자외선이 산개구리 (Rana dybowskii) embryo의 시원생식세포 수에 미치는 영향 (UV Effect on the Number of Primordial Germ Cells in the Embryo of Rana dybowskii)

  • 산개구리의 germ plasm 및 축결정 세포질에 미치는 자외선의 영향을 수량적, 시간적으로 조사하였다. 산개구리 난의 식물극을 수정 후 60분경에 1,600 $ergs/mm^2$이상의 자외선을 조사하였을때 40%이상의 시원생식세포의 수가 감소하였으나, 그 이하의 양에서는 전혀 영향이 없었다. 그러나 1,600 $ergs/mm^2$부터는 시원생식세포 수의 감소 정도는 자외선의 양에 비례하였다. 또 신경관 형성에 전혀 영향을 미치지 않으면서 시원생식세포의 형성을 완전히 억제시키는 자외선의 양은 4,800 $ergs/mm^2$이었다. 이에 4,800 $ergs/mm^2$의 자외선을 수정 후 일정한 시간간격으로 조사하여 germ plasm에 미치는 자외선의 영향을 조사하였다. 그 결과 자외선이 조사된 시기가 이를수록 시원생식세포수의 감소가 크며, 수정후 60분부터 차츰 감소율이 줄어 들다가 3분열 이후에는 자외선의 효과가 거의 없음을 관찰하였다. 또한 시원생식세포와 축형성에 미치는 자외선의 영향을 시기적으로 비교한 결과 산개구리난의 축결정이 다른 무미양서류에서와 같이 수정에서 제1분열 사이의 $0.7\\sim0.8$시기 이전에 이루어지므로 그 이후에는 자외선의 감응도가 급격히 저하되었다. 그러나 germ plasm의 자외선에 대한 감응도는 훨씬 늦게까지 지속되어 8구기에 이르기까지 그 영향이 나타났다.

Trichoderma koningii의 Protoplast Reversion에 대하여 (Protoplast Reversion of Trichoderma koningii)

  • Protoplasts from Trichoderma koningii were fractioned at varing digestion periods. There were distinct differences between the fractionated protoplasts in CMCase activity and protein content ; namely, protein contents and CMCase activities in the early-protoplasts were higher than the late-produced protoplasts. Reversion frequencies of the early-produced protoplasts. Reversion frequencies of the early-produced protoplasts (0-1hr.) and the late-produced protoplasts (1-2.5 hr.)were 1.25% and 0.56%, respectively. From these results it was assumed that the early-produced protoplasts were more active in physiological metabolism. Meanwhile, in osmotically stabilized liquid medium two distinct reversion patterns of these protoplasts were found. In the first, normal germ tube was developed from the opposite side of the protoplast after the production of an aberrent tube. In the second, reversion occurred through germination of protoplast, but this patterns was uncommon. It is suggested that the the physiological and morphological heterogeneities in protoplast reversion are related to the heterogeneous origins of protoplasts from highly differentiated mycelium.

Antifungal Mechanism and Properties of Antibiotic Substances produced by Bacillus subtilis YB-70 as a Biological Control Agent

  • Antibiotic substances were produced by Bacillus subtilis YB-70, a potential biocontrol agent found to suppress root-rot of eggplant (Solanum melonggena L) caused by Fusarium solani, in a dextrose glutamate medium and isolated by isoelectric precipitation. Partial purification was performed by column chromatography on silica gel with two solvent systems: chloroform-methanol and methanol-chloroform-water as eluting solvents, This active fraction YBS-1 s contained antifungal activity were soluble in ethanol, methanol, and water, but were not soluble in other solvents including acetone, butanol, ethyl ether, dimethylformamide, propanol, and etc. High performance liquid chromatography and thin layer chromatographic separation of YBS-1s showed that they have been composed of three biological active bands that were named YBS-1A, -1B, and -1C. The substances were stable to heat and resistant to protease. YBS-1s were active against a wide range of plant pathogenic fungi but did not inhibit the growth of bacteria and yeasts. They were not only fungicidal but also fungistatic against chlamydospores of F. solani. The $ED_{50}$ values for the chlamydospore germination and the germ-tube growth of F. solani were $O.725\mu\textrm{m}/ml\;and\;O.562\mu\textrm{m}/ml$, respectively. Microscopic observations proved the substances restricted the growth of phytopathogenic fungus F. solani by spore burst followed by dissolving of its germ-tube, and caused abnormal hyphal swelling after application to chlamydospores or growing hyphae. Cultural filtrate of B; subtilis YB-70 also suppressed the development of root-rot of eggplant in pot tests.

Allomyces macrogynus의 유주자와 반응하는 단일클론항체의 준비 (Preparation of the Monoclonal Antibodies against the Zppspores of Allomyces macrogynus)

  • Allomyces macrogynus의 유주자에 대한 단일클론항에를 만들었다. 고정된 유주자를 주사하거나 유주자 단백질 용액을 주사함으로서 생쥐를 면역화 하였으며, 하이브리도마 세포들은 효소면역흡착법을 이용하여 검색하였다. 약 30개의 하이브리도마 클론이 유주자에 대한 항체를 생산하는 것으로 확인 되었으며, 이들중 일부는 단일세포클론으로 분리되었다. 이들이 만들어내는 항체는 정제되어, 간법면역형광법에 의하여 유주자의 표면에 반응하는 항체로 확인되었다. 또한, 하이브리도마 배양상등액을 이용하여 유주자의 성장에 영향을 미칠 수 있는 클론을 조사하여 보았다. 조사한 배양5상등액중 두가지의 하이브리도마에서 얻은 배양상등액이 germ tube 의 성정을 촉진하는 것으로 확인되었다.

노랑느타리버섯의 원형질체(原形質體) 재생(再生) 및 환원(還元)에 관한 연구(硏究) (Protoplast Regeneration and Reversion in Pleurotus cornucopiae)

  • 원형질체(原形質體)를 이용한 버섯의 육종연구(育種硏究)를 하는데 있어서 원형질체(原形質體) 재생(再生) 및 환원(還元)은 필수적인 과정이라고 할 수 있다. 식용(食用)버섯의 일종(一種)인 P. cornucopiae 노랑 느타리버섯의 원형질체(原形質體) 재생(再生)에 적합한 제조건(諸條件)에 대해서 조사한 바를 요약하면 다음과 같다. 원형질체(原形質體) 재생(再生)에 적합한 삼투압 조절제(調節劑)와 농도(濃度)는 0.6M sucrose로 MCM에서 5.21%의 높은 재생률(再生率)을 보였고, agar를 포함하지 않은 0.6M sucrose 삼투압 조절제(調節劑)나 0.6M液 sucrose+MMM 체배지(體培地)만을 overlaying한 경우에 재생률(再生率)이 높았다. 노랑 느타리버섯의 균사(菌絲) 생장(生長)을 촉진시킨 아미노산, 핵산 구성 성분의 종류(種類)와 원형질체(原形質體) 재생률(再生率)을 증가시킨 종류(種類)는 유사하며 큰 차이가 없었으나 비타민에서는 다소 다른 경향을 나타냈다. 재생률(再生率)을 증가시킨 영양원(營養源)들을 혼합했을 때 무첨가 배지 MMM에 비해 아미노산 혼합, 핵산 구성 성분 혼합에서는 재생률(再生率)이 증가했으며 비타민 혼합에서는 오히려 감소하였고 아미노산, 핵산 구성 성분, 비타민 모두가 첨가된 데서 가장 높은 재생률(再生率)을 나타냈다. 고체(固體) 배지(培地)에서 원형질체(原形質體)가 $1{\sim}2$일 배양(培養)되었을 때 광학 현미경하에서 재생(再生) 형태(形態)를 첫째, one or two directgerm tube, 둘째, bud like structure, 세째, yeast-likecell chain, 네째는 한 원형질체(原形質體)에서 direct germ tube와 yeast-like cell chain을 형성하는 형태를 관찰할 수 있었으며 $3{\sim}4$일 배양(培養)때는 균사(菌絲)가 branch를 형성하며 성장하였고 결국 완전한 균사체(菌絲體)로 되어 노랑 느타리 자실체(子實體)를 형성하였다.

Artificial Reestablishment of the Kelp and Red Algal Symbiosis

  • A type of symbiosis was previously described from nature in which the gametophytes of Laminariales were endophytic in filamentous red algae. Here we reconstruct this symbiosis for the first time in laboratory culture using zoospores of the kelp, Undaria pinnatifida, and the red alga, Aglaothamnion oosumiense. Zoospores of U. pinnatifida readily attached to A. oosumiense. In 48 h these spores germinated and the initial germ tube penetrated into the host cell wall leaving only an empty zoospore wall outside the host. Within ten days, four to five-celled endophytic gametophytes were present. Zoospores of Laminaria religiosa which were also inoculated into cultures of A. oosumiense rarely attached to the red alga and never became endophytic. Within ten days the free-living gametophytes of L. religiosa on cover slips became fertile and produced young sporophytes. These observations demonstrate the ability of U. pinnatifida to become endophytic, and show differences in host specificity among kelp species.

Trichoderma koningii의 conidiospore로부터의 원형질체 생성에 관한 전자현미경적 연구 (Electron Microscopic Study of Protoplast Formation from the Conidiospore of Trichoderma koningii)

  • Fine structure of dormant and swollen conidiospore from Trichoderma koningii and the mechanism of protoplasting from the conidiospore were studied by scanning and transmission electron microscopy. The cell wall of dormant conidiospore was two-layered structure which consisted of electron dense outer layer and electron transparent inner layer. After 8.5 hrs incubation. the conidiospore was swollen and the outer layer of cell wall shown unequal thickness and partial breakage. Protoplast was released through the pore which has been formed by the breakage of outer layer and dissolution of newly synthesized cell wall for germ-tube formation. Swollen conidiospore and protoplast in releasing process contained various cell organelles and vacuoles with electron dense materials. The protoplast contained looser cytoplasm and had no cell wall materials outside of plasmamembrane.

Role of Siderophores in Biocontrol of Fusarium solani and Enhanced Growth Response of Bean by Pseudomonas fluorescens GL20

  • Plant growth-promoting Psudomonas fluorescens GL20 was isolated from a ginseng rhizosphere on chrome azurol Sagar. P. fluorescens GL20 produced a large amount of hydoxamate siderophore in an iron-deficient medium. The siderophore showed significantly high specific activity of 20.2 unit. Using an in vitro antifungal test, P. fluorescens GL20 considerably suppressed growth of phytopathogenic fungus Fusarium solani, inhibiting spore germination and germ tube elongation. In pot trials of kidney beans with P. fluorescens GL20, disease incidence was remarkably reduced up to $68{\%}$ compared with that of F. solani alone, and plant growth was also increased nearly 1.6 fold as compared to that of the untreated control, promoting elongation and development of the roots. These results indicate that the plant growth-promoting activity of P. fluorescens GL20 can play an important role in biological control of soil-borne plant disease in a rhizosphere, enhancing the growth of plants.

소 부루세라병 잠복기간과 방역대책;-제주도 소 부루세라병 근절대책을 중심으로- (The latent period and anti-epidemic measure of bovine brucellosis)

  • The 37,000 holstein which had been raised in Jeju island were examined for the purpose of brucellosis detection and so on. We investigated the relationship between the farms showing positive outbreak and the number of examination, incubation period, and the outbreak rate of positive cattle. We endeavored to separate germs from brucellosis positive cattles blood and negative bulls semen against brucellosis serum test and concluded as follows. We intend to offer you institutional supplements and preventions of brucellosis for the future. 1. Outbreak of brucellosis could not lead the outbreak rate of positive cattle to decrease through 1-2 examination per year, For the past 4 years, the outbreak rate of brucellosis positive cattle had a tendency to show a gradual decrease through at least 5 examinations per year. 2. As we examined live-in cattle with positive cattle in mass outbreak farms for 9 months, we found out positive cattle every month. In a grazing land, the group of cattle producd enormous positive cattle in 9-12 months. 3. Annual brucellosis positive outbreak rate was 20-25% among live-in cattle with brucellosis positive cattle, but the rate might be 50-100% depending on extent of pollution. 4. 94's brucellosis examination showed that 200 positive cattle of 71,153 cattle and most of them were live-in cattle with confirmed brucellosis positive cattle. 5. 1 head of 200 positive cattle showed positivity against serum of bulls test and the rate of bulls was about 1%. 6. Brucellosis germ was separated from 2 cattle's semen among 52 negative bulls against brucellosis serum test and there was the one brucellosis positive cattle against tube agglutination of semen test, so the positive rate of brucellosis was 5.8%. 7. Brucellosis germ was not separated from 15 brucellosis positive cattle's blood.

순수 에탄올을 이용한 재발된 좌골부위 압박궤양의 경화요법적 치료 (Treatment of Recurrent Ischial Pressure Sore Using Sclerotherapy with Absolute Ethanol)

  • Purpose: Ischial region is common site of pressure sore as well as greater trochanteric area. In general, ischial pressure sore associated with a large subcutaneous bursa often requires radical surgical treatment. The authors performed sclerotherapy using absolute ethanol which was considered as an alternative in treating recurrent ischial pressure sore. Methods: From may 2005 to February 2006, 11 ischial pressure sore patients were treated sclerotherapy using absolute ethanol. The authors performed sclerotherapy using absolute ethanol in 11 patients in whom the ischial sore has recurred despite of multiple radical surgical treatment. The patients' original disorders were spinal cord injury in 9 patients, cerebral palsy in 1 patient and giant cell tumor in thoracic vertebrae 1 patient. Results: Recurrence of pressure sore was not found in any patient during the follow-up period. The swap of the bursa taken before the surgery was germ cultured and compared with the discharge from an end of the inserted drain tube. The germ cultured results after the surgery were tested negative in all patients. Conclusion: This method involves causing the bursa to become scarred and closing it up by sterilizing, fixing, and denaturing by the pharmacologic effect of absolute ethanol instead of surgical excision of the bursa. We felt that aforementioned treatment modality may be considered as an alternative in treating recurrent ischial pressure sore.
