Protein Expression Profiling of Infected Murine Macrophage Cells (RAW 264.7) by Bacillus anthracis Spores

  • Current therapeutic strategies far anthrax have had no significant impact on anthrax mortality over the last several decades. This study used a matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) discovery platform to generate protein expression profiles in search of overexpressed proteins in murine macrophage cells (RAW264.7) which infected with Bacillus anthracis spores as potentially novel molecular targets. Two differentially expressed proteins were identified in infected murine macrophage cells as Syndapin and CDC46, respectively. Syndapins are potential links between the cortical actin cytoskeleton and endocytosis. Other two proteins were identified from murine macrophage cells infected with avirulent spores as ITBG-2 (CD18) and HSPA5, respectively. These data demonstrate the feasibility of using a MALDI-TOF platform to generate protein expression profiles and identify potential molecular targets for anthrax therapeutics.

Molecular Cloning and Structural Analysis of the Antibacterial Gene from the Common Cutworm, Spodoptera litura

  • The cDNA clone encoding the antibacterial peptide (SL-1) was isolated from the fat body of the common cutworm, Spodoptera litura, immunized with E. coli K12. The primary structure analysis revealed that its deduced amino acid sequence showed the characteristics of the cecropin family antibacterial peptides and that the amino acid residues highly conserved in the antibacterial peptides from moths and flies were also conserved, implying that SL-1 was a cecropin-like, and especially cecropin B-like, peptide. The predicted secondary structure of the mature SL-1 consists of three domains: (i) an amphiphilic ${\alpha}$-helical domain (Ile-4 to Gly-18); (ii) the hinge region (Gly-23 and Pro-24); and (iii) a hydrophobic domain (Ala-25 to IIe-38).

  • 마이크로어레이는 전체 유전체 수준의 mRNA 발현 여부에 대한 측정이 가능하다는 점에서 분자생물학의 실험 도구로서 가장 강력한 도구 중에 하나로 부각되어 있다. 현재까지 마이크로어래이의 결과로부터 유사한 발현 패턴을 찾기 위한 여러 가지 바이클러스터링 알고리즘들이 개발되어 왔다. 하지만 대다수의 알고리즘들이 최적의 바이클러스터들을 찾기보다는 일정 수준의 가능한 바이클러스터의 결과만을 제시하고 있다. 본 논문에서는 다른 개체집단들과 상호 진화하는 공진화적 학습에 의한 진화연산 기법을 통하여 유전자-조건의 매트릭스로부터 열과 행을 동시에 클러스터링하는 공진화적 바이클러스터링 알고리즘(co-evolutionary biclustering algorithm: CBA)을 제안하고자 한다. CBA는 유전자발현 데이터에서 유전자-조건의 상호의존적인 부성분들로 구성된 최적화 문제에 적합한 계산방식이라고 할 수 있다. 인간 유전자 발현 데이터에 대한 실험 결과. 제시한 알고리즘은 이전의 알고리즘에 비해 발견한 바이클러스터의 패턴 유사도에 있어서 우수한 성능을 보이고 있다.

  • 제초제 저항성 형질전환 목초 생산을 위하여 유전자총(Gene-gun) 및 Agrobacterium 기법을 이용하여 형질전환을 시도한 결과를 요약하면 다음과 같다. 1. 공시재료 : 알팔파 (cv. Vernal, Anker), 오차드그라스 (합성19호) 2. 캘러스의 유도 및 증식 : 알팔파 종자를 SH-3 배지 (SH기본배지, 2,4-D 3mg/L 포함) 그리고 오차드그라스 종자를 MS-5 배지 (MS기본배지, 2,4-D 5mg/L, 2g casein hydrolysate 포함)에 치상하여 캘러스를 유도하였다. 유도된 캘러스는 증식을 위하여 SH-3 및 MS-3 배지 (MS기본배지, 2,4-D 3mg/L, 2g casein hydrolysate 포함)에서 2주 간격으로 subculture 하였다.(중략)

Anti-inflammatory Activities of Chopi (Zanthoxylum piperitum A.P. DC) Essential Oil: Suppression of the Inducible Nitric Oxide Synthase and Cellular Adhesion

  • The aim of this study is to elucidate the anti-inflammatory activities of chopi (Zanthoxylum piperitum A.P. DC.) essential oil. Essential oil (EO) of chopi was extracted by steam distillation method, and its major constituents were limonene and geranyl acetate. Chopi-EO decreased approximately 38% of nitrite production, as compared to the lipopolysaccharde (LPS)-induced nitrite production. However, chopi-EO and its components did not quench nitric oxide (NO) chemically in cellfree system, and markedly inhibited approximately 40.4% of inducible nitric oxide synthase (iNOS) mRNA transcription. In addition, the inhibition of E-selectin gene transcription by chopi-EO caused the suppression of cellular adhesion. These results suggest that chopi-EO may exert potential anti-immunological inflammatory activity.

  • 유전자 발현은 많은 전자조절인자에 의해서 조절된다. 이러한 조절인자들은 각각 DNA 상에 존재하는 특정한 모티프에 결합하여 그 기능을 수행한다. 따라서 DNA 상의 특정한 서열 정보가 유전자 발현과 직접적으로 연관되어 있다고 생각할 수 있다. 본 논문에서는 두 가지 서로 다른 데이터들에 대한 관계를 알아보기 위하여 사용되는 방법인 Kernel CCA를 이용하여 DNA 상의 특정한 모티프와 유전자 발현 사이의 관계를 알아보았다. 이를 이용한 실험 결과, 유전자 발현과 밀접하게 관련되어 있는 모티프들을 발견할 수 있었고, 기존에 중요한 것으로 알려져 있는 모티프가 실제로도 유전자 발현에 밀접한 영향을 미친다는 것을 알 수 있었다.

Expressed sequence tags analysis of immune-relevant genes in rock bream Oplegnathus fasciatus gill stimulated with LPS

  • We constructed a rock bream (Oplegnathus fasciatus) gill cDNA library and a total of 1450 expressed sequence tag (EST) clones were generated. Gene annotation procedures and homology searches of the sequenced ESTs were locally done by BLASTX for amino acid similarity comparisons. Of the 1450 EST clones, 1022 EST clones showed significant homology to previously described genes while 428 ESTs were unidentified, and 259 clones were hypothetical, or unnamed proteins. Encoding 313 different sequences were identified as putative bio-defense genes or genes associated with immune response.

Gene annotation procedures and homology searches of the sequenced ESTs were analyzed using BLASTX. Of the 1533 EST clones, 1165 different ESTs showed significant homology to previously described genes while 368 ESTs were unidentified, hypothetical, or unnamed proteins. Encoding 106 different sequences were identified as putative bio-defense genes or genes associated with immune response.

Cell Cycle Regulated Expression of Subcloned Chicken H3 Histone Genes and Their 5' Flanking Sequences

  • We subcloned two chicken H3 histone genes and transfected them into Rat 3 cell line. One contains 300 bp 5' to its cap site and the other contains 130 bp 5' to its cap site when cloned into plasm ids. Both of them showed 5' phase specific expression of their mRNA about 8 fold higher (during 5' phase) than during Gl phase. This means that only 130 bp 5' to its cap site was enough to confer cell cycle regulated expression of the latter gene. The DNA sequences of their 5' flanking region did not reveal any particular homologies or subtype-specific sequences. The DNA sequence data also showed that even though the protein coding regions of the histone genes have been conserved exceptionally well throughout evolution, their 5' untranslated regions have not been conserved as well.

New record of the family Porcellidiidae Boeck, 1865 (Harpacticoida, Copepoda) in Korea

  • Kushia zosteraphila Harris V.A. & Iwasaki, 1996 is newly collected and described from macroalgae in the intertidal region of Gijang-gun, along the southeastern coastal region of Korea. Kushia zosteraphila can be distinguished from congeners by following morphological characteristics: the length of the first dorsal seta similar with the second dorsal seta of female P5, the length to width ratio of the female caudal ramus, and the presence of a conspicuous comb on the accessory lobe of the male antennule. Although there are some minor discrepancies, the main diagnostic characteristics of the specimen from the study area are well-matched with the original description. We herein provide detailed morphological descriptions and illustrations of this species. According to a survey of the location of the reported porcellidiid species in Korea, this specimen is the second record in Korean waters of the genus Kushia. A key to species of the family Porcellidiidae in Korea is provided. A partial sequence of the mitochondrial COI gene was obtained and provided as a DNA barcode for this species.
