THE EFFECTS OF FIBRONECTIN & GROWTH FACTOR ALONE OR COMBINED APPLICATION ON THE ACTIVITY OF GHUMAN GINGIVAL FIBROBLASTS AND PERIODONTAL LIGAMENT CELLS (Fibronectin과 성장인자의 단독 혹은 복합투여가 배양 인체 치은섬유모세포 및 치은인대세포의 활성에 미치는 효과)

  • The selective migration, attachment and proliferation of periodontal ligament cells are the desired goal of periodontal regeneration therapy. Fibronectin is well known for an attachment protein for dentin surface. Also, Fibroblast growth factor (FGF) is well known to enhance the periodontal regeneration. The purpose of this study was to evaluation the effect of fibronection and FGF on the attachment rate and the cellular activity. Human gingival fibroblast and periodontal ligament cells were cultured from the teeth extracted for non-periodontal reson. Cultured human gingival fibroblast and periodontal ligament cells in vitro were treated with fibronectin and FGF a various dosage and culture times. Cellular activity was examined by MTT assay. The results of this study was demonstrated that cell attachment rate of experimental group was under the control value at 1st, 2nd, 3rd incubation day. But, at 3rd incubation day, attchment value tended to return to the control value. In case of fibronectin alone application, cellular activity was decreased than that of control at 1st, 2nd incubation day. But 3rd day, cellular activity was returned to the control value. The activity of gingival fibroblast in FGF alone application was decreased thatn that of control at each incubation day. But activity of periodontal cell group was increased cell activities at 2nd, 3rd day. Additionally cellular activity of fibronectin & FGF combined application on gingival fibroblast group was similar to control value at incubation day. But activity of periodontal ligament cell group was increased at 2nd, 3rd day compared with control group.This study demonstrated that combined application of fibronectin & FGF induced the selective chemotaxis for periodontal ligament cell in vitro.

Antidiabetic Activity and Enzymatic Activity of Commercial Doenjang Certified for Traditional Foods (전통식품 품질인증 일부 시판 된장의 효소활성 및 항당뇨 활성)

  • We investigated the anti-diabetic activity and enzymatic activity of 24 commercial doenjang samples certified for traditional foods. Twenty four doenjang samples showed the wide ranges in enzymatic activities (protease activities 0-50.45 unit/g, ${\alpha}$-amylase activities 0-675.9 unit/g, ${\beta}$-amylase 13.6-308.6 unit/g), and there were no difference in enzymatic activity by the producing region. To evaluate the potential anti-diabetic activity of 24 doenjang samples, we examined the effect of doenjang methanol extract (DME) on 2-[n-(7-nitrobenz-2-oxa-1, 3-diazol-4-yl) amyno]-2-deoxy-d-glucose (2-NBDG) uptake. Ten samples among 24 samples significantly stimulated the uptake of 2-NBDG. When the cells were treated with DME at 400 ug/mL, No. 17 and 23 specially stimulated 2-NBDG uptake by 1.23-fold and 1.25-fold, respectively, compared with untreated control cell. And there were no cytotoxicity in the C2C12 cells treated with DME at concentration of 500 ug/mL. Among 24 samples, No. 6, 7, 12, 21 and 24 showed the ${\alpha}$-glucosidase inhibitor activity at concentration of 10 mg/mL; however, they were less effective than acarbose which is a commercial ${\alpha}$-glucosidase inhibitor.

The Effect of Fatigue on EMG Activity and Dynamic Balance of Subjects with Functional Lumbar Instability

  • Purpose: The aim of the current study was to investigate EMG activity on dynamic balance of subjects with functional lumbar instability following fatigue of low back. Methods: The subjects (24 university students) were divided into 2 groups; functional lumbar instability group (6 males and 6 females) and lumbar stable group (7 males and 5 females) who could complete a questionnaire and undergo a prone instability test. All participants were evaluated for distribution of muscle activity using the TeleMyo DTSTM system. Dynamic balance was tested by Y balance test. This study was conducted for measurement of EMG activity on dynamic balance with the difference between FLIG and control group following muscle fatigue. Results: The functional lumbar instability group (FLIG) showed a significantly lower YBT score (%) of anterior, posterolateral direction on Y-balance test (YBT) in dynamic balance than the lumbar stable group (LSG) (p<0.05). The FLIG was significantly lower than the LSG in anterior direction in EMG activity(%) of MF, RA, ES, GMX, GME, RF, and posteromedial direction in EMG activity(%) of IO, ES and then posterolateral direction in EMG activity(%) of IO, ES in dynamic balance (p<0.05). There was significant correlation of MF, RA, and GMX in anterior reach direction (p<0.05) and ES, GME (p<0.01) and IO, ES in posteromedial reach direction (p<0.05) and EO, ES, GMX in posterolateral reach direction (p<0.05) there was positive correlation. Conclusion: This study showed that FLIG effected EMG activity by dynamic balance following muscle fatigue. Further study is needed for measurement of various ages and work with lumbar instability for clinical application.

Effect of Prostaglandin $E_1$ and Acetazolamide upon Carbonic Anhydrase Activity of Whole Blood in Rat (Prostaglandin $E_1$과 Acetazolamide가 흰쥐 전혈(全血)의 Carbonic Anhydrase 활성에 미치는 영향)

  • This study was undertaken to investigate the influence of prostaglandin $E_1(PGE_1)$ upon the activity of carbonic anhydrase and upon the inhibitory action of acetazolamide on carbonic anhydrase. The heparinized blood was sampled by cardiac puncture from Sprague-Dawley strain rats under ether anesthesia and was hemolysed by adding distilled water 1,000 times the amount of the blood. The activity of carbonic anhydrase of 0.1 ml of the hemolysate was measured by Maren's simplified micro-method. In the first experiment, the 7 rats were used, and the activity was measured by adding 0.1 ml of various concentrations of $PGE_1$(0.5, 1.25, 2.5, 5.0, 10 and $20\;{\mu}g/ml$). In the second experiment, the 6 rats were used and the activity was measured by adding 0.1 ml of $PGE_1(5\;{\mu}g/ml)$ and 0.1 ml of acetazolamide$(6{\times}10^{-7}M/l)$ respectively or simultaneously. Obtained results were as follows: 1) The activity of carbonic anhydrase was significantly inhibited by $PGE_1$ at doses of $0.5{\sim}10\;{\mu}g/ml$, maximally at a dose of $2.5\;{\mu}g/ml$, but inhibition was no more observed at a dose of $20\;{\mu}g/ml$. 2) The activity of the acetazolamide group was significantly less than that of the control group. 3) The activity of the $PGE_1+acetazolamide$ group was significantly less than those of the $PGE_1$ group and the acetazolamide group. It is inferred from the above results that the $PGE_1$ inhibits the activity of carbonic anhydrase dose-dependently and strengthens the inhibitory effect of acetazolamide on carbonic anhydrase.

  • This study was conducted to investigate the effects of multibanded orthodontic appliances on dental caries activity. The subjects consisted of 50 males and females with multibanded orthodontic appliance and the same number of caries free group. The following conclusions were obtained, 1. There was significant difference between the experimental group and control group in the tests of Snyder colorimetric test, acid :buffering capacity and oral glucose clearance test (at $5\%$ level of significance). 2. There was no significant difference between the experimental group and control group in the tests of unstimulated salivary flow fate, stimulated salivary flow rate and salivary viscosity test (at $5\%$ level of significance). 3. Dental caries activity was higher in experimental group than in control group.

Comparative Analysis of Maximum Vertical Reaction Force and Lower Limbs on Drop Landing between Normal and Flat Foot Group

  • With comparison of maximum vertical reaction force and lower limb on drop landing between normal and flat foot group, this study is to provide fundamental data of the prevention of injury and the treatment of exercise which are frequently occurred on flat foot group's drop landing. The surface electrodes were sticked on lateral gastrocnemius muscle, medial gastrocnemius muscle, tibialis anterior and the drop landing on a force plate of 40cm was performed with a normal group who had no musculoskeletal disease and a flat foot group of 9 people who had feet examinations. Vertical reaction force were significantly statistically different between two groups(p<.001). Muscle activity of lower limbs in all three parts were not statistically different but showed high tendency on average in the flat foot group. The flat foot group had difficulties in diversification of impact burden and high muscle activity. Therefore, it was suggested that muscular strengthening of knee joints and plantar flexions of foot joints which were highly affected in impact absorption will be required.

The Effect of Activities-Centered Reading based on Korean Fairy Tales on Children's Reading Attitude and Comprehension (한국전래동화를 통한 활동중심 독서지도가 아동의 읽기 태도 및 읽기 이해에 미치는 영향)

  • This research analyzed the effect of activity-centered reading band on Korean fairy tales on children's reading attitude and comprehension. The reading altitude of the experimental-group children exhibited more meaningful change in such subordinate areas as academic reading attitude. The academic reading attitude of the male experimental-group had higher improvement than that of the female experimental-group. In addition, the reading comprehension scores of the experimental-group children were higher than those of the control-group children. The reading program effects on reading comprehension were more meaningful in girls in the experimental group than boys in the group. Therefore, the results of this study have demonstrated the necessity of the development and application of a Korean fairy tales program for reading and an activity-centered reading program.

Effects of MMP-2 activation and FSH or LH Hormone Supplementation on Embryo Development in In Vitro Fertilization of Porcine

  • The purpose of this study was to analyze whether FSH and LH hormone treatment directly or indirectly affect embryo development in embryonic development. To determine this, we compared the development of embryonic cells through the expression pattern of MMPs. As a result, 33.8% of blastocysts were formed in FSH added group, 20.8% in LH added group and 10% in FSH + LH added group. In addition, the activity of MMP-9 was highly detected in the FSH-added group, and the expression of Casp-3 was much lower than that of the other groups. These results suggest that the addition of FSH seems to increase the activity of MMP-9 in embryonic cells, and that LH, on the contrary, may activate MMP-2 activity. In addition, the expression level of MMP-2 in the FSH-added group was high in the Trophoblast cell group and in the LH-added group, the hormone ideal secretion might affect the development of the embryonic cell.

Effects of Moxibustion to Zusanli(ST36) on Alteration of Natural Killer cell Activity in rat spleen (족삼리혈 뜸자극이 흰쥐 비장 자연살해세포 활성에 미치는 영향)

  • Moxibustion is one of major healing technique in oriental medicine. It has been widely used in many diseases such as rheumatoid arthritis, Hashimoto disease, breech presentation, etc. However, till now, effects of moxibustion on NK cell activity and relations between sympathetic nerve system(SNS) and the immune alteration induced by moxibustion were not well studied. This study was designed to evaluate effects of moxibustion on NK cell activity and the intervention of SNS in the alteration of NK cell activity induced by moxibustion. Splenic NK cytotoxity was measured in a standard 4-h 51Cr release assay. We measured the NK cytotoxity at after moxibustion stimulation for 1,3,5, and 7 days, and also measured the NK cell cytotoxity after 3 and 7 days burn stimulation with similar temperature. IL-2, IL-4, INF-γ in serum were measured by rat IL-2, IL-4, INF-γ ELISA TEST KIT. To evaluate the effects of sympathectomy on alteration of NK cell cytotoxity, 6-hydroxydopamine(6-OHDA : 5Omg/kg) was used. We showed that NK cell activity of moxibustion stimulation group increased at the 3rd day, and declined at 7th day in comparison with that of contol group. In moxibustion stimulation group, NK cell activity of 3 day stimulation group was significantly higher than sham group. On the contrary, in burn stimulation group, NK cell activity was significantly higher than that of sham groups at 3rd, 7th days. Patterns between moxibustion and burns were different. INF- γ level of 3 days moxibustion stimulation group significantly higher than sham group. IL 2 level among groups were not different. IL-4 was not detected in serum with this method. Sympathectomy abolished the NK cell activity alteration induced by moxibustion. The results suggest that moxibustion induces the alteration of NK cell activity, along with INF-γ and SNS is related to these effects.

Comparative Study on the Quality of Life Between Senior Citizens and Disabled People (노인과 장애인의 삶의 질 비교연구)

  • The purpose of this study is to reveal the similarity and difference by comparing the quality of life between senior citizen group and disabled people group. For this purpose, I surveyed the questionnaire composed of sociological variable of population, social support, society activity participation, activity of daily living, self-efficacy, and quality of life to 113 senior citizens and 107 disabled people and analyzed the actual proof relationship of cause and effect in between variables. I executed the analysis of structural equation model by using SPSS 14.0 and AMOS 7.0 for the analysis of data. As the result of the study, firstly, the variables highly co-related with the quality of life to both senior citizen group and disabled people group were self-efficacy and society activity participation, and the variables highly co-related with self-efficacy were the quality of life and society activity participation. Secondly, the variables which show the meaningful difference between senior citizen group and disabled people group were society activity participation and activity of daily living. Thirdly, in the senior citizen group, social support, society activity participation, and activity of daily living have meaningful effect on self-efficacy, and all variables have meaningful effect on the quality of life. However, in the disabled people group, social support did not have meaningful effect on self-efficacy, and the social support and self-efficacy did not have meaningful effect on the quality of life. Based upon these study results I discussed the meaning and implications of the study, and suggested the successive study tasks.
