Effects of environmental factors on growth and morphology of mycoplasma pneumoniae (Mycoplasma pneumoniae의 성장과 형태에 미치는 환경요소적 영향)

  • Mycoplasma pneumoniae was examined for growth characteristics and morphology when cultivated in several media supplemented with a variety of sera and under different atmospheric conditions. different formula of the medium as well as different sources of lot numbers of the serum in the same medium exhibited varying effects on growth rate and adherence. When the organisms were cultivated in SSR-2 medium in a normal atmospheric environment or under a facultative anaerobic condition provided with carbon dioxide, they developed filamentous cells with heavy growth, whereas mainly round-shaped cells were produced under strict anaerobic conditions of hydrogen and carbon dioxide. Both morphologies of the organism were transformed by switching the incubation environments. An inverted phasecontrast microscopy using modified petri dishes was excellent to observe single cells and useful to follow the development of the cells. Growth, turbidity, and pneumoniae colonies developed on a solid medium and produced clear when overlaid with sheep blood agar.

Low Serum Medium for CH07E02 Hybridoma (하이브리도마 배양을 위한 저혈청배지의 조성 결정)

  • Up to now, 10% Fetal Bovine Serum(FBS(V/V)) was added to basal medium for the cultivation of hybridoma. For the cultivation of hybridoma cell line, CH07E02, against colon cancer, serum concentration was reduced to 3% FBS without influence on cell growth and maximum cell concentration. By the addition of cell growth promoting substances-insulin (I), pyruvate (P), oxaloacetate(O), Pluronic F-68(P) and 2-mercaptoethanol(2-ME)-to 1% FBS medium, a cell density higher than that with 1% FBS medium alone was achieved. FBS 3% medium was replaced by very cheap 2% Calf Serum (CS) medium without influence on cell growth rate and concentration. Cells grew vigorously in 0.5% CS+IPOP medium. This composition was used during suspension culture and exhibited good viability and high specific growth rate.

  • The effects of post-cultural addition of liquid medium addition to stimulate in vitro bulblet growth in Lilium oriental hybrid 'Casa Blanca' were investigated. The sections of bulblets with swollen basal plate (7 mm$\times$12 mm) were cultured on medium containing 60 g/L sucrose and 1 g/L activated charcoal for two months in dark, and then, liquid medium was added into the same vessels. The addition of liquid medium stimulated the growth of bulblets remarkably, compared to no addition of liquid medium. The liquid medium supplemented with 120 g/L sucrose and double strength of MS salts were the most effective on the growth of in vitro bulblets.

  • Tabtoxin produced in Pseudomonas syringae pv. tabace irreversibly inhibits its known physiological target, glutamine synthetase so that causes wildfire disease on leaves of host plant. In this study, we examined a rapid and sensitive microbiological method for tabtoxin assay in several media. In minimal A agar medium nd minimal glucose agar medium, growth inhibition zone of Agrobacterium tumefaciens was larger than that of other indicator strain. However, mostly, growth inhibition zone of indicator strains on the minimal glucose agar medium was smaller than that of on the miniaml A agar medium. In complex agar medium, growth inhibithiton zone was not observed in all the tested indicator strains. Pseudomonas syringae pv. tabaci produced more tabtoxin according to the incubation time. When glutamine was added to the minimal glucose agar medium, growth inhkbition zone of Agrobacterium tumefaciens was reduced.

The Influences of Extracts from Ceratium spp, on the Growth of Harmful Microalgae (유해성 적조생물에 대한 Ceratium 추출물 영향 평가)

  • The growth response of the fish-killing dinoflagellate Cochlodinium polykrikoides was studied in cultures, using the treatment of Ceratium extracts by a methanol, a water-soluble, and a cell-free medium. The cell-free medium had the most increasing on the growth of C. polykrikoides cultures, enriched with $\geq$ 25% Ceratium, whereas the methanol and water-souble fractions did not affect the growth of C. polykrikoides exposed to even higher concentration. In particular, the cell-free medium also increased the growth of Gyrodinium impudicum and Chaetoceros sp., similar species to C. polykrikoides. In contrast to C. polykrikoides, G. impudicum and Chaetoceros sp., the growth of Alexandrium tamarense was inhibited significantly, and there was no great effect on the growth of Prorocentrum minimum. These results imply that Ceratium extracts may play an important role in the stimulatory effect of C. polykrikoides, and they have to affect the interaction between C. polykrikoides and Ceratium when co-existing.

The effect of root zone environment on the growth of shoot and root of tobacco plant (연초의 근권환경이 뿌리와 지상부의 생육에 미치는 영향)

  • This experiment was conducted to investigate the effect of growth medium in pots composed of upland soil(S), rice straw manure(M), carbonated rice hull(CRH), and their mixture on growth of tobacco cv. NC 82. The growth of shoot and root was vigorous in order of medium S+M+CRH>M>CRH>S. In M+S medium, root growth in the part of manure was superior to upland soil. But root growth of upland soil part in M+S plot was more vigorous than that in upland soil only. It is possibly due to be influenced by manure in M+S plot. Total length and weight of root, number of roots, and especially for development of adventitious root were closely related to shoot growth. Roots grown in upland soil part was brownish gray in color, while the roots in manure part was milky white. The milky white colored roots had longer life than others. It was concluded that root zone environments derived from several media in pots closely related to shoot growth and disease tolerance of tobacco plant.

Effect of Inorganic Salts in MS Medium, Sucrose, and Activated Charcoal on Bulblet Formation from in Vitro Bulbscales in Lilium Oriental Hybrid 'Casa Blanca' (MS 배지 무기물, 당 및 활성탄의 농도가 Lilium Oriental Hybrid 'Casa Blanca'의 기내인편으로부터 자구형성에 미치는 영향)

  • The effects of MS salt strength, sucrose, and cultural conditions on bulblet formation and growth were investigated to optimize the conditions for micropropagating bulblets from in vitro bulb scales of Lilium Oriental Hybrid 'Casa Blanca'. There was no difference on bulblet formation in the range of 1/2~2 $\times$ strength of MS salt, but it was inhibited remarkably in 3 $\times$ strength of MS salt. The growth of regenerated bulblets was most stimulated on MS basal medium. Favorable bulblet formation and its growth from bulb scales were achieved when grown on the media with 1 : 2 or 1 : 3 in the ratio of NH$_4$^+ : NO$_3$^- , as well as on MS basal medium (NH$_4$^+ : NO$_3$^- = about l : 2). Therefore, MS basal medium was very suitable for bulblet formation and growth from bulb scales. Bulblet formation was inhibited but its growth was stimulated with increase sucrose concentration in the medium. The growth of regenerated bulblets was very effective on the media with 9~12% sucrose. Addition of activated charcoal (AC) to the medium inhibited bulblet formation from bulb scales, but enhanced the growth of regenerated bulblets. Especially, the medium containing 1 g/L AC was most effective on the growth of bulblets. No difference was found on bulblet formation and growth from bulb scales under light and dark conditions. In vitro micropropagation of L. Oriental Hybrid 'Casa Blanca' was supposed very reasonable to enhance the growth of the bulblets after forming of bulblets from in vitro bulb scales, and then, subculture the bulb scales from the grown bulblets on MS medium with 9% sucrose and 1 g/L AC.

Development of a Selective Medium for the Fungal Pathogen Cylindrocarpon destructans Using Radicicol

  • The soil-borne ascomycete fungus Cylindrocarpon destructans causes ginseng root rot disease and produces various secondary metabolites such as brefeldin A and radicicol. The slow growth of this fungus compared with other plant pathogenic and saprophytic fungi in soil disturbs isolation of this fungus from soil and infected ginseng. In this study, we developed a selective medium for C. destructans using radicicol produced by this fungus. Supplementing 50 mg/L of radicicol to medium inhibited the mycelia growth of other fungi including Botrytis cinerea, Rhizoctonia solani and Alternaria panax, but did not affect the growth of C. destructans. In addition, conidia germination of other fungal species except for C. destructans was inhibited in submerged culture supplemented with radicicol. This medium provides a very efficient tool for isolating C. destructans and also can be used as an enrichment medium for this fungus.

Studies on the Cultural Characteristics of Hohenbuehelia petaloides (Hohenbuehelia petaloides의 배양학적 특성에 관한 연구)

  • This study was carried out to obtain the basic data on artificial culture of Hohenbuehelia petaloides. The optimum medium are glucose peptone medium (GP), Hennerberg medium, Phellinus igniarius medium (PIM), Lentinus edodes medium (LEM), Czapek dox medium. The optimum condition for the mycelial growth was $30^{\circ}C$ and pH 6.0. The carbon sources such as dextrine, fructose and lactose were favorable to mycelial growth. The optimal concentrations of carbon sources are 10% dextrin and fructose. As nitrogen sources, tryptone, casamino acid and histidine appeared to be favorable. The optimal concentrations of nitrogen sources are 1% soy tone and 0.3% ammonium nitrate. The optimal concentration of yeast extract is 0.4%. The mineral nutrients of $KH_2PO_4$, $K_2HPO_4\;and\;MgSO_4{\cdot}7H_2O$ were effective and the optimal concentrations were 0.046, 0.1 and 0.05%, respectively.

Effects of Medium , Plant Growth Regulators , and Explant Sources on Plant Regeneration of Rehmannia glutinosa (배지, 생장조절물질 및 치상조직이 지황 체세포조직으로부터 식물체분화에 미치는 영향)

  • When the leaf and stem tissues of Rehmannia glutinosa were cultured on MS medium with plant growth regulators, shoots were regenerated on MS medium with TDZ(0.01 to $2.0{\mu}M$), and root initiation was better on MS medium treated with NAA(0.01 to 2.0mg/$\iota$). On $B_5$ medium, shoot regeneration was better on medium with TDZ than those with Quincrac, NAA, and 2.4-D. The addition of Quincrac, NAA, and 2.4-D inhibited shoot regeneration on MS medium, but promoted shoot regeneration on $B_5$ medium. Shoot regeneration and growth was better on MS medium with the combination treatments of 2.4-D 0.01mg/$\iota$ and TDZ 0.01, 0.1 and $2{\mu}M$ compared with other treatments. Also shoot regeneration and growth on B_5 medium showed the similar results to that on MS medium.
