Effect of Lignans Isolated from Schisandra chinensis Baillon on Seed Germination and Seedling Growth in Radish (오미자 (Schisandra chinensis Baillon)로부터 순수분리한 리그난이 무의 종자발아 및 유묘생육에 미치는 영향)

  • The aim of this study was to develop a rapid and cheap bioassay of four lignans (schisandrin, schisandrin C, the gomisin A and gomisin N) isolated from Schizandra chinensis Baill on seed germination and seedling growth of the radish. Its structure was determined by analysis of MS and NMR spectroscopic data. Radish seeds immersed for 1 hr in the solutions of $10^{-5}M$, $10^{-6}M$ and $10^{-7}M$ of schisandrin, schisandrin C, gomisin A, and gomisin N, seed germination was observed with in 60 hr after all of the treatments. Also, the seeds were germinated faster compared to untreated controls. At early germination stage, 48 hr after the treatment of the lignans, the suppression of germination was observed from all treatments; the suppression due to schisandrin and gomisin A was the highest at the concentration of $10^{-6}M$. The level of suppression increased as the concentration increased in the treatment of schisandrin C and gomisin N. Percent germination of seed after 184 hr was increased 90% at all treatments. For the effects of lignan treatment on seedling growth in radish, the growth of hypocotyl was promoted by gomisin A and gomisin N at all concentrations. Root elongation was significantly promoted by schisandrin and gomisin N at $10^{-5}M$ and $10^{-6}M$, respectively. Fresh and dry weights of the seedlings were high at a low concentration of $10^{-7}M$, but significantly reduced by schisandrin C at a high concentration of $10^{-5}M$. The results of the germination activity and seedling growth of the lignans from S. chinensis suggest their potential use as natural growth regulators.

  • This experiment was conducted to obtain the basic information for the increase ofgermination percent by the seed diameter, specific gravity, different grain filling period and stratification in Bupleurum falcatum seeds. The seed size of B.falcatum ranged from 2.0 to 3.9mm in diameter, The large seed showed the higher germinability, The optimum specific gravity of salt selection for seed screening was 1.05 resulting in screening out seeds with high germinability over 70%.The seeds needed grain filling period for 60 days or longer after flowering to keep high germinability The optimum period of stratification treatment to promote seed germination was about 80 days.

Germination and Seedling Growth Affected by Seed Specific Gravity

  • The amount of salt to make seed sorting solution of the specific gravity of 1.13 was reconsidered and determined as 3.8 kg salt in 18 L water, which is lower amount than currently used. Five rice cultivars were examined. Percent germination and seedling emergence were not similar. Seedling emergence rate of Japonica varieties, Nampyungbyeo and Daerypbyeo-1 were 87% and 95% under specific gravity of 1.13, respectively. Seedling emergence rate of Tongil type variety, Dasanbyeo was as high as 67% in specific gravity of 1.06. Seedling emergence rate of waxy rice, Hwasunchalbyeo and Aranghangchalbyeo were examined. Seedling emergence rate was 94% in both cultivars in specific gravity of 1.04. Seedling emergence rate was same in specific gravity of 1.08 which is generally used for selecting seed currently. Early growth (plant height, leaf number, and dry weight) were not significantly different by specific gravity within species. In all cultivars except waxy rice, highest seedling emergence rate was observed in specific gravity of 1.13 which is currently used for selection and decreased as specific gravity is lowed. However, considering total amount of seeds in each group of specific gravity, amount of seed in lower specific gravity group is relatively small and total seedling emergence rate within variety dose not show big difference. However, if seeds with low speicific gravity are produced due to the bad grain filling condition and consequently total seed content of low specific gravity increases, results will be differnt. Reduction in total growth and yield could occur. It will be important to comply with the seed sorting criterion of 1.13 for Japonica, 1.06 for Tongil, and 1.04 for waxy rice variety to ensure the maximum rice growth and yield.

  • Three varieties of Echinochloa crus-galli (L.) Beauv. and five collections different in gross morphology from the E. crus-galli species were obtained in a lowland rice field located in Jukjeol-Ri, Soyang-Myeon, Wanju-Kun, Jeonbug to compare the ecological and physiological characteristics. There were great variations in seed dormancy, relative growth rates, days required to panicle emergence and panicle morphology among the collections. On the basis of the cluster analysis using the different characteristics, the collections were divided into three groups; E. crus-galli var. frumentaceae and its two ecotypcs, E. crus-galli var. echinata and its two ecotypes, and E crus-galli var. crus-galli and its one ecotype. There were differential responses in percent germination and post-germination growth of the collections to butachlor (2-chloro-2', 6'-diethyl-N-butoxymethyl acetanilide).

Effect of Temperature, Soil Water Potential and Osmoconditioning on Germination and Seedling Elongation of Corn and Soybeans (온도ㆍ토양수분 포텐셜 및 삼투처리가 옥수수와 대두의 발아 및 묘신장에 미치는 영향)

  • Germination and seedling elongation of maize (Dekalb XL 72B), and soybeans (Williams) were measured at two temperatures (15 and 35$^{\circ}C$), three soil water potentials (-1.50, -0.5, and -0.05 MPa), and four polyethy-lene glycol 8000 (PEG) levels (0, 20, 30, and 50 percent). Twenty conditioned seeds of each cultivar were treated with 0.2% thiram and planted 2 cm deep in sterilized Mexico silt loam soil which was subsequently compacted to a bulk density of 1.20 g/㎤. Seedling moisture content, dry weight, and length were measured for each treatment combination. Osmoconditioning with PEG showed little effect at high temperature or low soil water potential conditions. Soybeans had higher seedling moisture content than corn and both crops in-creased moisture uptake as soil water potential and temperature increased. Seedling length of corn was longer than soybeans at 35$^{\circ}C$ but shorter at 15$^{\circ}C$. Seedling dry weight of corn decreased at 35$^{\circ}C$ and that of soybeans decreased as soil water potential increased.

Performance of Direct Seeded Paddy Rice in Dry Soil at Different Seeding Depths (건답직파재배에서 부토심에 따른 벼 품종의 생육가 수량)

  • Performance of 3 indica ${\times}$ japonica(Chilseungbyeo, Samgangbyeo, and Gayabyeo), 7 japonica(Yeongsanbyeo, Yeongdeugbyeo, Hwajinbyeo, Palgongbyeo, Seumjinbyeo, Tamjinbyeo, and Milyang 95), and 2 indica(Tebonnet and Lemont) rice varieties was tested at 1, 3, 5, and 7cm seeding depths in a growth chamber and field. In the growth chamber the number of days from seeding to seedling emergence increased as seeding depth increased, while percent germination and emergence and emergence / germination ratio decreased. However, in the field seedling emergence at 1em seeding depth delayed slightly compared to 3cm probably due to lack of soil moisture. The number of seedlings and maximum tillers at 1, 3, and 5cm seeding depths was similar, but it was higher compared to 7cm seeding depth. Yield and yield components were similar among the seeding depths when all the varieties averaged although 1,000-grain weight was slightly higher at 7cm deeding depth compared to others. At 1cm seeding depth the length of mesocotyl of all varieties was similar and ranged 0.1-0.2cm and that of coleptile ranged 0.8-1.1cm. As seeding depth increased up to 7cm Tebonnet which showed the highest percent emergence emerged by elongation of both mesocotyl and coleoptile, while the other varieties emerged largely by elongation of coleoptile.

Changes of Seed Chemical Component during Grain Filling in Job′s Tears (율무의 등숙에 따른 종실 성분함량 변화)

  • The reports on the chemical composition of developing job's tears seeds is very insufficient by this time. The purpose of this study was to obtain the fundamental information on chemical component of the developing job's tears seed, including starch, sugar, oil, protein, fatty acid, Alkali Digestibility Value(ADV), 100-grain weight. Two job's tears varieties in different by maturity types were analyzed for those components from 10 days after heading to maturity, with 5-day intervals. Total sugar content in seeds followed a pattern of linear increase after heading, but it started to decreased around 15 days after heading. On the other hand, starch content continued to increased until maturity and the highest increasing occurred between 15 and 20 days after heading. The protein content of seeds was decreased, while oil content was increased through the ripening process. Starch content showed a positive correlation with 100-grain weight, ADV, oil and germination percent, but it had negative correlation between total sugar and protein contents in ripening process. Maximum starch content, 100 grain weight, and germination percent obtained by quadratic equation were attained 38 to 43 days after heading. So that the physiological maturity of job's tears seeds was considered to be 40 days after heading.

Differences of Germination and Emergence rate among Isogenic Lines of Waxy and Hulless Gene Combinations in Barley (보리의 찰ㆍ메 및 피ㆍ과성 동질유전자 계통내 발아 및 출아율 차이)

  • On the purpose to increase the cultivation stability of the hulless waxy barley which is unstable in the northern region of Korea, the effects of genes of hulled /hulless and waxy /nonwaxy on seedling emergency was studied by using four isogenic lines of hulled -waxy, hulless -waxy, hulled -nonwaxy and hulless -nonwaxy barley. The germination rates of hulless lines lowered by four to six percent compared with those of hulled counterparts with one day's rainfall just before harvest, and were much deteriorated with raining prolonged. The gaps between emergency rates of hulless lines and their counterparts were greater than those of germination rates, and the differences increased as the seeding depths were deepened. The emergency rates of hulless lines were affected in the order of seeding depth, rotation speed of thresher and preharvest rainfall. Waxy endosperm gene did not show any significant effects on the damage of grain during threshing. It seems that the embryos of hulled lines were less damaged than those of hulless ones during threshing due to the protecting effects by husks. When grains were threshed with thresher of 600rpm or 900rpm and dehulled with dehuller of 750rpm, any significant depreciations such as broken seeds and decreased germination rate did not occured in hulled lines. But the grains of hulless lines received some damages with the rotation speeds, especially germination rates decreased seriously when the rotation speed of de huller was high. Prehavest sprout rates of the lines incorporated with the waxy genes were higher than those with nonwaxy genes whereas the genes of hulled and hulless did not give any significant effects.

Effects of Seed Specific Gravity by Germination and Seeding Emergence in Proso millet (Panicum miliaceum L.) (기장의 염수선 종자 정선법에 따른 발아 및 출현율 특성)

  • The experiment was implemented to gain basic data for stable proso millet (Panicum miliaceum L.) production through germination viability and establishment uniformity based on seed refining under seed selection with specific gravity of proso millet seed. Seeds of three proso millet cultivars were carefully selected by adjusting NaCl into 9 levels (1.000, 1.005, 1.010, 1.015, 1.020, 1.025, 1.030, 1.035, 1.04 g·L-1), and their standard germination rate, seeding quality and field emergence rate were investigated. Proso millets showed heavier thousand seed weight as specific gravity high. In standard germination, all of the 3 cultivars of selected seeding showed at least 91% germination rate. With respect to the 1.040 g·L-1 selection by specific gravity, 'Hwanggeum' had 98.9%, 'Ibaekchal', 99.4%, and 'Hwangsilchal', 98.9%, recording the highest. Seeding quality was investigated and it was found that, compared with the non-selected seeds, selected seeds had longer plant heights and averaged 2.0, 2.2 and 1.9 cm, respectably. In terms of leaf length, only 'Hwanggeum' and 'Ibaekchal' showed distinctive differences. No significance was found in stem diameter and SPAD. The seeds selected using specific gravity all had longer or heavier average root length, root weight and stem weight. Compared with non-selected seeds, the specific gravity of 1.040 g·L-1 had longer root lengths of 2.9, 3.0 and 2.9 cm. 1.040 g·L-1 was also found to have the heaviest root weight, and stem weight. The field emergence viability investigation found significance solely in the mean emergence time (MET) by the cultivar. High significance was found in percent emergence (PE) and emergence rate index (ERI) in the specific gravity for the selection of seeds. All of the three cultivars showed higher PE and ERI as specific gravity selection high. The 1.040 g·L-1 specific gravity selection had the highest PE of 90.7, 85.3 and 92.8%. For productivity increase based on improved proso millet germination rate and enhanced establishment uniformity, it seems favorable to employ 1.040 g·L-1 specific gravity selection using salt solution.

Effect of Osmotic and Solid Matrix Priming to Improve Germination and Early Growth of Carrot and Onion Seed (Osmotic Priming 및 Solid Matrix Priming 처리에 의한 당근과 양파종자의 발아증진과 초기생육에 미치는 영향)

  • The objective of this study was carried out to determine which method is effective for improving seed germination in carrot and onion among osmotic or solid matrix priming. Seeds were osmotic primed with -0.5 MPa polyethylene glycol 8000 (PEG) solution and solid matrix primed with Micro Cel E (seed, Micro Cel E, and water mixed with the ratio of 4.0:2.0:8.0 by weight for carrot, and the ratio of 4.0:1.2:5.0 for onion at $20^{\circ}C$ for 4 days). Seeds were rapidly absorbed water after osmotic priming and solid matrix priming treatment. The moisture content of solid matrix primed seeds was lower than that of osmotic primed seeds in carrot and onion. Osmotic priming and solid matrix priming did not influenced on increasing of percent germination, but $T_{50}$ of treated seeds was shorter than those of untreated seeds at four temperature regimes. The effects of osmotic or solid matrix priming in reducing the $T_{50}$ was greater when the seeds were germinated at $15^{\circ}C$ than at $25^{\circ}C$ and $30^{\circ}C$ temperature. Solid matrix primed seeds germinated faster than osmotic primed seeds at all temperature in both carrot and onion. After priming, surface-dried seeds germinated faster than dried-back seeds in carrot and onion. Emergence speed of carrot and onion seeds was faster than untreated seeds by osmotic priming or SMP treatment although the final emergence percentage was not significantly influenced. On the other hand, early growth was not significantly influenced by osmotic priming or SMP treatment of carrot and onion seeds.
