Emulsification Characters of COY (Cooking Oil and Egg Yolk Mixture) and Mixing Application with Sulfur Wettable Powder for Enhancing the Control Efficacies against Paprika Powdery Mildew (파프리카 흰가루병 방제용 난황유의 유화특성과 유황수화제와의 혼용 시 방제효과)

  • Emulsification characters were investigated for COY (Cooking oil and Egg Yolk Mixture) and control efficacies of COY was enhanced by supplemented Sulfur wp. against paprika powdery mildews. Amount of water added to one egg yolk and 100 ml olive oil affected tbe homogenization of mixtures to emulsion profoundly; those premixes with 5~10 ml water were too concentrated to be homogenized, hence not dispersible in water; those with 15~20 ml water were homogenized, but tended to flocculation and not readily dispersible in water, suggesting the instability of emulsion; and those with 50 or 100 ml water were homogenized well and was readily dispersible in water to stable emulsion over long period of time. It was further confirmed that those fruits sprayed with not-fully emulsified COY revealed the oily membranes or blotches on their surfaces, whereas the ones with fully emulsified COY revealed normal clean surfaces. Treatment of COY either alone or in supplementation with Sulfur wettable powder (sulfur wp) were also effective in suppressing the recurrence of powdery mildew signs, wbich had disappeared in three days of foliar application. Still, the COYs supplemented with sulfur wp (COY+sulfur wp) at 1,000x or 5,000x were significantly effective in suppression against the symptomlsign recurrence over COY or sulfur wp alone.

  • The non infecting, plant associated bacteria have attracted increased attention for stimulating plant growth and as environmental friendly plant protecting agents. Pink-pigmented facultatively methylotrophic bacteria (PPFMs), classified as Methylobacterium spp., are persistent colonizers of plant leaf surfaces. As the leaves of most or all plants harbor PPFMs that utilize leaf methanol as their sole source of carbon and energy, which is a specific attribute of the genus Methylobacterium. Although they are not well known, these bacteria are co-evolved, interacting partners in plant metabolism. This claim is supported, for example, by the following observations: (1) PPFMs are seed-transmitted, (2) PPFMs are frequently found in putatively axenic cell cultures, (3) Low numbers of seed-borne PPFMs correlate with low germinability, (4) Plants with reduced numbers of PPFM show elevated shoot/root ratios, (5) Foliar application of PPFMs to soybean during pod fill enhances seed set and yield, (6) Liverwort tissue in culture requires PPFM-produced vitamin B12 for growth, (7) treated plants to suppress or decrease disease incidence of sheath blight caused by Rhizoctonia solani in rice, and (8) the PPFM inoculation induced number of stomata, chlorophyll concentration and malic acid content, they led to increased photosynthetic activity. Methylobacterium spp. are bacterial symbionts of plants, shown previously to participate in plant metabolism by consuming plant waste products and producing metabolites useful to the plant. There are reports that inform about the beneficial interactions between this group of bacteria and plants. Screening of such kind of bacteria having immense plant growth promoting activities like nitrogen fixation, phytohormone production, alleviating water stress to the plants can be successfully isolated and characterized and integration of such kind of organism in crop production will lead to increased productivity.

Effect of Location, Season, and Variety on Yield and Quality of Forage Oat

  • Forage oat (Avena sativa L.) is grown extensively in a double-cropping system on dairy farms. Four oat varieties ('Swan', 'Targa', 'Foothill', and 'Nugene') were evaluated for forage production and forage quality during two growing seasons (spring and autumn) at two locations (central and southern region) in South Korea. The heading stage of four oat varieties was observed during spring, but the autumn season did not produce heading until harvest time except for the 'Swan' variety (early-maturing variety). The heading stage of 'Swan' in both locations was earlier compared to other varieties. The four varieties were resistant to both foliar disease and insects. Lodging resistance was higher during autumn except in 2002 at the central region, and late-maturing varieties ('Foothill' and 'Nugene') have lower lodging resistance. Dry matter (DM) content was significantly different between varieties (p<0.001). Comparing different varieties, 'Swan', an early-maturing variety, was highest in DM content. In DM, total digestible nutrients (TDN) and crude protein (CP) yield, the yield of oat varieties in the southern region was higher than in the central region, and forage yield of the oat varieties in the spring season was higher than during the autumn season. The DM and TDN yield showed significant differences between oat varieties. The CP content of oats grown in the central region (Cheonan) was lower than oats grown in the southern region (Daegu), and the spring season produced oats with lower CP compared to the autumn season. Among the four oat varieties, the CP content of late-maturing varieties was higher than the Swan variety (early-maturing variety). The acid detergent fiber (ADF) and neutral detergent fiber (NDF) contents were higher for the varieties grown in Cheonan and during the spring season. The ADF and NDF contents of late-maturing varieties were lower than the early-maturing variety. TDN and relative feed value (RFV) were higher for the varieties grown in Daegu and during the autumn season. Late-maturing variety had higher TDN and RFV than early-maturing variety. Our study showed differences in forage production and forage quality of oats grown in different locations, seasons and varieties. Forage quality as well as forage production was better in the southern region than in the central region. Forage quality was better during autumn, but forage production was better during spring. Late-maturing variety had better forage quality than the early-maturing variety. Therefore, late-maturing varieties are more suitable for use in the southern region.

Improvement of Shelf-life and Quality in Fresh-Cut Tomato Slices:

  • Quality of fresh-cut tomato slices was compared during cold storage under various modified atmosphere packaging conditions. Chilling injury of slices in containers sealed with Film A was higher than with Film B; these films had oxygen transmission rates of 87.4 and 60.0 ml $h^{-1}\;m^{-2}\;atm^{-1}$ at $5^{\circ}C\;and\;99\%$ RH, respectively. While slices in containers with an initial atmospheric composition of air, $4\%\;CO_2+1\;or\;20\%\;O_2,\;8\%\;CO_2+1\;or\;20\%\;O_2,\;or\;12\%\;CO_2+20\%\;O_2$ showed fungal growth, slices in containers with $12\%\;CO_2+1\%\;O_2$ did not. Low ethylene in containers enhanced chilling injury. Modified atmosphere packaging provided good quality tomato slices with a shelf-life of 2 weeks or more at $5^{\circ}C$. Experiments were conducted to compare changes in quality of slices of red tomato (Lycopersicon esculentum Mill. 'Sunbeam') fruit from plants grown using black polyethylene or hairy vetch mulches under various foliar disease management systems including: no fungicide applications (NF), a disease forecasting model (Tom-Cast), and weekly fungicide applications (WF), during storage at $5^{\circ}C$ under a modified atmosphere. Slices were analyzed for firmness, soluble solids content (SSC), titratable acidity (TA), pH, electrolyte leakage, fungi, yeasts, and chilling injury. With both NF and Tom-Cast fungicide treatments, slices from tomato fruit grown with hairy vetch (Vicia villosa Roth) mulch were firmer than those from tomato fruit grown with black polyethylene mulch after 12 days storage. Ethylene production of slices from fruit grown using hairy vetch mulch under Tom-Cast was about 1.5- and 5-fold higher than that of slices from WF and NF fungicide treatments after 12 days, respectively. The percentage of water-soaked areas (chilling injury) for slices from tomato fruit grown using black polyethylene mulch under NF was over 7-fold that of slices from tomato fruit grown using hairy vetch under Tom-Cast. When stored at $20^{\circ}C$, slices from light-red tomato fruit grown with black polyethylene or hairy vetch mulches both showed a rapid increase in electrolyte leakage beginning 6 hours after slicing. However, slices from tomato fruit grown using the hairy vetch mulch tended to have lower electrolyte leakage than those grown with black polyethylene mulch. These results suggest that tomato fruit from plants grown using hairy vetch mulch may be more suitable for fresh-cut slices than those grown using black polyethylene mulch. Also, use of the disease forecasting model Tom-Cast, which can result in lower fungicide application than is currently used commercially, resulted in high quality fruit for fresh-cut processing. Experiments were conducted to determine if ethylene influences chilling injury, as measured by percentage of slices exhibiting water-soaked areas in fresh-cut tomato slices of 'Mountain Pride' and 'Sunbeam' tomato (Lycopersicon esculentum Mill.). Ethylene concentration in containers without ventilation significantly increased during storage at $5^{\circ}C$, whereas little or no accumulation of ethylene occurred in containers with one or six perforations. Chilling injury was greatest for slices in containers with six perforations, compared to slices in containers with one perforation, and was over 13-fold greater than that of slices in control containers with no perforations. An experiment was also performed to investigate the effectiveness of including an ethylene absorbent pad in containers on subsequent ethylene accumulation and chilling injury. While ethylene in the no-pad controls increased continually during storage of both 'Mountain Pride' and 'Sunbeam' tomatoes at $5^{\circ}C$ under modified atmosphere conditions, no increase in accumulation of ethylene was observed in containers containing ethylene absorbent pads throughout storage. The ethylene absorbent pad treatment resulted in a significantly higher percentage of chilling injury compared with the no-pad control. In studies aimed at inhibiting ethylene production using AVG during storage of slices, the concentration of ethylene in control containers (no AVG) remained at elevated levels throughout storage, compared to containers with slices treated with AVG. Chilling injury in slices treated with AVG was 5-fold greater than that of controls. Further, we tested the effect of ethylene pretreatment of slices on subsequent slice shelf-life and quality. In slices treated with ethylene (0, 0.1, 1, or $10\;{mu}L\;L^{-1}$) immediately after slicing, ethylene production in non-treated controls was greater than that of all other ethylene pre-treatments. However, pretreatment of slices 3 days after slicing resulted in a different pattern of ethylene production during storage. Ihe rate of ethylene production by slices treated with 1 L $L^{-1}$ ethylene 3 days after slicing was greater during storage than any of the other ethylene treatments. With slices pre-treated with ethylene, both immediately and 3 days after slicing, the rate of ethylene production tended to show an negative correlation with chilling injury. Effect of Exogenous Application of Salicylic Acid or Nitric Oxide on Chilling Tolerance and Disease Resistant in Pepper Seedlings (외생 살리실산과 일산화질소 처리가 고추묘의 저온 내성 및 병 저항성에 미치는 영향)

  • As an abiotic stress, chilling stress is one of the major factors limiting plant growth and increasing susceptibility to pathogens. Therefore, enhancing stress tolerance in plants is an important strategy for their survival under unfavorable environmental conditions. The objective of this study was to determine the effects of the exogenous application of salicylic acid (SA) or nitric oxide (NO) on chilling tolerance in pepper seedlings. Pepper (Capsicum annuum L. 'kidaemanbal') seedlings were grown under normal growing conditions ($20/25^{\circ}C$, 15 hours photoperiod, $145{\pm}5{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, fluorescence lamps) for 23 days after transplanting. The solution (3 mL) of 1 mM SA and 0.3 mM NO with surfactant triton 0.1% were sprayed two times a week, respectively. Right after the completion of chemical application, seedlings were subjected to chilling condition at $4^{\circ}C$ for 6 hours under dark condition and then the seedlings were recovered at the normal growing conditions for 2 days. In order to assess plant tolerance against chilling stress, growth characteristics, chlorophyll fluorescence (Fv/Fm), and membrane permeability were determined after chilling stress imposition. Total phenolic concentration and antioxidant capacity were measured during the whole experimental period. Disease incidence for pepper bacterial spot and wilt was also analyzed. Pepper seedlings treated with SA or NO were maintained similar dry mass ratio, while the value in control increased caused by chilling stress suggesting relatively more water loss in control plants. Electrolyte leakage of pepper seedlings treated with SA or NO was lower than that of control 2 days after chilling treatment. Fv/Fm rapidly decreased after chilling stress in control while the value of SA or NO was maintained about 0.8. SA increased higher total phenolic concentration and antioxidant capacity than NO and control during chemical treatment. In addition, increase in total phenolic concentration was observed after chilling stress in control and NO treatment. SA had an effect on the reduction of bacterial wilt in pepper seedlings. The results from this study revealed that pre-treatment with SA or NO using foliar spray was effective in chilling tolerance and the reduction of disease incidence in pepper seedlings.

  • Two species of Phytophthora were isolated from a leaf blight disease found on castor bean plants growing at a residential area of Hwanggeum-dong in Daegu city. One species was producing conspicuously papillate, noncaducous, ovoid to obpyriform sporangia, 31.2-58.5 ${\times}$ 25.4-44.1 ${\mu}{\textrm}{m}$ in range with an average of 46.4 $\pm$ 6.5 ${\times}$ 35.9 $\pm$ 4.7 ${\mu}{\textrm}{m}$ on simple sympodial sporangiophores, was heterothallic with oogonia globose, 22.5-35.0 ${\mu}{\textrm}{m}$ with an average of 30 $\pm$ 3.3 ${\mu}{\textrm}{m}$ in diameter, oospores plerotic, 18.8-30.0 ${\mu}{\textrm}{m}$ in range averaging 25.4 $\pm$ 2.8 ${\mu}{\textrm}{m}$, and with antheridia amphigynous. This species was identified as P. nicotianae Breda de Haan. The other species produced papillate, caducous, ovoid to ellipsoid sporangia with pedicel, 21.5-54.6 ${\times}$ 17.6-34.3 ${\mu}{\textrm}{m}$ in range with an average of 41.7 $\pm$ 7.1 ${\times}$ 28.4 $\pm$ 4.0 ${\mu}{\textrm}{m}$, and was heterothallic with oogonia globose, 21.3-26.8 ${\mu}{\textrm}{m}$ with an average of 23.0 $\pm$ 1.6 ${\mu}{\textrm}{m}$ in diameter, oospores plerotic, 17.5-23.8 ${\mu}{\textrm}{m}$ in range averaging 20.2 $\pm$ 1.8 ${\mu}{\textrm}{m}$, and with antheridia amphigynous. This species was identified as P. palmivora (Butler) Butler. Both species produced chlamydospores. Pathogenicity of the 2 speices was confirmed by foliar inoculation of castor bean seedlings. Both species have been reported to infect castor bean plants in a few foreign countries but it is the first that a disease of castor bean plants caused by either or both of the species was found in Korea.

Diagnosis and management of the early defoliation of Korean flowering cherry in Gurye (구례 왕벚나무 가로수의 조기낙엽 진단 및 친환경적 관리 방법)

  • The Korean flowering cherry is a popular tree. However, the trees have started to defoliate early, including those in Gurye. Thus, it is necessary to identify the causes of the early defoliation and how to manage them. Therefore, the purpose of this study was threefold: 1) to investigate the early defoliation of flowering cherry trees, 2) to identify the differences in growth and flowering of the trees in response to treatment, and 3) to suggest an optimal treatment for the trees. The experiment was conducted in Gurye at a site 3km long with 102 flowering cherry trees along a street. There were three treatments: control, an environmentally friendly insecticide and a disinfectant(treatment 1), and an environmentally friendly insecticide, a disinfectant, and irrigation (treatment 2). The trees in Gurye were compared to trees on Jeju Island. The defoliation rates of the flowering cherry in Gurye were significantly higher than those on Jeju Island at each measurement time. Within Gurye, the defoliation rate was significantly higher in the trees of the control than in the two treatments and only the trees in the control from Gurye had shot-hole disease. Post-treatment, twigs collected from the control during April showed significantly poorer growth, lower numbers of flowering buds per twig, and lower numbers of foliar buds. The trees given treatment 1 showed significantly greater growth, number of total buds, flowering buds per twig, and density of buds than the trees given treatment 2. After analyzing the correlations, it was established that early defoliation led to poorer growth, affecting the flowering of the trees.

Functional Analysis of Bacillus subtilis Isolates and Biological Control of Red Pepper Powdery Mildew Using Bacillus subtilis R2-1 (Bacillus subtilis 균주의 기능성 분석 및 이를 활용한 고추 흰가루병 방제)

  • The multi-function of 18 Bacillus subtilis isolates collected from agricultural extension centers of local government and National Academy of Agricultural Science was investigated by measuring their antifungal activities against five plant pathogens, such as Rhizoctonia solani, Colletotrichum acutatum, Fusarium oxysporum, Magnaporthe oryzae and Phytophthora capsici, phosphorus solubilization ability, production of indole acetic acid (IAA) and siderophore, and nitrogen fixation. The B. subtilis isolates showed antifungal activity against several plant pathogens and nitrogen fixation activity, and produced siderophore and IAA. They could control pepper powdery mildew (Leveillula taurica), but there was no difference in control efficacy among the B. subtilis isolates. In fields, the control efficacy of B. subtilis R2-1 ($10^8$ cells/ml) was compared with two microbial fungicides, Q-pect and Topsid. In 2009, the control efficacy of B. subtilis R2-1 (37.7%) was lower than that of Topsid (47.6%), but higher than that of Q-pect (25.7%). In 2010, the control efficacy of B. subtilis R2-1 (83.3%) was higher than that of Topsid (67.9%). In order to elucidate mode of action of B. subtilis R2-1 for controlling pepper powdery mildew, spore germination rates of pepper powdery mildew pathogen collected on treated leaves was investigated when suspensions of B. subtilis R2-1 and two microbial fungicides (Q-pect and Topsid) were foliar-sprayed. They highly suppressed spore germination of the pathogen with inhibition values of 84.2% for B. subtilis R2-1, 97.9% for Q-pect and 94.7% for Topsid. Further study on the mass-culturing method and formulation is needed for development of a microbial fungicide.

Characterization of an Isolate of Cucumber mosaic virus Isolated from Canna generalis Bailey (칸나에서 분리한 Cucumber mosaic virus의 특성)

  • An isolate of Cucumber mosaic virus(CMV), called as Can-CMV, was originally isolated from Canna generalis showing typical streak mosaic foliar symptoms, and its properties were investigated in this study. Whereas all known isolates of CMV could induce symptoms on their systemic hosts(four kinds of Nicotiana spp and a zucchini squash), Can-CMV induced no symptoms on its systemic hosts tested. Replication and movement of the virus on upper leaves as well as inoculated leaves-were confirmed by RT-PCR suggesting that Can-CMV could only infect systemically on N. benthamiana and N. glutinosa. Size of local lesions on the Can-CMV-inoculated leaves of Chenopodium amaranticolor was much smaller than that of Fny-CMV. Whereas Fny-CMV and LS-CMV could induce distinct necrotic local lesions on Vigna unguiculata 2 to 3 days postinoculation(dpi), chlorotic spots symptom was expressed by Can-CMV 4 to 5 dpi. Virus-specific 4 kinds of dsRNAs were isolated from leaves of N. benthamiana infected with Can-CMV, and these dsRNAs corresponded to the viral genomic RNAs and subgenomic RNAs and their patterns were indistinguishable to those of Fny-CMV and LS-CMV. By restriction mapping analysis of 950 bp of RT-PCR amplified products of coat protein gene of the virus as well as by serological analysis of gel diffusion test, Can-CMV belongs to a typical member of CMV subgroup IA. These results suggest that the Can-CMV isolated from C. generalis possesses unique pathological properties to understand further insight into the various interactions between virus and host.

Effect of Yacon (Smallanthus sonchifolius H. Robinson) Extracts on Herbicidal, Fungicidal, and Insecticidal Activities (야콘(Smallanthus sonchifolius H. Robinson) 추출물의 제초, 살균 및 살충활성 효과)

  • This research has been carried out to find out the highest effect on insecticidal, fungicidal and herbicidal activities in leaves, stem and tuber extracts of yacon (Smallanthus sonchifolius) and extraction methods such as water, boiling water and methanol. Characteristics of potential herbicidal components among extraction methods were investigated by solvent fractions such as hexane, chloroform, ethyl acetate, butanol, and water. Generally, methanol extract was the best on inhibition of germination rate, plant height, and root length in cucumber and barley. On the other hand, the inhibition effect on growth in cucumber and barley was the best in tuber among plant parts of yacon. Inhibition of germination rate, plant height, and root length in cucumber and barley in solvent fractions was the best in water fraction, but there were no differences in other fractions. Digitaria sanguinalis L. and Solanum nigrum L. by 5 and 10% extractions of yacon tuber were controlled by more than 70~80% and 95~100%, respectively. However, there was no inhibition effect on foliar treatment in cucumber and barley as affected by 5 and 10% extractions of yacon tuber. Mortality of green peach aphid (Myzus persicae Sulzer) was 50% at 3 days after treatment of 5% extracts of yacon leaves. Mortality of brown plant hopper (Nilaparvata lugens Stal) was only 24% in 5% extracts of stems and leaves with midrib, but was 57% in 5% extracts of leaves without midrib. There was no fungicidal effect on anthracnose (Colletotrichum truncatum), wilt disease (Fusarium oxysporum), verticillium wilt (Verticillium dahliae), bacterial blight (Xanthomonus oryzae) in 5% extracts of yacon leaves.
