Antiarthritic action of polar fraction from Sida rhombifolia aerial parts

  • Aerial parts of Sida rhombifolia Linn. (Malvaceae) were extracted successively using various solvents and screened for various parameters of antiarthritic activity such as adjuvant-induced arthritis, motor performance, and histopathological study. The ethanol and the aqueous extracts showed potent activity; further these extracts were fractionated by using column chromatography. The fraction ET1 isolated from ethanol extract showed the most potent antiarthritic activity.

Cytotoxic Effects of Chloroform Extracts and Fraction from Cornis fructus on Cancer Cell Lines

  • Cornis fructus were extracted by successive extractions and then fractionated with chloroform extract to get active fractions. This study was performed to determine the cytotoxic effect of chloroform extract from Cornis fructus on NIH 3T3 fibroblasts and cancer cell lines using MTT assay. All extracts did not exhibit cytotoxicity in HIH 3T3 fibroblasts. Chloroform extract exhibited antitumor activity in A549, MDA-MB-123, B16 melanoma and SNU-C4 cells. Futher fractionation with chloroform extract was performed to obtain effective fractions. (omitted)

  • The objective of this in vitro study is to screen a possible inhibitor, originated from some chinese herb medicines, of 3-hydroxy-3-methylglutaryl Coenzyme A (HMG-CoA) reductase that is the major regulatory enzyme of hepatic cholesterol biosynthesis. Fourteen kinds of herbal plant were extracted with water and evaporated for prescreening. The methanol extracts of the effective 3 kinds (9 species) were fractionated with chloroform, ethylacetate, butanol and water, and vacuum evaporated. The degree of inhibition of the extracts to HMG-CoA reductase activity was calculated by the spectrophotometric method using microsomal protein of Saccharomyces cerevisiae ATCC 42949 as an enzyme source. Among these samples, marked inhibitory effects were observed in the extracts of ethylacetate and chloroform fractions of the Rosa rugosa roots, and those of butanol, ehtylacetate and water fractions of pine leaves. Also, the inhibitory effects of the extracts obtained from buckwheat shell and the roots of Rosaceae were found.

  • This study was investigated to observe the cytotoxicity effect of Ligularia fischeri extracts against cancer cell lines including human lung carcinoma(A549), human cervix epitheloid carcinoma(HeLa) and human hepatocellular carcinoma(HepG2) using SRB(sulforhodamine B) method. The ethanol and methanol extracts of 1$\mu\textrm{g}$/${mu}ell$ showed approximately 79.2% and 86.4% cytotoxicity effects on HepG2 cell line and the ethyl acetate fracton fractionated from ethanol extracts showed the strongest cytotoxicity effect with 94% inhibition. The inhibitory effect of ethanol extract on HeLa cell line was somewhat low with 50~56% inhibition, but ethyl acetate fraction showed higher cytotoxicity effect with 91% and 91.9% inhibition on the HeLa and A549 cell line. On the contrary, the ethanol and methanol extracts showed the lower inhibition effects on the normal liver cell, WRL68, compared to human cancer cell lines.

Ultrafiltration Characteristics of Hot-Water Extracts from Siberian Larch Barks for Tannin-based Adhesives (타닌접착제를 위한 시베리아산 낙엽송 수피 온수추출물의 한외여과 특성)

  • Hot-water soluble extracts were prepared from medium-sized barks of Siberian larch (Larix gmelineii). The extracts were fractionated by ultrafiltration method for producing uniform quality of adhesives. Molecular weight distribution of the extracts was ranged of 100 to 300.000. pH of the extracts affected amounts of flux, and the range of pH 6~pH 8 was proper for wood adhesives, because of easy concentration and fast gelation time. Removal of particles greater than $0.45{\mu}m$ from the extracts increased both filtration speed (flux) and yields of solids in the filtrates. In ultrafiltration process. operating pressure, filtrate temperature, and flow rate significantly increased with the increase of individual condition. Ultrafiltration using PM10 membrane was very effective to fractionate and concentrate the extracts. Removal of large particles greater than 0.45 m from the extracts increased filtration speed(flux) and yields of solids in the filtrates. A gelation time was accelerated with the increasing pH of the extracts and its concentration. The Stiasny precipitate(26%) from the filtrate obtained by PM 10 membrane was very lower than that(78%) of the retentates. This ultrafiltration method was efficient for obtaining high yield purified phenolic compounds.

Anti-oxidative and Anti-inflammatory Effects of Draconis Resina in Mouse Macrophage Cells (마우스 대식세포에서 혈갈(血竭)(Draconis Resina)의 항산화 및 항염증 효과)

  • Objectives : Draconis Resina (DR) has been used as a traditional Korean herbal medicine since ancient times, and today it is used as a medication for wounds, tumors, diarrhea, rheumatism, in the itching of insect bites and with other conditions in the folk medicine. The aim of this study was to determine whether fractionated extracts of DR inhibit free radical generation, intracellular oxidation, production of nitrite, an index of NO, PGE2, iNOS, COX-2 and proinflammatory cytokines in lipopolysaccharide (LPS)-treated RAW 264.7 macrophages, Methods : DR extract prepared with methanol, and then fractionated with hexane, dichloromethane, ethylacetate, n-butanol and water. Inhibitory effect of DR onto free radical generation was determined by measuring DPPH, superoxide anions and nitric oxide scavenging activities in vitro. Cytotoxic activity of extracts on RAW 264.7 cells was measured using 5-(3-caroboxymeth-oxyphenyl)-2H-tetra-zolium inner salt (MTS) assay. Intracelluar oxidation was analysed by DCF-DA assay. The nitric oxide (NO) production was measured by Griess reagent system. The levels of iNOS and COX-2 expression were confirmed by western blot. And proinflammatory cytokines were measured by ELISA kit. Results : Our results indicated that fractionated extracts, especially dichloromethane and ethyl acetate extracts, significantly inhibited free radical generation, the LPS-induced H202, NO, PGE2 production and iNOS, COX-2 expression accompanied by an attenuation of TNF-${\alpha}$, IL-$1{\beta}$ and IL-6 formation in macrophages. Conclusions : Our results indicate that dichloromethane and ethyl acetate extracts of DR have potential as an agent of chronic inflammatory diseases.

  • Objectives : The aim of this study was to authenticate whether fractionated extract of Rumex japonicus HOUTT. (RJ) has anti-inflammatory effects in mouse macrophage, RAW264.7 cells. Methods : Roots of RJ were extracted by methanol for 48hours. The methanol that gained was filtered and freeze dried. The methanol extract was dissolved in water and dichloromethane (DCM). After that, two layers were separated. Ethyl acetate (EA) added to the water layer and separated again. All the layers were filtered and freeze dried and the extracts were tested. Cytotoxic activity of extracts on RAW 264.7 cells was measured using MTS assay. The nitric oxide (NO) production was measured and proinflammatory cytokines and $PGE_2$ were measured by ELISA kit. The levels of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), I ${\kappa}$-B-${\alpha}$ and nuclear NF-${\kappa}B$ p65 expression were detected by western blot. Results : Our results indicated that DCM and EA extracts of RJ inhibited the LPS-induced NO, $PGE_2$ production and iNOS, COX-2 expression accompanied by an attenuation of TNF-${\alpha}$, IL-$1{\beta}$ and IL-6 production in RAW 264.7 cells most effectively. DCM and EA extracts also had suppression effects of LPS-induced NF-${\kappa}B$ and MAPKs activation. Conclusions : This results demonstrate that fractionated extract of RJ has anti-inflammatory effects and among the fractioned extract, dichloromethane and ethyl acetate extract have best anti-inflammatory effects.

Inhibitory Effect of Oriental Herbal Medicines on Tumor Necrosis $Factor-{\alpha}$ Production in Lipopolysaccharide-stimulated RAW264.7 cells

  • Eleven out of 118 herbal medicines which are frequently used in Korean traditional prescriptions for inflammatory diseases exhibited more than 50% of inhibition on $TNF-{\alpha}$ production in LPS-stimulated RAW264.7 cells by their total ethanol extracts with 0.1 mg/ml as a final concentration. The active 11 total extracts were prepared from Angelica koreana, Coptis japonica, Cynanchum paniculatum, Magnolia frgesii, Magnolia officinalis, Panax ginseng, Patrinia scabiosaefolia, Pterocarpus santalius, Rhapontica uniflora, Saussurea lappa. Of them, Coptis japonica, Magnolia fargesii and Saussurea lappa also significantly inhibited $TNF-{\alpha}$ production in vivo. These total extracts were sequentially fractionated with n-hexane, methylene chloride, ethyl acetate, n-butanol and water. Among the solvent-fractionated extracts with 0.05 mg/ml as a final concentration, inhibitory effects of Angelica koreana, Magnolia fargesii, Magnolia officinalis, Pterocarpus santalinus, Rhapontica uniflora and Saussurea lappa markedly showed in one or two solvent fractions suggesting that the principles may be concentrated by subfractionation as the main compounds.

  • To develop antioxidant substances in plant, phenolic compounds in Bangah (Igodon japonicus, Hara) and antioxidative effects of cell extracts were investigated. Defatted samples of Bangah were extracted with acetone/methanol and the extracts were fractionated Into the free and bound types of phenolics. The equal amounts of fractions were dissolved In soybean oil and autoxidized at 45$^{\circ}C$ for 25 days. Peroxide value and TBA values were determined every 5 days during oxidation. Changes of peroxide and TBA values showed antioxidant effects, which were lower than control during the oxidation periods. The effects of cell extracts were evaluated as high as BHT, 0.02 ppm. The antioxidative effect of insoluble-bound phenolic extract was higher than that of free. The contents of phenolic compounds in insoluble-bound fraction were also higher than free fraction.

  • Growth inhibitory effect of doenjang(Korean soypaste) methanol extracts in SRB assay using AGS human gastric adenocarcinoma cell, Hep 3B human hepatocellular carcinoma cell and HT 29 human colon cancer cell was studied. The treatment of doenjang methanol extracts(2mg/assay) to the AGS, Hep 3B and HT 29 cancer cells inhibited the growth of the cancer cells by 55%, 60%, and 71%, respectively. Doenjang methanol extracts exhibited the highest inhibitory effect among other soybean fermented foods and original materials in the SRB assay. In addition, to separate active compounds of doenjang methanol extracts, we fractionated the doenjang with hexane, methanol, dichloromethane, ethylacetate and butanol. Growth inhibitory effect on the AGS, Hep 3B, HT 29 and MG 63 cancer cells was the highest in the fractions of dichloromethane and ethylacetate among other solvent fractions of the doenjang. These results showed that some compounds contained in the fractions of dichloromethane and ethylacetate might play a role on the anticanceric effect of doenjang.
