Neurotensin Enhances Gastric Motility in Antral Circular Muscle Strip of Guinea-pig

  • Many reports suggest that neurotensin (NT) in the gastrointestinal tract may play a possible role as a neurotransmitter, a circulating hormone, or a modulator of motor activity. NT exerts various actions in the intestine; it produces contractile and relaxant responses in intestinal smooth muscle. This study was designed to investigate the effect of NT on motility of antral circular muscle strips in guinea-pig stomach. To assess the role of $Ca^{2+}$ influx in underlying mechanism, slow waves were simultaneously recorded with spontaneous contractions using conventional intracellular microelectrode technique. At the concentration of $10^{-7}$ M, where NT showed maximum response, NT enhanced the magnitude $(863{\pm}198%,\;mean\;SEM,\;n=13)$ and the frequency $(154{\pm}10.3%,\;n=11)$ of spontaneous contractions. NT evoked a slight hyperpolarization of membrane potential, tall and steep slow waves with abortive spikes $(278{\pm}50%,\;n=4).$ These effects were not affected by atropine $(2\;{\mu}M),$ guanethidine $(2\;{\mu}M)$ and tetrodotoxin (0.2μM). NT-induced contractile responses were abolished in $Ca^{2+}-free$ solution and reduced greatly to near abolition by $10\;{\mu}M$ of verapamil or 0.2 mM of $CdCl_2.$ Verapamil attenuated the effects of NT on frequency and amplitude of the slow waves. Taken together, these results indicate that NT enhances contractility in guinea-pig gastric antral circular muscle and $Ca^{2+}$ influx through the voltage-operated $Ca^{2+}$ channel appears to play an important role in the NT-induced contractile mechanism.

The Spatial Characteristics of Playgrounds Supporting Children's Creativity (창의성 증진을 위한 국내놀이터 공간특성 및 개선방안 연구)

  • As the paradigm which emphasizes nurturing creative elites is introduced, more attention has been paid to children playing and their creativity. This change means that the importance of creativity is emphasized in the education of children, and furthermore, playgrounds which focus on creativity and imagination are needed. Therefore, this study examines the spatial characteristics of playgrounds for children and suggested strategies to improve them. There are five spatial characteristics of a creative playground: aesthetics, being eco-friendly, exploration, challenging and variable. Through the five characteristics, playgrounds were analysed based on ten expressive elements and the results are summarized: Creative playgrounds for children should have an attractive design with sculptural elements which stimulate children to be excited through associable shapes and abstract colors. Second, shelter type structures and a flexible flow design are needed so that children can feel free and be encouraged to explore in diverse ways. Third, as playgrounds for children provide spaces for rest in addition to play, organic facilities and eco-friendly materials for the rest space should to be used. Fourth, as childhood is a period when emotional, cognitive and physical development are achieved, organic geographical features and atypical facilities should be provided so that children can acquire extensive skills and join in physical activities. Finally, creative playgrounds for children should have mobile or variable facilities which consider the characteristics of children who want to do diverse activities and play different games. This study analysed playgrounds for children and suggested strategies to improve them. For successful design and use of creative playgrounds, it is suggested that intensive research on them and consultation with experts should be considered.

Apoptosis and remodeling in adriamycin-induced cardiomyopathy rat model

  • Purpose: The mechanism for the pathogenesis of adriamycin (ADR)-induced cardiomyopathy is not yet known. Different hypotheses include the production of free radicals, an interaction between ADR and nuclear components, and a disruption in cardiac-specific gene expression. Apoptosis has also been proposed as being involved in cardiac dysfunction. The purpose of this study was to determine if apoptosis might play a role in ADR-induced cardiomyopathy. Methods: Male Sprague-Dawley rats were separated into 2 groups: the control group (C group) and the experimental group (ADR 5 mg/wk for 3 weeks through intraperitoneal injections; A group). Echocardiographic images were obtained at week 3. Changes in caspase-3, B-cell leukemia/lymphoma (Bcl)-2, Bcl-2-associated X (Bax), interleukin (IL)-6, tumor necrosis $factor-{\alpha}$, brain natriuretic peptide (BNP), troponin I, collagen 1, and collagen 3 protein expression from the left ventricle tissues of C and A group rats were determined by Western blot. Results: Ascites and heart failure as well as left ventricular hypertrophy were noted in the A group. Ejection fraction and shortening fraction were significantly lower in the A group by echocardiography. The expression of caspase-3, Bax, IL-6, BNP, collagen 1, and collagen 3 were significantly higher in the A group as compared with the C group. Protein expression of Bcl-2 decreased significantly in the A group compared with the C group. Conclusion: ADR induced an upregulation of caspase-3, Bax, IL-6, and collagen, as well as a depression in Bcl-2. Thus, apoptosis and fibrosis may play an important role in ADR-induced cardiomyopathy.

A Sense of Place of Kindergartens as Existential Space in which Children and Teachers Experience (유아와 교사가 경험하는 실존적 공간으로서 유치원의 장소성)

  • The purpose of this study is to explore the meaning of place that is formed by children experiencing the space of kindergarten existentially. For this purpose, the indoor and outdoor spaces of kindergarten, which is a real place where infants experience, are selected as the study observation category. In order to carry out the research, video recording was done focusing on children's free choice activity time and outdoor play through infant observation and non - formal interview. As a result of research, it is found that the place nature of kindergarten as an existential space experienced by infant and teacher as a space that is formed in experience and relation, place as a common production space of play, place as a space where education and life coexist, It was shown that it represents a special space created by experienced kindergarten, and a space characteristic as a space shared by communication. The place of kindergarten has a relationship with the existential experiences of infants, suggesting that teachers need a reinterpretation of space.

  • The future of human life is expected to be innovative by increasing social, economic, political and personal, including all areas of life across the multi-disciplinary skills. Particularly, in the field of robotics and next-generation games with robots, by multidisciplinary contributions and interaction, convergence between technology is expected to accelerate more and more. The purpose of this study is that by new interface model beyond the technical limitations of the "human-robot interface technology," until now and time and spatial constraints and through fusion of various modalities which existing human-robot interface technologies can't have, the research of more reliable and easy free "human-robot interface technology". This is the research of robot game system which develop and utilizing real time synchronization engine linking between biped humanoid robot and the behavior of the position value of mobile device screen's 3D content (contents), robot (virtual robots), the wireless protocol for sending and receiving (Protocol) mutual information and development of a teaching program of "Direct Teaching & Play" by the study for effective teaching.

Effect of the Brotherhood Group Sensory Integration Intervention to Social Interaction of the Children With Pervasive Developmental Disorders (형제그룹 감각통합치료가 전반적 발달장애 아동의 사회적 상호작용에 미치는 효과)

  • Objective : This study aims to examine the Effect of the Brotherhood Group Sensory Integration intervention to Social Interaction of the Children with Pervasive Developmental Disorders. Methods : The Group Sensory Integration intervention was implemented for 8 weeks period from July to September 2011 targeting 2 children of 7~8 ages diagnosed Pervasive Developmental Disorders. For the study A-B-A' design among Single subject research design was used, while baseline A 3 phases, intervention B 16 phases, baseline A' 3 phases in total 22 phases were applied. The Quality of Social Interaction was measured through ESI(Evaluation Social interaction) during the baseline between A and A' period, and the Frequency of Social Interaction was measured under the condition that non-availability of intervention during baseline period, while during intervention period, it was measured by utilizing Interval recording method upon filming by video of 10 minutes free-play after every intervention. Results : The Quality of Social interaction indicated a statistical significance by all targeted children after intervention. Also a statistical significance was indicated in the Frequency of Social interaction as there appeared consecutively more than two of the values of baseline A average and Standard deviation band from all targeted children at the free-play implemented after each intervention of every phase. Conclusion : It could be verified that Brotherhood Group Sensory integration intervention has a positive effect to Social interaction of the Children with Pervasive Developmental Disorders. Therefore, the Brotherhood Group Sensory integration intervention can be effectively applicable as being a therapy to improve Social interaction toward Children with Pervasive Developmental Disorders at the clinic.

The Taste Compounds in Fermented Entrails of Trepang, Stichopus Japonicus (해삼내장(內臟)젓의 맛성분(成分))

  • Fermented trepang entrails, Stichopus Japonicus, is widely used and occupied an important position in foods of this country. But little study on its taste compounds has been reported. This study was attempted to establish the basic data for evaluating taste compounds of fermented trepang entrails. Changes of free amino acids, free sugars, nucleotides and their related compounds as taste compounds during the fermentation of trepang entrails were analyzed by amino acid autoanalyzer and high speed liquid chromatography. Glutamic acid, alanine, glycine and proline were dominant amino acid in the fresh extracts, having 32.3%, 16.4%, 12.0% and 10.5% of the total free amino acid content, respectively. The content of leucine, valine, phenylalanine, isoleucine, methionine and tyrosine were low. The free amino acid were not changed in composition but changed in amounts during the fermentation of trepang entrails. Glutamic acid, alanine, glycine, proline, lysine, arginine and leucine were abundant in both fresh sample and fermented products. Free sugars in fermented trepang entrails, the results showed that galactose(933.7-988.0 mg%) was dominant and the content of arabinose, xylose were 78.7, 55.2-771mg% on moisture and salt free base respectively but glucose was detected in trace amount. Nucleotides and their related compounds were increased during the fermentation and hypoxanthine(47.1-$62.5{\mu}mole/g$, on moisture and salt free base) were dominant, IMP was abundant in fermented trepang entrails. TMA was increased while TMAO was decreased during the fermentation. The content of TMAO nitrogen in fermented trepang entrails was 30.0mg% on moisture and salt free base. The content of betine was increased during the fermentation and was ranged from 734.2 to 934.2mg% on moisture and salt free base. It is believed that such amino acids as glutamic acid, alanine, glycine, lysine, proline, arginine, leucine, such free sugars as galactose, arabinose, xylose, glucose, such nucleotides and their related compounds as IMP, hypoxanthine play an important role as taste compounds in fermented trepang entrails.

ADVANTAGE OF USING FREE NETWORK ADJUSTMENT TECHNIQUE IN THE CRUSTAL MOVEMENT MONITORING GEODETIC NETWORKS

  • There are numerous adjustment techniques that deal with the adjustment of geodetic networks but the least squares adjustment is the most common one. During the network adjustment procedure two techniques can be used, the free network adjustment technique and the constrained network adjustment technique. In order to determine the optimum technique for adjusting the geodetic networks, which used for the geodynamical purposes, data from two different geodetic networks "Sinai geodetic network, Egypt, and HGN network, South Korea" had been examined. The used networks had a different configuration and located in different areas with different seismic activity. The results show that both techniques have a high accuracy and no remarkable differences in terms of RMS. On the contrary, the resulted coordinates shows that the constrained network adjustment technique not only cause a remarkable distortion in the station final coordinates but also if the fixed points that define the datum parameters are changed different solutions for the coordinates will be determined. This distortion affect not only in the determination of point displacement but also in the estimation of the deformation parameters, which play a significant role in the geodynamical interpretation of results. Comparing the results which obtained from both techniques with the widely known geodynamical models of the area reviles that the free network adjustment technique results are clearly match with these models, while those obtained from the constrained technique didn’t match at all. By considering the results it seams to be that the free network adjustment technique is the optimum technique, which can be used for the geodetic network adjustment.

Free-air anomaly from Airborne Gravity Surveying (항공중력측정에 의한 프리에어 이상 산출)

  • The gravity data collected and reserved in Korea is seriously biased in its distribution. That is, only the west-southern part of the peninsula including Chungcheong and Jeonla area has dense distribution while only a part is covered in Gyoungsang area. Especially, the low density of the gravity data in mountainous area basically limits the accuracy of the gravimetric geoid in Korea. As one of the solution to overcome the problem, an airborne gravity survey were conducted from Dec. 2008 $\sim$ Jan. 2009. In this study, free-air gravity anomaly derived from the airborne gravity data which has consistent quality are presented. The data processing for the airborne gravity is composed of several corrections of errors such as errors from gravity measurement, errors from flight dynamics, errors from GPS, and errors from time synchronization. We presented detailed explanations on the data processing with the final cross-over results. The free-air anomaly from airborne gravity finally shows the cross-over accuracy of 2.21mGal which reflects the precision of each track is 1.56mGal. It is expected that the result from this study will play a role as input data in precision geoid determination with ground and ship-borne gravity data after appropriate fusion process.

A Novel Ubiqutin C-terminal Hydrolase (UCH-9) from Chick Skeletal Muscle: Its Purification and Charaterization

  • We have previously shown that chick muscle extracts contained at least 10 different ubiquitin C-terminal hydrolases (UCHs). In the present studies, one of the enzymes, called UCH-9, was purified by conventional chromatographic procedures using $^{125}l$-labeled ubiquitin-${\alpha}$NH-MHISPPEPESEEEEE HYC (Ub-PESTc) as a substrate. The purified enzyme behaved as a 27-kDa protein under both denaturing and nondenaturing conditions, suggesting that it consists of a single polypeptide chain. It was maximally active at pHs between 7 and 8.5, but showed little or no activity at pH below 6 and above 10. Lice other UCHs, its activity was strongly inhibited by sulfhydryl blocking reagents, such as iodoacetamide, and by Ub-aldehyde. In addition to Ub-PESTc, UCH-9 hydrolyzed Ub-aNH-protein extensions, including Ub-${\alpha}NH$-carboxyl extension protein of 80 amino acids and Ubo-${\alpha}NH$-dihydrofolate reductase. However, this enzyme was not capable of generating free Ub from mono-Ub-${\varepsilon}NH$-protein conjugates and from branched poly-Ub chains that are ligated to proteins through ${\varepsilon}NH$-isopeptide bonds. This enzyme neither could hydrolyze poly-His-tagged di-Ub. These results suggest that UCH-9 may play an important role in production of free Ub and ribosomal proteins from their conjugates.
