Neurospora의 생육시기에 따른 호흡능의 변화와 자외선 감수성과의 상관관계

  • Using conidia of Neurospora, changes in respiratory activities and the sensitivity to the ultraviolet light of the cells at different growing stages were measured by manometric methods, and the correlation between them was observed. Efficiency in the utilization of various carbon sources, such as, glucose, sucrose, maltose, starch and sodium acetate, in growth and exogenous respiration of N. crassa was also determined. Growth rate of N. crassa was decreased considerably in the medium containing sodium acetate than in the glucose medium and was almost zero in the lactose medium, whereas the utilization of sucrose, maltose and starch was ve교 high, as that of glucose. Respiratory activities of the cells veried considerably depending upon their different growing stages. Actively growing hyphae exhibited the greatest activity in exogenous glucose respiration, followed by germinating and activated conidia in decreasing order. There was no proportional relationship between the dose of ultraviolet light irradiated and its effect on the respiratory activity of the cells, though the more the dose of ultraviolet light, the more the injury. The sensitivity of the cells to ultraviolet light varied with the different respiratory activities of the cells linked to the developmental stages. In general, the more actively growing cells having high respiratory activities exhibited the more serious injury.

Promotion strategy of Economic Base through Employment Growth Analysis (고용성장율 시차분석을 통한 산업기반의 확충방안 - 충남지역의 사례 -)

  • This paper primarily aims to analyze the growth rate differentials of the economic activities in chung-Nam area. The research has been undertaken to find out the potentials of the economic activities and economic bases in the area. To analyze the economic potentials of the industries, the study employed the economic Growth Rate Differential Analysis by Henderson which is revised form of Shift-Share Analysis Methods. The research employed the employment data according to the standardized two-digit-classification-system during the period of 10 years from 1981 through 1991. The Growth Rate Differential Analysis calculates Total Growth Differentials which are dicomposed into two parts: Weight Part and Rate Part. Total Growth Differential can be calculated as the difference between national growth rate and regional growth rate by industry. The foundings are as follows: First, the economic bases of Chung-Nam area were found to be very weak, largely depending on primary industries such as agriculture and fisheries. Second, there was a great decline in urban industries in Tae Joeon, Cheon An and other cities over period of 1971-1981. It is strongly recommended that the planned items and products of each industrial complex must be reorgnized in a fashion to match those with high competitive power found in this research.

Cadmium-Induced Phytotoxicity in Tomato Seedlings Due to the Accumulation of H2O2 That Results from the Reduced Activities of H2O2 Detoxifying Enzymes

  • Tomato (Lycopersicon esculentum) seedlings exposed to various concentrations of $CdC1_2$ (0∼100 $\mu$M) in the nutrient solution for up to 9 days were analyzed with the seedling growth, $H_2O_2$ production, glutathione levels and activity changes of enzymes related to $H_2O_2$ removal. The growth of seedlings was inhibited with over 50 $\mu$M Cd, whereas the levels of $H_2O_2$ and glutathione were enhanced with Cd exposure level and time. Meanwhile, Cd exposure increased the activities of catalase (CAT) and glutathione reductase (GR) but decreased the activities of dehydroascorbate acid reductase (DHAR) and ascorbate peroxidase (APX) in both leaves and roots. These results suggest that the altered activities of antioxidant enzymes particularly involved in the $H_2O_2$ removal and the subsequent $H_2O$$_2$ accumulation could induce the Cd-induced phytotoxicity.

Effects of Oxidative Stress on Growth Performance, Nutrient Digestibilities and Activities of Antioxidative Enzymes of Weanling Pigs

  • This study was undertaken to investigate the effects of oxidative stress on growth performance, nutrient digestibilities and activities of antioxidant enzymes of weanling pigs. In the experiment, 24 male $Landrance{\times}Yorkshire $weanling pigs were allotted to three groups of 8 animals each. Pigs were fed individually. According to a single factorial arrangement, pigs received diets with 5% of either fresh (group 1 and group 3) or oxidized fish oil (peroxide value was 786.50 meq $O_2/kg$ before inclusion in the diet, group 2). At the beginning of the experiment, pigs in group 3 received an intraperitoneal injection of diquat at 12 mg/kg of body weight. The trial lasted for 26 d. A metabolism test was carried out during the last 4 days of the second week. The results showed that feeding diets containing oxidized fish oil or injection with diquat depressed the growth performance and nutrient digestibilities of weanling pigs, decreased activities of antioxidant enzymes and increased concentration of malondialdehyde in plasma and liver. Intraperitoneal injection of diquat would induce more serious oxidative stress than oral intake of oxidized fish oil in the diet. In conclusion, administration of oxidized fish oil or diquat could induce oxidative stress in weanling pigs, and oxidative stress could depress growth performance and impact anti-oxidative ability of young pigs.

Antimicrobial Activities of Medicinal Herb Extracts (한약재추출물의 항균활성)

  • In this study, 18 kinds of Korean medicinal herb extracts were examined for anti-microbial activities against pathogenic microorganisms. The methanol (MeOH) extracts from Schizandra chinensis, Rhus javanica and Caesalpinia sappan exhibited antimicrobial activities against most pathogenic microorganisms at concentrations of 5 mg/mL, whereas the other 15 extracts exhibited anti-microbial activities at concentrations of 30 mg/mL. The minimum concentration at which Schizandra chinensis extracts inhibited for S. epidermidis and Bor. bronchiseptica was 0.6 mg/mL. The MeOH extracts from Schizandra chinensis, Caesalpinia sappan, Rhus javanica and Seutellaria baicalensis which had higher anti-microbial activities were subsequently fractionated using 5 different solvents, and further screened for anti-microbial activities. The inhibitory effects of ethyl acetate (EtOAc) extracts on microbial growth were greater compared to any other solvent extracts. In order to investigate the inhibitory effect of Korean medicinal herbs with high anti-microbial activities on microbial proliferation, the MeOH extracts at concentrations of 0, 100, 300 and 500 ppm were added to the media. No addition of extracts caused rapid growth of microbes after 12 hours incubation. As the concentration of extracts from Rhus javanica and Caesalpinia sappan increased, the growth-inhibiting effect on gram-positive bacteria including S. aureus, S. epidermidis, and L. monocytogenes was prominent. Rhus javanica extracts exhibited growth-inhibiting activity for gram-negative bacteria including Sal. Pullorum and Sal. Choleraesuis. The low concentration of extracts from Rhus javanica and Caesalpinia sappan exhibited the growth of Bor. bronchiseptica and E. coli serotype $O_8$. However, the higher concentration of extracts from Rhus javanica and Caesalpinia sappan exhibited a strong inhibitory effect on microbial proliferation.

Effect of Cadmium on Oxidative Stress and Activities of Antioxidant Enzymes in Tomato Seedlings

  • Leaves of two-week old seedlings of tomato (Lycopersicon esculentum) were treated with various concentrations (0∼100 M) of $CdCl_2$ for up to 9 days and subsequent growth of seedlings, symptoms of oxidative stress and isozyme activities of antioxidant enzymes including superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX) and peroxidase (POX) were investigated. Compared with the non-treated control, Cd exposure decreased biomass but increased Cd accumulation, hydrogen peroxide production and lipid peroxidation as malondialdehyde (MDA) formation in leaves and roots. Further studies on the developmental changes of isozyme activities showed that Fe-SOD, Cu/Zn-SOD and one of three APX isozymes decreased and CAT and one of four POX isozymes increased in leaves, whereas Fe-SOD, one of three POX isozymes and two of four APX isozymes decreased and CAT increased in roots, showing different expression of isozymes in leaves and roots with Cd exposure level and time. Based on our results, we suggest that the reduction of seedling growth by Cd exposure is the oxidative stress resulting from the over production of $H_2O_2$ and the insufficient activities of antioxidant enzymes particularly involved in the scavenging of $H_2O_2$. Further, the decreased activities of SOD and APX isozymes of chloroplast origin, the increased activities of CAT and POX and high $H_2O_2$ contents with Cd exposure might indicate that Cd-induced oxidative stress starts outside chloroplast.

  • KIPRIS patent database was analyzed for identifying the technological status of Daedeok Innopolis Biocluster. It was found that the pattern of activities among various technological areas in Daedeok Biocluster is similar to that of an advanced country rather than those of other cities in Korea. The technological growth in Daedeok Innopolis Biocluster is in the progressive stage, which may be due to the innovative activities rather than the rise in the number of new firms or institutes. The concentration of technology in Daedeok Innopolis Biocluster is a favorable condition for the innovation activities. The trend for the technological concentration was remarkably consistent with the growth curve that a population increases according to the logistic equation. The logistic growth may be represented by the result of competition due to the limited resource allocation and then innovation cluster is corresponding to the ecosystem composed by biological individuals. There is strong competition in Daedeok Innopolis Biocluster in around 2009, so the government might make a policy to encourage the technological diversity for healthy knowledge ecosystem.

  • Knowledge-based economy is the economy or economic structure based on production, accumulation and utilization of knowledge. With the emergence of knowledge-based economy, the importance to the corporate competitiveness of IP and IP-related activities has increased. This paper discuss the issues related to the mode of IP-related activities, including the comparison of SMEs and large firms. Especially, I focus on the role and impact of IP-related activities on innovation and growth of firm in machinery industry. The result of this study can help to set up strategy for supporting firm's technology innovation.

Effect of 1-[(2-Hydroxyethoxy)methyl]-6-(phenylthio)thymine (HEPT) Derivatives on Bacterial Growth

  • 6-Substituted derivatives of 1-[(2-hydroxyethoxy)methyl]-6-(phenylthio)thymine (HEPT) were synthesized by introducing alkyl groups with the aid of chlorotrimethylsilane, and then purified ranging 40 to 81 % of yield. Because of their peculiar structures, we presumed that HEPT derivatives would contain extra biological activities other than their already known anti-human immunodeficiency viral (HIV -1) activities. In this study, we investigated the possible effects of the HEPT derivatives on bacterial growth and found their selective antibiotic activities against gram-positive strains. We could not observe the corresponding activity from a disc-zone test, but confirmed the activity by liquid cultivation. Since the growth rate of cells was easily recovered, the antibiotic function was suggested to be bacteriostatic. We also suggested that the intracellular fate of HEPT derivatives would be fast. A HEPT derivative f-3 was shown to synergize unidirectionally toward chloramphenicol (Chr). With 0.1 mM f-3, the Chr-directed growth-inhibitory curve appeared 4 hours earlier than found without the additive. Interestingly, from the data of SDS-polyacrylamide gel electrophoresis (PAGE), we found that a membrane-bound protein having a molecular weight of 70-kDa was overexpressed by f-3 in S. aureus.
