar cartilage. 2. There were no significant histological changes in the articular disk and glenoid fossa through the experimental period. 3. The length and anterior height of the mandible subjected to the retractive force were significantly smaller and greater than those of the control group. 4. There were no significant differences in the mandibular length between 4-week force application - 4-week force removal and the control group. 5. It was concluded that a mandibular retractive force produced inhibitory effects in the growth of the mandible, but that these effects were not sustained during mandibular growth in this experimental model.
