ESR spectroscopy에 의한 감마선 조사된 수입 향신료의 검지 (Detection of Gamma-Irradiation Treatment in Imported Spices by ESR Spectroscopy)

  • ESR spectroscopy를 이용하여 방사선 조사된 수입산 향신료 3종에서 생성된 ESR 신호 크기에 조사선량과 저장기간이 미치는 영향을 조사하였다. 수입산 향신료 3종(겨자, 오레가노, 월계수)를 0, 1, 5 및 10kGy의 Co-60 감마선을 조사한 후, 이를 시료로 Bruker-EPR spectrometer를 이용하여 ESR 신호를 측정하였다. 실험결과 조사 직후 감마선 조사된 겨자와 월계수는 조사시료에서 특유의 신호를 나타내어 비조사시료와 뚜렷이 구별할 수 있었고 오레가노는 비조사시료에서 single- line을 보였으나 조사시료와 비교하여 신호 크기가 매우 커서 비조사시료와 쉽게 구별할 수 있었다. 모든 적용선량$(1{\sim}10kGy)$의 범위에서 조사선량이 증가함에 따라 ESR 신호 크기가 증가하였으며, 조사선량과 생성된 ESR 신호 크기와의 상관계수인 $R^2$ 값은 $0.9939{\sim}0.9993$의 높은 값을 나타내었다. 또 상온에서 12주 후에도 ESR spectroscopy를 이용하여 방사선 조사의 여부를 확인할 수 있었다.

Thermoluminescence (TL) of Minerals Separated from Irradiated Mussel

  • This study was carried out to determine whether detection of minerals separated from irradiated mussel could be could be done by thermoluminescence (TL) method. After the minerals were separated by sodium polytungstate solution (2.0 g/mL) from irradiated mussel, organic compounds remaining in the minerals were removed by acid-base treatment and dried at 50℃ overnight, and then the minerals were measured through TL. The TL intensities of separated minerals at different irradiation doses during storage conditions of room and darkroom were obtained. TL intensity of first glow curves for minerals separated from irradiated mussel showed linear increase from the control to 5 kGy and slight increase from 5 kGy to 10 kGy. Since glow curve ratios of G2, G3 and G4, calculated from re-irradiated minerals measured immediately after irradiation and after storage of three months were over 0.5, detection of irradiation was possible. G1, which showed the glow curve ratios above 0.1, was classified as non-irradiated samples because the unique first glow curve was not found within the recommended temperature interval (150-230℃). Hence, on the basis of TL intensity, and glow curve ratio and shape, it is possible to correctly identify irradaited mussels after mineral separation during storage.

  • The development of methods for the identification of irradiated foods helps enforce national and international regulations on labelling to ensure the consumer's free choice to buy irradiated or unirradiated foods. and the availabilityof such methods may assist the promotion of international trade in irradiated food products and help prevent abuse of the technology. A number of approaches to determine the physical , chemical, microbiological and biological changes that occur in foods treated with ionizing radiation have been studied. However no single method is universally applicable. Among physical measurements, the leading methods of indentification are electron spin resonance (ESR) spectroscopy and thermoluminescence(TL). ESR is an established non-destructive method for the analysis of free radicals from their traps and TL is the emission of light from irradiated mineral extracts by heating. Viscosity of carbohydrate polymers by causing chain breaks by irradiation, measuring the impedance of potatoes and detection of gases produced radiolytically are promising techniques for identification purposes. Irradiated water-containing foods show significant supercooling when monitored with a differential scanning calorimeter (DSC), which can be applied to identifying irradiated ones.

Effects of Irradiation Temperature on the Sensory Quality Improvement of Gamma-irradiated Ganjang-gejang, Korean Traditional Marinated Raw Crab Portunus trituberculatus in Soybean Sauce

  • This study was conducted to confirm quality properties of sterilized Ganjang-gejang (marinated crab Portunus trituberculatus) with Korean soy sauce using by gamma irradiation and to improve quality of sterilized Ganjang-gejang. The Ganjang-gejang was irradiated at dose of 3, 6, 9, 12, and 15 kGy by gamma irradiation and there was evaluated in microbiological, physicochemical, and sensory properties. Total aerobic bacteria and fungi contents of non-irradiated samples were about 6 and 4 log CFU/g level, respectively. Gamma-irradiated samples at above 9 kGy did not contain aerobic bacteria or fungi at detection limit less than 2 log CFU/g, but sensory scores were significantly decreased depending on the irradiation dose. To improve the sensory qualities of gamma-irradiated Ganjang-gejang, the temperature was adjusted during sample irradiation. When samples were irradiated under freezing temperatures, especially on dry ice, the TBARS and the deterioration of sensory qualities of Ganjan-Gejang were reduced. Different odor patterns were observed among samples, as observed using electronic nose analysis system. The results of this study indicated that treatment with irradiation under low temperatures may help to preparing high-quality Ganjang-gejang.

PSL, TL 및 ESR 분석을 통한 수입산 들깨, 아몬드 및 대두의 방사선 조사 여부 판별 특성 (Detection of Gamma-irradiated Imported Foods (Perilla seeds, Almonds, and Soybeans) by Using PSL, TL, and ESR)

  • This study examined radiation detection properties of imported perilla seeds, almonds, and soybeans. Food samples were irradiated at doses of 0.5, 1, 2, and 4 kGy and analyzed by photo-stimulated luminescence (PSL), thermoluminescence (TL), and electron spin resonance (ESR). The results of PSL showed that photon counts of irradiated perilla seeds and soybeans were $32,959counts{\cdot}60sec^{-1}$ and $7,234counts{\cdot}60sec^{-1}$, respectively, at an irradiation dose of 0.5 kGy, and the photon count of irradiated almonds was $5,581counts{\cdot}60sec^{-1}$ at an irradiation dose of 1 kGy. The results of TL showed that this technique is useful for detecting irradiated samples; the TL ratios ($TL_1/TL_2$) measured for the food samples at an irradiation dose of 0.5 kGy were 0.2301 for perilla seeds, 0.4595 for almonds, and 0.4827 for soy beans. Lastly, results of ESR spectroscopy for only soybeans revealed specific signals derived from free radicals captured in the cellulose. In conclusion, PSL, TL, and ESR methods can be used for the detection of gamma-irradiated soybean samples, whereas gamma-irradiated perilla seeds and almonds can be confirmed by a serial detection with a TL method after PSL.

Thermoluminescence 측정에 의한 조미분말식품의 방사선 조사유무 확인 (Detection of Irradiation Treatment for Seasoned-Powdered Foods by Thermoluminescence Measurement)

  • 조개추출물분말, 해물조미베이스, 라면분말수프, 정어리추출물분말 등 조미분말식품에 대한 TL 검지방법의 적용가능성을 확인하였다. 이들 시료의 방사선 조사선량과 TL response 간의 $R^2$는0.5966 이상을 보여 주었고, 특히 라면수프는 0.9500 이상의 상관계수를 보이면서 방사선 조사선량이 증가할수록 TL intensity도 증가하였다. 따라서 라면수프 시료를 대상으로 re-irradiation step에 의한 TL response의 검증을 실시하였고, 미지시료의 검지에 필요한 역치(threshold value)를 측정하였다. 이때 비조사 시료의 역치는 최고 1.37 이하였고, 감마선 조사시료(2.5 kGy) 역치는 최저 6.06 이상이였다. 그리고 1.37과 6.06 사이의 역치를 나타내는 미지시료는 재조사하여 방사선 조사유무를 검지하였고, 검지시료의 호기성 전세균의 수를 측정함으로써 검지 결과를 검증하였다. Blind test의 일환으로 실시한 미지시료 검지실험에서는 본 실험에서 설정한 역치의 적용으로 총 167개 시료(감마선 82, 전자선 85)의 방사선 조사유무를 정확하게 확인할 수 있었다. 방사선 조사선량을 예측하기 위하여 3가지 예측모델식을 설정하고 조사선량을 예측하여 본 결과, 2차회귀식이 가장 적합하였으나 실제 조사선량과는 상당한 오차를 보였다.

광자극발광법과 열발광법을 이용한 국내 방사선 조사 허용 외 식품에 대한 실태 조사 (Monitoring on the Foods not Approved for Irradiation in Korea by PSL and TL Detection Method)

  • 영 유아용이유식, 견과류, 조미건어포류, 천연향신료, 건조과실류, 과 채가공품, 곡류가공품, 수산물가공품 등 국내 방사선 조사가 허용되지 않은 8품목에 대하여 PSL과 TL 분석법의 적용가능성을 검토하고 325건의 식품을 대상으로 방사선 조사여부 실태조사를 실시하였다. 선행 연구가 수행된 천연향신료, 과 채가공품 및 수산물가공품을 제외한 5개의 식품군에 대하여 PSL과 TL 시험법 적용 가능성 검토 결과, PSL의 경우 비조사구는 모두 700 count보다 낮아 음성 시료로 나타났고 1 kGy로 조사 시 중간 및 양성 시료로 확인되었다. TL 분석결과 비조사구는 모두 자연방사선에 의해서 $300^{\circ}C$ 부근에서 peak를 가지는 비 특이적 glow curve를 나타냈고 TL ratio는 0.1보다 낮게 나타나 음성 시료로 확인되었다. 1 kGy로 조사 시 TL ratio는 영 유아용이유식을 제외하고 모든 시료에서 0.1 이상으로 나타났으며 $150{\sim}250^{\circ}C$ 사이에서 peak가 나타나는 특이적 glow curve를 보여 확인 시험법으로 적용 가능하였다. 또한 영 유아용이유식은 2가지 이상의 원료가 혼합되어있어 TL ratio가 비록 0.1 이하지만 $150{\sim}250^{\circ}C$ 사이에서 peak가 나타나는 특이적 glow curve를 통해 조사 여부를 확인할 수 있었다. 국내 방사선 조사 허용 외 식품 8품목에 대하여 PSL과 TL 시험법의 적용가능성을 확인한 후 325건의 식품에 대하여 실태조사 결과, PSL 경우 15건에서 중간시료가 나타났으나 TL 분석 시 TL ratio가 0.1 이상이거나, $150{\sim}250^{\circ}C$ 부근에서 최대강도를 보이는 glow curve를 나타내는 양성시료는 없는 것으로 확인되었다.

Identification of Irradiated Granule-Type Ramen Soup Powder by Pulsed Photostimulated Luminescence and Thermoluminescence during Storage

  • This study was carried out to establish a method for determining if granule-type Ramen soup powder has been irradiated. Thermoluminescence (TL) and pulled photostimulated luminescence (PPSL) were used as the detection methods through observed changes of TL and PPSL intensities after storage under differing conditions. PPSL intensities increased with increases in irradiation doses. The threshold level of PPSL was below 412$\pm$58 photon counts regardless of storage conditions (room and darkroom) after 10 months. TL intensities also increased with increasing irradiation doses. The coefficients ($R^2$) of PPSL (0.74~0.94) and TL intensities (0.92~0.58) were very highly correlated with irradiation dose. The PPSL and TL intensities were decreased after 10 months of storage. These results indicate that discrimination of irradiated from non-irradiated granule-type Ramen soup powder is possible using TL and PPSL methods despite the decrease in intensities of TL and PPSL with increasing storage times.

Comparison of Gamma Irradiation and Sodium Hypochlorite Treatments to Inactivate Staphylococcus aureus and Pseudomonas aeruginosa Biofilms on Stainless Steel Surfaces

  • Biofilm formation on various surfaces is a well-known phenomenon and it has caused pollution problems, health and safety hazards, and substantial economic loss in many areas including the food industry. In the present study, Gamma irradiation at a dose of 2.0 kGy reduced the bacterial counts of Staphylococcus aureus and Pseudomonas aeruginosa suspensions by 6.7 and >6.5 log CFU/mL, respectively, and 30 ppm of sodium hypochlorite effectively reduced the counts of both bacterial suspensions to below the limit of detection ($<2\;log\;CFU/cm^2$). However, in bacterial biofilms attached to stainless steel, gamma irradiation at a dose of 10.0 kGy reduced the counts of S. aureus attached fur 1 hr and overnight by ${\geq}5.1\;and\;5.0\;log\;CFU/cm^2$, respectively. Gamma irradiation at a dose of 1.0 kGy reduced the counts of P. aeruginosa counts to below the limit of detection ($<2\;log\;CFU/cm^2$). On the contrary, S. aureus and P. aeruginosa cells attached to stainless steel chips were difficult to eliminate using sodium hypochlorite. Four hundred ppm of sodium hypochlorite reduced the counts of S. aureus and P. aeruginosa attached for 1 hr by 2.5 and $3.3\;log\;CFU/cm^2$, respectively.
