The present study was conducted for the production of transgenic cloned cows by somatic cell nuclear transfer (SCNT) that secrete human prourokinase into milk. To establish an efficient production system for bovine transgenic SCNT embryos, the offset was examined of various conditions of donor cells including cell type, size, and passage number on the developmental competence of transgenic SCNT embryos. An expression plasmid far human prourokinase (pbeta-ProU) was constructed by inserting a bovine beta-casein promoter, a green fluorescent protein (GFP) marker gene, and a human prourokinase target gene into a pcDNA3 plasmid. Three types of bovine somatic cells including two adult cells (cumulus cells and ear fibroblasts) and fetal fibroblasts were prepared and transfected using a lipid-meidated method. In Experiment 1, developmental competence and rates of GFP expression in bovine transgenic SCNT embryos reconstructed with cumulus cells were significantly higher than those from fetal and ear fibroblasts. In Experiment 2, the effect of cellular senescence in early (2 to 4) and late (8 to 12) passages was investigated. No significant differences in the development of transgenic SCNT embryos were observed. In Experient 3, different sizes of GFP-expressing transfected cumulus cells [large (>30 ${\mu}{\textrm}{m}$) or small cell (<30 ${\mu}{\textrm}{m}$)] were used for SCNT. A significant improvement in embryo development and GFP expression was observed when small cumulus cells were used for SCNT. Taken together, these results demonstrate that (1) adult somatic cells could serve as donor cells in transgenic SCNT embryo production and cumulus cells with small size at early passage were the optimal cell type, and (2) transgenic SCNT embryos derived from adult somatic cells have embryonic development potential.
This paper provides basic data for product developers by investigating the consumption status, consumer perception, and consumer demand on products using Aster Yomena. Two hundred ninety-five people over 20 years of age were analyzed according to gender and age. In the purchase of Aster Yomena products, men were the higher purchasers except for powder. In age, noodles, wild vegetables, jangajji, tteok, and powder were consumed by those older than 30 years. In an analysis of purchase when developing Aster Yomena products, the male purchase intention was higher in all items except for kalguksu in the staple, and male purchase intention was higher in only bibimbapseasoning among aster powder addition sauce and all items in dessert. The purchase intention of those older than 30 years was higher in all items of staples, aster powder addition sause and sause The purchase intention of those older than 30 yearse was higher in green vegetables juice and milk tea among drink, and all items except ice cream among dessert. An analysis of the popularization of Aster Yomena revealed higher perceptions of men in the medium (TV, internet, online cafe, agro-fishery market), experience program (area tourism, exposition), and product development (convenience food, recipe development). The medium (agro-fishery market), experience program, and product development were more recognized in their 30s or more. The products with aster yomana require the most effective marketing to men 30 years and older, and require a strategy that will interest those in their 20s.

Generally, in terms of the development of irrigation scheme, the efficient water resource management that supplies the irrigation water in consideration of the required time and accurate quantity to grow the crop should be conducted. The water resource assessment should precede to supply the irrigation water efficiently. The water resources assessment is divided into the water requirement analysis and the water availability assessment. In case of Korea, the major crop is paddy rice unlike crops of Africa, such as sugarcane, maize, and cassava, etc. Because it is not familiar with the method for upland irrigation development in tropical area, it needs to know the water resources assessment for irrigation scheme development about these crops. The Natama Scheme in Chiradzulu District of the Southern Malawi was selected as study area, which has tropical climate. From the collected meteorological data, the evapotranspiration was analyzed by Penman-Monteith Method and the effective rainfall was analyzed by USDA Soil Conservation Service Method. This study displays the results that for study area, the evapotranspiration varies from 2.80 mm/day to 5.51 mm/day and the effective rainfall varied from 2.1mm to 149.0mm. According to the selected crop (Green Maize, Dry Maize), the unit water requirement (UWR) and water demand (WD) considering the irrigation efficiency, irrigation time and irrigation area were estimated to be $0.00122m^3/s/ha$ and $0.0122m^3/s$ respectively. For the water availability assessment, the runoff of Natama scheme was calculated by specific yield method. The water availability was evaluated through reviewed differences of discharge between $Q80_{intake}$ and Total WD, and the irrigation water can be supplied sufficiently in the existing 10ha of Natama scheme. As a result of reviewing the extensibility of irrigable area, total WD of scheme is $0.02313m^3/s$, and $Q80_{intake}$ is $0.02387m^3/s$ ($Q80_{intake}$ > Total WD). Therefore, Natama scheme can be extended from 10 ha to 17 ha in the dry season in consideration of the $Q80_{intake}$.
The magnetic polishing is the useful method to finish using magnetic power of magnet. That method is one of precision polishing techniques and has an aim of the clean technology using for the pure of gas and inside of the clean pipe. The magnetic abrasive polishing method is not so common for machine that it is not spreaded widely. There are rarely researcher in this field because of non-effectiveness of magnetic abrasive. Therefore, in this paper deals with development of the magnetic abrasive using Sr-Ferrite. In this development, abrasive grain GC used to resin bond fabricated low temperature. And Sr-Ferrite of magnetic abrasive powder fabricated that Sr-Ferrite was crused into 200 mesh. The XRD analysis result show that only GC abrasive and Sr-Ferrite crystal peaks detected which explains resin bond was not any more chemical reaction. From SEM analysis it is found that GC abrasive and Sr-Ferrite were strong bonding with each other by bond. The magnetic polishing is performed by polishing the surface of pipe by attracting magnetic abrasives with magnetic fields. This can be widely applied for finishing machinery fabrications such as various pipes and for other safety processes. In this paper, we could have investigated in to the changes of the movement of magnetic abrasive grain. In reference to this result, we could have made the experiment which is set under the condition of the magnetic flux density, polishing velocity according to the form of magnetic brush.

In 2004, the Incheon Urban Development Corporation held a design competition for the Song-Do apartment complex development. Several projects that won the prize in the recent design competitions were reviewed to look around the current design tendency, and current conditions of the site were surveyed. The authors tried to combine digital and environment-friendly factors, considering the site condition. Because the site is on reclaimed land and is located at the center of the information-technology industrial area, these two factors inform the main concept of this project. The site was divided into two different zones : the Ocean Zone and the Digital Zone. The 'community spaces'(community street, community plaza and so on) were designed for the various activities of the residents. The Patios, 'private' community spaces, included many facilities for play, appreciation, rest, exercise, refreshment and gathering. To avoid common landscape styles in the patios, simple and minimal forms were designed. The environmental sculptures were carefully planned, allocated and shaped with digital technology, based on the 'Present and Future of Song-Do'. The authors also tried to introduce environment-friendly factors, such as the green roof system, wall planting, and the water recycling system. This project is following the M.A. (Master Architect) process, and is still being modified to make the hi-tech and environment-friendly apartment complex.
While the global food system prevails in America, the community food system is also spreading rapidly. The former, which is based on industrial agriculture, has negative impacts on family farms, food safety, and community development. In contrast, the latter has positive effects on farmers, consumers, communities, and the environment. Community food systems have been able to grow successfully through the participation of farmers and consumers, support of the federal and state governments, and the initiatives of NGOs. Food Circle, upon which this study focuses, is one of the community food systems based on the regional level. It is the outcome of the green movement in Kansas City, Missouri. The goal of Food Circle in Missouri is to retain and support rural family farms by connecting them with regional consumers. Its main activity is to collect data, including a directory of farmers and their regional products, and distribute this data to regional consumers. It is informally organized and entirely self funded. Although community food systems have several problems, their development is to continue in America. The food system in Korea has become a global one in recent decades, and agricultural problems and food safety issues are being generated as a result. The development of community food systems would be helpful in resolving these problems. In order to activate community food systems in Korea as seen in America's case, it would be necessary to have the imperative participation of both farmers and consumers, the support of the central and provincial governments, and the initiatives of NGOs.

The objective of this study is to analyze the distribution of urban parks and its problems in a city in Gyeonggi-do where new towns and old towns are mixed. The study will also analyze and understand the unbalanced deployment of urban parks - rest areas and carbon absorbers of an entire city - between old towns and new towns, suggest improvement opportunities and examine and suggest plans to create and expand urban parks in old cities that lack urban parks. Findings showed that first, new towns were 1.2~1.6 higher than old towns in the number of parks, park area, and per capita park area, indicating that parks were unbalanced across towns. Second, as for a plan for improving the unbalanced deployment of urban parks, when the use area needs to be changed in an urban development project, it was suggested to donate 10% of the project site to the city to create it into a park and it was suggested to identify and proactively improve alternative park resources that can replace parks such as rivers and recreational sites. Third, regarding a plan for improving urban parks in old towns, it was suggested to secure urban parks by attracting various urban development projects to old town areas. The amendment of related laws was proposed to double required park area to $6m^2$ per household. Amendments were also suggested for one law and four guidelines to specifically define the location of urban parks as well.

This study has focused 302 environmental ordinances enacted from 1974 to 2008 by 16 upper-local governments (7 deignated cities and 9 provinces) and the relationships between them and the development of the Korean local autonomy system. The ordinances have been categorized into three types, the voluntary ones without any obligatory stipulations in laws, the optional ones based on laws, and the law based obligatory ones, which have been examined the enactment years and the contents of them. The local ordinances have been enacted in four periods with a few characteristics, which are "the pre-local-autonomy period (1974-1991)", "the introductory-local-autonomy period (1991-1995)", "the local-autonomy-developing period (1995-1999)", and "the mature-local-autonomy period (after 2000)", along the local autonomy development. 57 ordinances were enacted in the first period, 20 enacted in the second period, 46 enacted in the third period and 179 ordinances in the fourth period. The obligatory ordinances were the most in the first period and in the second period, while the voluntary ordinances were enacted most in the third period. 7 designated cities have pretty more ordinances, 160 in all, than 9 provinces, 142. The tendency to enact earlier in the metropolitan area, SeoulCity, IncheonCity and Gyeonggi province, than the others can be seen. It can be said that the establishment of the local autonomy in Korea had accelerated the development of the local environmental management.

Hundreds of contaminated sites have been generated due to the past mismagement of toxic substances, the lack of adequate environmental controls and ignorance of the potential environmental impacts of general activities in Canada. The general public, industry and governments have been addressing the contaminated sites with a number of cleanup responses. Environmental protection and remediation have become top priorities for the public and private sectors alike in Canada. Between the late 1980s and the early 1990s, the Canadian Environmental Protection Act and Canada's Green Plan were followed by provincial and territorial laws and policies to regulate contaminated sites. The National Contaminated Site Remediation Program(NCSRP) was initiated in 1989. It has been administered through bilateral agreements between the federal and participating provincial/territorial governments. They have committed a total of $250 million toward orphan site cleanup and technology development/demonstration over a five year period. The federal government has committed an additional $25 million to assess contaminated sites on federal crown land. Over 40 orphan high-risk contaminated sites, over 230 federal sites and over 35 technology development/demonstration projects have been addressed. The Canadian Council of Ministers of the Environment has developed a series of guidance documents to ensure a consistent and successful implementation of the Program. The management/regulation scheme of contaminated sites generally consists of: 1) identifying and investigating sites, 2) determining site contamination, 3) recognizing responsibility and liability, 4) assessing priority for remediation, 5) activation, evaluation and implementing remediation options, and 6) documenting remediation completion. The NCSRP supported the successful development/demonstration of a wide range of innovative remedial technologies. They are related to stabilization/solidification, thermal washing/flushing, advanced oxidation, sonics, and groundwater contaminated with hydrocarbons, PAHS, PCBs, heavy metals, and other hazardous pollutants in a variety of site environments.

Green shoot cuttings of 10 different grapevine species were inoculated with Agrobacterium vitis to find disease-related metabolites in the grapevine. Crown galls formed 60 days after inoculation varied in gall severity (GS) evaluated by gall incidence (GI) and gall diameter (GD), which were classified into three response types as RR (low GI and small GD), SR (high GI and small GD), and SS (high GI and large GD), corresponding to resistant, moderately resistant, and susceptible responses, respectively. In this, 4, 4, and 2 Vitis species were classified into RR, SR, and SS, respectively. Gas chromatography mass spectrometry (GC-MS) analysis of the grapevine stem metabolites with A. vitis infection showed 134 metabolites in various compound classes critically occurred, which were differentially clustered with the response types by the principal component analysis. Multivariate analysis of the metabolite profile revealed that 11 metabolites increased significantly in relation to the response types, mostly at post-inoculation stages, more prevalently (8 metabolites) at two days after inoculation than other stages, and more related to SS (7 metabolites) than RR (3 metabolites) or SR (one metabolite). This suggests most of the disease-related metabolites may be rarely pre-existing but mostly induced by pathogen infection largely for facilitating gall development except stilbene compound resveratrol, a phytoalexin that may be involved in the resistance response. All of these aspects may be used for the selection of resistant grapevine cultivars and their rootstocks for the control of the crown gall disease of the grapevine.
