Guar gum이 type-II 당뇨병 환자의 혈액성분에 미치는 영향 (Effect of guar gum on the blood composition in type-II diabetic subjects)

  • Guar gum, a storage polysaccharide galactomannan was administered to 11 patients with type-II diabetes mellitus for 7 days and 3 weeks. They took 5 grams of guar gum 30 mins before each of three meals daily. The dinner 2-h postprandial values of their blood glucose were significantly lowered (P<0.05) after their guar treatment for 7 days compared with before taking guar gum. The 2-h postprandial values of blood glucose were significantly lowered(P<0.05) after 3 weeks of gual treatment compared with before taking guar gum. In an oral glucose tolerance test, their blood glucose values were significantly lowered at 120 mins(P<0.02) and 180 mins(P<0.05) after guar treatment. Total-lipid(P<0.01) and triglycerides(P<0.02) of their blood were significantly decreased and HDL-cholesterol(P<0.02) was significantly increased after guar treatment. HbA1C was significantly reduced (P<0.05) from 11.3% to 10.1% The body weight, total-cholesterol and insulin activity of the patients after guar treatment were not significantly changed and the satiety ratings of the patients with guar treatment was not significantly changed and the satiety ratings of the patients with guar treatment was not significantly changed, however, the subjects that answered from 'Want to eat but can wait' to 'No desire to eat' were 81.1%. It is concluded that guar gum improves their carbohydrate and lipid metabolism in Korean type-II diabetic subjects with high fiber diets.

삼기지황탕가음양곽이 Streptozotocin으로 유발된 흰쥐의 당뇨병성(糖尿病性) 신증(腎症)에 미치는 영향 (Beneficial Effect of Epimedii Herba combined Samgijiwhang-Tang on Diabetic Nephropathy Rats)

  • The present study was carried out to investigate the preventive effect of Epimedii Herba combined Samgijiwhang-Tang(SJTE) on streptozotocin(STZ)-induced diabetic nephropathy. SJTE was given to rats with oral administration. The experimental animals were divided into normal group of rats, control group of STZ-induced diabetic rats, and sample group with SJTE administration. Experimental diabetic nephropathy was induced by the injection of STZ(60mg/kg) to the rat via the peritoneum. The effect of SJTE on STZ-induced diabetic nephropathy was observed by measuring the serum level of insulin, glucose, creatinine and BUN. Urine secretion of albumin for 24 hours and urine level of glucose measures too. Anti-oxidative stress of STZ administration in living body was estimated by measuring lipid peroxide in cortex of kidneys. STZ induced increase of serum glucose. creatinine, urine albumin secretion and renal cortical lipid peroxidation were lowered by SJTE administration. In conclusion, the SJTE treatment showed protective effect on rat diabolic nephropathy model, and action mechanism of the effect was thought to be concerned with internal glucose metabolism.

Perilipin 5 is a novel target of nuclear receptor LRH-1 to regulate hepatic triglycerides metabolism

  • Liver receptor homolog-1 (LRH-1) has emerged as a regulator of hepatic glucose, bile acid, and mitochondrial metabolism. However, the functional mechanism underlying the effect of LRH-1 on lipid mobilization has not been addressed. This study investigated the regulatory function of LRH-1 in lipid metabolism in maintaining a normal liver physiological state during fasting. The Lrh-1f/f and LRH-1 liver-specific knockout (Lrh-1LKO) mice were either fed or fasted for 24 h, and the liver and serum were isolated. The livers were used for qPCR, western blot, and histological analysis. Primary hepatocytes were isolated for immunocytochemistry assessments of lipids. During fasting, the Lrh-1LKO mice showed increased accumulation of triglycerides in the liver compared to that in Lrh-1f/f mice. Interestingly, in the Lrh-1LKO liver, decreases in perilipin 5 (PLIN5) expression and genes involved in β-oxidation were observed. In addition, the LRH-1 agonist dialauroylphosphatidylcholine also enhanced PLIN5 expression in human cultured HepG2 cells. To identify new target genes of LRH-1, these findings directed us to analyze the Plin5 promoter sequence, which revealed -1620/-1614 to be a putative binding site for LRH-1. This was confirmed by promoter activity and chromatin immunoprecipitation assays. Additionally, fasted Lrh-1f/f primary hepatocytes showed increased co-localization of PLIN5 in lipid droplets (LDs) compared to that in fasted Lrh-1LKO primary hepatocytes. Overall, these findings suggest that PLIN5 might be a novel target of LRH-1 to mobilize LDs, protect the liver from lipid overload, and manage the cellular needs during fasting.

Streptozotocin으로 유도한 당뇨쥐의 대퇴신경에서 Hematoxylin의 당대사 개선 메커니즘 (The Mechanism of Hematoxylin on Glucose Metabolism Improvement in Sciatic Nerves from Streptozotocin-induced Diabetic Rats)

  • Hematoxylin은 한국의 남부지방에서 당뇨합병증을 치료하기 위하여 사용해오던 민간약제인 Hematoxylon campechianum의 주성분이다. 본 논문에서는 hematoxylin의 혈당저하 메커니즘을 연구하기 위하여 3개 군의 흰쥐-정상군, 당뇨군, hematoxylin 처치군-에서 분리한 대퇴신경을 대상으로 2-deoxyglucose 수송능과 인지질 대사능을 조사였다. 실험결과 hematoxylin은 당뇨군에서 혈당치를 현저하게 감소시키는 것으로 나타났다. 또한 무게단위의 기준으로 당뇨군의 신경조직의 경우, 총 인지질의 양은 20% 감소하였으나 상대적으로 phosphatidylinositide의 감소는 작은 것으로 나타났다. 이 경우, hematoxylin을 처치하면 2-[3H] myo-inositol이 총 phosphoinositids로 대사되는 비율이 증가하는 것으로 밝혀졌다. 이러한 효과는 낮은 농도의 hematoxylin 처치군보다 높은 농도의 처치군에서 훨씬 큰 것으로 나타났다. 이상의 실험결과는 hematoxylin의 당뇨개선 메커니즘이 myo-inositol 대사를 증가시킴으로써 체내 당 수송과 인지질 대사를 정상화시킨다는 점을 제시하고 있다. 따라서 본 연구결과를 토대로 저자들은 hematoxylin은 향후 당뇨 치료제로서 후보 물질이 될 수 있음을 제시하고자 한다.

남빙양 krill이 흰쥐의 혈청 지질성분 및 알부민/글로불린 비에 미치는 영향 (Influences of Antarctic Ocean Krill (Euphausia superba) on Lipid Components and A/G Ratio in Rats)

  • The aim of the study were to investigate the influences of krill (Euphausia superba) meal on the body weight, lipid metabolism functional improvement, blood glucose level, protein component in the sera of rats which fed experimental diets for 5 weeks. Serum concentrations of total cholesterol, Low-Density Lipoprotein (LDL)-cholesterol, free cholesterol, triglyceride (TG), phospholipid (PL) and blood glucose were higher in the control diet group (G1 group) than the control diet plus 10% krill meal group (G2 group), the control diet plus 20% krill meal group (G3 group), the control diet plus 30% krill meal group (G4 group), and a general dose and time independent one-way analysis of variance was performed to assess efficacy. Conversely depending on the content of krill meal for the High-Density Lipoprotein (HDL)-cholesterol level, it showed higher results. The concentrations of total protein, albumin and globulin in sera, there were not significant difference among the groups (p<0.05). The results indicate that a krill meal diet effectively inhibited increases in lipid elevation, blood glucose level in the sera of rats.

Effects of a Butanol Fraction of Alisma canaliculatum and of Selenium on Blood Glucose Levels and Lipid Metabolism in Streptozotocin-Induced Diabetic Rats

  • The purpose of this study was to investigate the effects of a butanol fraction of fraction of Alisma canaliculatum All. Braun et Bouche (Ac), and of selenium (Se), on plasma gllucose and lipid levee in streptozotocin (STD-induced diabetic rats. Male Sprague-Dawley rats, fed the AIN-93 recommended diet, were divided into five groups: a non-diabetic control group (no STZ treatment), and four 572-induced diabetic groups which consisted of a diabetic-control group, an Ac-treated group, an Ac-Se treated group, and a Se-treated group. Diabetes was induced in the rats by an injection of STZ into the tail vein at a dose of 45 mg/kg body weight. The butanol (BuOH) fraction of Ac was orally administered at a rate of 400 mg/kg body weight for 21 days to both the Ac and Ac-Se groups. The supplementation of selenium in the Se and Ac-Se groups was achieved by adding (freshly, every day) 2 mg of Se as Na$_2$SeO$_3$ per kg of feed. The rats'body weights and hematocrit (Hct) levels were measured, along with plasma levels of glucose, insulin, cholesterol, HDL-cholesterol, triglyceride (TG), and free fatty acids (FFA). Aminotransferase activities were also analyzed. The non-diabetic rats gained weight, while the diabetic rats lost weight - except in the Ac-Se group, which maintained their initial weight. The blood glucose levels of the Ac group and the Se group were significantly lower than for the diabetic-control group. The plasma triglyceride levels were lowered when both Ac and Se were administered to diabetic rats. The concentrations of plasma FFA in the Ac-Se group were significantly lower compared with the diabetic-control group. Plasma cholesterol levels and alanine aminotransferase activity in the Ac, Ac-Se, and Se groups were significantly lower when compared with the diabetic-control group. Aspartate aminotransferase activity was significantly lower in the Se group compared to the other diabetic groups. These data show that treatment with a butanol fraction of Ac in combination with Se has no synergistic effect. Plasma glucose levels tended to be low when Se was administered to diabetic rats. Supplementation of Se in diabetic rats did not elicit a significant increase in plasma insulin levels or result in hypolipemic effects.

Crosstalk between FXR and TGR5 controls glucagon-like peptide 1 secretion to maintain glycemic homeostasis

  • Though bile acids have been well known as digestive juice, recent studies have demonstrated that bile acids bind to their endogenous receptors, including Farnesoid X receptor (FXR) and G protein-coupled bile acid receptor 1 (GPBAR1; TGR5) and serve as hormone to control various biological processes, including cholesterol/bile acid metabolism, glucose/lipid metabolism, immune responses, and energy metabolism. Deficiency of those bile acid receptors has been reported to induce diverse metabolic syndromes such as obesity, hyperlipidemia, hyperglycemia, and insulin resistance. As consistent, numerous studies have reported alteration of bile acid signaling pathways in type II diabetes patients. Interestingly, bile acids have shown to activate TGR5 in intestinal L cells and enhance secretion of glucagon-like peptide 1 (GLP-1) to potentiate insulin secretion in response to glucose. Moreover, FXR has been shown to crosstalk with TGR5 to control GLP-1 secretion. Altogether, bile acid receptors, FXR and TGR5 are potent therapeutic targets for the treatment of metabolic diseases, including type II diabetes.

The Short-Term Effects of Soft Pellets on Lipogenesis and Insulin Sensitivity in Rats

  • The purpose of the present study was to investigate the short-term effects of a 12-day, soft pellet (SP) diet with a 3-h restricted feeding schedule on caloric intake, body weight, lipid metabolism, and insulin sensitivity. Glucose and insulin levels were measured pre-, mid-, and post-feeding. The SP rats exhibited postprandial hyperglycemia compared to rats fed control pellets (CP). The insulin response of SP rats during a meal was significantly higher than that of CP rats. There were no significant differences in the hepatic triacylglycerol contents and lipogenesis gene mRNA levels of SP and CP rats. However, the hepatocytes of SP rats were slightly hypertrophic. In addition, histological analysis revealed that the pancreases of SP rats had more islet areas than those of CP rats. This study demonstrated that feeding an SP-only diet for 12 days induces glucose intolerance, suggesting that the consumption of absorbable food, like a soft diet, may trigger glucose metabolism insufficiency and lead to life-threatening diseases.

Metabolic Activity of Desalted Ground Seawater of Jeju in Rat Muscle and Human Liver Cells

  • Ground seawater in the east area of the volcanic Jeju Island contains abundant minerals. We investigated the metabolic activity of electrodialyzed, desalted ground seawater (EDSW) from Jeju in both cultured cells and animals. The addition of EDSW to the culture medium (up to 20%, v/v) reduced the leakage of lactate dehydrogenase and increased MTT activity in CHO-IR cells. EDSW (10%) promoted insulin-induced glucose consumption in L6 muscle cells as well as the activities of the liver ethanol-metabolizing enzymes, alcohol dehydrogenase and aldehyde dehydrogenase. Moreover, EDSW suppressed palmitate-induced intracellular fat accumulation in human hepatoma $HepG_2$ cells. Activities of AMP-stimulated protein kinase and acetyl CoA carboxylase, enzymes that modulate fat metabolism, were altered by EDSW in $HepG_2$ cells toward the suppression of intracellular lipid accumulation. EDSW also suppressed hepatic fat accumulation induced by a high-fat diet in mice. Taken together, EDSW showed beneficial metabolic effects, including the enhancement of ethanol metabolism and insulin-induced glucose consumption, and the suppression of intrahepatic fat accumulation.

고수 에탄올 추출물의 고지방식이 비만 동물모델에서의 항비만효과 (Ant-Obesity Effect of Coriandrum sativum L. Ethanol Extract in High Fat-Induced Obesity Animal Model)

  • This study investigated the anti-obesity effects of Coriandrum sativum L. ethanol extracts in a high fat diet-induced obesity model (DIO). We confirmed the anti-obesity effects by analysing the expression of the related proteins, weight gain, dietary intake, dietary efficiency, blood biochemistry, histological analysis and western blot analysis. After oral administration of Coriandrum sativumL. ethanol extracts at concentrations of 250 and 500 mg/kg, a significant improvement in dietary efficiency, reduction in weight gain, triglycerides, total cholesterol and LDL-cholesterol in blood lipid was observed for 8 weeks. In addition, improvement in blood glucose and metabolism confirmed through glucose tolerance test was observed. Further, the concentration of alanine transaminase (ALT) in blood was significantly decreased, which improved the fatty liver caused by high-fat diet intake as confirmed by liver tissue analysis. This phenomenon was confirmed to decrease the expression of fat accumulation-related PPARγ and FAS protein in the liver tissue. Especially, it is believed that FAS, a liposynthetic enzyme, has a stronger inhibitory effect than PPARγ. Therefore, Coriandrum sativum L. ethanol extract is thought to improve obesity by reducing blood lipids levels, improving glucose metabolism and inhibiting synthesis of the fat that accumulates in the liver in high-fat diet-induced obesity animal models.
