Endophytic fungi are microorganisms inhabiting living plant tissues without causing apparent harm to the host. They are drawing increasing attention due to their ability to produce various bioactive compounds as well as their effects on host growth and resistance to biotic and abiotic stresses. As a first step to assess biodiversity of plant associated fungi in Korea and the following evaluation on diverse biological activities, we are collecting endophytic fungi from plant in wild followed by systematic long-term storage in liquid nitrogen. Molecular identification using ITS sequences was also incorporated for pure culture by hyphal tip isolation. As of April 2015, about 1,400 fungal strains had been isolated from about 170 plant taxa. Fungal isolates belonging to Pleosporales, Diaporthales, Glomerellales, Hypocreales, and Xylariales were the most abundant. These collections are being used for several complementary researches, including screening of isolates with novel bioactive compounds or conferring drought stress resistance, phylogenetic and genomic study. Genome sequencing was performed for 3 isolates, one Xylaria sp. strain JS573 producing griseofulvin, an antifungal compound, and two Fusarium spp. strains JS626 and JS1030, which are assumed to be new species found in Korea. More detailed analysis on these genomes will be presented. These collections and genome informations will serve as invaluable resources for identifying novel bioactive materials in addition to expand our knowledge on fungal biodiversity.

This study evaluated the airborne concentrations of fungi in university laboratories, hospital diagnostic laboratories in Seoul. The incubated fungi was identified by lactophenol cotton blue (LPCB) staining method. Variables such as types of ventilation, temperature and relative humidity were investigated to explain laboratory airborne fungal concentrations. A total of 97 air samples were collected from 10 facilities in two institutions. Aspergilus spp., including Aspergilus niger, Aspergillius flavos and Penicillium spp. were found as predominant species. Airborne fungal concentrations ranged from not detected (ND) to 1,890 CFU/$m^3$. Airborne fungal concentrations were high in general-ventilated facilities and in laboratories where relative humidity ( > 60 %) were high ( p < 0.001). Therefore, we suggest that relative humidity should be maintained to properly reduce the concentration of fungal in university and hospital laboratories.

The morphological optimization of Trichoderma harzianum was carried out using several surfactants to achieve increased cellulase production. Addition of the surfactants to the culture medium successfully modified the fungal morphology from an aggregated form to a dispersed form. Optimization of the fungal morphology increased cellulase activity up to 177%. The morphologically optimized conditions enhanced the accessibility of the fungus to substrates and thus promoted cellulase production.
An imperfect fungus Fusiococcum species was isolated from Quercus dentata. A naturally infected Daimyo oak tree was collected and showed elongate wounds on the stem. The fungal cultures were initially white and cottony, and later turned dark gray. Numerous solitary pycnidia were developed on the medium surface, and typically spherical. Yellowish conidial masses were exuded from pycnidia on the culture plates. Conidial masses were swollen and measured as approximately 100 to $300{\mu}m$ in length. It appeared that conidia were usually held together in globose to oval drops. Conidia were hyaline, single-celled (nonseptate), ellipsoid to fusoid, and measured as approximately $8.0{\times}2.7{\mu}m$. Based on these cultural and morphological characteristics, the fungal isolate was identified as a species of Fusicoccum Corda. To preserve and examine fungal spores exuded from pycnidia on the medium surface, a vapor fixation procedure for scanning electron microscopy was employed in this study. The specimens were exposed to the vapor of 2% (v/v) glutaraldehyde and 2% (w/v) osmium tetroxide each for 2 h. With the vapor fixation we obtained excellent retention of conidial masses in this study. The simple and versatile procedure for demonstrating fungal spores and their exudation from fruiting bodies would facilitate characterization of diverse pathological and environmental isolates as they are in native environments.
Mucor sp. KCTC 8405P was cultivated in a jar fermentor for the production of fungal lipid containing ${\gamma}-linolenic$ acid with feeding the glucose solution periodically. The transition of the fungal growth into the mycelial phase from yeast-like growth was achieved by pH shift after the first two day of cultivation in the low pH medium and then lipid accumulation was accelerated until the seven day of cultivation, when the glucose in the culture broth was almost consumed. With the culture conditions applied in this experiment, biomass of 99.3 g/l by the dry cell weight and the total extractable lipid of 38.0 g containing 3.5 g/l ${\gamma}-linolenic$ acid were obtained.
Endophytic fungal strains were isolated from the sterilized roots of a medicinal plant, Ligusticum chuanxiong and identified based on morphological characteristics and molecular analysis of internal transcribed spacer, large subunit rDNA, and beta-tubulin DNA regions. Our results confirmed the presence of Pithomyces chartarum and Plectosphaerella niemeijerarum in the fungal strains. To the best of our knowledge, the fungal strains have not been reported in Korea. In this report, we describe morphological characteristics and phylogenetic trees of these endophytic fungal strains.
During an investigation of fungi of the order Mucorales from freshwater and sediment samples in Korea, we isolated six strains, NNIBRFG6649, NNIBRFG6255, NNIBRFG1498, CNUFC-YJ13, CNUFC-YR7, and NNIBRFG2739. The morphology and phylogeny of these strains were analyzed. Based on the morphological characteristics and molecular data from internal transcribed spacer (ITS) region, the isolates NNIBRFG6649 and NNIBRFG6255 were identified as Mucor abundans, and M. aligarensis, respectively. The isolates NNIBRFG1498 and CNUFC-YJ13 were identified as M. moelleri, whereas the isolates CNUFC-YR7 and NNIBRFG2739 were identified as M. heterogamus. To the best of our knowledge, M. abundans, M. aligarensis, M. moelleri, and M. heterogamus have not yet been reported in Korea.

The aim of this study was to compare the results of morphological phenotypic examination and molecular biological method, randomly amplified polymorphic DNA (RAPD) using clinical isolates from Korean patients infected with dermatophytes. A total of 44 clinical isolates have been collected by Korean Collection Medical Fungi (KCMF) and those were subjected for the RAPD analysis. The results showed that the fungal strains that have RG1 band patterns were identified as Trichophyton rubrum by the neighbour-joining analysis. Additionally, other fungal strains that have RG3 band patterns were identified as Epidermophyton floccosum and RG5 band patterns were identified as Micorsporum gypseum. Even though molecular biological method such as RAPD are not necessary, it might be useful when some strains have similar morphological characteristics or when same species has phenotypic variations. In this study, therefore, we targeted internal transcribed spacer (ITS) regions of fungal rDNA, performed RAPD and then compared with morphological phenotypic examination.
Effects of silver nanoparticles on the phytopathogenic fungal growth were investigated. Fungal phytopathogens, especially for sclerotium-forming species Rhizoctonia solani, Sclerotinia sclerotiorum and S. minor, were selected due to their important roles in survival and disease cycle. Tests for the fungal hyphal growth revealed that silver nanoparticles remarkably inhibit the hyphal growth in a dose-dependent manner. Different antimicrobial efficiency of the silver nanoparticle was observed among the fungi on their hyphal growth in the following order, R. solani > S. sclerotiorum > S. minor. Tests for the sclerotial germination growth revealed that the nanoparticles showed significant inhibition effectiveness. In particular, the sclerotial germination growth of S. sclerotiorum was most effectively inhibited at low concentrations of silver nanoparticles. A microscopic observation revealed that hyphae exposed to silver nanoparticles were severely damaged, resulting in the separation of layers of hyphal wall and collapse of hyphae. This study suggests the possibility to use silver nanoparticles as an alternative to pesticides for scleotium-forming phytopathogenic fungal controls.

In recent years, there has been an intensive research on decolorization of dye and textile wastewater by various fungal strains. In this study, the decolorization ability of three commercial dyes, acid yellow 99, acid blue 350, and acid red 114, were investigated using 10 fungal strains. Among the fungal strains tested, Trametes versicolor KCTC 16781 completely decolorized all dyes in both solid and liquid experiments, and was also able to decolorize the mixture of those three dyes in liquid experiments. The secretion of the ligninolytic enzymes into the extracellular medium during decolorization by T versicolor KCTC 16781 was also studied. No lignin peroxidase activity was detected, and manganese peroxidase and laccase activities were investigated.
