Antioxidant Activity of Ginseng Extracts Prepared by Enzyme and Heat Treatment (효소처리와 열처리에 의한 인삼 추출물의 항산화 활성)

  • In this study, extraction yield of ginseng and contents of phenolic compounds were increased through enzyme and heat treatment, and antioxidant activities of each treatment group were investigated. In the case of enzyme treatment groups, the extraction yields showed remarkable difference: pectinase (76.0%)> cellulase (44.7%)> ${\alpha}-amylase$ (43.1%)> protease (34.3%). Heat treatment groups did not show much difference in extraction yields $(51.1{\sim}53.1%)$ except for $90^{\circ}C-treated$ group (48.7%). Pectinase-treated group (2.21%) and $90^{\circ}C$ heat treatment group (2.34%) showed the highest total phenol contents. DPPH radical scavenging activities of the solvent-treated, enzyme-treated and heat-treated extracts were $12.3{\sim}33.2%$, $29.7{\sim}40.3%$ and $57.8{\sim}75.2%$, respectively, at 100 mg/mL concentration. The pectinase-treated extract exhibited the greatest ABTS radical scavenging activity (60%), followed by $150^{\circ}C$, $ 120^{\circ}C$, and $90^{\circ}C-treated$ groups.

Sampling Methods for the 'dark grey cutworm' (Agrotis tokionis B.) Larval Population and Effect of its Larval Density on Tobacco Yield (숯검은밤나방(Agrotis tokionis B.) 유충개체군의 밀도추정방법 및 유충밀도와 연초감수량의 관계)

  • As a primary study on the economic injury level of A. tokionis larval populations in tobacco fields, we carried out some experiments for the evaluation of sampling efficiency of 3 sampling methods (pit-fall traps of apple pomace and rice bran, and clover patches) for the larval population and the regressions between loss and infested larval density The $10\times10cm$ clover patch showed a better sampling efficiency $11.8\;to\;18.0\%$ than the others. The sampling efficiency of clover patch becomes higher when the plots did not have any green plants. The linear regression equation (Y=4.2+1.383x) between loss (Y:kg/10a) and infested larval density (X: no. of larvae/plot) which was obtained by substitution of damage ratio and corrected damage ratio fitted the observed data better than the one (Y=2.68X) obtained without substitioning.

  • Fruit bodies of Ganoderma lucidum have been used for most pharmacological studies, but pharmacological effects are likely variable because the habitats and strains of Ganoderma lucidum are different. Therefore, their fermentation is required to produce constant and reliable pharmacolo­gical constituents from Ganoderma lucidum. During the studies of medium for industrial application. it was found that ginseng root residues, remaining after being extracted with ethanol, were a good carbon source for a fermentation of Genoderma lucidum and a corn steep liquor was also economical for the nitrogen source. Yield of the mycelial cultured in ginseng root residues and corn steep liquor was 2.5 times higher than that in glucose and peptone, known as a conventional medium of Ganoderma lucidum. The polysaccharide content of the extracts from the cultured mycelia was higher than that from fruit bodies, but protein content was vice versa. Extracts of the cultured mycelia were more effective and lasting than extracts of the fruit bodies in decreased hypertention of spontaneously hypertensive rats (SHR).

  • This study was conducted to obtain basic information on growth characteristics and major components of three Korean endemic resource plant species, Artemisia sp., Gynostemma pentaphyllum Makino and Humulus japonicus S. et Z. , growing in different areas in Korea. Three geographical types of artemisia, namely Pangssuk, Ongjinssuk and Yakssuk which were collect in Suwon, Ongjin and Eumsong, respectively were compared in yield, and Pangssuk artemisia was higher in yield than the others. However, essential oil content was the highest in Yakssuk artemisia. Dry weight of areal parts of G. pentaphyllum was higher in the cultural method with props for tendril growth than in the conventional cultural method without props. Rutin was detected in areal parts of G. pentaphyllum. Dry weight of areal parts of H. japonicus collected at the Kyeryong mountain in Chungnam province was higher than that collected in Eumsong. A phenolic compound isoquercitrin was detected in areal parts of H. japonicus.

Characteristics of A New Flue-cured Tobacco Mutant Line KF 8832-85 (황색종 연초 돌연변이 계통 KF8832-85의 특성)

  • A new flue-cured tobacco mutant line BU 8832-85 was developed at Taegu Experiment Station, Korea Ginseng and Tobacco Research Institute in 1994. KF 8832-85 was resulted from a cross of flue-cured cultivars NC 95$\times$NC 2326, and developed by a pedigree system of breeding ; initial selection was made by plant type and resistance to bacterial wilt(BW) disease(heudomonas solanaceamm) in the F2 generation under the natural field conditions infested with the pathogen. One white flowered plant was occurred by spontaneous mutation in a certain line among the F3 generatioin while the others were pink. Six plants from the seeds by selfing were selected at the field infested with the pathogen among 240 populations with white flowering in the F4, KF 8832-85 was selected based on yield and leaf quality trials among 6 lines in Fs generation. BCF 8832-85 was compared with its Parent for certain agronomic and chemical characteristics at Taegu Experiment Station in 1993 and 1994. The results showed that KF 8832-85 have white flower, the stalk height was approximately that of NC 2326 but averaged about loom taller than NC 95. It produced ground suckers as much as NC 95, and did not breakdown leaf at the same as WC 2326. KF 8832-85 have high resistance to bacterial wilt disease. Yield of KF 8832-85 was 10 and 18% higher then that of NC 2326 and WC 95, respectively. Price per Kg was equal to that of NC 2326. The contents of nicotine and reducing sugar did not differ significantly from NC 95, while total nitrogen was significantly lower than NC 95. Therefore, the new mutant line is genetically stable for agronomic and chemical characteristics and provides a source of bacterial wilt disease resistance for use in breeding resistant flue-cured cultivars.   • The objectives of this investigation were to produce artificially an antitumor constituent by submerged culture of the mycelium of Coriolus versicolor (Fr.) Quel., to characterize the influence of various modifications of the nutrient and culture conditions with respect to the pro­duction, to determine chemical composition of the antitumor constituent, and to examine effects of the constituent on the immune response of mice. Submerged agitation of the mycelium in flasks containing a nutrient solution showed its adequate growth. Especially the mycelial growth in the medium containing glucose and yeast extract was abundant. The addition of cotton seed flour or ginseng waste to the medium increased the yield of mycelial growth and the production of the antitumor constituent. The replacement of glucose with starch also yielded the adequate growth. The antitumor constituent extracted from the mycelium and isolated from the culture filtrate was a protein-bound polysaccharide. The analyses of this constituent by GLC and amino acid autoanalysis showed that it contained four monosaccharides and fifteen amino acids. The protein-free polysaccharide of the constituent was also found to exert greater antitumor activity against sarcoma-180 in mice than the entire constituent. The antitumor constituent was found to potentiate the immune response of mice against sheep red blood cell. The protein-bound polysaccharide exerted more favorable influence on the immunity than the protein-free moiety.

  • Field test were conducted to select the best method for soil phosphorus test and to obtain the critical levels of each method for P fertilizer recommendations based on tobacco production. Truog method was recognized to be the mort suitable one for soil P test. N. Carolina, Bray No.1 and Olsen methods were followed in order. Bray No. 2 and Lancaster methods showed the least satisfaction. The critical levels of available P by lancaster, Bray No. 1, N. Carolina,01sen, and Truog methods were 55ppm, 32ppm, 21ppm, 19ppm, and 22ppm, respectively. Below the critical level a large yield response can be expected with 10kg $P_2O_5$/10a, above critical level little or no response is expected. In soils the more the initial available p, the more available P was carried over, while the more the total amount of silt and clay, the less the residual P was found. The increase in residual P by 10kg of $P_2O_5$ was greater than that of 5kg/10a. The chemical composition and the price of the cured leaf were not affected by the $P_2O_5$ application, indicating that the increase in the rate of $P_2O_5$ application would not exert on the quality of tobacco.

연초 버어리종 웅성불임 일대잡종 KB 111의 육성경과 및 농경적 특성

  • The vein necrotic strain of Potato Virus Y (PVY - VN) and black shank (Phyto-phthora parasitica var. nicotianae) are the two major diseases causing severe damages especially in burley tobacco (N. tabacum L.) area in Korea. A new tobacco variety, KB 111, resistant to PVY and black shank disease, was developed by Korea Ginseng & Tobacco Research Institute in 1997. It is a male sterile(MS) F$_1$ hybrid of the cross between MS TC 613 and KB 108. KB 111 was compared to Burley 21 on the agronomic characteristics and disease resistances in performance tests: It possessed upright growth habit and flowered two days later than Burley 21. It was resistant to both PVY and black shank and yielded about 3% more cured leaf than Burley 21, but other characteristics are very simiar to those of Burley 21. The chemical composition and physical properties of the cured leaf of KB 111 were as much acceptable as those of Burley 21 while it produced average yield of good quality leaf and appeared to resistant to PVY and black shank disease on regional farm test in 1998.

Fertilizer Placement Effect on Agronomic Characteristics of Burley Tobacco(N. tabacum L.) (시비방법이 버어리종 잎담배의 농경적 특성에 미치는 영향)

  • Recently the method of basal compound fertilizer (N : P : K = 18.2 : 9.8 : 35.0) Placement has been changed from banding to broadcasting in tobacco cultivation. The effects of the compound fertilizer placement, level of fertilizer and additional urea application on the agronomic characteristics, two chemical compositions and physical Properties of burley tobacco(Nicotiana tabacum L. cv. KB 108) were investigated at Chonju Experiment Station, Korea Ginseng and Tobacco Research Institute in 1995-1996. Six treatments consisted of (1) band (method of fertilizer Placement) + In kg/10a(compound fertilizer) + 0(additional urea applied), (2) broadcast+140+0, (3) broadcast+140+25, (4) band+180+0, (5) broadcast+180+0, and (6) broadcast +180+25. The additional urea was applied at hilling. No significant differences were detected between banding and broadcasting method of compound fertilizer Placement in field 9rowth, wield, organoleptic qualify(price Per kilogram), chemical composition and Physical properties of cured leaf, The increased compound fertilizer by 30%(40k/10a) or the additional urea application by 25kg per 10a produced slightly higher yield than the recommended amount of basal compound fertilizer without additional urea application did. It also increased the total alkaloid content of cured leaf. It is recommended that no more basal fertilizer above the recommended amount and no additional urea application are needed in burley tobacco fertilization, even though the method of basal fertilizer placement being chanced from banding to broadcasting. Development of a New Flue - cured Tobacco Variety KFl13 by Nicotiana africana Method (Nicotiana africana 방법에 의한 황색종 연초 신품종 KF113 육성)

  • A new flue - cured tobacco variety KF113 was developed by Nicotiana africana method from a cross of NC82 and Coker 347 at the Suwon Experiment Station. It was tested in the 0fficial Variety Test in 1991-1992 and the flue - cured Regional Farm Test in 1992. KFl13 flowers 4 days later than WC82 (standard variety in Korea) and its harvestable leaves are 2 more than those of NC82. The leaf type and shape of KFl13 resembles Coker 347. It has high resistance to bacterial wilt (Pseudomonas solanacearum) and black shank (Phytophthora parasitica var. nicotianae), and is susceptible to tobacco mosaic virus. It should adapt well to the flue - cured production area and can reduce premature flowering under unfavorable weather conditions. Yield of KFl13 is 5% higher than that of NC82, and nearly equal in value per kg compare with NC82. This variety met acceptable standards for chemical and physical characteristics of cured leaf and for smoking taste evaluated by panel members in Korea Ginseng & Tobacco Research Institute.
