Morphological, Physiological and Biochemical Responses of Gerbera Cultivars to Heat Stress

  • Heat stress is an agricultural problem for Gerbera jamesonii, and it often causes poor seedling growth, reduced flower yield and undesirable ornamental characteristics of flowers. However, little is known about the effect of heat stress on the morphological, physiological and biochemical characteristics of gerbera plants. Here, the responses of six cultivars of Gerbera jamesonii to heat stress were investigated. Under a 1-d heat treatment at $45^{\circ}C$, the leaves of gerbera cultivars showed yellowing, wilting, drying and death to varying degrees. The heat treatment also resulted in increased electrical conductivity, decreased soluble protein and chlorophyll contents, and the accumulation of malondialdehyde (MDA) and proline in leaves. Moreover, heat tolerance differed among the six tested gerbera cultivars. Our results demonstrated that among the six gerbera cultivars, 'Meihongheixin' is a heat-resistant cultivar, whereas 'Beijixing' is a heat-sensitive one. 'Shijihong' and 'Linglong' are relatively heat-resistant cultivars, and 'Dadifen' and 'Taiyangfengbao' are relatively heat sensitive.

  • 절화용 거베라 'Sunmyo'는 적색의 'Beauty'를 모본, 황색의 중형인 'Tamara'를 부본으로 하여 2001년에 교배하여 선발한 계통으로부터 육성하였다. 2007년부터 3년간 특성검정을 실시하였는데, 'Sunmyo'는 화색이 밝고 선명한 황색의 절화용으로 화형이 안정된 반겹꽃의 중륜화 품종이다. 절화 수량은 본당 70.0본/년으로 대비품종인 '비로'(51.0본/년)에 비해 37.0% 증수 되었으며 절화 수명은 7.5일로 나타났다. 월동 후 생존율은 85.0%이었으며 개화 소요일수는 95일이었다.

A New Gerbera Cultivar "Pink Swan"; High-yielding Cultivar with Pink and Semi-double for Cut-flower

  • A new gerbera variety "Pink Swan" was released by the National Horticultural Research Institute (NHRI) in 2007. A cross was made between "Royal" with orange and single and 'Himalaya' with white and semi-double in 2004. After investigation of the characteristics for 4 years (from 2004 to 2007), it was selected specially for the use of cut-flower. "Pink Swan" is a vigorous medium-sized cultivar with pink (RHS R48D), yellow green center and semi-double. The average flower diameter is 10.6 cm. And inner ray floret and disc diameter is 5.99cm and 1.83cm, respectively. It produces a large yield with continuous supply of flowers and has adequate peduncle height. We believe that "Pink Swan" has a great potential for expanding cultivated area of domestic gerbera cultivar and increasing farmers-income.

국내 거베라 육종계통 형질전환 기초 조건 확립 (Optimization of Genetic Transformation Conditions for Korean Gerbera Lines)

  • 본 연구에서는 거베라 형질전환 기초기술개발을 위한 연구를 수행하였다. 경남원예화훼시험장에서 육성된 국내 거베라 18품종 중, Agrobacferium 감염에 순응적인 품종을 선발하기 위하여 18개의 국내 거베라 품종을 스크린하여 011, 016, GM023 등 12개의 순응형 계통을 스크린 하였으며 효율적인 형질전환을 위한 제균 항생제로는 cefotaxime이 선택되었다. 더 많은 캘러스 유도를 위한 전처리 배지의 호르몬 농도는 BA 2 ppm, Zeatin 2 ppm, IAA 0.2 ppm (2 x media)이 효율적이었고 재분화 배지의 호르몬 농도도 전처리 배지 호르몬의 농도와 동일하게 BA 2 ppm, Zeatin 2 ppm, IAA 0.2 ppm (2 x media)에서 효율적이었으나 GUS 발현에는 다른 차이가 없었다. 고효율 형질전환 체계 확립을 위하여 네 가지 다른 전처리 기간 처리와 두 가지 다른 Agrobacterium tumerfaciens 사용, 두 가지 다른 공배양 기간을 처리한 결과 전처리 하지 않은 엽병과 엽신을 LBA4404 Agrobacterium tumerfaciens을 사용하여 5일 간 공배양 기간을 가졌을 때와 dipping 처리하여 접종한 엽병과 엽신에서 높은 GUS 발현빈도를 나타냈다.

In vitro shoot regeneration and genetic transformation of the gerbera (Gerbera hybrida Hort.) cultivar 'Gold Eye'

  • This research was conducted to improve the cold tolerance of the gerbera cv. Gold Eye by introduction of the Arabidopsis $Ca^{2+}/H^+$ antiporter gene (CAX1) via Agrobacterium-mediated transformation. Prior to genetic transformation, we optimized a combination of plant growth regulators; $1.0mgl^{-1}$ 6-Benzyladenine (BA) and $0.1mgl^{-1}$3-indole-acetic acid (IAA) were found to lead to proper in vitro shoot regeneration from petiole explants. In addition, $50mgl^{-1}$ kanamycin was determined to be the minimal concentration useful for selection of putative transgenic plants. In this study, transgenic gerbera expressing the Arabidopsis $Ca^{2+}/H^+$ antiporter gene (CAX1) were obtained using the optimized concentrations. We expect that introduction of the gene to the cultivar will improve cold tolerance, which will be important in the winter months.

Influence of basal medium formulations and silver nanoparticles on in vitro plant growth in gerbera

  • This study investigated the impact of two distinct MS basal media: one containing FeNaEDTA and the other FeEDDHA, on the growth of five unique gerbera cultivars (Shy Pink, Pink Holic, Breeze, Harmony, Snow Dream). Notably, the response to these media types varied significantly among the cultivars, particularly concerning leaf yellowing and plant growth. 'Shiny Pink' and 'Pink Holic' exhibited leaf yellowing on the FeNaEDTA-containing medium but displayed leaf greening on the FeEDDHA-containing medium. In contrast, 'Snow Dream,' 'Harmony,' and 'Breeze' remained unaffected on both medium types. However, the FeNaEDTA-containing medium promoted higher plant height and petiole length in 'Breeze,' 'Harmony,' and 'Snow Dream' than the FeNaEDTA-containing medium did. A promotive effect of silver nanoparticles (AgNPs) on plant growth and leaf greening was observed in 'Pink Holic,' particularly on the FeNaEDTA-containing medium, while the addition of AgNPs to the FeEDDHA-containing medium negatively affected plant growth. These results highlight the substantial influence of medium type, specifically the presence of FeNaEDTA or FeEDDHA, on gerbera growth responses, emphasizing the critical role of medium selection in gerbera propagation. Additionally, when contemplating the addition of AgNPs for in vitro gerbera propagation, it is crucial to consider the medium type.

화색이 선명한 황색 절화용 거베라 '조이풀' 육성과 유전적 분석 및 절화 수명 특성 (Breeding, Genetic Analysis, and Vase-life of Bright Yellow Gerbera Cultivar 'Joyful' for Cut Flower)

  • 경남농업기술원 화훼연구소에서 '포커스(Focus)'와 '초우(Chowoo)' 품종 간의 교배 후, 특성검정을 통해 선명한 황색의 반겹꽃 절화용 거베라(Gerbera hybrida Hort.) '조이풀(Joyful)'을 육성하였다. 인공교배로부터 얻은 실생계통을 2005년부터 2010년까지 조직배양과 포장재배를 통해 개체 증식과 생육 및 개화특성 검정, 농가 실증, 절화수명 등을 평가하였다. '조이풀'의 화색은 황색(12-A)으로 꽃의 직경이 12.8cm의 대륜화이며, 화경장은 60cm로 길었다. 개화 소요일수가 89일 정도이며, 절화 수량도 49.2개로 대조품종보다 개화가 빠르고 수량도 많았다. 절화수명에 있어서도 12.8일로 비교 품종보다 우수하였으며, 절화 보존 시 꽃크기 변화 등이 적고, 외형 등의 변화가 대조품종보다 우월하여 절화로서 높은 관상가치를 보였다. 또한 농가실증이나 기호도 평가 시 잘 조화된 절화품종으로 평가받았다. RAPD 분석에서 육성 품종이 모본과 부본의 밴드 패턴이 교배품종인 '조이풀'에는 모두 나타나서, 양친간 교배에 의해 육성된 품종임을 확인할 수 있었다. '조이풀'은 2013년에 신품종으로 품종보호등록(품종보호 제4574호)이 되었으며, 절화용 거베라로서 농가소득 창출에 기여할 수 있을 것으로 기대된다.
