A Study on Fatty Liver Syndrome in Periparturient Holstein Cows (분만전후 젖소의 지방간증에 관한 연구)

  • This study was carried out to investigate the Preventive and treatment effects of mixed compound of vitamin E and selenium, and ursodeoxycholic acid on the pre-parturient and post-parturient fatty liver of high. yielding dairy cows. Thirty pregnant dairy cows(2 weeks preparturition) were divided into clinical(13cows) and subclinical(17 cows) groups based upon blood chemical values, blood serum protein levels and clinical symptoms. The clinical group was subdivided into treated clinical group(6 cows) and non-treated clinical group(7 cows). The cows(treated clinical group) with fatty liver diagnosed before parturition were dosed with mixed compound of vitamin I(1head) and selenium(10mg/head) intramuscularly once before and after parturition, respectively. These cows(treated clinical group) were also given ursodeoxycholic acid(50g/head) intramuscularly 3 times before parturition. The cows(treated clinical group) with fatty liver diagnosed after parturition were also given the same compounds as dosed to the above cows at same dose rate once or 3 times, respectively after parturition. Blood samples were collected from the cows on 4th, 7th and 2nd day of preparturition and on 1st, 7th, 14th and 20th day of postparturition. Blood chemical values(non-esterified fatty acid : NEFA) concentration, serum aspartate aminotransferase activity, serum ${\gamma}$-glutamyl transpeptidase activity, bilirubin concentration, glucose level) were checked and clinical symptoms of the cows were also observed. 1. The cows in non-treated clinical group showed severe depression, anorexia and diarrhea from prepartum period, and retained palcenta, endometritis, ketosis, abomasal displacement and mastitis after parturition. Cows in treated clinical group showed moderate depression, anorexia, diarrhea during preparturient period, but they were relieved within a week from parturition. Cows in subclinical group showed mild depression, anorexia and diarrhea after parturition, but these symptoms were not observed in prepartum period. 2. NEFA concentration, serum aspartate aminotransferase activity, serum ${\gamma}$-glutamyl transpeptidase activity and bilirubin concentration observed In the cows of treated clinical group were significantly lower than those of the cows non-treated clinical group. The blood glucose concentration of the cows in treated clinical group were higher than those of the cows in non-treated clinical group. These results indicate that the preparturient and postparturient fatty liver of high-yielding dairy cows could be reduced by dosing the animals with appropriate amounts of mixed compound of vitamin I and selenium, and ursodeoxycholic acid at proper times of the preparturition and postparturition of the dairy cows.

The Chemical Composition of Taraxacum officinale Consumed in Korea (한국산 서양민들레(Taraxacum officinale)의 화학적 조성)

  • For centuries, dandelion(Taraxacum officinale) is widely used as a food and a medicinal herb. However, there is inadequate documented information on the chemical composition of T. officinale consumed in korea. This study was attempted to analyze proximate component, amino acid, free sugar, mineral and fatty acid composition in T. officinale to establish the value as an useful food material. Moisture, ash, crude protein and fat contents in leaf are 7.85, 11.35 21.6 and 5.12%, respectively. Moisture and ash contents of root were 7.73 and 4.82%. Crude protein and fat contents of root were 11.8 and 1.73%, respectively. The major free amino acid was asparagine, arginine, serine and proline etc. The total contents of free amino acid in leaf and root were 1476.98 and 2710.06 mg%, respectively. The major free sugar in T. officinale was glucose, fructose, sucrose and maltose. Sucrose was also detected in a large amount. The major fatty acid of T. officinale are palmitic acid, stearic acid, oleic acid, linoeic acid and linolenic acid. The fatty acid compositions of leaf and root were different each other. Linolenic acid(63.6%) was the major fatty acid of the leaf and linoleic acid(56.4%) was the major fatty acid of the root. Most of mineral contents were high in leaf and root. Especially K content was higher in leaf than root.

Cissus quadrangularis Extracts Decreases Body Fat Through Regulation of Fatty acid Synthesis in High-fat Diet-induced Obese Mice

  • The current study investigated the anti-obesity effect of Cissus quadrangularsis extracts (CQR-300) and its molecular action mechanism on obese mice induced high-fat diet (HFD). To induce the obesity, mice were fed a HFD for 6 weeks and then fed HFD only or HFD with CQR-300 at 50 and 200 mg/kg. Then, body weight gain and white adipose tissue weights were measured. We investigated the reduction in body fat and the regulation of fatty acid synthesis was measured by dual energy X-ray absorptiometry and real-time PCR with Western blot, respectively. In vitro study, CQR-300 inhibited pancreatic lipase activity. The CQR-300 treatment was significantly decreased the body weight gain and adipocytes size as well as white adipose tissues weights in HFD-induced obese mice. Furthermore, CQR-300 reduced the body fat and fat mass with regulating of adipose tissue hormones as leptin. Treatment with 50 mg/kg CQR-300 showed effectively lower expression levels of adipogenesis/lipogenesis related genes and proteins such as CCAAT/enhancer binding protein ${\alpha}$ ($C/EBP{\alpha}$), peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$), Sterol regulatory element binding protein-1c (SREBP-1c), and fatty acid synthase (FAS) in white adipose tissue (WAT) as compared with the HFD fed only mice. These results suggest that the CQR-300 has an anti-obesity effect via inhibition of lipase activity, decrease the body fat mass by regulating the adipogenesis and lipogenesis related genes and proteins in epididymal adipose tissue with evaluate body fat reduce in the HFD-induced obese mice.

  • Rapeseed (Brassica napus L.) is one of the most valuable oilseed crop in the world. It is widely used in various industries, such as food, animal feed, energy and chemical industries. In order to improve the industrial requirements for rapeseed, useful agronomic characteristics (higher yields and disease resistance etc.) and modified oil traits (fatty acid composition and fat content) are important in rapeseed. However, Korea has limiting genetic resources of novel traits in rapeseed. In this research, novel rapeseed mutant genotypes by mutation breeding was developed. The mutant lines were generated by the treatment of the seeds of the original cultivar 'Tamra' with 700 Gy of gamma-ray (60Co). Mutants showing varied in flowering time, crude fat content, seed yield and fatty acid content that exhibited stable inheritance of the mutated characteristics from M5 to M7 generations were selected. We investigated genetic variation using SNPs identified from GBS analysis in rapeseed mutant lines derived from the gamma-ray, and interactions between the major agronomic and the oil traits. Significantly associated SNP loci were explored along with candidate genes using SNPs obtained by GBS analysis. As a results of association mapping, a total of 322 SNPs were significantly associated with agronomic traits (155 SNPs) and oil traits (167 SNPs). A total of 70 genes were annotated from agronomic characteristics SNPs; among them 7 genes significantly enriched in developmental process, and a total of 70 genes were annotated from crude fat content and fatty acid compositions SNPs; among them, 11genes were significantly enriched in biosynthetic process. These results could be used for the selection of rapeseed cultivar with enhanced qualities and potential economic benefits.

Extraction of Brown Rice Oil Including Essential Fatty Acid Using Supercritical Fluid Process (초임계유체 공정에 의한 유효지방산이 풍부한 현미유의 추출)

  • Brown rice oil contains palmitic acid, linolenic acid, linoleic acid, oleic acid, stearic acid, tocopherol, squalene, etc. The oil including essential fatty acids was extracted from the domestic brown rice bran using supercritical carbon dioxide(SCC) process, and the extracts were analyzed with GC-MSD. The extraction amount of brown rice oil was dependent upon the operating pressure and temperature, and the fatty acid composition of oil was varied with the reduced density(${\rho}_{\gamma}$) of the SCC. About 70~80% of brown rice oil was extracted in 4 hours. Especially, squalene which was not found in solvent extract phase was identified in supercritical fluid extraction phase only.

Effect of Seaweeds added in preparation of Tofu (두부제조시 해조류 첨가 효과)

  • This study was performed to investigate some quality characteristics of 새려 prepared from soybean milk and various seaweed(Undaria pinnatifida, Laminaria japonica, Porpyra tenera, Enteromorpha sp., Codium sp.) pulps in the ration of 9:1(v:v) with 20% MgCl2. The yields of tofu containing Undaria pinnatifida, Laminaria japonica, or Enteromorpha sp. increased but porphyra tenera, Codium sp. decreased in comparison with tofu prepared from whole soybean milk. The protein content of tofu containing Undaria pinnatifida, Laminaria japonica, Porphyra tenera, or Codium sp. increased but Enteromprpha sp. decreased in comparison with the tofu prepared from whole soybean milk. The content of Ca in Undaria pinnatifida, Porphyra tenera added tofu was higher than that of the tofu prepared from whole soybean milk or other seaweeds added tofu. In sensory evaluation the texture, color, taste of tofu were favored with the addition of sea mustard(Undaria pinnatifida) pulp than that of the tofu prepared from whole soybean milk or tofu prepared other seaweed. Tofu prepared was possible with adding 0.5~1.5% sea mustard to soybean milk but the feasible added amount level was 1% of sea mustard. The yields, protein Ca, and K content of tofu were increased by the more adding amount of sea mustard tan tat of the tofu prepared from whole soybean milk. The hardness values of 1% sea mustard added tofu were decreased than that of the tofu prepared from whole soybean milk ; on the other hand, elasticity, cohensiveness, gumminess and brittleness of tofu with sea mustard increased. The L and a values of tofu were lower and b values were higher with the addition of 1% sea mustard. The content of histidine, tyrosine, leusine, and phenylalanine were decreased but the other amino acid were increased in tofu prepared from 1% sea mustard pulp added to soybean milk. The saturated fatty acid and monoene fatty acid content of tofu were increased and C18:2, C18:3(${\gamma}$), C18:3, C20:5 of polyene fatty acid were slightly decreased in tofu prepared from 1% sea mustard pulp added to soybean milk.

  • The concentrations of tocopherols and compositions of fatty acids during germination and maturation of soybean seeds were determined by HPLC and GC. In germination stages, when the length of seedling axis was about 10 cm, the contents of total tocopherols and lipids were the highest. At the early phase of pod filling on field condition, the concentration of ${\delta}-tocopherol$ in soybean seeds was the highest, but, in further pod filling, the content of ${\delta}-tocopherol$ decreased continuously, and ${\alpha}-$ and ${\gamma}-tocopherol$ increased. Accumulation of oil during pod filling seems to be determined at the beginning. There seem positive correlations between several tocopherol homologues and fatty acids.

Repression of $PPAR{\gamma}$ Activity on Adipogenesis by $17{\beta}$-estradiol in Differentiated 3T3-L1 Cell

  • In our previous report, we showed that $PPAR{\gamma}$ does not influence adipogenesis in females with functioning ovaries, indicating that $PPAR{\gamma}$ activity on adipogenesis is associated with sex-related factors. Among the sex-related factors, estrogen has been recognized as a major factor in inhibiting adiposgenesis in females. Thus, we hypothensized that $17{\beta}$-estradiol (E) inhibits 3T3-L1 cell adipogenesis by preventing $PPAR{\gamma}$ activity. E decreased triglyceirde accumulation in differentiated 3T3-L1 cells compared with control group. E also decreased the expression of $PPAR{\gamma}$ mRNA as well as $PPAR{\gamma}$ dependent adipocyte-specific genes, such as adipocyte fatty acid binding protein and tumor necrosis factor $\alpha$. In addition, E not only decreased luciferase reporter activity by $PPAR{\gamma}$, but also transfection of estrogen receptor $\alpha$ ($ER{\alpha}$) or $ER{\beta}$ led to decreases in $PPAR{\gamma}$ reporter gene activation. Moreover, E-activated ERs significantly decreased the luciferase reporter gene activation induced by $PPAR{\gamma}$ transfection, suggesting that estrogen-activated ERs inhibit $PPAR{\gamma}$-dependent transactivation. Accordingly, our results demonstrate that E inhibits the action of $PPAR{\gamma}$ on adipogenesis through E activated ER, providing evidence that lack of estrogen may potentiate $PPAR{\gamma}$ action on adipogenesis.

groES Expression Related to Antifungal Activity of Streptomyces sp. SAR01 (Streptomyces sp. SAR01 균주에서의 항진균 관련 groES의 발현)

  • To analyse proteins and gene related to antifungal activity, SAR01 strain was isolated from a brown seaweed and identified as Streptomyces sp. by FAME(fatty acid methyl ester) analysis. Antifungal activity deficient mutant(SAR535) of Streptomyces sp. SAR01 was induced by gamma radiation$({60}^Co)$. It was found that 6 specific protein spots appeared only in SAR01 by 2-D electrophoresis analysis. Among them, a protein of 10 kDa had homology of 96% with 10 kD chaperonin cpn 10 (GroES) by Basic Local Alignment Search Tool(BLAST, NCBI) analysis. SAR535 transformants into which groES was transferred by electroporation revealed antifungal activity newly similar with SAR01 It suggested that groES be supposed to be related to the antifungal activity of Streptomyces sp. SAR01.

Effect of Feeding Mugwort (Artemisia capillaris) TMR Fodder on Nutritional Composition of Hanwoo Beef (한우육의 성분조성에 미치는 인진쑥 TMR 사료 급여 영향)

  • TMR feed developed by adding mugwort originating from Jeju Island,was fed to Hanwoo to investigate the effects of feeding mugwort on nutritional composition, as well as the feasibility of producing Hanwoo beef with high quality and functionality. The samples for the experiment consisted of Hanwoo fed fattening cattle TMR feed without mugwort (control), and Hanwoo fed fattening cattle TMR feed with added mugwort (treatment). Feeding mugwort did not lead to a significant difference in the moisture, crude protein, crude fat, crude ash, calorie, cholesterol, and collagen contents. There was no significant difference between the control and treatment groups in mineral components. The control and treatment groups had total mineral amounts of 3,559.20 and 3,998.01 ppm, respectively. The total amount of structural amino acids did not show any significant difference between the control (14.56%) and treatment groups (13.90%). The total amount of free amino acids did not show any significant difference between the control (763.044 ppm) and treatment groups (720.885 ppm). The composition of saturated fatty acids was 42.956% for the control and 40.918% for the treatment group. Further, the composition of unsaturated fatty acids was 57.044% for the control and 59.082% for the treatment group. Myristic acid, myristoleic acid, pentadecenoic acid, palmitic acid, and ${\gamma}$-linolenic acid were increased by feeding mugwort. Arachidic acid was contained the most among the fatty acid of Hanwoo fed feed without mugwort (p<0.05).
