Chemical Compositions and Antioxidant Activity of Extract from a Extruded White Ginseng (압출성형 백삼추출물의 화학적 조성 및 항산화 활성)

  • Chemical components and antioxidative activities of white ginseng, red ginseng and extruded white ginseng (EWG) were evaluated. Extrusion condition was 20% moisture content, 100 and $140^{\circ}C$ barrel temperature. The results showed that total sugar and acidic polysaccharide contents of white ginseng powder were increased after extrusion treatment of which EWG at $140^{\circ}C$ barrel temperature had higher value than EWG at $100^{\circ}C$ barrel temperature. Free radical scavenging activity of EWG at $140^{\circ}C$ barrel temperature was 80.2 and 45.6% respectively. The butanol fraction of polyphenolic compound and acidic polysaccharide were $27.2{\pm}0.1\;mg/g$ and $217.6{\pm}0.7\;mg/g$, respectively. The ginsenosides were quantified by HPLC and the yield of ginsenoside-Rg3s and Rg3r were achieved by extrusion process.

  • The competition in root growth between plants was investigated under the field condition of Panax ginseng. Competition pattern was classified in three groups, interline, interrow and diagonal and the principal factor for competition was considered light, nutrients including water and both of them, respectively. Interline competition was always great while interrow and diagonal competition were varied to field. Diagonal competition was always great at the backside line. Competition appeared to be significant from the age of 4 years old at which leaf area index become greater than one. Above facts strongly suggest that main competition at present shading structure appears light and thus light intensity sufficient to decrease or eliminate light competition is the key factor to increase yield.

  • Ginseng is cultivated in Korea. Japan. China. the Soviet Union and North America. Studies of the macroclimate of each of these producing areas shows that ginseng has certain requirements for production. In each producing area the microclimate is modified in different ways. Comparisons of recent research data from North America. Korea and China is presented in order to define. more precisely. the various microclimate requirements for ginseng production. These include studies of light interception as influenced by different shade materials. In North America. wood. woven black polypropylene and knitted polyethylene shade are used. whereas in China. dried grasses are bound together in layers with wire and polyvinylchloride is inserted between the layers. The influence of these various shade materials in terms of crop grow1h and root yield are presented. The major effect of temperature seems to be on root growth. During much of the growing season optimum temperatures for root grow1h are not reached. Growth analysis data for different age plants are being used to show the effects of different soil temperature regimes on distribution of dry matter between the shoot and root.

Functional characterization of gibberellin signaling-related genes in Panax ginseng

  • Gibberellins (GAs) are essential phytohormones for plant growth that influence developmental processes and crop yields. Recent functional genomic analyses of model plants have yielded good characterizations of the canonical GA signaling pathways and related genes. Although Panax ginseng has long been considered to have economic and medicinal importance, functional genomic studies of the GA signaling pathways in this crucial perennial herb plant have been rarely conducted. Here, we identified and performed functional analysis of the GA signaling-related genes, including PgGID1s, PgSLY1s, and PgRGAs. We confirmed that the physiological role of GA signaling components in P. ginseng was evolutionarily conserved. In addition, the important functional domains and amino acid residues for protein interactions among active GA, GID1, SCFSLY1, and RGA were also functionally conserved. Prediction and comparison of crystallographic structural similarities between PgGID1s and AtGID1a supported their function as GA receptors. Moreover, the subcellular localization and GA-dependent promotion of DELLA degradation in P. ginseng was similar to the canonical GA signaling pathways in other plants. Finally, we found that overexpression of PgRGA2 and PgSLY1-1 was sufficient to complement the GA-related phenotypes of atgid1a/c double- and rga quintuple-mutants, respectively. This critical information for these GA signaling genes has the potential to facilitate future genetic engineering and breeding of P. ginseng for increased crop yield and production of useful substances.

Difference of Ginsenoside Contents in Roots Cultivated under Blue and Red Polyethylene Shading Net in Panax ginseng C. A. Meyer (청색과 적색 해가림 재배에 따른 인삼의 진세노사이드 함량 차이)

  • This study was carried out to investigate the effect of light quality on root yield and ginsenoside contents of 4-year-old ginseng by using the fourfold polyethylene shading net with different colors, blue and red color, compared to blue-black (3:1) mixed shading net as control. Control and blue shading net occurred higher root yield, especially, in tap root growth than red one, whereas transmitted quantum in red shading net was higher than those in blue one or control. However, red shading net caused the highest content of total ginsenoside, especially, Rg1 content, as compared to blue and control. We assumed that the increased content of ginsenoside is not caused by light quality such as red, but is due to the increase of relative ratio of ginsenoside in whole root tissue arising from the reduced root growth.

  • This experiment was conducted to determine economic application method of Yakto among 6 application method : amount of Yakto application 3 plots(70 l, 25.5 l, 13.3l/kan), and added side dressing(3 l/kan) each plot. The reduced Yakto application(25.5 l/kan) with side dressing(3 l/kan) showed similar root diameter, fresh weight of roots and yield of good seedlings to standard Yakto application(70 l/kan). And also this application method of Yakto gave similar content of Ca and Mg, but the lower content of N, P, K in roots to standard Yakto application(70 l/kan). These suggested that 25.5 l Yakto application added side dressing(3 l/kan) per kan might be economical application method of Yakto.

Studies on the Extraction of Polyacetylene from Korean Ginseng Using Supercritical Carbon Dioxide (초임계 $CO_2$를 이용한 고려인삼으로부터 Polyacetylene 추출에 관한 연구)

  • Polyacetylenes were extracted from Korean ginseng using supercritical $CO_2$ Yield of extraction of panaxydol and panaxynol was increased as the pressure of supercritical $CO_2$ increased at the range from 200 to 300 bar. The optimal yields of panaxydol and panaxynol was achieved at 65 and $55^{\circ}C$, respectively. Methanol was applied as a modifier. The highest yield of panaxydol and panaxynol were 0.230 and 0.054 mg/g-dry weight at modifier concentration of 10%(w/w), 300 bar, and $65^{\circ}C$. When these results were compared to that of methanol-extraction, the recoveries of panaxydol and panaxynol by supercritical $CO_2$ extraction in SFE were 37.8 and 55.1%, respectively.

Effect of Extraction Temperature and Time on Saponin Composition of Red Ginseng Extract (추출온도(抽出溫度) 및 시간(時間)이 홍삼(紅蔘)엑기스의 사포닌 조성(組成)에 미치는 영향)

  • The saponin yield and its compositional changes of red ginseng extract (RG-EXT) was investigated during extraction at various temperature for 5 times of 8 hours. The higher temperature resulted an increase in solids yield while the total saponin recovered was decreased, particularly at $100^{\circ}C$. A relatively lower thermal stability was found for protopanaxadiol saponin, one of the saponin fractions, than protopanaxatriol saponin. The compositional ratio of saponin at ginsenoside level was little affected by extraction time. The yields data showed that more than 94% of total saponin was recovered by 3$\sim$4 times of 8 hours extraction. Extraction at $80^{\circ}C$ for 4 times of 8 hours were suggested for preparation of RG-EXT from the result of this work.

Effects of Light Intensity and Temperature on Growth and Root Yield of Valeriana fauriei var. dasycarpa HARA (쥐오줌풀의 생육 및 뿌리수량에 미치는 광도와 온도의 영향)

  • This study was conducted to know the effects of light intensity and temperature on growth of V. fauriei plants. Photosynthesis of V. fauriei had highly significant relations to light intensity and temperature in a quadratic regression model, from which the optimum light intensity and temperature for the plant growth were estimated to be 40,000lux and $17.7^{\circ}C.$ Root was produced less by shading at Jinbu where is located in alpine region, but root yield is increased by shading at Umsong where is located in plane region. Roots were produced more in Jinbu than in Umsong. A highly significant quadratic regression was noted between temperature and leaf width or root weight of V. fauriei. It was estimated from the regression equation that the optimum temperature for root growth was $20.3^{\circ}C.$

Studies of the effects and mechanisms of ginsenoside Re and Rk3 on myelosuppression induced by cyclophosphamide

  • Background: Ginsenoside Re (Re) is one of the major components of Panax ginseng Meyer. Ginsenoside $Rk_3$ ($Rk_3$) is a secondary metabolite of Re. The aim of this study was to investigate and compare the effects and underlying mechanisms of Re and $Rk_3$ on cyclophosphamide-induced myelosuppression. Methods: The mice myelosuppression model was established by intraperitoneal (i.p.) injection of cyclophosphamide. Peripheral blood cells, bone marrow nucleated cells, and colony yield of hematopoietic progenitor cells in vitro were counted. The levels of erythropoietin, thrombopoietin, and granulocyte macrophage colony-stimulating factor in plasma were measured by enzyme-linked immunosorbent assay. Bone marrow cell cycle was performed by flow cytometry. The expression of apoptotic protein bcl-2, bax, and caspase-3 was detected by Western blotting. Results: Both Re and $Rk_3$ could improve peripheral blood cells, bone marrow nucleated cell counts, thymus index, and spleen index. Furthermore, they could enhance the yield of colonies cultured in vitro and make the levels of granulocyte macrophage colony-stimulating factor, erythropoietin, and thrombopoietin normal, reduce the ratio of $G_0/G_1$ phase cells, and increase the proliferation index. Finally, Re and $Rk_3$ could upregulate the expression of bcl-2, whereas they could downregulate the expression of bax and caspase-3. Conclusion: Re and $Rk_3$ could improve the hematopoietic function of myelosuppressed mice. The effect of $Rk_3$ was superior to that of Re at any dose. Regulating the levels of cytokines, promoting cells enter the normal cell cycle, regulating the balance of bcl-2/bax, and inhibiting the expression of caspase-3 may be the effects of Re and $Rk_3$ on myelosuppression.
