감마선 조사 미역귀(Undaria pinnatifida Sporophyll) 열수추출물을 첨가한 초콜릿의 품질 및 항산화 활성 (Quality Characteristics of Chocolate with Added Hot Water Extracts of Undaria pinnatifida Sporophylls under Gamma Irradiation)

  • 미역귀를 이용한 가공식품개발을 위해 비조사구와 감마선 50 kGy 조사구의 미역귀 열수추출 분말을 첨가(1, 3, 5%)한 초콜릿을 제조하여 품질 특성을 알아보았다. 초콜릿의 수분함량에서는 첨가량이 증가할수록 증가하는 반면, pH는 미역귀 열수추출 분말 첨가에 의해 유의적으로 감소하였다. 색도의 L과 b값은 비조사구 및 조사구에서 첨가량이 증가할 수록 유의적으로 감소하였으나, a값은 증가하는 경향을 보였다. 경도는 첨가량이 증가할수록 유의성을 보이며 높아졌고, 비조사구보다 조사구에서 경도가 높았다. DPPH 라디칼 소거능 및 총 페놀함량에서는 조사 5% 첨가구에서 가장 높은 값을 나타내었으며, 비조사구보다 조사구에서 높은 항산화 활성이 나타났다. 초콜릿의 활성이 결과 냄새와 맛, 달콤함과 전반적인 기호도 측면에서 조사 1% 첨가구가 대조구 보다 높은 점수를 얻은 것으로 보았을 때, 감마선 조사 열수추출 분말 1% 첨가의 비율로 미역귀 초콜릿을 제품개발 함으로써 대중화가 가능할 것으로 사료된다.

나리 생장, 자구 발달 및 화분과 종자발아에 미치는 방사선의 영향 (Effect of Irradiation on Growth, Bulblet Formation, and Germination of Pollen and Seed of Several Lily Cultivars)

  • 나리 돌연변이 육종을 위한 기초자료로서 구근, 종자, 화분의 방사선 감수성을 구명하고자 하였다. 'New Wave'외 6품종의 구근은 감마선에 매우 민감하였다. 무조사구는 개화하였으나 조사구에서는 생육이 크게 지연되었으며 화서는 전혀 형성되지 않았다. 'New Wave'와 'Tiny Dino'는 125Gy 조사구에서 생존은 하였으나 생육은 저조하였다. 'New Wave'와 'Tiny Dino'를 제외한 다른 품종들은 잎의 일부 또는 전부가 고사하였다. 품종별 인편의 수분함량은 72~78%이었다. 인편으로부터 자구 형성은 무조사구에서 100%, 조사된 구에서는 'Siberia' 50Gy 처리구에서만 8.5% 일어났다. 화분의 발아율은 감마선 조사에 의해 일정한 경향을 나타내지 않았다. $F_1$ 'Augusta' 종자는 선량이 높아질수록 생존율은 급격히 떨어졌다. 자구는 무조사구에서만 형성되었으며 $LD_{50}$는 100Gy 미만이었다. 방사선 조사된 기내 $F_1$ 'Augusta'의 자구는 배양방법에 관계없이 구고와 구폭이 선량 증가와 함께 감소하였다. 신초 형성은 무조사구에서 100% 되었으나 다른 처리구에서는 15~16%로 떨어졌다.

폴리프로필렌의 내방사선성에 미치는 산화방지제와 열처리의 영향 (Effects of Antioxidant and Thermal Treatment on the Radiation Resistance of Polypropylene)

  • 의료용 등급의 폴리프로필렌 수지에 감마선을 조사한 후 내방사선성에 대한 산화방지제와 열처리의 효과에 대해 연구하였다. 폴리프로필렌과 여러 종류의 산화방지제를 혼합하고 이를 시이트 형태로 제작하여 질소분위기에서 25 kGy로 감마선 조사하였다. 감마선을 조사한 폴리프로필렌 시이트에 대해 산화방지제 종류 및 함량에 따른 색도 변화와 기계적 강도 변화를 조사하였으며 감마선 조사 후의 열처리 조건에 따른 라디칼 잔존량과 산화지수의 변화를 측정하였다. 페놀계 및 아민계 산화방지제보다는 인계 산화방지제를 폴리프로필렌에 혼합하였을 때 감마선 조사 후 색도 변화를 최소화할 수 있었다. $130\;^{\circ}C$에서 30분 동안의 열처리에 의해 산화의 원인이 되는 자유라디칼을 충분히 제거할 수 있어 열처리하지 않은 폴리프로필렌보다 낮은 산화지수를 얻을 수 있었다.

  • Kim, Jong Hyup., (Div. of Biology, Atomic Energy Research Institute,Korea.;) Studies on the Cellulor Metabolism in Microorganisms as influenced by Gamma-irradiation(III): On the Changes of Free Amino acid Pool and content of Protein in Yeast clls irradiated by .gamma.-ray. 1. The strain of Saccharomyces cerevisiae had been cultured synchronously in aerobic condition and irradiatel by gamma-ray from the source of cobalt-60. Drying in vacuum oven at $90^{\circ}C$ C over 12 hours, then changes of protein content (Kjeldahl) and free amino acid pool have been assayed with use of spectrophotometer. Results obtained were compared with those of unirradiated normal cells. 2. It is proved that amount of protein content in the irradiated cells increases to seven percent more than those of normal cells in the same weight of dried samples. It seems like carbohydrate breakown had been stimulated by irradiation and that relative contents of protein shows higher values than those of normal in the same weight of samples. 3. The amount of free amino acid pool in the irradiated cells shows less value about ten percent than those of normal cells, and rate of decreasing is also weak than those of standard reagent solution of amino acid. We may assume that free amino acid pool would be protected against radiation damage in living cells and more stable than in vitro. 4. The component of free amino acid pool have been assayed on second dimensional paper chromatogram, and the identified amino acids are as follows; aspartic acid, serine, glutamic acid, cystine, lysine, glycine, threonine, histidine, arginine, tyrosine, phenylalanine, valine and leucine. 5. Distributional presence of free amino acids are identical to that of normal cells except arginine, it is cosumable that radiation effect is univerlsal to all amino acid. However it is obvious that there are differences in radiolabilities of amino acids in irradiated cells.

실험적 간 발암모델에서 감마선 조사 쇠고기 섭취가 쥐의 항산화 방어체계에 미치는 효과 (Effects of $\gamma$-Irradiated Beef Feeding on Antioxidative Defense System in Experimental Hepatocarcinogenesis)

  • This study was done to investigate the effect of ${\gamma}$ irradiated beef feeding on antioxidant vitamin levels and defense enzyme activities in diethylnitrosamine(DEN) initiated rats. Weaning Sprague Dawley male rats were fed the diet containing ${\gamma}$ irradiated ground beef at the dose 0, 3, 5 kGy as a 20% of protein source for 8 weeks. One week after feeding, rats were intraperitoneally injected twice with a dose of DEN(50mg/kg BW). As a promoter, 0.05% phenobarbital was fed in drinking water from one week after DEN treatment until the end of experiment. At the end of 8th week, serum level of vitamin C, serum and hepatic levels of retinol and tocopherol were determined. In addition, activities of cytosolic glutathione peroxidase, glutathione reductase, glutathione S transferase, catalase and hepatic superoxide dismutase(SOD) were measured. By ${\gamma}$ irradiation, there was no significant effect on serum and hepatic levels of vitamin C and tocopherol except a significant decreasing effect on hepatic retinol level. There was also no significant effect on the activities of enzymes involved in antioxidative defense system, However, DEN treatment led to a significant increase in activities of glutathione reductase and glutathione S transferase while the activity of glutathione peroxidase was decreased. The activities of hepatic SOD and catalase were not changed by DEN treatment. Overall results indicate that the consumption of low dose of ${\gamma}$ irradiated beef does not affect antioxidative defense system.

감마선 조사가 베타글루칸의 미백활성에 미치는 영향 (Effect of Gamma-Irradiation on the Whitening Activity of β-Glucan)

  • This study evaluated the change in whitening activity of ${\beta}-glucan$ by gamma-irradiation. Tyrosinase inhibition was significantly increased in the samples with 30, 50, 100 kGy irradiated ${\beta}-glucan$. Melanin synthesis of irradiated ${\beta}-glucan$ was measured from B16BL6 melanoma cell line treated with ${\alpha}-melanin$ stimulating hormone. Melanin synthesis was increased in the ${\alpha}-melanin$ stimulating hormone added group. However, it was decreased in the groups of 30, 50 and 100 kGy gamma-irradiated ${\beta}-glucan$ treated with ${\alpha}-melanin$ stimulating hormone. These results indicate that gamma irradiated ${\beta}-glucan$ may elevate the whitening activity. Therefore, gamma-irradiated ${\beta}-glucan$ could be used for nutraceutical foods in cosmetic industry.

Effects of Low Dose Gamma Radiation on the Root Growth of Soybean Cultivars

  • ${\gamma}-Radiation$ at very low doses frequently has a stimulating or hormetic effect on the growth of organism. Effects of low dose ${\gamma}-ray$ irradiation on the root growth of soybean cultivars were investigated and hormetic effects by environmental conditions were compared with the occurrence of increased economic yield, seeds of cultivars were irradiated with the dose of $0.5{\sim}20Gy$ and cultivated in growth chamber controling temperature, humidity, light, greenhouse and field respectively. To understand hormetic effect on root growth of cultivars and the difference of hormetic effect by cultivation environment, harvested root of soybean cultivars were scanned with image file, and root surface area, root length, root average diameter etc. were examined by WinRhizo program. Also, dry weight of cultivars was examined. Root growth and dry weight of soybean cultivars showed apparently hormetic effect at cultivation of growth chamber condition. In field experiment executed for whole life cycle, yields of pea were not different significantly in each ${\gamma}-ray$ irradiated cultivars but weight of one hundred peas increased in whole ${\gamma}-ray$ irradiated cultivars. Increment of yield was assumed to be induced through shortening of maturing stage caused by ${\gamma}-ray$ hormesis in early growth stage.

방사선 조사된 콩류의 선별 검지를 위한 발아특성 확인 (Identification of Germination Properties for the Screening of Gamma-irradiated Beans)

  • 본 연구는 콩류의 방사선 조사 여부를 검지하기 위해서 생물학적 방법인 발아법으로 확인하였다. 콩류는 대두, 강남콩, 그리고 팥으로 0.1, 0.3, 0.5, 0.7, 1.0 kGy의 조사선량으로 방사선 조사하고 5일 동안 뿌리의 길이를 측정하여 방사선 조사에 의한 발아 특성을 살펴보았다. 모든 시료는 5일 동안 지속적인 길이 성장을 하였지만, 방사선 조사에 의해서 길이 성장이 억제되어 발아 3일째 부터는 뿌리길이에 유의적인 차이를 보이지 않았다. 대두와 팥은 발아 2일째, 강낭콩은 발아 3일째 비조사 시료와 조사 시료간에 1 cm 단위의 길이 차이를 보였다. 발아 5일째는 조사선량간에 유의적인 차이를 나타내는 경향이었지만 0.5 kGy 이상 조사 시료간에는 유의적인 길이 차이를 보이지 않았다. 또한 모든 시료에서 4일부터 길이 성장정도가 현저히 낮아지고 발아 5일 동안 비조사 시료의 성장정도가 가장 큰 것으로 나타났다. 콩류의 방사선 조사 여부는 발아 5일 동안 뿌리 길이의 유의적인 길이 성장과 비조사 시료와 조사 시료들의 성장정도를 측정함으로써 방사선 조사 여부의 선별검지가 가능한 것으로 판단되었다.

A Study on the Germanium Radiation Detector Compensated by Gamma-ray Irradiation

  • n형 Ge 결정체를 보상하기 위해 상온에서 $^{60}$Co 감마선으로 647 Mrad 조사하였다. 감마선으로 조사된 Ge 결정체로 Ge(${\gamma}$) 검출기를 제작하였다. Ge(${\gamma}$) 검출기의 검출기 특성은 얇은 Ge(Li) 검출기나 고순도 Ge 검출기와 비슷하였다. Ge(${\gamma}$) 검출기의 온도에 대한 안정도는 상온에서의 저장이 가능함을 보여 주었다.

Quality characterization of gamma-irradiated fresh oyster mushrooms (Pleurotus ostreatus) during low temperature storage

  • Fresh oyster mushrooms (Pleurotus ostreatus) were gamma-irradiated at 0, 1, 2, and 3 kGy. The effects on various quality attributes were determined during storage at $5{\pm}1^{\circ}C$. Color changes were more prominent in the cap region than the stem part. At the start of storage increase of Hunter's L-value (lightness) was observed in the caps of 2 and 3 kGy-irradiated samples. The L-value was higher in the all irradiated samples during storage. The trend was different in the case of stem region, where L-value decreased upon irradiation, but remained high throughout storage. The ${\alpha}$-value declined, whereas the b-value increased following irradiation. Irradiation showed a dose-dependent effect on the firmness, which was clearer during storage, but the samples irradiated at 1 kGy maintained an overall better texture than other irradiated samples. The weight loss was also higher in the all irradiated samples during storage. The samples irradiated at 1 kGy showed good physical appearance without any fungal attack at the end of storage; however color change in cap region was quite apparent. The ultra-structural drastic effect of irradiation was understandable using scanning electron microscopy. E-nose analysis demonstrated a clear change in the volatile profiles of all irradiated samples. Although the effect of irradiation on quality characteristics was quite clear but the all irradiated samples were free from fungal attack that was observed in the case of control sample.
