Screening of Anti-Adhesion Agents for Pathogenic Escherichia coli O157:H7 by Targeting the GrlA Activator

  • Enterohemorrhagic Escherichia coli (EHEC) is a foodborne pathogen that produces attaching and effacing lesions on the large intestine and causes hemorrhagic colitis. It is primarily transmitted through the consumption of contaminated meat or fresh produce. Similar to other bacterial pathogens, antibiotic resistance is of concern for EHEC. Furthermore, since the production of Shiga toxin by this pathogen is enhanced after antibiotic treatment, alternative agents that control EHEC are necessary. This study aimed to discover alternative treatments that target virulence factors and reduce EHEC toxicity. The locus of enterocyte effacement (LEE) is essential for EHEC attachment to host cells and virulence, and most of the LEE genes are positively regulated by the transcriptional regulator, Ler. GrlA protein, a transcriptional activator of ler, is thus a potential target for virulence inhibitors of EHEC. To identify the GrlA inhibitors, an in vivo high-throughput screening (HTS) system consisting of a GrlA-expressing plasmid and a reporter plasmid was constructed. Since the reporter luminescence gene was fused to the ler promoter, the bioluminescence would decrease if inhibitors affected the GrlA. By screening 8,201 compounds from the Korea Chemical Bank, we identified a novel GrlA inhibitor named Grlactin [3-[(2,4-dichlorophenoxy)methyl]-4-(3-methylbut-2-en-1-yl)-4,5-dihydro-1,2,4-oxadiazol-5-one], which suppresses the expression of LEE genes. Grlactin significantly diminished the adhesion of EHEC strain EDL933 to human epithelial cells without inhibiting bacterial growth. These findings suggest that the developed screening system was effective at identifying GrlA inhibitors, and Grlactin has potential for use as a novel anti-adhesion agent for EHEC while reducing the incidence of resistance.

Resistance to fluoroquinolone of Staphylococcus pseudintermedius isolated from dogs and cats in Daegu (대구지역 개와 고양이에서 분리된 Staphylococcus pseudintermedius의 fluoroquinolone 내성)

  • The purpose of this study was to investigated the fluoroquinolone (FQ) resistance and presence of gyrA and grlA gene in 87 Staphylococcus pseudintermedius isolates obtained from clinical samples of dogs and cats. Also, the profiles of FQ resistance compared with methicillin resistant S. pseudintermedius (MRSP) isolates. FQ resistance was observed for enrofloxacin (41.4%), ciprofloxacin (39.1%), norfloxacin (36.8%), ofloxacin and levofloxacin (32.2%, respectively), and moxifloxacin (31.0%). Thirty-eight (43.7%) of 87 S. pseudintermedius isolates were resistant to more than one FQ. Twenty-six (64.5%) of 38 FQ resistant isolates were resistant to all the six FQ tested. Of 38 FQ resistant isolates, gyrA gene was detected in all isolates but grlA gene was not found. Moreover, 19 MRSP isolates were resistant to enrofloxacin (63.2%), ciprofloxacin (57.9%), norfloxacin (52.6%), and ofloxacin, levofloxacin and moxifloxacin (47.4%, respectively). FQ resistance were highly prevalence in S. pseudintermedius isolates from dogs and cats. Our results emphasize the prudent use of antimicrobial agents to companion animals is necessary for prevent antimicrobial resistance.

Cell-SELEX Based Identification of an RNA Aptamer for Escherichia coli and Its Use in Various Detection Formats

  • Escherichia coli are important indicator organisms, used routinely for the monitoring of water and food safety. For quick, sensitive and real-time detection of E. coli we developed a 2'F modified RNA aptamer Ec3, by Cell-SELEX. The 31 nucleotide truncated Ec3 demonstrated improved binding and low nano-molar affinity to E. coli. The aptamer developed by us out-performs the commercial antibody and aptamer used for E. coli detection. Ec3(31) aptamer based E. coli detection was done using three different detection formats and the assay sensitivities were determined. Conventional Ec3(31)-biotin-streptavidin magnetic separation could detect E. coli with a limit of detection of $1.3{\times}10^6CFU/ml$. Although, optical analytic technique, biolayer interferometry, did not improve the sensitivity of detection for whole cells, a very significant improvement in the detection was seen with the E. coli cell lysate ($5{\times}10^4CFU/ml$). Finally we developed Electrochemical Impedance Spectroscopy (EIS) gap capacitance biosensor that has detection limits of $2{\times}10^4CFU/mL$ of E. coli cells, without any labeling and signal amplification techniques. We believe that our developed method can step towards more complex and real sample application.

Transformation of Orchardgrass (Dactylis glomerata L.) with Glutathione Reductase Gene (Glutathione Reductase 유전자의 도입에 의한 오차드그래스의 형질전환)

  • To develop transgenic orchardgrass resistant to reactive oxygen species produced from environmental stresses, a vector with the cytosolic glutathione reductase cDNA (BcGRl) from Chinese cabbage was constructed under the control of the cauliflower mosaic virus 35S promoter and was introduced into orchardgrass using Agrobacterium tumefaciens EHA101. Transgenic plants from hygromycin-selected calli of orchardgrass did not show any morphological difference from wild-type plants. The results of PCR amplification and genomic Southern blot analysis confirmed the integration of foreign gene into the chromosome of transgenic orchardgrass. Northern blot analysis with total RNA from leaves also confirmed the constitutive expression of BcGR1 in transgenic orchardgrass.

Comparative Superiority of in vitro Activity of DW-224a Supported by the Downward MIC Distribution in Ciprofloxacin-resistant Staphylococcus aureus (시프로플록사신 내성 황색 포도상 구균에서 MIC의 하향 분포로 입증된 DW-224a의 in vitro 항균 활성의 비교 우수성)

  • The comparative superior in vitro activity of DW-224a was supported by the downward MIC distribution due to the weakened influence of alterations within target enzymes in ciprofloxacin-resistant Staphylococcus aureus. The MI$C_{50}$ for DW-224a was 4 $\mu$g/mL, similar to that of gemifloxacin, 8-fold less than that of sparfloxacin and 16-over-fold less than that of ciprofloxacin. We constructed combinations of amino acid changes, located at codon 80, 83 or 84 within GrlA and 84, 85 or 88 within GyrA, which were associated with MIC increase. The amino acid changes were less influential to the MIC of DW-224a compared to those of other fluoroquinolones, and it was verified from the requirement of a total of two GrlA- and two GyrA-alterations to reach the MIC of DW-224a over 32 $\mu$g/mL.

  • In this paper, a free-space material measurement technique is discussed in W-band(75~110 GHz). For the accurate measurement of S-parameters of an MUT(Material Under Test) in free space, a W-band quasi-optical free-space material measurement system, less affected by the measurement environments, is discussed, and GRL(Gated Reflect Line) method for calibrating the measurement system is described. Proposed technique is verified for 'Air' and measurement results for arystal plates of thickness 1.1 mm, 2 mm, 2.75 mm and 5 mm are also shown.

  • This paper presents the methods to solve the problem about Power System Protection Program(PROSET). PROSET is a overall program system to protect power system and correct it. In the context of the PROSET, it is so difficult to edit database data when topology is changed. When the renewal of data is performed, all changed values are disappeared. Therefore, it will not be expected to know the present situation by using the information of the databases data. So it is necessary to solve this problem. Solution is copy of the user. Database of GRL,GRZ,KYP2D1,KYD2X1,L90 Relay module were developed. We made a database management system for PROSET, therefore I will show this management system briefly.

Simplified Failure Mechanism for the Prediction of Tunnel Crown and Excavation Front Displacements

  • This case study presented a simplified failure mechanism approach used as a preliminary deformation prediction for the Mexico City's metro system expansion. Because of the Mexico City's difficult subsoils, Line 12 project was considered one of the most challenging projects in Mexico. Mexico City's subsurface conditions can be described as a multilayered stratigraphy changing from soft high plastic clays to dense to very dense cemented sands. The Line 12 trajectory crossed all three main geotechnical Zones in Mexico City. Starting from to west of the City, Line 12 was projected to pass through very dense cemented sands corresponding to the Foothills zone changing to the Transition zone and finalizing in the Lake zone. Due to the change in the subsurface conditions, different constructions methods were implemented including the use of TBM (Tunnel Boring Machine), the NATM (New Austrian Tunneling Method), and cut-and-cover using braced Diaphragm walls for the underground section of the project. Preliminary crown and excavation front deformations were determined using a simplified failure mechanism prior to performing finite element modeling and analysis. Results showed corresponding deformations for the crown and the excavation front to be 3.5cm (1.4in) and 6cm (2.4in), respectively. Considering the complexity of Mexico City's difficult subsoil formation, construction method selection becomes a challenge to overcome. The use of a preliminary results in order to have a notion of possible deformations prior to advanced modeling and analysis could be beneficial and helpful to select possible construction procedures.

A comparative study on applicability of nano-sized iron(II, III) oxide in ultrasonicated Fenton process

  • Fenton process is one of the most effective advanced oxidation processes for the removal of pollutants from wastewater. In this study, while ferrous iron was used in conventional Fenton process (CFP); nano-sized iron(II, III) oxide was experienced in modified Fenton process (MFP) as a new catalyst alternative. In order to enhance their oxidation efficiencies, both CFP and MFP were combined with ultrasonication at 53 kHz fixed frequency. Thus, the influences of both catalyst iron species and ultrasonication on color and chemical oxygen demand (COD) removals from synthetic textile wastewater including Maxilon Red GRL 200% dyestuff were investigated experimentally. While the COD and color removal rates were found as 72.5% and 69.7% via CFP; they were 87% and 75.8% by ultrasonicated CFP, respectively. The color and COD removals were 40.6% and 64.8% via MFP, and 49.9 and 73.1% by ultrasonicated MFP, respectively. Therefore, it was found that the simultaneously usage of ultrasonication with CFP and MFP was improved the COD and color removal efficiencies and oxidation rates even at lower H2O2 dosages, compared to individual CFP and MFP. Moreover, the color and COD removal kinetics were also modelled mathematically and compared in the study.

A Case Study of large diameter steel pipe pile Foundation for Offshore LNG Facility (해상 LNG 인수시설 대구경 강관말뚝 시공 사례 연구)

  • In this paper, a case study of drivability and bearing capacity of large diameter steel pipe piles at PTT LNG site in Thailand is introduced. The LNG facilities were designed to be founded on steel pipe pile foundations driven into the weathered rock formation overlaid by sand layers. The drivability analyses of open ended pipe piles were carried out using GRL WEAP program and the bearing capacities of the piles were estimated. Dynamic load tests were performed to evaluate end bearing resistance, and it is shown that the measured end bearing resistance is smaller than the calculated end bearing because the plugging does not develop sufficiently in case of large diameter pipe piles.
