Morphological Study on the Epithelial Cells in the Gall Blader of Vertebrates (각급 척추동물의 담낭 상피세포에 대한 형태학적 연구)

  • These experiments were performed in order to study histologically and histochemically on the epithelial cells of gall bladder in Carassius carassius, Bufo bufo gargarizans, Natrix tigrina lateralis, Urloncha striata var. domesticus and Bos taurus var. domesticus. The results of the observation were as follows: 1. There were different cell types in the epithelium of gall bladder in each animal and it could not be supported histochemically that the epithelia cells of gall bladder were divided into two cell types of the rod-shaped and barrel-shaped ones. 2. The epithelium of gall bladder in Carassius carassius, Bufo bufo gargarizans, Natrix tigrina lateralis, Urloncha striata var. domesticus and Bos taurus var. domesticus was simple columnar epithelium. 3. The eosinophilities of cytoplasm in the epithelial cells of gall bladder were in uniform stronger in the upper portion of nucleus in Carassius carassius, Bufo bufo gargarizans and Natrix tigrina lateralis than its other portions, and in Urloncha striata var. domesticus and Lepus cuniculus var. domesticus existed uniformly in all portions, but there were many non-eosinophilic cells in Bufo bufo gargarizans and many cells that weakly eosinophilic around nucleus in Bos taurus var. domesticus. 4. The periodic acid Schiff's reactivities in the epithelial cells of gall bladder were different in each other and the epithelial cells in PAS reaction were divided into two cell types of the dark and light ones. There presented the light cells of 6.4%, 4.3% and 3.7% of epithelial cell of gall bladder in Carassius carassius, Bufo bufo gargarizans and Urloncha striata var. domesticus for each other, but were not presented in Natrix tigrina lateralis and Bos taurus var. domesticus. 5. The ninhydrin-Schiff-active proteins were much in the epithelial cells of gall bladder in Bos taurus var. domesticus, Carassius carassius, Urloncha striata var. domesticus and Natrix tigrina lateralis in order and were much in epithelial cells in the upper portion of mucosal folds in Carassius carassius, Urloncha striata var. domesticus and Natrix tigrina lateralis, and the ninhydrin-Schiff-active protein of the epithelium of gall bladder in Bufo bufo gargarizans was uniformly distributed. 6. The epithelial cells of gall bladder in Carassius carassius, Natrix tigrina lateralis, Urloncha striata var. domesticus and Bos taurus var. domesticus had no stain reactivity or weak stain reactivity to neutral fat and all epithelial cells in Bufo bufo gargarizans had strong stain reactivity, though they were different in quantity of epithelial cell portion. 7. The stain reactivities to RNA and DNA were stronger in the epithelial cells of the upper portion of mucosal fold than in those of other portions.

  • Objective : Thread embedding acupuncture has become popular as a minimally invasive treatment for facial wrinkles and laxity. However, there is little published clinical practice guidelines. This study is to developing a specific procedure of thread embedding acupuncture for facial wrinkles and laxity. Method : We reviewed and summarized 6 studies on thread embedding acupuncture for facial wrinkles and laxity. And, four practitioners who have more than four year of clinical experience and one hundred of cases were participated in developing a thread embedding acupuncture procedure for facial wrinkles and laxity. Result and conclusion : We developed a thread embedding acupuncture procedure for facial wrinkles and laxity which consists of correction of lower jaw, facial laxity, nasolabial fold and eye wrinkles.

A Case of Adenomyoma of the Stomach in a Child Presenting Epigastric Pain (심와부 복통으로 내원한 5세 소아에서의 위 선근종 1례)

  • Adenomyomas of the stomach are rare tumors characterised by duct/gland-like structures embedded within a smooth muscle stroma. A 5-year-old female patient was admitted to the department of Pediatrics, Ewha Womans University MokDong Hospital with the history of severe epigastric pain and vomiting for 1 day. Esophagogastroduodenoscopy showed bridging fold with central dimpling on posterior wall of prepyloric antrum. Endoscopic biopsy was nondiagnostic. The patient complained epigastric pain continuously and underwent wedge resection. Pathologic examination showed an adenomyoma of the prepyloric antrum. After wedge resection, the patient did not complain epigastric pain during the postoperative follow-up. We report an unusual case of an adenomyoma of stomach.

First Record of Gobiid Fish, Astrabe fasciata (Perciformes: Gobiidae) from Dokdo, Korea (독도에서 채집된 망둑어과 어류 1미기록종, Astrabe fasciata)

  • The one specimen of Astrabe fasciata, belonging to the family Gobiidae was collected from the Dokdo of East Sea in Korea, and described as the first record from Korea. The specimen is characterized by having two dorsal fins, pectoral fin with free rays (five upper, two lower), protrusion on upper posterior part of supraorbital dermal fold, white spots dispersed on head and five white transverse bends on the body. We proposed its Korean name "Dok-do-eol-ruk-mang-duk-sok" and "Dok-do-eol-ruk-mang-duk" for the genus and species, respectively.

Anti-wrinkle Activity of $\beta$-carotene Extracted & Purified from Recombinant Escherichia coli (재조합 대장균으로부터 추출.정제된 베타-카로틴의 주름개선 활성)

  • This paper described the extraction/purification of $\beta$-carotene from recombinant E.coli and evaluation of anti-wrinkle activity of purified $\beta$-carotene. No significant differences in extraction yields were observed when hexane or isobutyl acetate was used. However, extraction from wet-cell cake resulted in 2-fold higher amount of $\beta$-carotene than that from dry cells. Disruption of 5 g-wet cells by ultrasonic homogenizer, acetone dehydration, extraction with isobutyl acetate resulted in 36 mg of $\beta$-carotene corresponding to 61.2% of recovery. The formation and separation of $\beta$-carotene crystal improved the purity. 633 mg of $\beta$-carotene crystal with 93% purity was obtained from 223 g/L of wet-cell cake harvested from 2.5-L fed-batch culture broth. The cultures of normal human primary fibroblast were performed to investigate the effect of $\beta$-carotene on cytotoxicity as MTT assay and anti-wrinkle activity as collagen synthesis assays. $1.7{\mu}M$ of $\beta$-carotene was found to be optimal concentration at which 1.4-fold higher amount of collagen was synthesized than that in absence of $\beta$-carotene. This indicates that highly purified $\beta$-carotene can be obtained from recombinant E.coli by applying simple method with less toxic solvent and can be used in functional cosmetics as anti-wrinkle agent.

The Structure of Digestive Tract and Histological Features of the Larvae in Sevenband Grouper, Epinephelus septemfasciatus (능성어 (Epinephelus septemfasciatus) 자어의 소화기관 구조 및 조직학적 특징)

  • Histological and morphological development of the digestive tract of sevenband grouper were observed from after hatching to 60 days. Fishes were fed with rotifer (Brachionus rotundiformis) and chlorella (Chlorella ellipsoidea) after hatching from 2 to 20 days rotifer and brine shrimp (Artemia salina) in after 20 days rotifer, brine shrimp and semi-dry artificial diet in after 23 days. Histological and morphological development of ten larvae was observed by paraffin embedding method after fixing in 10% neutral buffered formalin. Sevenband grouper RLG showed characteristics of carnivorous fish by average 0.87. Larvae after hatching can't open the mouth and anus digestive tract was observed in a straight line following yolk sac. Larvae was observed feeding activity by opening the mouth and anus. Metamorphosis started 8 days after hatching. Esophagus divided four layer, and goblet cell was observed in esophagus, mid intestine and rectum. Larvae started cannibalism and it was caused by difference of growth. The inside of stomach was differentiated to cardiac orifice, body of stomach, pyloric stomach, and pyloric caeca. Goblet cell was observed all intestine. Gastric gland differentiated after hatching 28 days in stomach. Secretion of gastric juice was found at stomach and mucosal fold pyloric caeca. Even thought the inside of stomach expended and the number of gastric gland increased consistently and goblet cell in intestine and mucosa became longer, histochemical changes follow couldn't be found during transforming juveniles 38 days after hatching.

Influence of The Neck acupressure massage and The Neck Cream Including Korean Herbal Extract on The Neck Wrinkles and Facial Blood Circulation. (경항부 수기요법과 한방 목 크림이 목 주름과 안면 혈행에 미치는 영향 연구)

  • Objectives: We investigated the influence of the neck acupressure massage and the neck cream including korean herbal extract on the neck wrinkles and blood circulation. Methods: Korean female($37.47{\pm}3.66$ years, n=18) aged 30-39 years were participated in this study. We measured the neck wrinkles with PRIMOS(GFMesstechnik GmbH, Germany) and blood circulation of neck and facial skin with Laser Doppler Imaging-Periscan PIM II(Perimed AB, Sweden). Results: The neck wrinkles were significantly decreased after applying neck cream. But when with the neck acupressure massage, the neck wrinkles were decreased 2-fold than applying only the neck cream. The skin blood circulation was significantly increased on the neck and facial skin immediately after the neck acupressure massage. But this effect was not significantly different after 8 weeks Conclusions: This result suggests that the neck acupressure massage can improve the anti-wrinkles effect of the neck cream including Korean herbal extract.

Developmental Study on the Argentaffin and Argyrophile Cells in the Gastroinestinal Mucosae of Rana nigromaculata (개구리(Rana nigromaculata) 위장관 점막내의 은친화성 세포와 은호성 세포에 대한 발생학적 연구)

  • This experiment was performed in order to study the morphological changes of the argentaffin and argyrophile cells in the gastrointestinal mucosae of Rana nigromaclata during development. The specimens from the stomach and the small intestine at different developmental stages were fixed in neutral 10% formalin, sectioned at a thickness of 3 microns, and impregnated by Masson's method for argentaffin cells and by Bodian's method for argyrophile cells. The results of observation were as follows: 1. In the stomach, the argentaffin cells appeared at XIII stage of metamorphosis and the argyrophile cells at X stage and they rapidly increased in number at XXV stage. 2. In the small intestine, the argentaffin cells appeared at XXV stage of metamorphosis and the argyrophile cells at XVII stage and they rapidly increased in number at XXV stage. 3. The argentaffin and argyrophile cells in the gastrointestinal mucosae, appeared prior to forming gastrointestinal mucosal fold following to development of muscle layer. 4. The rapid numeral increase of the argentaffin and argyrophile cells in the last stage of metamorphosis would be due to ecological changes and differentiations of gastrointestinal mucoae in amphilbia.

Early Vegetation Succession in Abandoned Field in Cheju Island (제주도 저지대 묵밭 식물군락의 2차 천이)

  • In order to clarify the successional trend and its characteristics of plant community in abandoned field in Cheju Island, we investigated the seral changes of vegetation height, coverage, growth form, species richness, diversity and dominance index along with the time lapse, and then compared with those of mainland researches. And in order to seek the overall trend of vegetation changes, we used cluster, TWINSPAN and PCA ordination techniques. The succession was characterized by the lower species number, lower vegetation height, longer period of herb dominant and later invasion of tree species. These results may come from that ecological traits of Cheju Island itself, and ecosystem fragmented influences that block a seed (propagule) transport. Sere of the dominant species was shown as follows: Mazus japonicus(0.5∼1 years)→Conyza sumatrensis (2 years)→Artemisia princeps var. orientalis(4 years)→Artemisia princeps var. orientalis, indigofera pseudotinctoria(5 years)→Rosa multiflora, Miscanthus sinensis, etc.(8 years)→Miscanthus sinensis(12 years)→Boehmeria nivea, Pueraria thunbergiana etc.(15 years)→Mallotus japonicus(20 years)→Litsea japonica, Machilus thunbergii (20 years<). Abandoned fields were classified into three groups according to time lapse; earlier stage(0∼1 years), middle stage(2∼8 years) and later stage(8∼20 years).

Antitumor and Immuno-potentiating Activities of Crude Polysaccharides from Fruiting Body of Agaricus brasiliensis (신령버섯(Agaricus brasiliensis) 자실체 추출 조다당류의 항암 및 면역증강 작용)

  • Agaricus brasiliensis, one of edible mushroom belonging to Basidiomycota, has been used for curing gastric ulcer and stomach cancer of human beings and also known to have good inhibitory effects on sarcoma 180 and Ehrlich carcinoma of mice. Neutral saline soluble (0.9% NaCl), hot water soluble and methanol soluble substances (hereinafter referred to Fr. NaCl, Fr. HW and Fr. MeOH, respectively) were prepared from fruiting body of the mushroom. ${\beta}$-glucan and total protein contents were identify from fractions of edible mushrooms extract. The ${\beta}$-glucan and protein contents of all fractions of the mushrooms ranged from 21.54~32.31% and 0.16~9.34%, respectively. In vitro cytotoxicity tests, crude polysaccharides were not cytotoxic against cancer cell lines such as Sarcoma 180, HT-29, NIH3T3 and RAW 264.7 at the concentration of 10~2000 ${\mu}g/ml$. Intraperitoneal injection with crude polysaccharides exhibited life prolongation effect of 18.8~50.6% in mice previously inoculated with Sarcoma 180. Fr. HW increased the numbers of spleen cell by 1.2 fold at the concentration of 200 ${\mu}g/ml$ compared with control. Fr. MeOH and Na improved the immuno-potentiating activity of B lymphocyte by increasing the alkaline phosphatase activity by 1.6 fold compared with control at the concentration of 50~500 ${\mu}g/ml$. Fr. Na generated 15.9 ${\mu}M$ of nitric oxide (NO) when cultured with RAW 264.7 at the concentration of 200 ${\mu}g/ml$, while lipopolysaccharide, a positive control, produced 3.7 ${\mu}M$. The Fr. NaCl, Fr. HW and Fr. MeOH increased the secretion of TNF-${\alpha}$, IL-$1{\beta}$, Il-2 and IL-6 by 2.2 times compared with the control group. Fr. Na increased the numbers of peritoneal exudate cells by 4 folds at the concentration of 50mg/kg compared with control. Circulating leukocytes increased by 2.7 folds when Fr. HW from A. brasiliensis was inoculated at the concentration of 50 mg/kg body weight. The hematological and blood chemical analysis of the 3 fractions did not show any difference in blood compositions and enzyme activities compared with the control group (p<0.05). Therefore, the experimental results suggested that crude polysaccharides extracted from A. brasiliensis contain antitumor and immuno-potentiating activities against Sarcoma 180 in ICR mice.
