Copolymers composed of hyaluronic acid (HA) and poly(N-isopropylacrylamide) (PNIPAAm) were prepared to create temperature-sensitive injectable gels for use in controlled drug delivery applications. Semi-telechelic PNIPAAm, with amino groups at the end of each main chain, was synthesized by radical polymerization using 2-aminoethanethiol hydrochloride (AESH) as the chain transfer agent, and was then grafted onto the carboxyl groups of HA using carbodiimide chemistry. The result of the thermo-optical analysis revealed that the phase transition of the PNIPAAm-grafted HA solution occurred at around 30$\∼$33$^{circ}C$. As the graft yield of PNIPAAm onto the HA backbone increased, the HA-g-PNIPAAm copolymer solution exhibited sharper phase transition. The short chain PNIPAAm-grafted HA ($M_{w}$=6,100) showed a narrower temperature range for optical turbidity changes than the long chain PNIPAAm-grafted HA ($M_{w}$=13,100). PNIPAAm-grafted HA exhibited an increase in viscosity above 35$^{circ}C$, thus allowing the gels to maintain their shape for 24 h after in vivo administration. From the in vitro riboflavin release study, the HA-g-PNIPAAm gel showed a more sustained release behavior when the grafting yield of PNIPAAm onto the HA backbone was increased. In addition, BSA released from the PNIPAAm-g-HA gels showed a maximum concentration in the blood 12 h after being injected into the dorsal surface of a rabbit, followed by a sustained release profile after 60 h.
In this study, a series of highly swelling hydrogels based on sodium alginate (NaAlg) and polymethacrylamide (PMAM) was prepared through free radical polymerization. The graft copolymerization reaction was performed in a homogeneous medium and in the presence of ammonium persulfate (APS) as an initiator and N,N'-methylenebisacrylamide (MBA) as a crosslinker. The crosslinked graft copolymer, alginate-graft-polymethacrylamide (Alg-gPMAM), was then partially hydrolyzed by NaOH solution to yield a hydrogel, hydrolyzed alginate-graft-polymethacrylamide (H-Alg-g-PMAM). During alkaline hydrolysis, the carboxamide groups of Alg-g-PMAM were converted into hydrophilic carboxylate anions. Either the Alg-g-PMAM or the H-Alg-g-PMAM was characterized by FTIR spectroscopy. The effects of the grafting variables (i.e., concentration of MBA, MAM, and APS) and the alkaline hydrolysis conditions (i.e., NaOH concentration, hydrolysis time, and temperature) were optimized systematically to achieve a hydrogel having the maximum swelling capacity. Measurements of the absorbency in various aqueous salt solutions indicated that the swelling capacity decreased upon increasing the ionic strength of the swelling medium. This behavior could be attributed to a charge screening effect for monovalent cations, as well as ionic cross-linking for multivalent cations. Because of the high swelling capacity in salt solutions, however, the hydrogels might be considered as anti-salt superabsorbents. The swelling behavior of the superabsorbing hydrogels was also measured in solutions having values of pH ranging from 1 to 13. Furthermore, the pH reversibility and on/off switching behavior, measured at pH 2.0 and 8.0, suggested that the synthesized hydrogels were excellent candidates for the controlled delivery of bioactive agents. Finally, we performed preliminary investigations of the swelling kinetics of the synthesized hydrogels at various particle sizes.
열응답성 고분자인 폴리(N-이소프로필아크릴아미드) (PNIPAAm) 말단에 아민기를 갖는 PNIPAAm-$NH_2$ 및 이들 알긴산 나트륨에 그래프팅시킨 공중합체를 합성하고 이들의 여러 가지 특성들을 분석하였다. PNIPAAm-$NH_2$를 합성하기 위하여 N-이소프로필아크릴아미드를 라디칼 중합할 때 2-aminoethanethiol hydrochloride (AESH)를 연쇄이동제로 사용하였다. AESH 농도를 높이면 PNIPAAm-NH2의 분자량이 작아지며 PNIPAAm-$NH_2$ 수용액의 하한임계용액온도(LCST)가 낮아졌다. N-(3-dimethylaminopropyl)-N'-ethylcarbodiimide 및 N-hydroxysuccinimide를 사용하여 PNIPAAm-$NH_2$를 graft-onto 방법으로 알긴산 나트륨에 그래프팅시킨 alginate-g-PNIPAAm 공중합체 역시 온도에 따라 팽윤-수축 거동을 하였으며, PNIPAAm-$NH_2$보다 약간 높은 온도에서 LCST를 나타내었고, 그래프트된 PNIPAAm의 양이 많아질수록 팽윤비가 커졌다.
필름 형태의 Polydimethylsiloxane (PDMS)은 표면 개질을 하기 위해 Plasma 처리 또는 Corona 처리를 하여 표면을 -OH기로 활성화시키는 공정이나, 피착제와 PDMS 필름의 접착 또는 Adhesion promoter와 축합 반응을 통해 PDMS 표면을 다른 작용기로 개질시키는 공정, Grafting polymerization을 이용하는 PDMS 개질 공정이 주로 이용된다. 그러나 Plasma나 Corona 처리 후에 친수성이 오래가지 못하고, 보관에 어려움이 있다. 따라서, 본 연구에서는 코팅 공정을 통하여서 PDMS표면 개질을 하기 위해서, 먼저 새로운 형태의 실란 기능화 아크릴 고분자 전구체를 합성하고 이를 Hydroxyl-terminated PDMS와의 축합 반응을 통해 아크릴 고분자와 PDMS 고분자가 결합된 형태의 표면 개질제를 제조한 후, 이를 PDMS 필름 위에 코팅하였다. 제조한 표면 개질제의 구조와 분자량을 확인하기 위하여 1H-NMR과 GPC를 분석하였고, 표면 개질제가 코팅된 PDMS표면 특성 변화를 확인하기 위하여 XPS, ATR, WCA를 이용하여 표면 특성을 조사하였으며, PDMS 필름과의 부착을 확인하기 위해 Cross cutting test를 진행하였다. 그 결과 PDMS 필름 표면이 아크릴 고분자층이 형성된 것으로 확인하였고, PDMS 필름과 표면 개질제와의 부착성 (4 - 5B) 또한 우수한 것을 확인하였다.
직접 메탄올 연료전지(direct methanol fuel cell, DMFC)는 연료의 개질 없이 메탄올 연료를 공급하여 수소이온과 전자 생성을 통해 전류를 생산하는 에너지 변환 장치이다. 현재 DMFC에 적용되고 있는 고분자 전해질 막(polymer electrolyte membrane, PEM)은 높은 수소이온 전도도와 물리화학적 안정성을 갖는 과불소화계 이오노머를 활용한 PEM이지만, 높은 메탄올 투과율과 분해 시 발생되는 환경 오염 물질 등의 문제로 인해 신규 소재 개발이 요구되고 있다. 최근 들어, 과불소화계 이오노머에 비해 낮은 연료 투과율 및 우수한 물리화학적 안정성을 갖는 탄화수소계 고분자 기반 PEM을 DMFC에 적용하는 연구들이 보고되고 있다. 본 총설에서는 탄화수소계 고분자 기반 PEM 중 1) 친수성/소수성 영역의 뚜렷한 나노 상분리 구조를 나타내는 가지형 공중합체를 합성하여 수소이온 전도성과 메탄올의 선택도를 향상시킨 연구, 2) 제막 단계에서 가교 구조를 도입하여 메탄올 투과율을 감소시키고 치수 안정성을 향상시킨 연구, 3) 유/무기계 첨가제 및 다공성 지지체를 도입하여 성능을 개선한 복합 막 개발 연구에 대해 소개하고자 한다.
본 연구에서는 실리카원으로 Tetraethyl orthosilicate (TEOS)를 사용하고 주형으로 트리블럭 공중합체(P123)를 사용하여 산성 조건에서 자기조립 방법과 수열합성 과정을 거쳐서 잘 배열된 육방체 구조의 메조세공 배열구조를 가지는 다공성 실리카 물질(Surfactant-extracted SBA-15)을 합성하였다. Surfactant-extracted SBA-15는 약 980 nm의 크기를 가지는 짧은 로드의 입자 모양을 보여주었다. 그리고 표면적과 세공 직경은 각각 730 m2g-1와 70.8 Å이었다. 한편, 포스트-합성방법(post-synthesis method)을 이용하여 메조세공 내에 아미노실란(3-aminopropyltriethoxysilane, APTES)을 그래프팅(grafting) 하였다. 아미노실란으로 개질된 메조다공성 실리카(APTES-SBA-15)는 잘 배열된 세공구조(p6mm)를 가지고 짧은 로드의 입자모양을 잘 유지 하였다. APTES-SBA-15의 표면적과 세공 직경은 각각 350 m2g-1와 60.7 Å으로 감소하였다. APTES가 개질된 메조 다공성 실리카에 희토류 금속이온(Eu3+, Tb3+) 용액을 처리하여 메조세공 내에 희토류 금속 착물이 도입된 메조다공성 실리카 물질을 합성하였다. (Eu/APTES-SBA-15, Tb/APTES- SBA-15) 이들 물질은 λex=250 nm 광에 의해 특징적인 광발광 스펙트라를 나타내었다. (Tb/APTES-SBA-15를 위하여 5D4→7F5 (543.5 nm), 5D4→7F4 (583.5 nm), 5D4→7F3 (620.2 nm) 전이; Eu/APTES-SBA-15를 위하여 5D0→7F0 (577.7 nm), 5D0→7F1 (592.0 nm), 5D0→7F2 (614.9 nm), 5D0→7F3 (650.3 nm) and 5D0→7F4 (698.5 nm) 전이)
