Chromatographic Fractionation of Protease Inhibitors from Fish Eggs (어류 알로부터 Protease Inhibitors의 크로마토그래피법에 의한 분획)

  • A protease inhibitor from fish eggs was fractionated using chromatographic methods. The fractionation efficiency was evaluated in terms of specific inhibitory activity (SIA, U/mg), purity (fold), total inhibitory activity (TIA, U), and recovery (%). The protease inhibitor (PI) from egg extracts of skipjack tuna (ST Katsuwonus pelamis), yellowfin tuna (YT Thunnus albacares) and Alaska pollock (AP Theragra chalcogramma) was fractionated using Sephadex G-50 gel filtration and DEAE-Sepharose CL-6B anion exchange chromatography based on protein size exclusion and net charge, respectively. Fractions exhibiting strong inhibitory activity were contained in the 30-50 kDa fraction on gel filtration and in the range of 0.4-0.7 M NaCl gradient fraction on anion exchange chromatography. The respective TIA and percent recovery of the fraction obtained with gel filtration toward trypsin and $N{\alpha}$-benzoyl-L-arginine-p-nitroanilide (BAPNA) were 2,758.7 U and 29.6% for ST, 1,005.5 U and 25.6% for YT, and 1,267.5 U and 26.0% for AP. Gel filtration chromatography was more effective at fractionating PI than using ion exchange chromatography. These results suggest that fish eggs act as serine protease inhibitors and might be useful for protease inhibition in foodstuffs.

Protein Profiles of Major Korean Rice Cultivars

  • The protein profiles among Korean rice cultivars were assessed by total protein determination, solubility fractionation, SDS-PAGE analysis and scanning densitometry. In the extraction of protein, the SDS/urea system at a neutral pH was more efficient than that at alkaline pH. The determination of total protein showed that the protein content was similar among cultivars, ranging from 87.9 to 92.7 mg/g dry weight. Additionally, the water/NaCl-soluble protein fraction, containing 14${\sim}$16 kDa albumin and 22 kDa globulin ${\alpha}$-globulin, was also similar among cultivars, with a range of 9.94 to 11.98 mg/g dry weight. The SDS-PAGE/densitometry of total protein showed that there was no discernable difference in proteins of higher molecular weights among various cultivars, whereas the amount of lower molecular weight proteins (14${\sim}$16 kDa) is somewhat variable among cultivars. Furthermore, SDS-PAGE analysis of water/NaCl-soluble and propanol-soluble fractions indicates that there is a discernible change in the content of albumin, globulin or prolamin among cultivars. Thus, the PAGE/densitometry method, preceded by solubility fractionation, is useful for examining differences in protein profiles of rice cultivars.

  • To access the result of radiation therapy for 8 years experiences, 21 patients who were treated with superior vena cava syndrome had been analysed according to dose fractionation and total dose. The results are as follows; 1. In high fractionate dose group, six of eleven patients $(54.5\%)$ exhibited relief of symptoms in 1-2 days, and additional three patients of nine $(81.7\%)$ within 34 days, while standard fractionated dose treatment is not effective to achieve initial relief of symptoms. 2. Graded response by total dose was correlated with total dose rather than dose fractionation. 3. Overall one year survival rate with superior vena cava syndrome was $9.1\%$ and mean survival was 4.2 months.

Applications of Cryogenic Method to Water Vapor Sampling from Ambient Air for Isotopes Analysis (수증기 동위원소 측정을 위한 저온채집법에 대한 연구)

  • Stable water vapor isotopes have been utilized as a tracer for studying atmospheric global circulations, climate change and paleoclimate with ice cores. Recently, since laser spectroscopy has been available, water vapor isotopes can be measured more precisely and continuously. Studies of water vapor isotopes have been conducted over the world, but it is the early stage in south Korea. For vapor isotopes study, a cryogenic sampling device for water vapor isotopes has been developed. The cryogenic sampling device consists of the dewar bottle, filled with extremely low temperature material and impinger connected with a vacuum pump. Impinger stays put in the dewar bottle to change the water vapor which passes through the inside of impinger into the solid phase as ice. The fact that water vapor has not sampled completely leads to isotopic fractionation in the impinger. To minimize the isotopic fractionation during sampling water vapor, we have tested the method using a serial connection with two sets of impinger device in the laboratory. We trapped 98.02% of water vapor in the first trap and the isotopic difference of the trapped water vapor between two impinger were about 20‰ and 6‰ for hydrogen and oxygen, respectively. Considering the amount of water vapor trapped in each impinger, the isotopic differences for hydrogen and oxygen were 0.33‰ and 0.06‰, respectively, which is significantly smaller than the precision of isotopic measurements. This work can conclude that there is no significant fractionation during water vapor trapping.

  • To understand the mass-independent isotopic fractionation effects, thermal decomposition of ozone was performed. Initial oxygen gas was converted to ozone completely. Then, the ozone was decomposed to oxygen at various temperatures ($30~150^{\circ}C$). Isotopic compositions of product oxygen and residual ozone were measured using a stable isotope mass spectrometer. The experimental results were compared with the studies which were performed at the similar conditions. From the raw experimental data, the functions of the instantaneous fractionation factors were calculated by the least square fit. The results clearly showed the temperature dependence. They also showed the pressure dependence and the surface effect. This study may play an important role in the study of ozone decomposition mechanism. It can be applied to explain the mass-independent isotopic pattern found in stratospheric ozone and in meteorites.

Fluorescence-labelling for analysis of protein in starch using asymmetrical flow field-flow fractionation (AF4)

  • Starch is a mixture of amylose (AMY) and amylopectin (AMP) which are different in physical properties such as molar mass (M), rms radius ($R_g$) and hydrodynamic diameter ($d_H$). The rheological and functional properties of starch are influenced by various factors including the molecular size, molar mass distribution (MD) and the concentration ratio of AMY and AMP. It is also important to analyze proteinaceous material in starch as they affect the flavor and texture of food to which starch is added. In this study, asymmetrical flow field-flow fractionation (AF4) was employed for separation and quantitation of AMY and AMP in starches (Amaranth, potato, taros and quinoa). AF4 was coupled with a multi-angle light scattering (MALS) and a refractive index (RI) detector for determination of the absolute M, MD and molecular structure. It was found that AMP has the M and $R_g$ ranging $3.7{\times}10^7{\sim}6.5{\times}10^8g/mol$ and 84 ~ 250 nm, respectively. Also the existence of branch was confirmed in higher M. In addition, proteinaceous material in starch was analyzed by AF4 coupled with a fluorescence detector (FS) after fluorescence-labeling. AF4-FS with fluorescence-labelling showed a potential for investigation on existence of proteinaceous material and the interaction between proteinaceous material and polysaccharide in starch.

Size Determination of Pollens Using Gravitational and Sedimentation Field-Flow Fractionation

  • Pollens are known to be an allergen. They penetrate human respiratory system, triggering a type of seasonal allergic rhinitis called pollen allergy (hey fever). The purpose of this study is to test two field-flow fractionation (FFF) techniques, gravitational FFF (GrFFF) and sedimentation FFF (SdFFF), for their applicability to sizecharacterization of micron-sized pollens. Both GrFFF and SdFFF are elution techniques, providing sequential elution of particles based on size. They allow the size distribution as well as the mean size of the sample to be determined from the elution time. In this study, GrFFF and SdFFF were used to determine the size distribution of Paper Mulberry and Bermuda Grass pollens. For the Paper Mulberry pollen, the mean size obtained by GrFFF is 12.7 μm, and agrees rather well with the OM data with the relative error of 8.0%. For the Bermuda Grass pollen, the mean size obtained by GrFFF is 32.6 μm with the relative error of 12.3%. The mean sizes determined by SdFFF are 12.4 (relative error = 10.1%) and 27.1 μm (relative error = 5.2%) for the Paper Mulberry and the Bermuda Grass pollen, respectively. Although SdFFF tends to yield more accurate size distribution due to lower band broadening under the field strength higher than 1 G, the sizes determined by GrFFF were not significantly different from those by SdFFF.

  • 마이야르반응(反應) 생성물(生成物)인 Premelanoidin 중(中) 주정발효(酒精醱酵)의 속도(速度)를 촉진(促進)하는 물질(物質)을 분리(分離)하기 위(爲)하여 양(陽)이온 교환수지를 이용(利用)하여 Premelanoidin을 fractionation하고 얻어진 각(各) fraction의 주정발효(酒精醱酵)의 활성(活性)여부를 시험(試驗)하였다. 양(湯)이온 교환수지로는 Dowex $50{\times}8$, 50-100을 사용(使用)하고 elution solvent로는 $2N-NH_4OH$ 용액(溶液)을 사용(使用)하였으며 얻어진 각(各) 5ml의 fraction은 냉동건조(冷凍乾操)해서 다시 본래(本來) 시료(試料)의 농도(濃度)(0.2N)로 희석하여 활성시험(活性試驗)에 사용(使用)하였다. Fractionation의 결과(結果)는 water filtrate 구(區)에 glucose, 5-HMF 및 소량(少量)의 갈색색소의 fraction을 얻었고 ammoniacal eluate 구(區)에 대부분(大部分)의 갈색색소와 glycine N-glycoside의 fraction을 얻을 수 있었다. 주정발효(酒精醱酵)의 활성(活性)은 glucose, 갈색색소 및 glycine-N-glycoside 구획에서 관찰할 수 있었으나 glucose 는 전(前) 실험(實驗)에서 활성(活性)이 없는 물질(物質)로 인정(認定)되었으며 본(本) 실험(實驗)에 나타난 활성(活性)은 Dowex 50에 의(依)한 fractionation 과정중 glucose 자체(自體)의 변질(變質)에 의(依)한 것으로 추측된다. 결국(結局) 활성물질(活性物質)이 존재(存在)하는 fraction은 Ammoniacal eluate 구(區)의 갈색색소 fraction과 glycine 및 N-glycoside를 함유하는 fraction이라고 인정(認定)된다.

Current Status of High Dose Rate Brachytherapy in Cervical Cancer in Korea and Optimal Treatment Schedule (자궁 경부암 고선량율 강내조사 치료의 국내 현황과 적정 치료방법)

  • Brachytherapy is an essential part of radiotherapy for uterine cervical cancer. The low dose rate (LDR) regimen has been the major technique of intracavitary therapy for cervical cancer. However, there has been an expansion in the last 20 years of high dose rate (HDR) machines using Ir-192 sources. Since 1979, HDR brachytherapy has been used for the treatment of uterine cervical cancer in Korea. The number of institutions employing HDR has been increasing, while the number of low dose rate system has been constant. In 1995, there was a total 27 HDR brachytherapy units installed and 1258 cases of patients with cervical cancer were treated with HDR Most common regimens of HDR brachytherapy are total dose of 30-39 Gy at point A with 10-13 fractions in three fractions per week. 24-32 Gy with 6-8 fractions in two fractions per week, and 30-35 Gy with 6-7 fractions in two fractions per week. The average fractionation regimen of HDR brachytherapy is about 8 fractions of 4.1 Gy each to Point A. In Korea, treatment results for HDR brachytherapy are comparable with the LDR series and appears to be a safe and effective alternative to LDR therapy for the treatment of cervical carcinoma. Studies from the major centers report the five-year survival rate of cervical cancer as. 78-86$\%$ for Stage 1, 68-85$\%$ for stage 11, and 38-56$\%$ for Stage III. World-wide questionnaire study and Japanese questionnaire survey of multiple institutions showed no survival difference in any stages and dose-rate effect ratio (HDR/LDR) was calculated to be 0.54 to 0.58. However the optimum treatment doses and fractionation schemes appropriate to generate clinical results comparable to conventional LDR schemes have yet to be standardized. In conclusion, HDR intracavitary radiotherapy is increasingly practiced in Korea and an effective treatment modality for cervical cancer. To determine the optimum radiotherapy dose and fractionation schedule, a nation-wide prospective study is necessary in Korea. In addition, standardization of HDR application (clinical, computer algorithms, and dosimetric aspects) is necessary.

  • As the size of semiconductors becomes smaller, it is necessary to perform high precision polishing of nanoscale. Ceria, which is generally used as an abrasive, is widely used because of its uniform quality, but its stability is not high because it has a high molecular weight and causes agglomeration and rapid precipitation. Such agglomeration and precipitation causes scratches in the polishing process. Therefore, it is important to accurately analyze the size distribution of ceria particles. In this study, a study was conducted to select dispersants useful for preventing coagulation and sedimentation of ceria. First, a dispersant was synthesized and a ceria slurry was prepared. The defoamer selection experiment was performed in order to remove the air bubbles which may occur in the production of ceria slurry. Dynamic light scattering (DLS) and asymmetrical flow field-flow fractionation (AsFlFFF) were used to determine the size distribution of ceria particles in the slurry. AsFlFFF is a technique for separating nanoparticles based on sequential elution of samples as in chromatography, and is a useful technique for determining the particle size distribution of nanoparticle samples. AsFlFFF was able to confirm the presence of a little quantities of large particles in the vicinity of 300 nm, which DLS can not detect, besides the main distribution in the range of 60-80 nm. AsFlFFF showed better accuracy and precision than DLS for particle size analysis of a little quantities of large particles such as ceria slurry treated in this study.
