Stachys tibetica Vatke (Lamiaceae) is an important medicinal plant in the folk medicine of Ladakh, India and Tibet for the treatment of various mental disorders. Infusion and decoction of the whole plant is used as a cup of tea for a severe fever, headaches and to relieve tension. The recent study is aimed to evaluate the preliminary pharmacognostical and phytochemical nature of Stachys tibetica Vatke. The whole plant material was subjected to successive soxhlet extraction with petroleum ether (40 - $60^{\circ}C$), chloroform, ethyl acetate, methanol and finally decocted with water to get the respective extracts. The fluorescence characteristics of the powdered materials were analysed under ultraviolet light and ordinary light. Different physicochemical parameters such as ash value, extractive value, foaming index, pH values, loss on drying and determination of foreign matter were carried out as per WHO guidelines. The total fat, flavonoid, saponin and volatile contents were also determined. Macroscopical studies revealed the authentication of the plant drug. Physicochemical parameters helped to standardize the plant material while preliminary qualitative chemical tests of different extracts showed the presence of Glycosides, Carbohydrates, Phytosterols/triterpenoids, Saponins, Fixed oils, Fats and phenols/tannins. Quantification of the total flavonoids and saponins and contents were determined as $54.66{\pm}0.58mg/g$ and $75.42{\pm}0.48mg/kg$ respectively, while the volatile and fat contents were 6.5% and 0.7% respectively. Results may lay the foundation for the standardization of the drug and discovery of new molecules from S. tibetica for the treatment of various diseases.
Angelica decursiva has been utilised as remedy for controlling the airway inflammatory diseases in folk medicine. We investigated whether nodakenin, columbianadin, and umbelliferone isolated from the roots of Angelica decursiva inhibit the gene expression and production of MUC5AC mucin from human airway epithelial cells. Confluent NCI-H292 cells were pretreated with nodakenin, columbianadin or umbelliferone for 30 min and then stimulated with epidermal growth factor (EGF), phorbol 12-myristate 13-acetate (PMA) or tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) for 24 h. The MUC5AC mucin gene expression was measured by reverse transcription - polymerase chain reaction (RT-PCR). Production of MUC5AC mucin protein was measured by enzyme-linked immunosorbent assay (ELISA). The results were as follows: (1) Nodakenin did not affect the expression of MUC5AC mucin gene induced by EGF, PMA or $TNF-{\alpha}$. Columbianadin inhibited the expression of MUC5AC mucin gene induced by EGF or PMA. However, umbelliferone inhibited the expression of MUC5AC mucin gene induced by EGF, PMA or $TNF-{\alpha}$; (2) Nodakenin also did not affect the production of MUC5AC mucin protein induced by EGF, PMA or $TNF-{\alpha}$. Columbianadin inhibited the production of MUC5AC mucin protein induced by PMA. However, umbelliferone inhibited the production of MUC5AC mucin protein induced by EGF, PMA or $TNF-{\alpha}$. These results suggest that, among the three compounds investigated, umbelliferone only inhibits the gene expression and production of MUC5AC mucin stimulated by various inducers, by directly acting on airway epithelial cells, and the results might explain the traditional use of Angelica decursiva as remedy for diverse inflammatory pulmonary diseases.
손바닥선인장 추출물은 한의학에서 항염증 작용, 화상, 부종, 소화 불량 등 몇 가지의 생리학적 기능이 확인되었다. 본 연구에서는 손바닥선인장 줄기를 메탄올을 이용하여 추출한 후 그 추출물을 이용하여 항균활성을 확인한 결과 병원균 등 일반 세균에서 항균활성을 확인하였다. 특히, 약제내성균 및 혐기성 균인 여드름 원인균 뿐만 아니라 효모, 곰팡이 등 진균류에서도 항균활성을 나타내어 넓은 항균 스펙트럼을 가지고 있음을 확인할 수 있었다. 또한 열 처리, 저장온도 및 저장기간이 항균활성에 영향을 미치지 않았으므로 온도 및 기간에 안정함을 확인할 수 있었고, 식품 보존제로서의 가능성 확인 시험에서도 기존의 식품 보존제보다 안전성이 있는 항균활성 물질로 평가되었다.
Zizyphi Spinosi Semen (ZSS), a traditional Chinese folk medicine, has been used for treatment of insomnia and anxiety. This experiment was performed to investigate the anxiolytic-like effect of methanol extract of ZSS (MEZSS) in mice by using the experimental paradigms of anxiety and compared with that of a known anxiolytic, diazepam. In the elevated plus-maze test, it showed that MEZSS (100 mg/kg, p.o.) and diazepam (2.0 mg/kg, p.o.) increased the percentage of time spent on the open arms and the number of open arms entries. MEZSS (50, 100 and 200 mg/kg, p.o.) and diazepam (0.5 mg/kg, p.o.) significantly increased the number of head dips compared with that of control group in the hole-board test. However, MEZSS has no effect on decreasing the locomotor activity, while diazepam (2.0 mg/kg, p.o.) significantly inhibited locomotor activity. MEZSS did not decrease the strength force in the grip strength test, either. In addition, GABAergic involvements were also investigated to understand the possible mechanisms. $GABA_{A}$ receptors subunits and glutamic acid decarboxylase (GAD) were not over expressed, compared with that of the saline group. We also found that MEZSS did not increase chloride influx in cultured cerebellar granule cells. It is concluded that MEZSS might have anxiolytic-like effects, but these effects might not be mediated by GABAergic transmission.
엽산을 다량 함유하고 있는 수벌 번데기와 수벌 번데기에서 생산한 밀리타리스동충하초의 혈청 테스토스테론 상승효과를 확인하기 위하여 실험동물 쥐에 4주간 경구 투여하여 체중변화, 테스토스테론의 수치를 검사하였다. 대조군 및 투여군에서 체중의 증가량은 그룹간, 날짜별 유의적인 차이는 보이지 않았다. 혈청 테스토스테론은 대조군에 비하여 시료를 투여한 모든 군에서 증가경향을 보였다. 더욱이 밀리타리스동충하초 투여구의 호르몬 수치는 대조군에 비하여 통계학적 유의하게 증가하였다.
Night-break, 일장조절, 월동가온 실험으로부터 얻어진 바위솔에 포함된 flavonol glycoside, $kaempferol-3-O-{\beta}-D-glucoside$(1), $kaempferol-3-O-{\beta}-D-glactoside$ (2), $kaempferol-3-O-{\beta}-D-rhamnoside$ (3), $quercetin-3-O-{\beta}-D-glucoside$ (4) and $quercetin-3-O-{\alpha}-D-rhamnoside$ (5)의 함량이 분석되었으며 자연산 바위솔의 함량과 비교되었다. 월동가온 시험에서 얻은 flavonol glycoside 1-5의 함량은 자연산보다 감소하였으며 일장조절 실험 및 night-break 시험에서 얻은 flavonol glycoside 함량은 조사한 빛의 양이 증가함에 따라 증가하였다. 월동가온은 flavonol glycoside의 생성에 부정적으로 작용하는 반면 조사되는 빛의 양의 증가와 같은 경작조건은 flayonol glycoside의 생성에 유리하다.
In oriental folk medicine, Artemisia messes-schmidiana var viridis(Compositae) has been used for jaundice, hepatitis, diuretic and liver cirrhosis etc. 1-naphthylisothiocyanate(ANIT) has been used for more than 20 years as a model compound to study mechanisms of intrahepatic cholestasis in laboratory animals as rat and mouse. Various biochemical and morphological changes including biliary epithelial and parenchymal cell necrosis occur in the liver of animals treated with ANIT. The purposes of present study are to examine pharmacological effects of Artemisia messes-schmidiana var viridis water extract(AMWE) on alterations of secretion volume and total bile acids level in bile juice, and that of serum AST, ALT, ALP, bilirubin, and glucose levels in rat. AMWE stimulated bile secretion and recovered ANIT-induced cholestasis. Bile acid concentrations increased to more than 60% compared with normal by ANIT, which were returned toward normal value with AMWE treatment. Serum AST and ALT activities were increased by ANIT and yet which were significantly decreased with AMWE treatment. In addition, this effect was apparent in AMWE pretreatment group. Serum glucose levels were increased with AMWE and ANIT, while were decreased compared with control in AMWE posttreatment group. Increased serum total bilirubin contents and ALP activities by ANIT were significantly decreased with AMWE posttreatment. In conclusion, AMWE exerted bile acid-independent choleresis effect and then improved to normal conditions ANIT-induced cholestatic syndromes. Also, AMWE have protective and regenerative effect of hepatocytes in rat.
Phytoestrogens, especially Yak-kong or soybean-derived isoflavones have been traditionally used as a supplement of estrogen for preventing postmenopausal osteoporosis in oriental folk medicine. In a previous study, we demonstrated that as Yak-kong and soybean increased MG-63 human osteoblastic cell proliferation, the expression of estrogen receptor $\alpha\;and\;beta\;(ER\;\alpha:\;ER\;\beta$) both were increased. However, the increased level of ER $\alpha$ is much higher than that of ER$\beta$. To determine whether the altered level of ER $\alpha$ expression affects Yak-kong or soybean induced MG-63 cell proliferation, we established cell lines stably expressing either ER $\alpha$ or antisense ER $\alpha$ RNAs. Increased expression of ER a in MG- 63 cells (ER $\alpha$-MG63) enhanced Yak-kong or soybean induced proliferation which paralleled with the enhanced expression of IGF-I. Inhibition of ER $\alpha$ expression by antisense $ER\;\alpha\;RNAs\;(As-ER\;\alpha-MG63$) caused these cells to insensitize Yak- kong or soybean induced proliferation and IGF-I expression. Furthermore, the comparable effects between Yak-kong and the combined treatment of genistein and daidzein at $0.5\;{\times}\;10^{-8}M$, which is a concentration of these two isoflavones similar to Yak-kong at 0.001 mg/ml, on cell proliferation and IGF-I expression in $ER\;\alpha-MG63\;or\;As-ER\;\alpha-MG63$ cells demonstrate that ER $\alpha$ plays an important, active role in MG-63 cell proliferation induced by phytoestrogens, especially Yak-kong or soybean derived isoflavones.
The aerial parts of Eclipta prostrata is used as a traditional medicine and vegetable. In traditional folk medicine, it is used for treatment of hemorrhages, hepatic, disease, renal injuries, hair loss, tooth mobility, and viper bites. In this study, ten compounds (1 - 10) were isolated from the aerial parts of E. prostrata. A reliable high performance liquid chromatography equipped with photometric diode array detector (HPLC-PDA) method was developed to simultaneously quantitate 10 marker compounds [chlorogenic acid (1), paratensein 7-O--ᴅ-glucoside (2), quercetin 7-O--ᴅ-glucoside (3), luteolin 7-O--ᴅ-glucoside (4), apigenin 7-O--ᴅ-glucoside (5), apigenin 4'-O--ᴅ-glucoside (6), apigenin (7), luteolin (8), wedelolactone (9), and paratensein (10)]. In addition, compounds 5 and 6 showed considerable inhibitory effects against protein-tyrosine phosphatase 1B (PTP1B) enzyme. Moreover, compounds 6 - 8, and 10 exhibited potent α-glucosidase inhibitory effects with IC50 values of 24.5 ± 1.9, 33.0 ± 0.5, 45.5 ± 0.1, and 23.8 ± 1.0 µM, respectively. All compounds (1 - 10) showed considerable acetylcholinesterase (AChE) inhibitory effects with IC50 ranging from 30.1 to 75.2 µM.

Background Rosa damascena, a type of herb, has been used for wound healing in Eastern folk medicine. The goal of this study was to evaluate the effectiveness of rose placenta from R. damascena in a full-thickness wound model in mice. Methods Sixty six-week-old C57BL/6N mice were used. Full-thickness wounds were made with an 8-mm diameter punch. Two wounds were made on each side of the back, and wounds were assigned randomly to the control and experimental groups. Rose placenta ($250{\mu}g$) was injected in the experimental group, and normal saline was injected in the control group. Wound sizes were measured with digital photography, and specimens were harvested. Immunohistochemical staining was performed to assess the expression of epidermal growth factor (EGF), vascular endothelial growth factor (VEGF), transforming growth factor-${\beta}1$ (TGF-${\beta}1$), and CD31. Vessel density was measured. Quantitative analysis using an enzyme-linked immunosorbent assay (ELISA) for EGF was performed. All evaluations were performed on postoperative days 0, 2, 4, 7, and 10. Statistical analyses were performed using the paired t-test. Results On days 4, 7, and 10, the wounds treated with rose placenta were significantly smaller. On day 2, VEGF and EGF expression increased in the experimental group. On days 7 and 10, TGF-${\beta}1$ expression decreased in the experimental group. On day 10, vessel density increased in the experimental group. The increase in EGF on day 2 was confirmed with ELISA. Conclusions Rose placenta was found to be associated with improved wound healing in a mouse full-thickness wound model via increased EGF release. Rose placenta may potentially be a novel drug candidate for enhancing wound healing.
