Fungal Elicitor와 기계적 상해에 의한 토마토 HMGR mRNA 증가 (Increase in 3-Hydroxy-3-Methylglutarly Coenzyme A Reductase mRNA Level in Tomato by Fungal Elicitors and Mechanical Wounding)

  • 3-hydroxy-3-methyglutaryl coenzyme A reductase (HMGR)는 phytoalexin을 포함하는 수 많은 isoprenoid화합물의 생합성을 조절하는 효소이다. 토마토의 경우 sesquiterpenoid phytoalexin류가 식물방어를 위한 반응산물로서 축적되는 것이 알려져 있다. Verticil-lium albo-atrum이나 Fusarium oxysporum으로부터 추출한 elicitor를 토마토의 배양세포에 처리하는 경우 처리량의 증가에 따른 2.7kb 크기의 HMGR mRNA의 상당한 유도증가가 토마토의 HMG2 DNA를 이용한 northern hybridization에 의해 관찰되었다. 토마토의 잎, 뿌리, 줄기 등에 기계적 상해를 가하는 경우에서도 HMGR mRNA는 2단계를 걸쳐 증가함이 관찰되었다. HMGR mRNA는 양 실험의 경우 모두 9시간에서 12시간 사이에서 최대발현됨이 관찰되었다.

Thalicrtrum rugosum 세포배양에 의한 berberine 생산에 미치는 여러 가지 elicitor의 영향 (Effects of Various Elicitors on the Production of Berberine in Plant Cell Suspension Cultures of Thalicrtrum rugosum)

  • Thalicrtrum rugosum 식물세포 현탁배양에 의한 berberine 생산에 미치는 여러 가지 종류의 elicitor들의 영향에 관해 연구 하였다. 효모 유래의 elicitor, 15가지의 서로 다른 종류의 무생물 종류 elicitor, 3가지 곰팡이로부터 얻은 elicitor 들을 처리하여 세포의 생장 및 berberine 생산에 미치는 영향을 비교 분석해 본 결과, 이들 elicitor들은 T. rugosum에 의한 berberine 생산을 크게 증대시키지 못함을 확인하였다.

미치광이풀 모상근 배양에서 Tropane Alkaloids 생산성 증진을 위한 최적 생물학적 엘리시터 선발 (Selection of Optimal Biotic Elicitor on Tropane Alkaloid Production of Hairy Roots in Scopolia parviflora Nakai)

  • 미치광이풀의 모상근 뱌양에 의해 의학적으로 중요한 물질인 scopolamine과 hyoscyamine 생산을 증진시키기 위해 3종의 곰팡이와 1종의 효모 유래의 생물학적 elicitor를 supernatant와 homogenate의 형태로 처리하였다. Elicitor 중 Candida albicans는 supernatant와 homogenate 모두 scopolamine 생산성을 증가시켰다. Fusarium solani의 homogenate와 C. albicans의 supernatant 처리로 hyoscyamine 함량이 증가되었다. 그러나 나머지 fungus strain도 대조구에 비해 tropane alkaloids의 생산을 다소 증가시켰다. 본 연구에서는 C. albicans가 tropane alkaloids 생산에 적합한 elicitor임으로 구명되었다. 본 연구 결과는 미치광이풀 모상근 대량배양에 의한 tropane alklaoids 대량생산에 기여할 것이다.

당근의 모상근 배양에서 Fungal Elicitor에 의한 Anthocyanin 생산의 향상 (Improved Anthocyanin Production in Hairy Root Culture of Daucus carota by Fungal Elicitor)

  • In order to improve anthocyanin production, effects of fungal elicitation in hairy root culture were investigated. fungal elicitor prepared from Fusarium moniliforme was the best in enhancement of anthocyanin production among the eight fungal elicitors tested. The optimum treat-ment time and concentration of treated elicitor for anthocyanin production were 12 hours and 3.28 mg carbohydrates per liter medium. Also, fungal elicitor was treated to hairy root culture in flat-bottomed fluidized-bed bioreactor. The anthocyanin production of elicited culture was enhanced 227% than non-treated.

The effects of various biotic elicitors on the accumulation of scopolamine and hyoscyamine by adventitious hairy root cultures of Scopolia parviflora

  • Hyoscyamine and scopolamine are the pharmaceutically valuable anticholinergic drugs. The aim of our study is to increase the contents of both hyoscyamine and scopolamine by means of elicitation. Various biotic elicitors derived from 3 fungi and 1 yeast were inoculated to the hairy root cultures. When homogenate and supernatant of elicitors treated with hairy root cultures, that of yeast elicitor was only increased scopolamine production. However, all of the fungal elicitors did not influenced the productions of scopolamine and hyoscyamine. Our results will contribute to mass production of tropane alkaloids by hairy root cultures of Scopolia parviflora.

Enhancing Production of Terpenoids in Metabolically Engineered Transgenic Spearmint (Mentha spicata L.) by Salt and Fungal Elicitors

  • Forest tree species usually takes for long periods to be harvested and cultivated but spearmints are a good model system for woody plant because of reducing and shortening cultivation time. Spearmints are good model plants (Mentha species) for research about terpenoids production and industrial essential oil manufacture. Isopentenyl pyrophosphate isomerase (Iso) and limonene synthase (Limo) are the key enzymes of terpenoid biosynthesis pathway. Transgenic and wild spearmints (Mentha spicata, MS) were cultured in vitro and assessed for the essential oil contents. The content of essential oil of transgenic spearmint also was enhanced slightly depending on the target terpenoid genes. In an attempt to increase productivity of terpenoids further, salt and fungal elicitation strategy was adopted on transgenic Mentha spicata. The salt (800 mM NaCl) as abiotic and two fungi (Botrytis cinerea and Glomerella cingulata) as biotic were used for elicitors. In the absence of salt stress four terpenoids were detected from the spearmint extracts, all of them being monoterpenes. On the other hand, the transgenic (MSIso) extracts contained eleven terpenoids (10 monoterpenes and 1 phenylpropene) while transgenic (MSLimo) extracts contained seven monoterpenes. After 3 days of fungal infection, the resistance indices further increased to 4.38, 3.89 and 2.04 for wild type, MSIso and MSLimo, respectively. The salt and fungal elicitators proved beneficial towards modifying both the terpenoids profile and improvement in the composition of essential oil. These results have important applications for the large-scale production of essential oils and forest biotechnology with respect to spearmint.

Baicalin Production in Transformed Hairy Root Clones of Scutellaria baicalensis

  • A transformed hairy root clone of Scutellaria baicalensis was established following infection with Agrobacterium rhizogenes ATCC15834. Three root clones of S baicalensis were selected by growth habit and baicalin content. The most active strain-the SR-03 clone-was examined for its growth and baicalin content under various culture conditions. The root growth and baicalin content were maximized in a Schenk and Hildebrandt medium supplemented with 4 and 6% sucrose, respectively. The accumulation of baicalin in transformed hairy roots was enhanced through exposure to various elicitors. Elicitation was attained by the addition of methyl jasmonate, salicylic acid, and various concentrations of fungal cell wall elicitors to the medium. The accumulation of baicalin in the elicited cultures ranged from 10.5 to 18.3 mg/g dry weight of the roots, which was 1.5- to 3-fold the amount attained in controls.

참당귀 현탁세포배양에 의한 면역증강성 다당 생산 (Production of Immunostimulating Polysaccharide in Angelica gigas Nakai SusDension Cell Cultures)

  • 참당귀(Angelica gigas Nakai) 현탁세포의 면역 증강성 다당의 생산을 증진시키기 위하여 초기 당농도와 생장조절제인 2,4-D의 농도를 변화시켰다. 당의 농도를 높일수록 세포의 생장과 ECP의 생산은 각각 1.6배, 1.1배 증가하여 효과적이었다. 2,4-D는 첨가하지 않은 것이 세포의 생장이 대조군에 비해 1.2배 높았지만, ECP의 생산의 경우 2mg/L 2,4-D를 첨가한 경우 대조군에 비해 10% 이상 더 생산되었다. 또한 배양 중 여러 가지의 elicitor를 첨가함으로써 ECP의 생산을 증진시키기 위하여 fungal elicitor들을 처리한 결과 Verticillium dahliae 90 mg/L을 배양 4일째에 처리한 경우 세포의 생장을 다소 저해시킨 반면, ECP 생산량이 1.7배나 증가함을 확인하였다.

Increased lignan biosynthesis in the suspension cultures of Linum album by fungal extracts

  • Linum album accumulates anti-tumor podophyllotoxin (PTOX) and its related lignans, which were originally isolated from an endangered species Podophyllum. In the present study, we examined the effects of five fungal extracts on the production of lignans in L. album cell cultures. Fusarium graminearum extract induced the highest increase of PTOX [$143{\mu}g\;g^{-1}$ dry weight (DW) of the L. album cell culture], while Rhizopus stolonifer extract enhanced the accumulation of lariciresinol up to $364{\mu}g\;g^{-1}$ DW, instead of PTOX. Typical elicitors, such as chitin, chitosan, or methyl jasmonate (MeJA), were shown to be less effective in lignan production in L. album cell cultures. These results verified the advantages of fungal extracts to increase lignan production in L. album cell culture, and suggested potential on-demand metabolic engineering of lignan biosynthesis using differential fungal extracts.

Effect of Phytohormones and Chemical Inhibitors on Pathogenesis-related Genes Identified by Differential Hybridization in Rice Suspension Culture Cells

  • In order to study disease resistance mechanisms in rice against the rice blast fungus Magnaporthe grisea, we screened fungal elicitor-responsive genes from rice suspension-cultured cells treated with fungal elicitors employing differential hybridization (DH). By DH screening, 31 distinct rice clones were isolated and a majority of them were full-length cDNAs encoding pathogenesisrelated (PR) genes. Sixteen of the 31 genes were upregulated at 4, 8, and 12 h following fungal elicitor treatment. To elucidate the effect of signal molecules and biotic elicitors on the regulation of rice defense genes, we further characterized the transcriptional expression patterns of representative isolated PR genes; OsGlu1, OsGlu2, OsTLP, OsRLK, and OsPR-10, following treatment with fungal elicitor, phytohormones, cycloheximide, and inhibitors of protein phosphorylation. Jasmonic acid (JA) induced transcriptional expression of OsGlu1, OsTLP, and OsRLK, but not of OsGlu2 and OsPR-10 at any of the tested time points. Salicylic acid (SA) and abscisic acid weakly induced the expression of OsTLP and OsRLK. SA showed an antagonistic effect with fungal elicitor and JA. Cycloheximide suppressed all these genes upon elicitor treatment, except for OsGlu2. Staurosporine only induced the expression of OsRLK. Application of calyculin A strongly induced OsRLK expression, but suppressed the expression of OsGlu2. Our study yielded a number of PR genes that play a role in defense mechanisms against the rice blast fungus, as well as contribute towards the elucidation of crosstalk between phytohormones and other modifications during defense signaling.
