Expression and Characterization of RNA-dependent RNA Polymerase of Dendrolimus punctatus Tetravirus

  • Dendrolimus punctatus tetravirus (DpTV) has been identified as a new member of the genus Omegatetravirus of the family Tetraviridae that may be related serologically to Nudaurelia capensis virus ($N{\omega}V$). To establish the function of DpTV RNA genome and to better understand the mechanism of viral replication, the putative RNA-dependent RNA polymerase (RdRp) domain has been cloned and expressed in Escherichia coli. The recombinant protein was purified on a Ni-chelating HisTrap affinity column and demonstrated to initiate viral RNA synthesis in a primer-independent manner but not by terminal nucleotidyle transferase activity in the presence of $Mg^{2+}$ and RNA template. Mutation of the GDD to GAA interferes with the residues at the polymerase active site and metal ions, and thus renders the polymerase inactive.

회전각이 큰 터빈 동익 누설유동 영역에서의 열(물질)전달 특성 (Heat(Mass) Transfer Characteristics in the Tip-Leakage Flow Region of a High-Turning Turbine Rotor Blade)

  • The heat (mass) transfer characteristics in the tip-leakage flow region of a high-turning first-stage turbine rotor blade has been investigated by employing the naphthalene sublimation technique. The heat transfer data in the tip-leakage flow area for the tip clearance-to-span ratio, h/s, of 2.0% are compared with those in endwall three-dimensional flow region without tip clearance (h/s : 0.0 %). The result shows that the thermal load in the tip-leakage flow region for h/s = 2.0% is more severe than that in the endwall flow region for h/s : 0.0%. The thermal loads even at the leading and trailing edges for h/s = 2.0% are found larger than those for h/s = 0.0%. The tip-leakage flow results in heat transfer augmentations near the tip on both pressure and suction sides in comparison with the mid-span results.

  • In this paper, steering systems for mobile robot with permanent magnet wheels are discussed. The mobile robot with permanent magnet wheels can have three different types of steering and driving configurations; two-wheels, three-wheels, four-wheels. By a Two-WD(Wheel Driving) system, driving and steering characteristics are controlled by ratio of each wheel speeds. Three-WD system is steered by a front wheel and driven by rear wheels. Four-WD system has better stability than two wheel system. Usually the permanent magnet wheel has nearly none slip. Thus turning radius of the mobile robot with three-WD and four-WD System will be increased and the steering and driving system will be complicated. To solve this problem, two magnet wheels with two dummy wheels are used in this study. fuming radius of the developed mobile robot is small and the structure of the robot is simple. It is possible to move forward, backward, to turn left and right, and to rotate freely with two-WD. This study proved that two-WD system is very suitable fur the mobile robot with permanent magnet wheels.

Dual graph-regularized Constrained Nonnegative Matrix Factorization for Image Clustering

  • Nonnegative matrix factorization (NMF) has received considerable attention due to its effectiveness of reducing high dimensional data and importance of producing a parts-based image representation. Most of existing NMF variants attempt to address the assertion that the observed data distribute on a nonlinear low-dimensional manifold. However, recent research results showed that not only the observed data but also the features lie on the low-dimensional manifolds. In addition, a few hard priori label information is available and thus helps to uncover the intrinsic geometrical and discriminative structures of the data space. Motivated by the two aspects above mentioned, we propose a novel algorithm to enhance the effectiveness of image representation, called Dual graph-regularized Constrained Nonnegative Matrix Factorization (DCNMF). The underlying philosophy of the proposed method is that it not only considers the geometric structures of the data manifold and the feature manifold simultaneously, but also mines valuable information from a few known labeled examples. These schemes will improve the performance of image representation and thus enhance the effectiveness of image classification. Extensive experiments on common benchmarks demonstrated that DCNMF has its superiority in image classification compared with state-of-the-art methods.

  • 방사성 낙진에 의한 우리나라 식품의 오염도를 파악할 목적으로 우리나라의 우유중의 스트론튬-90의 양을 측정하였다. 스트론튬의 분석은 발연질산법에 의하였으며, 스트론튬 단위로 결과를 정리하기 위하여 칼슘의 함량도 동시에 정량하였다. 방사능의 측정은 $^{90}$ Sr+$^{90}$ Y의 방사평형이 이루어진 후에 $^{90}$ Y의 방사능을 측정하였다. 액체 우유의 스트론튬-90의 함량이 가장 많았던 것은 1966년의 34.9 pCi$^{90}$ Sr/g.Ca이었으며, 1966년을 고비로 현저한 감소를 보여주고 있다. 분유의 스트론튬-90의 함량은 액체 우유와 같은 값을 보여 주고 있으며, 방사선 장해면에서 고려할때 안전한 함량임을 알 수 있다.

Controlled Conversion of Sodium Metal From Nuclear Systems to Sodium Chloride

  • A series of three bench-scale experiments was performed to investigate the conversion of sodium metal to sodium chloride via reactions with non-metal and metal chlorides. Specifically, batches of molten sodium metal were separately contacted with ammonium chloride and ferrous chloride to form sodium chloride in both cases along with iron in the latter case. Additional ferrous chloride was added to two of the three batches to form low melting point consolidated mixtures of sodium chloride and ferrous chloride, whereas consolidation of a sodium-chloride product was performed in a separate batch. Samples of the products were characterized via X-ray diffraction to identify attendant compounds. The reaction of sodium metal with metered ammonium chloride particulate feeds proceeded without reaction excursions and produced pure colorless sodium chloride. The reaction of sodium metal with ferrous chloride yielded occasional reaction excursions as evidenced by temperature spikes and fuming ferrous chloride, producing a dark salt-metal mixture. This investigation into a method for controlled conversion of sodium metal to sodium chloride is particularly applicable to sodium containing elevated levels of radioactivity-including bond sodium from nuclear fuels-in remote-handled inert-atmosphere environments.

하이퍼골릭 이온성 추진제 연구 개발 동향 (A Review of the Technical Development on Ionic Liquids for Hypergolic Propellants)

  • 1990년대부터 기존 맹독성 이원추진제를 대체하기 위해 수행되어 오던 친환경 혹은 저독성 이원추진제 조합에 관한 연구는 최근 우주 추진기술 개발과정에서 경제성, 안정성, 효율성 등이 강조되며 더욱 주목받고 있다. 연구활동은 주로 저독성 하이퍼골릭 연료를 개발하는 것에 집중되어 왔으며 백연질산 혹은 고농도 과산화수소 산화제로 사용하였다. 저독성 하이퍼골릭 연료의 종류는 촉매성 연료, 반응성 연료, 이온성 연료로 구분할 수 있다. 본 논문에서는 하이퍼골릭 이온성 연료의 연구개발 동향을 소개하고 선행연구에서 보고된 주요 결과들에 대해 분석하고자 한다.

  • 본 연구는 대용량의 공간자료에서 의미 있는 지식을 탐색 및 발견하고, 인터넷 환경에서 공간적 의사결정과정을 지원하기 위한 방안을 설계하였다. 그리고 설계를 기반으로 실험적 원형 시스템(Pilot Tested System)을 구현하여 의사결정자와 분석가 사이에서 특정주제에 적합한 다차원적 분석을 위해 양방향 통신이 가능하도록 공간 데이터 웨어하우스의 눈송이 스키마모델(snowflake schema model)을 활용하였다. 눈송이 스키마는 공간자료와 연계할 경우 기존 스타 스키마모델(star schema model)보다 확장성과 유연성이 뛰어난 분석중심의 설계방안이다. 또한 눈송이 스키마모델의 평가를 위해 백화점 거래사례를 통해 실험적 시스템을 구현하여 평가하였다. 이 시스템의 목적은 목표마케팅 지역설정이며, 의사결정자와 분석가의 양방향통신이 가능한 인터넷상담시스템이다.

  • 환경시료 중 토양에 대한 방사성 스트론튬인 $^{89}Sr$$^{90}Sr$의 분석을 위해 Ca과 Sr을 분리하기 위한 방법으로 Crown ether를 이용한 추출방법을 사용하여 기존의 발연질산법과 비교하여 보았다. Crown ether를 이용한 추출방법을 기존의 발연질산법과 비교하여 보았을 때 높은 Sr 수율을 보였을 뿐만 아니라 분석에 소요되는 시간과 절차가 훨씬 빠르고 간단했다. Crown ether을 이용한 추출방법으로 원전주변 토양에 대한 방사성 스트론튬의 분석을 수행하였으며, 방사성핵종 분석에 의한 환경감시의 효율적인 수행을 위한 스트른튬 분석법을 제시하였다

  • The present study investigates heat/mass transfer and flow characteristics in a ribbed rotating passage with turning region. The duct has an aspect ratio (W/H) of 0.5 and a hydraulic diameter ($D_h$) of 26.67 mm. Rib turbulators are attached in the parallel arrangement on the leading and trailing surfaces of the passage. The ribs have a rectangular cross section of 2 m (e) $\times$ 3 m (w) and an attack angle of $70^{\circ}$. The pitch-to-rib height ratio (p/e) is 7.5, and the rib height-to-hydraulic diameter ratio (e/$D_h$) is 0.075. The rotation number ranges from 0.0 to 0.20 while the Reynolds number is constant at 10,000. To verify the heat/mass transfer augmentation, internal flow structures are calculated for the same conditions using a commercial code FLUENT 6.1. The results show that a pair of vortex cells are generated due to the symmetric geometry of the rib arrangement, and heat/mass transfer is augmented up to $Sh/Sh_0=2.9$ averagely, which is higher than that of the cross-ribbed case presented in the previous study for the stationary case. With the passage rotation, the main flow in the first-pass deflects toward the trailing surface and the heat transfer is enhanced on the trailing surface. In the second-pass, the flow enlarges the vortex cell close to the leading surface, and the small vortex cell on the trailing surface side contracts to disappear as the passage rotates faster. At the highest rotation number ($R_O=0.20$), the turn-induced single vortex cell becomes identical regardless of the rib configuration so that similar local heat/mass transfer distributions are observed in the fuming region for the cross- and parallel-ribbed case.
