The aim of this study was to evaluate effects of modified glucomannan ($Mycosorb^{TM}$) on the antioxidant profile of egg yolk and tissues of newly hatched quail after aurofusarin inclusion in the maternal diet. Fifty-four 45 day-old Japanese quail were divided into three groups and were fed a corn-soya diet balanced in all nutrients ad libitum. The diet of the experimental quail was supplemented with aurofusarin at the level of 26.4 mg/kg feed in the form of Fusarium graminearum culture enriched with aurofusarin or with aurofusarin plus $Mycosorb^{TM}$ at 1 g/kg feed. Eggs obtained after 8 weeks of feeding were analysed and incubated in standard conditions of $37.5^{\circ}C$/55% RH. Samples of quail tissues were collected from newly hatched quail. The main carotenoids, retinol, retinyl esters and malondialdehyde were analysed by HPLC-based methods. Inclusion of aurofusarin in the maternal diet was associated with decreased carotenoid and vitamin A concentrations in egg yolk and liver of newly-hatched quail. Furthermore, lipid peroxidation in quail tissues was enhanced. Inclusion of modified glucomannan ($Mycosorb^{TM}$) in the toxin-contaminated diet provided a significant protective effect against changes in antioxidant composition in the egg yolk and liver. It is suggested that a combination of mycotoxin adsorbents and natural antioxidants could be the next step in counteracting mycotoxins in animal feed.
Acemannan, a highly acetylated glucomannan, was extracted from fresh Aloe vera leaves by ethanol fractionation, resulting in a concentration increase of more than threefold. The presence of acemannan was confirmed using FTIR and 1H NMR analysis, revealing an average molecular weight of 780 kDa. The flocculating activity of the fractionated aloe gel polysaccharide was assessed through settling tests in a 1% (w/v) bentonite suspension. The results demonstrated that the aloe polysaccharide exhibited remarkable stability within a temperature range of 20~70 ℃. The maximal flocculation rate at different pH levels ranged from 93% to 97%, with an optimal dose for maximum flocculation rate between 0.25 mg/mL. Notably, the minimum dose required for flocculation was achieved at a pH of 3, attributed to the compression of electrostatic repulsion on the surface of bentonite particles. However, the flocs obtained under acidic conditions were less dense and compact, exhibiting lower sedimentation velocity compared to those formed under neutral and alkaline pH conditions. Additionally, the addition of salt showed a slight synergistic effect on flocculation, significantly enhancing the sedimentation velocity. This investigation highlights the potential of Aloe vera polysaccharide as a natural and edible flocculant, offering promising applications in various industries.
A feeding trial was conducted on commercial broilers for a period of 35 days to determine the individual and combined effects of aflatoxin (AF) and T-2 toxin (T-2) on performance, organ weights and immune status. The efficacy of dietary glucomannan-containing yeast product (GYP) ($Mycosorb^{(R)}$) and hydrated sodium calcium aluminosilicate (HSCAS) in preventing the adverse effects of aflatoxin and T-2 toxin was also evaluated. Twelve dietary treatments ($4{\times}3$ factorial) comprising two dietary levels each of AF (0 and 2 mg/kg), T-2 toxin (0 and 1 mg/kg), GYP (0 and 1 kg/ton) and HSCAS (0 and 10 kg/ton) were tested on 720 commercial broiler chickens divided at random into 36 replicates of 20 chicks each (10 males and 10 females). Weight gain and feed intake were recorded weekly. Organ morphology and antibody titers for Newcastle disease (ND) and infectious bursal disease (IBD) were measured on the $35^{th}$ day. AF and T-2 toxin individually decreased weight gain and increased feed conversion ratio (FCR) (p<0.05). AF alone (p<0.05) increased weights of liver, kidney, gizzard and spleen and reduced thymus and bursal weights. T-2 toxin (p<0.05) increased liver and gizzard weights and decreased thymus weight. Both AF and T-2 toxin when fed individually affected ND and IBD titers in a significant manner. Significant interactions between AF and T-2 toxin were observed for their additive effects on weight gain, FCR, organ weights and antibody titers. Addition of GYP (p<0.05) improved weight gain, feed conversion efficiency and restored the organ weights. Antibody titers against ND and IBD were significantly improved with the supplementation of GYP. Supplementation of HSCAS (p<0.05) resulted in improvement in weight gain and restored organ weights in the groups fed AF alone, but not in T-2 toxin fed groups. HSCAS inclusion did not influence FCR in toxin fed groups. Addition of HSCAS (p<0.05) improved the antibody titers against ND and IBD only in AF fed groups. Thus, the results indicate that addition of GYP is effective in averting the individual and combined toxicity of aflatoxin and T-2 toxin in commercial broilers, while HSCAS is effective only against aflatoxin.
Aloe gel의 동결건조에 농축도가 서로 다른 액상 시료를 사용하였을 때 건조분말의 기능성 변화를 비교분석하였다. Aloe의 많은 기능성 중에서 유효 다당류의 농도, 기능성 작용기에 대한 FT-IR 특성, Jack bean urease 저해 효과, FAC(fat adsorption capacity)를 대상으로 하였다. 농축도가 높은 경우에 유효 다당류의 농도, urease 저해 효과, FAC 모두 상대적으로 크게 나타났다. FT-IR의 분석에서는 농축도가 높은 경우에 glucomannan의 acetyl 기에 대한 spectrum 영역인 1600-1550 $cm^{-1}$와 1450-1400 $cm^{-1}$에서 흡광도가 크게 관찰되었다. 결과적으로 Aloe gel의 동결건조시 전처리과정인 농축 공정에서는 농축도가 높을수록 유효다당류의 양과 acetyl기 함량의 수준이 크며, 간접적으로 urease 저해 효과에 따른 Helicobacter pylori의 억제 및 FAC의 증가 효과를 보였다.
Environmental and energy issues have always been a hot topic of global research. Oil leakage has caused great damage to the environment, affecting a wide area and it is difficult to clean up. In most cases, carbon-based adsorbents are typically utilized to remove oil spills because of their economic benefits and high adsorbent efficiency. At the same time, its excellent material properties can also be used for the preparation of supercapacitors. In this paper, the carbon aerogels were prepared by the one-step method. The prepared materials endowed a 3D network structure with a huge number of micropores and mesoporous, and the material is light-weight, stable, hydrophobic and has affinity for oil (17.02 g/g) to the KGM carbon aerogel. Through the physicchemical characterization, the KGM carbon aerogel shows specific surface area is $689m^2/g$, high water contact angle ($136.64^{\circ}$) and excellent reusability (more than 15 cycle times). In addition, we also discussed the electrochemical properties of the material and obtained the specific electrical capacity of 139 F/g under the condition of 1 A/g.
Constipation is one of the most common symptoms that a child visits pediatrician. The general approach to the child with functional constipation includes the following steps; disimpaction, maintenance and withdrawal of medication. There are many drugs which can be applied to children; osmotic agents (lactulose, sorbitol, magnesium hydroxide/citrate, polyethylene glycol with/without electrolytes, sodium phosphate, glycerin), stimulants (senna, bisacodyl, caster oil), lubricant (mineral oil), bulking agent (psyllium, cellulose, glucomannan). At each stage of treatment, one or some of these drug can be applied to the purpose. The author tries to summarize recent studies on drugs for constipation in child, and finally introduces new dugs for constipation which is under investigation.
Xylan in hemicellulose has been reported as one of the reasons of the yellowing of chemical pulp and paper. But little relevant information is available in literature. In this study, we examined into the influence that glucuronoxylan or glucomannan and metallic ion($Cu^{2+},\;Fe^{2+},\;Fe^{3+},\;Mn^{2+}$) residued in pulp get each or when mixed at yellowing. Filter paper was treated with each metallic ion, glucuronoxylan and the mixture of glucuronoxylan and metal ions, and brightness and CIE $L^*a^*b^*$ was measured after accelerated aging. As the results of measurements, the filter paper processed by 10% glucuronoxylan was dropped $2{\sim}3%$ on the brightness after accelerated aging for 24 hours. Also, the filter paper treated with glucuronoxylan and $Fe^{2+}$ was dropped 7% on the brightness.
Generally, aging of paper is evaluated by folding endurance and degree of yellowing. It was known as that lignin, hemicellulose and metallic ion affect yellowing of mechanical or chemical pulps. Among these elements, especially, the study of hemicellulose that affect at yellowing of pulp is insufficient. In this study, we examined into the influence that glucuronoxylan or glucomannan and metallic ion($Cu^{2+},\;Fe^{2+},\;Fe^{3+},Mn^{2+}$) residued in pulp get each or when mixed at yellowing. Paenibacillus woosongensis의 유전체 부분 염기서열로부터 mannanase를 코드하는 것으로 유추되는 open reading frame을 중합효소연쇄반응으로 증폭하여 대장균에 클로닝하고 염기서열을 결정하였다. Mannanase 유전자는 man26AT로 명명하였으며 1,053 아미노산으로 구성된 단백질을 코드하는 3,159 뉴클레오티드로 이루어졌다. 아미노산 잔기배열을 분석한 결과 Man26AT는 glycosyl hydrolase family 26의 mannanase와 상동성이 높은 활성영역, 탄수화물 결합영역 CBM27과 CBM11로 구성되어 있었다. Man26AT의 아미노산 배열은 P. ihumii의 유추된 mannanase와 상동성이 81%이고 다른 Paenibacillus 속 균주의 여러 mannanases와 57% 이하의 상동성을 보였다. man26AT 유전자를 함유한 재조합 대장균의 균체 파쇄상등액은 $55^{\circ}C$와 pH 5.5에서 최대의 mannanase 활성을 보였고, $50^{\circ}C$에서 1시간 열처리한 후에 80% 이상의 잔존활성을 보였다. Man26AT는 locust bean gum (LBG) galactomannan과 konjac glucomannan에 대한 분해활성이 유사하였으며, carboxymethylcellulose, xylan과 para-nitrophenyl-${\beta}$-mannopyranoside는 분해하지 못하였다. Man26AT에 의해 mannotriose, mannotetraose, mannopentaose와 mannohexaose 등의 만노올리고당이나 LBG로부터 공통의 최종 가수분해 산물로 mannose, mannobiose와 mannotriose가 생성되었다. 또한 mannotriose 보다 큰 만노올리고당이 LBG와 guar gum의 분해산물로 각각 생성되었다. 그러나 Man26AT는 mannobiose를 분해하지는 못하였다. 활성염색을 통해 Man26AT는 균체 내에서 3개 이상의 크기가 다른 활성 단백질로 분해된 것이 확인되었다.
