복강경을 이용한 한국흑염소의 성주기 판정 (Laparoscopy Application to Determine Estrous Cycle in Korean Black Goats)

  • 본 연구는 복강경을 이용하여 한국흑염소의 난소를 연속적으로 관찰하여 그 형태적 변화를 확인함으로써 한국흑염소의 성주기를 판정하고 그 결과를 한국흑염소의 수정란 이식시에 활용하고자 하는 목적으로 수행하였다. $CIDR^(R)$ 제거 2일 후부터 복강경 수술을 통해 난소의 형태적 변화를 관찰하였다. 난소의 크기와 황체의 변화를 관찰하였으며, 황체는 그 성숙과 퇴축 과정에 따라 corpus hemorrhagicum(CH), corpus luteum(CL), corpus albicans(CA)로 나누었다. 배란 전일(Day 0)에는 배란 직전의 대형난포(Graafian follicle)가 존재하였고, 배란일(Day 1)에는 배란 직후의 함몰 부위(OD)를, 그리고 배란 2일째(Day 2)에는 초기 출혈체$(CH_1)$가 관찰되었다. 배란 3일 및 4일째에는 배란된 자리로부터 형성된 출혈체가 성숙하여 $CH_2,\;CH_3$로 발육되며 점차 그 크기가 커지고 주위에 잘 발달된 혈관의 분지들을 관찰할 수 있었다. 배란 5일째와 6일째에는 완전히 성숙하여 난소의 절반 이상을 차지하는 성숙 황체$(CL_3)$를 관찰하였다. 배란 10일째에는 더욱 발달한 성숙 황체와 함께 같은 쪽 또는 반대편 난소에 작은 난포가 생성된 것을 확인하였고, 배란 15일째에는 10일째 생성된 난포와는 다른 새로운 난포가 나타나 발육되는 것을 확인하였다. 배란 17일과 18일째에는 점차 황체의 크기가 작아져 $CL_2,\;CL_1$의 단계로 퇴행하였고, 배란 20일과 22일째에는 완전히 퇴행된 황체, 즉 백체(CA)의 존재와 함께 같은 쪽 또는 반대편 난소에 발육된 우세난포를 확인하였다. 본 연구에서 복강경을 통한 난소의 변화상을 관찰한 결과, 한국흑염 소의 발정주기는 estrus기(성숙 난포기)가 1일(성주기제 0일), metestrus기(배란 및 황체 발육기)는 4일(성주기 제 $1{\sim}4$일), diestrus기(황체기)는 $12{\sim}13$일(성주기 제$5{\sim}16/17$일), proestrus기(황체퇴행 및 난포 형성기)는 5일(성주기 제 $17/18{\sim}21/22$일)인 것으로 확인되었다. 난소의 육안적 관찰을 위해 사용한 복강경 시술은 개복술에 비래 수술 상처와 감염이 적고 수술 후 회복도 빨랐으며 최소한의 유착과 수술 시간의 단축 등 많은 장점을 가지고 있어 한국흑염소의 수정란이식 등에 그 활용 가치가 클 것으로 사료된다.

Estradiol Valerate에 의해 유도된 다낭성난소와 CHO세포에서 NGF발현 (Expression of NGF in Estradiol Valerate-Induced Polycystic Ovary and CHO Cells)

  • 다낭성 난소증후군(polycystic ovary syndrome, PCOS)은 내분비 및 대사장애로 호르몬 불균형 상태를 야기하여 가임기 여성의 불임 및 여러 합병증을 유발한다. 폐경기 여성의 호르몬대체 치료에 사용되는 estradiol valerate (EV)는 과다 투여 시 PCOS를 유발하는 것으로 알려졌다. 신경성장인자(nerve growth factor, NGF)는 발생 중인 신경세포의 생존과 성숙을 조절하는 인자로 난소에서도 합성되고, EV 처리에 의해 유도된 다낭성난소(polycystic ovary, PCO)에서 발현이 크게 증가하는 것으로 보고됐다. 따라서 본 연구에서는 난소유래 세포인 CHO 세포주와 다낭성 난소를 이용해 호르몬 불균형 상태인 PCOS 진단 시 NGF가 생물학적 표지인자로서 사용될 수 있는 가능성을 규명하고자 하였다. CHO 세포에 EV 2 mg과 3 mg 처리는 초기에 세포증식을 억제했으나 1 mg 처리는 영향을 미치지 않음으로써 실험 시 최적농도로 사용하였다. 하지만 농도별 EV 처리에 따른 CHO 세포의 형태학적 차이는 관찰되지 않았다. CHO 세포에 EV 처리 후 NGF mRNA 및 단백질 발현은 대조군에 비해 30분 후 크게 증가하였고, 다수의 난포낭이 관찰된 PCOS 조직에서도 정상군에 비해 크게 증가하였다. 따라서 이상의 결과들을 종합하면 EV 처리 후 난소에서 유도되는 NGF 과발현은 비정상적 난포발달을 유도하는 인자로 작용할 것으로 보이며, 다낭성난소 진단 시 표지인자로서 사용될 수 있을 것으로 생각된다.

OPU 유래 수정란의 대량생산을 위한 고능력 공란우 반복사용 가능성에 관한 연구 (Possibility of Repeated Use of Elite Donor Cows for Mass Production of OPU-Derived Embryos)

  • This study was designed to know the possibility in repeat uses of elite donor cows for getting mass production of OPU-derived embryo production (OPU-IVP). Ultrasound transvaginal ovum pick-up (OPU) performed in 6 Korean native cows was aged 4 to 10 years old. The aspiration of immature oocytes for OPU derived embryo was carried out 2 times per week, and OPU-IVP of $1^{st}$ period was carried out 22~48 sessions from each donors. And the break time for OPU-IVP of $2^{nd}$ period after $1^{st}$ OPU from each donors were 2~25 months. The OPU-IVP of $2^{nd}$ period each donors conducted total 15~65 times for 2~8 months by an ultrasonographic, was guided follicular aspiration system. The average numbers of collected oocytes, grade 1 + grade 2(G1+G2) oocytes and cleavage embryo from $1^{st}$ period OPU-IVP were significantly differences between donors (p<0.05). Total collected oocytes of donor D were significantly higher compared with donors of A, B, C, E and F (average 17.0 per session vs. 11.2, 10.1, 8.5, 10.2 and 9.6; p<0.05) and also oocytes of G1+G2 were significantly higher compared with r A and D and subsequently to donors of B, C, E and F (average 7.9 and 8.5 per session vs. 5.0, 2.7, 6.0 and 1.6; p<0.05). Cleavage rate of donor D was significantly higher compared with donors of A, B, C, E and F (average 13.1 per session vs. 10.1, 9.1, 6.9, 8.9 and 6.7; p<0.05). The average numbers of OPU-IVP for $1^{st}$ period was significantly higher from donors of B, D and E than those from donors of A, C and F (average 6.5, 7.1 and 6.5 per session vs. 3.5, 4.2 and 2.8; p<0.05). The possibility investigation of $2^{nd}$ OPU-IVP was carried out after 2~25 months rest periods from $1^{st}$ period OPU session. Total average numbers of collected oocytes, cleavages and blastocyst development rates were significantly higher from $1^{st}$ period OPU compared with $2^{nd}$ period one (p<0.05). The OPU-IVP efficiency by break for more embryo production from elite cow was analysis comparing without rest of donor A, under 6 months rest period as B and over 6 months rest period as C and then the average numbers of collected oocytes, cleavages and blastocysts were significantly higher from A group (11.8, 9.5 and 5.2 per session) than those from B and C groups (7.9, 6.2 and 2.6 vs. 9.2, 7.5 and 3.9, p<0.05), and also C group was significantly higher than B group. In conclusion, $1^{st}$ period OPU-IVP was more efficient compared with $2^{nd}$ period repeated uses of donor, and the break times for additional production of embryo on donor were needed more than over 6 months after $1^{st}$ period OPU-IVP. This repeating uses of elite donor cows given more emphasis for getting the opportunity on mass production of elite cow OPU-IVP embryo should be increased G1+G2 possibility of genetic improvement of livestock within short period.

배양액과 산소농도가 돼지난포란의 체외성숙과 배발달에 미치는 영향 (Effects of Different Media and Oxygen Concentrations on In Vitro Maturation and Development of Porcine Follicular Oocytes)

  • 본 실험은 돼지체외수정란을 생산하는데 적합한 배양액과 산소조건을 구명하기 위하여 NCSU(North Carolina State University)-23, PZM(Porcine Zygotes Medium)-3, PZM-4 및 TCM(Tissue Culture Medium)-199 배양액과 5% 산소조건(39, 5% $O_2$, 5% $CO_2$ , 90% $N_2$ 및 포화습도) 및 20% 산소조건(39, 5% $CO_2$ 및 포화습도)에서 돼지 미성숙난포란의 체외성숙과 체외배발달을 실시하였다. 본 연구에서 얻어진 결과를 요약하면 다음과 같다. 1) 돼지 난포란을 5 및 20% 산소조건하에서 NCSU-23, PZM-3, PZM-4 및 TCM-199 배양액에 44시간 동안 성숙을 유도한 결과 난핵포붕괴율과 핵성숙률에 있어서 산소조건 및 배양액간 유의적(p>0.05)차이는 없었다. 2) 체외수정을 실시한 결과 정자 침투율, 자ㆍ웅전핵형성률, 다정자 침입률 및 평균정자수에 있어서 산소조건 및 배양액간에 유의적인 차이가 없었다. 3) 체외수정을 실시한 후, 5 및 20% 산소조건의 NCSU-23, PZM-3, PZM-4 및 TCM-199 배양액에서 배양하여 배발달에 미치는 영향을 조사한 결과, 난할률에 있어서는 유의적(p<0.05)인 차이가 없었으며. 배발달 7일째 배반포기 발달률에 있어서 5% 산소조건에서는 PZM-3 배양액이, 20% 산소조건에서는 NCSU-23 배양액이 유의적(p<0.05)으로 높은 결과를 나타냈다. 4) 배발달 7일째 배반포기 배의 세포수를 조사한 결과, 내부세포괴세포수와 총세포수는 산소조건과 배양액간에 통계적인 유의성이 인정되지는 않았으나, 5% 산소조건에서 PZM-3 배양액(36.8$\pm$6.5개)이 다소 높은 총세포수를 나타냈다. 위의 결과를 종합하면 돼지체외수정란의 생산에는 5% 산소조건에서 PZM-3 배양액에서 배발달을 유도하는 것이 배반포기 발달률과 세포수에 있어 효과적이라고 판단된다.

계화도조개 (Potamocorbula amurensis)의 생식주기에 관한 조직학적 연구 (Histological Study on the Reproductive Cycle of Potamocorbula amurensis (Bivalvia: Corbulidae))

  • 1996년 11월부터 1997년 10월까지 전북 부안군 문포 앞바다의 갯벌에서 채집된 계화도조개, p. amurensis를 재료로하여, 그들의 생식소발달과정, 생식세포형성과정, 생식주기, 난경의 월별변화, 비만도 및 군성숙도 둥을 조직학적으로 조사하였다. 계화도조개는 자웅이체이며 난생이다. 난소와 정소는 각각 수많은 난자형성소낭과 정자형성소낭으로 구성되어 있었다. 소낭에는 간충직과 호산성 과립세포들이 들어 있는데, 이들은 초기 생식세포의 형성과 발달에 영양을 공급하는 영양세포로 생각된다. 분열증식 중인 초기 활성기의 난원세포는 $9\~12\mu$m로서 6$\mu$m 내외의 크고 뚜켠한 핵과 호염기성 단일 인을 가지고 있었다. 직경 $30\~32\mu$m로 성장한 난모세포는 난병을 소낭벽에 부착한 채 소낭의 내강을 향해 긴 타원형으로 성장하였다. 난모세포가 $43\~45\mu$m로 성장하면 소낭벽으로부터 유리되며, 완숙란의 크기는 $50\~60\mu$m이었다. 정자형성소낭의 벽에는 정원, 정모, 정세포 및 완숙정자 순으로 충상배열을 하며 성숙되었다. 생식소의 발달, 생식세포형성과정, 난경의 월별변화, 조직분화과정 및 세포학적 특성에 따라, 생식주기를 초기 활성기, 후기 활성기, 완숙기, 부분 산란기 및 회복기 등으로 구분할 수 있었다. 산란기는 5$\~$7월과 9$\~$10월 사이이며, 주산란은 수온이 $18^{\circ}C$ 이상인 5$\~$6 및 10월에 2차례 일어났다. 비만도지수의 월별변화는 생식주기와 밀접한 관계가 있었다. 군성숙도가 $50\%$를 넘는 개체는 암, 수 모두 각장 8.1$\~$10.0 mm인 개체들로 조사되었다. 생식세포형성은 두 가지 방법 즉 1. 산란 후, 생식세포형성소낭내에 잔존하던 미방출란과 미방출정자는 퇴화, 흡수되지만 기존의 소낭은 크게 위축되지 않고, 2$\~$3개월 동안 (특히 여름)에 곧 회복되면서 새로운 생식세포형성에 참가하는 방법, 2. 산란 후 기존의 각 소낭은 위축되고 새로운 소낭의 형성과 더불어 새로운 생식세포가 형성되는 방법이 있다고 생각되었다.

클로미펜에 얇은 자궁내막을 보이는 환자에서 성선자극호르몬 병합 과배란유도시 클로미펜과 레트로졸의 임상적 효용성 (Clinical Efficacy of Clomiphene Citrate and Letrozole Combined with Gonadotropins for Superovulation in Patients with Clomiphene-Induced Thin Endometrium)

  • 목 적: 클로미펜을 사용한 배란유도시 얇은 자궁내막을 보였던 환자들에서 성선자극호르몬에 클로미펜 또는 레트로졸을 병합 투여하는 과배란유도 방법의 임상적 효용성을 비교 분석하고자 하였다. 연구방법: 이전의 클로미펜을 사용한 배란유도 주기에서 8 mm 미만의 얇은 자궁내막을 보였던 불임 환자들에서 시행된 성선자극호르몬 병합 과배란유도/인공수정 51주기가 연구에 포함되었다. 월경주기 제3일째부터 5일 동안 클로미펜+성선자극호르몬 군은 일일 클로미펜 100 mg을 투여하였고 (n=26) 레트로졸+성선자극호르몬 군은 일일 레트로졸 2.5 mg 또는 5 mg을 투여하였다 (n=25). 양 군에서 월경주기 제5~7일째부터 우성난포의 크기가 18 mm 이상에 도달할 때까지 이틀에 한 번씩 성선자극호르몬은 75~150 IU를 투여하였다. 양 군에서 성선자극호르몬 총 사용량, 자궁내막의 두께, 자궁내막의 형태, hCG 투여일의 14 mm 이상 난포의 수, hCG 투여일, 임신율, 다태 임신율을 비교하였으며 통계 분석은 Mann-Whitney U test or Fisher's exact test 등을 이용하였다. 결 과: 연령, 불임기간, 이전 인공수정 횟수, 기저 혈중 LH, FSH, $E_2$ 농도, 불임의 원인 등의 임상적 특성은 양 군간 차이가 없었다. 성선자극호르몬 병합 과배란유도시 배란전 자궁내막의 두께는 이전의 클로미펜을 사용한 주기와 비교하여 유의하게 증가되었다. 성선자극호르몬 총 사용량, hCG 투여일, 자궁내막의 삼중선 비율, 임신율 및 다태 임신율은 유의한 차이가 없었다. 클로미펜+성선자극호르몬 군에 비하여 레트로졸+성선자극호르몬 군에서 14 mm 이상 난포의 개수는 유의하게 적었고 ($3.7{\pm}1.7$ vs. $2.8{\pm}1.7$, p=0.03). 배란 전 자궁내막 두께는 유의하게 두꺼웠다 ($7.7{\pm}1.5\;mm$ vs. $9.1{\pm}1.7\;mm$, p<0.05). 결 론: 배란유도를 위하여 클로미펜 사용시 얇은 자궁내막을 보였던 환자들에서 인공수정을 위한 과배란유도시클로미펜 또는 레트로졸을 성선자극호르몬과 병합하여 사용하는 방법은 클로미펜의 자궁내막에 대한 부정적인 효과를 피할 수 있는 것으로 사료된다. 적절한 자궁내막의 발달 및 적절한 난포의 성장 측면에서 성선자극호르몬에 레트로졸을 병합하는 과배란유도 방법이 클로미펜을 병합하는 방법에 비하여 더 유용할 수 있으나 추가적인 대규모 전향적 연구가 필요할 것으로 사료된다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. Under our experimental conditions, the presence of Photolfrin II increased surface temperature by at least 40% and the temperature rise due to 300 mW/cm2 irradiation exceeded values for hyperthermia. Light and temperature distributions with depth were estimated by a computer model. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm.
