Correlation Coefficient of Leaf Breakdown with Some Agronomic Characters in Flue- Cured Tobacco (Nicotiana tabacum L.) (황색종 연초(Nicotiana tabacum L.)에서 엽탈락과 농경적 형질과의 상관)

  • This study was conducted to obtain the breeding information for the leaf breakdown, and correlation coefficients between leaf breakdown and some agronomic characters in flue-cured tobacco at Taegu Experiment Station, Korea Ginseng & Tobacco Research Institute. NC 95 and NC 2326 were used as parents. The bulk method procedure was used from the F2 to the F4 generations, 54 plants were selected among F4, and 11 lines were selected among 54 F5 which based on high resistance to bacterial wilt compared with NC2326. The entries were composed of parents and 11 F6 advanced lines, and designed in randomized block with 3 replications. Plant seedlings were transplanted in 16 April. The number of breakdown leaf were investigated at flowering stage, 18 June. There were significant difference between NC 95 and NC 2326 for the number of breakdown leaf, number and weight of ground suckers, days to flower, yield and resistance to bacterial wilt, and similar results were appeared among lines. Estimated heritability in the broad-sense ranged from 59.3% to 87.4% for all characters. Most of genetic correlations were higher than phenotypic coefficients between characters. Number of breakdown leaf was positively correlated with days to flower, the number and weight of ground suckers, while it was negative to yield. One line among 11 had few breakdown leaf, low producted ground suckers and high resistance to bacterial wilt. Above results suggest that the number and weight of ground suckers can be use as a selection criterion for leaf breakdown even under the condition without leaf breakdown.

  • High salt concentrations in the ginseng nursery soil environment of Korea is one of important reducing factors for the stable production of quality ginseng. These studies were accomplished for check the response on germination of ginseng seed, somatic embryogenesis of zygotic embryo, and biosynthesis of ginsenoside from ginseng hairy root against NaCl. Ratio of germination was at the $3\%\;and\;84.5\%$ on the basic media with 0.1M and free of NaCl repectedly, but $0\%$ at the upper of 0.2M NaCl. Somatic embryogenesis from zygotic embryo were the highest when immatured embryo was cultured on free of NaCl concentration, and which was intend to decrease at treatment of NaCl. However, in case of using the matured embryo, treatment of 0.05M NaCl resulted in better embryogenesis than NaCl free media. Red pigment was synthesized from ginseng hairy root cultured on the medium with various NaCl concentration(from 0.04 to 0.08M) and its pigment was analyzed as spectrum of anthocyane by spectrophoto- meter scanning. This cell line biosynthesized lots of crude saponin and total ginsenoside than other cell lines, also had 2 times of panaxadiol than panaxatriol.

Characteristics of A New Flue-cured Tobacco Mutant Line KF 8832-85 (황색종 연초 돌연변이 계통 KF8832-85의 특성)

  • A new flue-cured tobacco mutant line BU 8832-85 was developed at Taegu Experiment Station, Korea Ginseng and Tobacco Research Institute in 1994. KF 8832-85 was resulted from a cross of flue-cured cultivars NC 95$\times$NC 2326, and developed by a pedigree system of breeding ; initial selection was made by plant type and resistance to bacterial wilt(BW) disease(heudomonas solanaceamm) in the F2 generation under the natural field conditions infested with the pathogen. One white flowered plant was occurred by spontaneous mutation in a certain line among the F3 generatioin while the others were pink. Six plants from the seeds by selfing were selected at the field infested with the pathogen among 240 populations with white flowering in the F4, KF 8832-85 was selected based on yield and leaf quality trials among 6 lines in Fs generation. BCF 8832-85 was compared with its Parent for certain agronomic and chemical characteristics at Taegu Experiment Station in 1993 and 1994. The results showed that KF 8832-85 have white flower, the stalk height was approximately that of NC 2326 but averaged about loom taller than NC 95. It produced ground suckers as much as NC 95, and did not breakdown leaf at the same as WC 2326. KF 8832-85 have high resistance to bacterial wilt disease. Yield of KF 8832-85 was 10 and 18% higher then that of NC 2326 and WC 95, respectively. Price per Kg was equal to that of NC 2326. The contents of nicotine and reducing sugar did not differ significantly from NC 95, while total nitrogen was significantly lower than NC 95. Therefore, the new mutant line is genetically stable for agronomic and chemical characteristics and provides a source of bacterial wilt disease resistance for use in breeding resistant flue-cured cultivars. Development of Liquid Phase Product from Red Ginseng and Medicinal Herbs for Alcoholic Beverage (홍삼 및 기능성 소재를 이용한 주류첨가용 액상제제 개발)

  • This study was conducted to investigate the effect of crude saponin of red ginseng and aqueous extracts from several medicinal herbs on the activity of alcohol dehydrogenase(ADH) and aldehyde dehydrogenase(ALDH) in alcohol metabolism. In order to develop a liquid phase product a model product was prepared using functional plants. In case of red ginseng crude saponin, and some medicinal herbs(Puerariae radix, the flower of Puerariae lobata, the fruits of Hovenia dulcis Thunb., Morus alba L.) the relative activity of ADH was significantly increased, which was from 104~114%. The relative activity of ALDH was also significantly increased in the cases of the fruits of Hovenia duicis Thunb., Morus alba L. which was from 102∼106%. To improve of soju flavours by addition of liquid phase product, it is prepared three samples added a given content (0.28%) of chik, ssanghwa and kyungokgo flavour. The total acceptability test showed significant differences among three samples, and it is observed that the sample added ssanghwa flavour get the best grade. As the second acceptability test, the preference of ssanghwa flavoured sample, sample without flavour and a similar product in market was compared. There was no significant difference among three samples.

Ginseng Transformation of Betaine Aldehyde Dehydrogenase Gene Relative Salt Resistant through Somatic Embryogenesis (염류내성관련 유전자 Betaine Aldehyde Dehydrogenase Gene의 인삼 체세포 배발생을 통한 형질전환)

  • Korean ginseng(Panax ginseng C.A. Meyer) is very difficult to obtain stable production of qualified ginseng roots because of variable stresses in soil environments. In transformation of ginseng with betain aldehyde dehydrogenase gene, compounds synthesized for controlling osmotic pressure such as proline, glycine, betaine, polyols and sugar were accumulated in cell for salt resistance in transgenic plants. 2 Agrobactgerium conjugants were acquired with bet A and bet B genes for solt resistant plants. A. tumefaciens MP90/pBetA and A. tumefaciens MP90/pBetB were recombined for increasing the tolerance to salt stress. To confirm the transformation of the binary vector, tobacco plant was transformed, and the transformant can grow on media containing high concentration of kanamycin. To identify NPT 11, BetA and BetB genes of the transformants, the band on the agarose was confirmed by PCR and RT-PCR techniques. The transformants of ginseng with bet A and bet B genes were acquired on the phytohormone free basic MS media containing only antibiotics and 1M mannitol used for selection of transgenic plant, but the transfomation efficiency for BetA and BetB was very low.

Varietal Difference for Cool Temperature and Short-day in Nicotiana tabacum L. (연초 품종간 저온단일 감응성의 차이)

  • Varietal response to cool temperature(18$^{\circ}C$) and short (8hr.)-day treatment were investigated under controlled conditions of Phytotron in the Suwon Agronomy Experiment Station. Five flue-cured and one burley tobacco varieties (Nicotiana tabacum L.) were used, and the length of treatment ranging from 8 to 32 days at 3-day intervals and also includes continuous treatment up to the plants were flowered. The days to flower decreased significantly than the no treatment when the length of treatment was 11 to 14 days for the varieties NC82, Br.21 and NC22NF, and 17 to 26 days for SPG-28, Mc.944 and TC499, respectively. And also the number of leaves decreased significantly than the no treatment when the length of treatment was 8 days for NC22NF, 11days for NC82 and Br.21, 14 days for Mc.944, and 17 days for SPG-28 and TC499, respectively. The maximum decreasing ratio of the leaf number by the cool temperature and short-day treatment were 47.7 to 58.5% for NC82, Br.21 and NC22NF, and 38.9% for Mc.944, 33.4% for SPG-28 and 29.0% for TC499, respectively.

Quality characteristics of muffins added with fresh ginseng and different amounts of Gryllus bimaculatus powder

  • Edible insects, such as the two-spotted cricket (Gryllus bimaculatus, GB), have high nutritional value but are not widely consumed because of their appearance and smell. Consequently, the development of foods containing these insects in less recognizable forms, e.g., flour-like powders, has drawn considerable attention. Herein, we investigated the quality characteristics of muffins prepared from wheat flower supplemented with fresh ginseng (5%) and GB (0, 10, 20, and 40%) powders. GB loading was negatively correlated with muffin volume, height, moisture content, and textural properties (hardness, springiness, cohesiveness, and chewiness) and positively correlated with crude protein content and antioxidant properties. Significant (p<0.05) color differences were observed between samples with different GB loadings. The contents of hexanal and nonanal, which are the major volatiles responsible for off-flavor, increased with increasing GB loading, and the number of volatiles maximized at 40% GB. Sensory preference decreased in the order of 0% GB>10% GB≈ 20% GB>40% GB. Based on these results, a GB loading of 20% offered the best trade-off between attractiveness and nutritional value. Thus, this study promotes the widespread use of GB in the food industry and the development of various edible-insect-based food products.

A Study on the Consumers' Recognition of Regional Cuisine in Kyonggi Province (경기지방 향토 음식의 소비자 인지도 연구)

  • This study focused on the consumers' recognition of regional cuisine in Kyonggi province. Above all, the study was conducted being related to the tourism merchandising strategy based on the purposes of this study such as the establishment of concept of regional cuisine in Kyonggi province, its recognition and interest, and literature study was also conducted with empirical study. The results are as follows: First, to the question about whether they know the regional cuisine in Kyonggi province or not, 44% of respondents answered they don't know it well, which means recognition is very low. Second, to the question about the recognition of regional cuisine in Kyonggi province, Sujebi(a clear soup with wheat flakes in it) and Nengkonggugsu(iced bean noodles) of main dish, Samgyetang (chicken broth with ginseng and other ingredients) and Galbitang(beef-rib soup) of side dish, Moguachunghwachae(honeyed juice mixed with a Chinese quince as a punch) of drinks, and Kaedduck(a pie-shaped cake made of some rough flower) of desserts were highly recognized.

Varietal Difference of Sucker producing Characteristics in Burley Tobacco(Nicotiana tabacum L. var. burley) (버어리종 연초 곁순발생의 품종간 차이)

  • This study was carried out to obtain the basic information about the sucker producing characteristics of certain burley tobacco varieties(Nicotiana tabacum L. var. burley) at Suwon and Chonju Experiment Station, Korea Ginseng & Tobacco Research Institute in 1999. The number and weight of suckers for 12 varieties or breeding lines were investigated three times, 45 days after transplanting for ground sucker, 10 days after topping, and 10 days after maleic hydrazide(MH) applying for upstalk sucker. KB 103 and KB 9416-1 produced less ground suckers and upstalk suckers than other entries did. KB 9210-8 produced less ground suckers and (Male sterile TC 612 x KB 108)F$_1$ also produced less upstalk suckers than other entries did. There was no differences among entries in sucker production at 10 days after MH treatment. Between the weight of total sucker and days to flower revealed the negative correlation, but not significant. The low sucker producing entries described above could be used as a parental line in the breeding program for the low sucker producing variety.

  • Mesophyll protoplasts derived from young leaves of Nicotiana rustica and N. tabacum cv Burley 21 were fused with the aid of polyethylene glycol(PEG). Cytological examination of protoplasts after PEG treatment revealed 12.8 % heterokaryocytes. After 7 weeks culture, the hybrid calli showing greenish white with a compact appearance were selected in contrast to parental type calli tinged with white or green color. The somatic hybrid plants were verified by morphological, biochemical and cyclological analysis. A heterosis effect for plant vigor and height was observed but the shape of leaves and flower characteristics were intermediate between N. tabacum and N. rutstica. The isozyme banding patterns for peroxidase of somatic hybrid lines were compared with the parent species. A number of isozyme bands derived from both parental species were found in the hybrids. Somatic hybrid plants have been successfully backcrossed to the parental N. tabacum particularly with somatic hybrid plants as female parents. These hybrid plants yielded small seeds, only few which were germinable.
