In the present study, we investigated the physicochemical characteristics and antioxidant activity of kimchi during the fermentation process. Kimchi was fermented at 18.5 ℃, then after one day, the storage temperature was changed to 5 ℃ without fresh kimchi (Fresh; pH 5.6, total acidity 0.3%), which obtained optimum-ripened kimchi (OptR; pH 4.3, total acidity 0.64%), and over-ripened kimchi (OvR; pH 3.8, total acidity 1.24%). As a result, the glucosinolates content of the kimchi was increased during the fermentation process. Among the glucosinolates, glucoraphanin possesses the highest amounts in kimchi. In addition, the contents of sulforaphane and total polyphenol, which are common antioxidant compounds, were increased during the fermentation process. To evaluate the antioxidant activities of Fresh, OptR, and OvR, we measured 1,1-diphenyl-2-picrylhydrazyl (DPPH) and hydroxyl (·OH) radicals radical scavenging activity in vitro. Fresh, OptR, and OvR exerted DPPH and ·OH radical scavenging activities dose-dependently. In particular, the ·OH radical scavenging activities of OptR and OvR were higher than that of Fresh. Therefore, we suggest that kimchi at the ripe and over-ripe stage is considered to have high antioxidant activity by increasing glucosinolate, sulforaphane, and total polyphenols, compared with fresh kimchi.
Isothiocyanates are naturally occurring small molecules that are formed from glucosinolate precursors of cruciferous vegetables. Many isothiocyanates, both natural and synthetic, display anti-carcinogenic activity because they reduce activation of carcinogens and increase their detoxification. This minireview summarizes the current knowledge on isothiocyanates and focuses on their role as potential anti-cancer agents.
유채박의 사료화를 위한 성분개량을 목표한 육종의 기초자료를 얻고자 유채박의 OZT함량이 낮은 영산유채와 그 함량이 고함유인 품종들을 정역교배하여 얻은 F$_2$세대의 종실을 대상으로 OZT함량에 관여하는 유전자수와 세포질의 영향에 관하여 조사분석한 결과는 다음과 같다. 1. F$_2$세대에서 OZT함양을 영산유채에 기준한 낮은 개체의 출현율은 OZT함량이 저함유인 품종을 모본으로 교배한 조합에서 높게 나타났다. 2. OZT함량의 분리비는 63 : 1에 가장 적중도가 높아 3쌍의 열성유전자가 OZT생성에 관여하고 있는 것으로 추정되었다. 3. OZT함량의 유전에 세포질의 영향이 인정되었다.
An experiment was carried out to evaluate the nutritive values of the varieties of rapeseed meal (Canola, Yudal and Yeongsan). Canola and Yeongsan contained more sulphur-amino acid than soybean meal and the metabolizable energy values were 1821 Kcal in Canola, 1586 Kcal in Yudal and 1683 Kcal in Yeongsan. There were no significant differences in body weight gains among the diets of substitute 10% of Canola, 5% of Yudal and Yeongsan for soybean meal and control broiler diet. The weight of thyroid glands increased by increasing the ratio of rapeseed meal in the diets. Based on the results of this study, it might be concluded that Conola meal, Yudal and Yeongsan rapeseed meal can be used up to 10%, 5% and 5% in broiler diets, respectively.
Two completely randomized block design experiments were conducted to evaluate the effect of gamma irradiation processing of canola meal on performance parameters of broiler chicks (Ross 308) and protein quality of canola meal. Protein efficiency ratio (PER) and net protein ratio (NPR) were measured as indices of canola meal protein quality. Samples of canola meal were tested for nutritional value after being irradiated at dose levels 10, 20 and 30 kGy. Glucosinolate content was reduced 40, 70 and 89 percent at irradiation dose levels of 10, 20 and 30 kGy respectively (p<0.01). Percent of erucic acid in total fatty acid content increased 44, 58 and 48% as a function of radiation dose (p<0.01). Dose levels did not affect feed conversion ratio (FCR) and body weight gain of chicks (p>0.05). Liver weight was decreased by irradiation dose (p<0.05). The same trend was observed for kidney weights, but this trend was not significant (p>0.05). Gamma irradiation processing of canola meal had no significant effect on $T_3$ level in blood of chickens that consumed canola meal, but $T_4$ level of chicken blood at the 30 kGy dose decreased significantly (p<0.05). PER and NPR were not affected by radiation dose level (p>0.05). Gamma irradiation seems to be a good procedure to improve the nutritional quality of canola meal.
Indole glucosinolates (IG) play important roles in plant defense, plant-insect interactions, and stress responses in plants. In an attempt to metabolically engineer the IG pathway flux in Chinese cabbage, three important Arabidopsis cDNAs, CYP79B2, CYP79B3, and CYP83B1, were introduced into Chinese cabbage by Agrobacterium-mediated transformation. Overexpression of CYP79B3 or CYP83B1 did not affect IG accumulation levels, and overexpression of CYP79B2 or CYP79B3 prevented the transformed callus from being regenerated, displaying the phenotype of indole-3-acetic acid (IAA) overproduction. However, when CYP83B1 was overexpressed together with CYP79B2 and/or CYP79B3, the transformed calli were regenerated into whole plants that accumulated higher levels of glucobrassicin, 4-hydroxy glucobrassicin, and 4-methoxy glucobrassicin than wild-type controls. This result suggests that the flux in Chinese cabbage is predominantly channeled into IAA biosynthesis so that coordinate expression of the two consecutive enzymes is needed to divert the flux into IG biosynthesis. With regard to IG accumulation, overexpression of all three cDNAs was no better than overexpression of the two cDNAs. The content of neoglucobrassicin remained unchanged in all transgenic plants. Although glucobrassicin was most directly affected by overexpression of the transgenes, elevated levels of the parent IG, glucobrassicin, were not always accompanied by increases in 4-hydroxy and 4-methoxy glucobrassicin. However, one transgenic line producing about 8-fold increased glucobrassicin also accumulated at least 2.5 fold more 4-hydroxy and 4-methoxy glucobrassicin. This implies that a large glucobrassicin pool exceeding some threshold level drives the flux into the side chain modification pathway. Aliphatic glucosinolate content was not affected in any of the transgenic plants.

Glucosinolates(GSLs) are major secondary products($\beta$-thioglucoside N-hydroxysulfates) containing sulfates that are found mainly in Cruciferae family such as cabbage, broccoli, radish, turnip, kale, mustard and rapeseed as the important crops in agronomic and economic aspects. Especially, isothiocyanates(ITCs) have been shown to inhibit carcinogenesis with regulation cancer cell development followed by regulating target enzymes, inducing cell cycle arrest and apoptosis against various human cancer cell lines. Total desulfo(DS)-GSLs of nine type in 210 Korean leaf mustard accessions were isolated and confirmed using electrospray ionization mass spectrometric(ESI-MS/MS) method and DS-GSLs powerful library maked from several reference data. The isolated individual DS-GSLs were identified by removed $\beta$-D-glucopyranose residue ($C_6H_{11}O_5$, MW 163) from MS/MS data. Among them sinigrin(41.7%), glucoiberverin(21.7%) and gluconasturtiin(12.6%) were major components, Especially, despite gluconapin was minor component, accession K046197-1 and K046197-2 showed higher content of 4.11 and 3.31 mg/g(DW), respectively. The total GSLs contents in 210 accessions were ranged from 5.3 to 23.2 mg/g(DW) with a mean value of 13.0 mg/g(DW). As a result of principal component analysis(PCA), the individual GSLs loading plots were composed of three groups and components belonged to each group showed correlationship in quantitative pattern.
Glucosinolates (GSLs), beneficial secondary metabolites for human health are abundantly present in radish vegetable. Radish is a member of Brassicaceae family and its seed, leaf and root contain very important GSLs. The objective of this study was to determine the variation of individual and total GSL contents in leaves and roots of 44 radish (Raphanus spp.) germplasm (26 R. sativus L., 3 R. raphanistrum, and 15 R. sativus L. var. raphanistroides Makino), and compare the GSL contents between leaves and roots among three Raphanus species. Thirteen GSLs were identified, being the glucoraphasatin (GRS) and glucobrassicin (GBS) the most abundant aliphatic and indolyl GSLs in both the leaves and roots. Variation in individual and total GSL contents was found among the germplasm of three Raphanus species. The GRS content was higher in roots than that of leaves in all three Raphanus species but the GBS content was higher in leaves than roots. GRS was represented 87.0%, 92.7% and 94.7% of the total GSL in roots of R. sativus L., R. raphanistrum and R. sativus L. var. raphanistroides (Makino) germplasm, respectively. Germplasm of R. raphanistrum exhibited the highest (average, $79.5{\mu}mol/g\;dw$) total GSL with a ranged from 62.7 to $92.9{\mu}mol/g\;dw$. The germplasm IT119288, Joseonmu and IT119262 from R. sativus L., RA 504 and K046542 from R. raphanistrum, and Gyeongju-2003-32 (G2003-32) and IT302373 from R. sativus L. var. raphanistroides (Makino) had high total GSL contents and these could be good candidates for developing the functional compounds-rich varieties in radish breeding program.

The released amount of thiocyanate ion in Cruciferous vegetables treated by wet heat, increased as the reaction time goes by and was maximum value after treatment for 30min, but it was not changed by dry heat treatment. When samples were autolyzed by myrosinase, the amount of thiocyanate ion increased gradually as time goes by, was maximum value after 3hrs and much higher than those in the sample treated by wet. The released amount of thiocyanate ion in each sample showed much in cabbage, Chinese cabbage, radish, kale and mustard in the order. The generated amount of indoleacetonitrile by heat treatment increased as time goes by and the generated amount in each sample determined was high in the order of cabbage, Chinese cabbage and radish.
