Winter Wheat Grain Yield Response to Fungicide Application is Influenced by Cultivar and Rainfall

  • Winter wheat is susceptible to several fungal pathogens throughout the growing season and foliar fungicide application is one of the strategies used in the management of fungal diseases in winter wheat. However, for fungicides to be profitable, weather conditions conducive to fungal disease development should be present. To determine if winter wheat yield response to fungicide application at the flowering growth stage (Feekes 10.5.1) was related to the growing season precipitation, grain yield from fungicide treated plots was compared to non-treated plots for 19 to 30 hard red winter wheat cultivars planted at 8 site years from 2011 through 2015. At all locations, Prothioconazole + Tebuconazole or Tebuconazole alone was applied at flowering timing for the fungicide treated plots. Grain yield response (difference between treated and non-treated) ranged from 66-696 kg/ha across years and locations. Grain yield response had a positive and significant linear relationship with cumulative rainfall in May through June for the mid and top grain yield ranked cultivars ($R^2=54%$, 78%, respectively) indicating that a higher amount of accumulated rainfall in this period increased chances of getting a higher yield response from fungicide application. Cultivars treated with a fungicide had slightly higher protein content (up to 0.5%) compared to non-treated. These results indicate that application of fungicides when there is sufficient moisture in May and June may increase chances of profitability from fungicide application.

Chemical Resistance of Diaporthe citri against Systemic Fungicides on Citrus

  • Citrus melanose, caused by Diaporthe citri, has been one of the serious diseases, and chemical fungicides were used for protection in many citrus orchards of Jeju Island. Establishing a disinfectant resistance management system and reducing pesticide usage would be important for contributing to safe agricultural production. In this study, monitoring of chemical resistance was performed with 40 representative D. citri isolates from many citrus orchards in Jeju Island. Four different fungicides, kresoxim-methyl, benomyl, fluazinam, and prochloraz manganese, with seven different concentrations were tested in vitro by growing the mycelium of the fungal isolates on the artificial medium potato dextrose agar. Among the 40 fungal isolates, 12 isolates were investigated as resistant to kresoxim-methyl which could not inhibit the mycelium growth to more than 50%. Especially isolate NEL21-2 was also resistant against benomyl, whose hyphae grew well even on the highest chemical concentration. However, any chemical resistance of fungal isolates was found against neither fluazinam nor prochloraz manganese. On the other hand, in vivo bio-testing of some resistant isolates was performed against both kresoxim-methyl and benomyl on young citrus leaves. Typical melanose symptoms developed on the citrus leaves pre-treated with both agrochemicals after inoculation with the resistant isolates. However, no or less symptoms were observed when the susceptible isolates were inoculated. Based on these results, it was suggested that some resistant isolates of D. citri occurred against both systemic fungicides, which may be valuable to build a strategy for protecting citrus disease.

  • 세계 양서류 감소의 주요 요인으로 감염성 질병이 활발하게 연구되고 있으며, 야외 병원균 전파의 가능한 근원지로서 양서류 증양식장들이 언급되고 있으나, 국내에서 관련된 연구수행은 매우 미진한 상태이다. 본 연구에서는 인제, 괴산, 공주에 위치한 북방산개구리 증양식장의 내 외부에서 채집된 성체와 유생의 피부와 내부 장기들에서 확인되는 세균과 곰팡이 콜로니 수의 지역 간, 증양식장 내 외부 간 차이 여부를 파악하고, 나아가 유생으로부터 발견되는 세균의 종류를 16s rDNA 서열 비교를 통해 규명하고자 하였다. 연구결과, 성체의 경우 괴산에서 채집된 성체의 피부와 소화관으로부터 표집된 세균 콜로니 수와 피부와 간으로부터 표집된 곰팡이 콜로니 수가 인제에서 확인된 콜로니 수보다 의미 있게 많았으나 증양식장 내 외 간에는 두 지역 모두에서 어느 부위에서도 차이가 없었다. 유생의 경우 공주에서 채집된 유생의 내부로부터 표집된 곰팡이 콜로니 수가 인제에서 확인된 콜로니수보다 많은 경향을 보였다. 증양식장 내 외부의 비교 결과, 피부에서의 박테리아 수는 인제 증양식장 내부, 개체 내부에서의 박테리아 수는 공주 증양식장 외부에서 더 많은 콜로니 수가 확인되었다. 성체의 건강지수가 좋을수록 피부의 박테리아 콜로니 수와 피부와 심장의 곰팡이 콜로니 수가 적었으나, 유생에서는 관련성이 없었다. 유생으로부터 검출된 세균을 분류한 결과, 4개 문 내, 15개 속에 속하는 세균이 검출되었으나 증양식장 간 및 증양식장 내 외부 간 차이는 분명하지 않았다. 우리의 결과는 증양식장의 지역과 증양식장 내 외부 간의 서로 다른 조건들이 양서류에 있는 세균과 곰팡이의 군집크기 차이를 유발하며, 이것이 증양식 북방산개구리의 건강상태에 영향을 미칠 가능성이 있음을 보여준다. 더불어, 우리의 결과는 성공적인 양서류 증식장 운영과 증식장으로부터 질병의 가능한 야외전파를 막기 위하여, 증식장 내 효율적인 질병관리의 필요성이 높음을 시사한다.

Bacillus oryzicola sp. nov., an Endophytic Bacterium Isolated from the Roots of Rice with Antimicrobial, Plant Growth Promoting, and Systemic Resistance Inducing Activities in Rice

  • Biological control of major rice diseases has been attempted in several rice-growing countries in Asia during the last few decades and its application using antagonistic bacteria has proved to be somewhat successful for controlling various fungal diseases in field trials. Two novel endophytic Bacillus species, designated strains YC7007 and $YC7010^T$, with antimicrobial, plant growth-promoting, and systemic resistance-inducing activities were isolated from the roots of rice in paddy fields at Jinju, Korea, and their multifunctional activities were analyzed. Strain YC7007 inhibited mycelial growth of major rice fungal pathogens strongly in vitro. Bacterial blight and panicle blight caused by Xanthomonas oryzae pv. oryzae (KACC 10208) and Burkholderia glumae (KACC 44022), respectively, were also suppressed effectively by drenching a bacterial suspension ($10^7cfu/ml$) of strain YC7007 on the rhizosphere of rice. Additionally, strain YC7007 promoted the growth of rice seedlings with higher germination rates and more tillers than the untreated control. The taxonomic position of the strains was also investigated. Phylogenetic analyses based on 16S rRNA gene sequences indicated that both strains belong to the genus Bacillus, with high similarity to the closely related strains, Bacillus siamensis KACC $15859^T$ (99.67%), Bacillus methylotrophicus KACC $13105^T$ (99.65%), Bacillus amyloliquefaciens subsp. plantarum KACC $17177^T$ (99.60%), and Bacillus tequilensis KACC $15944^T$ (99.45%). The DNA-DNA relatedness value between strain $YC7010^T$ and the most closely related strain, B. siamensis KACC $15859^T$ was $50.4{\pm}3.5%$, but it was $91.5{\pm}11.0%$ between two strains YC7007 and $YC7010^T$, indicating the same species. The major fatty acids of two strains were anteiso-$C_{15:0}$ and iso $C_{15:0}$. Both strains contained MK-7 as a major respiratory quinone system. The G+C contents of the genomic DNA of two strains were 50.5 mol% and 51.2 mol%, respectively. Based on these polyphasic studies, the two strains YC7007 and $YC7010^T$ represent novel species of the genus Bacillus, for which the name Bacillus oryzicola sp. nov. is proposed. The type strain is $YC7010^T$ (= KACC $18228^T$). Taken together, our findings suggest that novel endophytic Bacillus strains can be used for the biological control of rice diseases.

국내 주요 버섯류의 병해 발생과 재배사의 미생물 밀도 조사 (Occurrences of Major Mushroom Diseases and Microbial Densities of Mushroom Cultivation Facilities)

  • The occurrences of the major diseases and the densities of air-born microbes were surveyed in the cultivation facilities for oyster mushroom (Pleurotus ostreatus), king oyster mushroom (Pleurotus eryngii), and enoki mushroom (Flammulina velutipes) in different areas of Korea. Green mold disease was most often developed in oyster mushroom bed cultivation with the disease incidence rate of approximate 10% while the disease incidences from bottle and plastic envelop cultivation were less than 1~2%. In the bed cultivation, the major air-born microbes in the growth room were Aspergillus, Penicillium, Trichoderma, and Curvularia with the total fungal population density of 567~1,297 CFU/$m^3$ . However, only Trichoderma and Penicillium were detected in the growth rooms and innoculation rooms of bottle and plastic envelop cultivation with the densities of 350~700 CFU/$m^3$ and 160~260 CFU/$m^3$, respectively. The bacterial diseases become evident in the growth rooms of bottle and plastic envelop cultivation with the approximate incidence rate of 10%. The identified bacterial species were Brevibacillus levelkil, Rhizobium radiobacter, Brevundimonas vesicularis, Pseudomonas mosselii, Microbacterium testaceum. Sphingomonas panmi, Sphingomonas yabuuchiae, Paracocus dinitrificans, Curtobacterium flaccumfaciens pv. flaccumfaciens and some unidentified bacteria with the densities of 40~6,359 CFU/$m^3$ in the growth rooms and 9 CFU/$m^3$ in the inoculation room. This study indicated that the green mold disease by fungal strains was the major mushroom disease in the bed cultivation and suggested that the contamination of bacteria and fungi together in the growth media could result in severe production loss. The plastic envelope and bottle cultivation were evidenced to be less susceptible to such contaminations.

  • Viral, bacterial and fungal infections can be transmitted via allografts such as bone, skin, cornea and cardiovascular tissues. Allogenic bone grafts have possibility of transmission of hepatitis C, human immunodeficiency virus (HIV-1), human T-Cell leukaemia virus (HTLV), tuberculosis and other bacterias. The tissue bank should have a policy for obtaining information from the patient's medical report as to whether the donor had risk factors for infectious diseases. Over the past several years, improvements in donor screening criteria, such as excluding potential donor with "high risk" for HIV-1 and hepatitis infection, and donor blood testing result in the reduction of transmission of these diseases. During tissue processing, many allografts are exposed to antibiotics, disinfectants and terminal sterilization such as irradiation, which further reduce or remove the risk of transmitting diseases. Because the effectiveness of some tissue grafts such as, fresh frozen osteochondral grafts, depends on cellular viability, not all can be subjected to sterilization and processing steps and, therefore, the risk of transmission of infectious disease remains. This article is review of the transmission of considering infectious disease in allogenic bone transplantation and the processing steps of reducing the risk. The risk of viral transmission in allografts can be reduced in several standards. The most important are donor-screening tests and the removal of blood and soft tissues by processing steps under the aseptic environment. In conclusion, final sterilizations including the irradiation, can be establish the safety of allografts.

  • Eosinophilic lung diseases are heterogeneous disorders characterized by varying degrees of pulmonary parenchyma or blood eosinophilia. Causes of eosinophilic lung diseases range from drug ingestion to parasitic or fungal infection as well as idiopathic. The exact pathogenesis of eosinophilic lung disease remains unknown. Urushiol chicken can frequently cause allergic reactions. Contact dermatitis (both local and systemic) represents the most-common side effect of urushiol chicken ingestion. However, there has been no previous report of lung involvement following urushiol chicken ingestion until now. A 66-year-old male was admitted to our hospital with exertional dyspnea. Serial chest X-ray revealed multiple migrating infiltrations in both lung fields, with eosinophilic infiltration revealed by lung biopsy. The patient had ingested urushiol chicken on two occasions within the 2 weeks immediately prior to disease onset. His symptoms and migrating lung lesions were resolved following administration of oral corticosteroids.

A Case of Endobronchial Aspergilloma Associated with Foreign Body in Immunocompetent Patient without Underlying Lung Disease

  • Aspergillus causes a variety of clinical syndromes in the lung including tracheobronchial aspergillosis, invasive aspergillosis, chronic necrotizing pulmonary aspergillosis, allergic bronchopulmonary aspergillosis, and aspergilloma. Aspergilloma usually results from ingrowths of colonized Aspergillus in damaged bronchial tree, pulmonary cyst or cavities of patients with underlying lung diseases. There are a few reports on endobronchial aspergilloma without underlying pulmonary lesion. We have experienced a case of endobronchial aspergilloma associated with foreign body developed in an immunocompetent patient without underlying lung diseases. A 59-year-old man is being hospitalized with recurring hemoptysis for 5 months. X-ray and computed tomography scans of chest showed a nodular opacity in superior segment of left lower lobe. Fiberoptic bronchoscopy revealed an irregular, mass-like, brownish material which totally obstructed the sub-segmental bronchus and a foreign body in superior segmental bronchus of the lower left lobe. Histopathologic examinations of biopsy specimen revealed fungal hyphae, characteristic of Aspergillus species.

Chemical Fungicides and Bacillus siamensis H30-3 against Fungal and Oomycete Pathogens Causing Soil-Borne Strawberry Diseases

  • Chemical and biological agents were evaluated to inhibit Colletotrichum fructicola, Phytophthora cactorum, and Lasiodiplodia theobromae causing strawberry diseases. Mycelial growths of C. fructicola were gradually arrested by increasing concentrations of fungicides pyraclostrobin and iminoctadine tris (albesilate). P. cactorum and L. theobromae were more sensitive to pyraclostrobin compared to C. fructicola, but iminoctadine tris (albesilate) was not or less effective to limit P. cactorum or L. theobromae, respectively. Bacillus siamensis H30-3 was antagonistic against the three pathogens by diffusible as well as volatile molecules, and evidently reduced aerial mycelial formation of P. cactorum. B. siamensis H30-3 growth was declined by at least 0.025 mg/ml of pyraclostrobin. The two fungicides additively inhibited mycelial growths of C. fructicola, but not of P. cactorum and L. theobromae. B. siamensis H30-3 volatiles led to less growth of C. fructicola than one reduced by the fungicides. Taken together, in vitro antimicrobial activities of the two fungicides together with or without B. siamensis H30-3 volatiles may be cautiously incorporated into integrated management of strawberry diseases dependent on causal pathogens.

Transcriptome Analysis of Early Responsive Genes in Rice during Magnaporthe oryzae Infection

  • Rice blast disease caused by Magnaporthe oryzae is one of the most serious diseases of cultivated rice (Oryza sativa L.) in most rice-growing regions of the world. In order to investigate early response genes in rice, we utilized the transcriptome analysis approach using a 300 K tilling microarray to rice leaves infected with compatible and incompatible M. oryzae strains. Prior to the microarray experiment, total RNA was validated by measuring the differential expression of rice defense-related marker genes (chitinase 2, barwin, PBZ1, and PR-10) by RT-PCR, and phytoalexins (sakuranetin and momilactone A) with HPLC. Microarray analysis revealed that 231 genes were up-regulated (>2 fold change, p < 0.05) in the incompatible interaction compared to the compatible one. Highly expressed genes were functionally characterized into metabolic processes and oxidation-reduction categories. The oxidative stress response was induced in both early and later infection stages. Biotic stress overview from MapMan analysis revealed that the phytohormone ethylene as well as signaling molecules jasmonic acid and salicylic acid is important for defense gene regulation. WRKY and Myb transcription factors were also involved in signal transduction processes. Additionally, receptor-like kinases were more likely associated with the defense response, and their expression patterns were validated by RT-PCR. Our results suggest that candidate genes, including receptor-like protein kinases, may play a key role in disease resistance against M. oryzae attack.
