Bacterial strains isolated from Jeotgal (salted seafood) induce maturation and cytokine production in mouse bone marrow-derived dendritic cells (마우스 골수 유래 수지상세포의 성숙과 사이토카인 생산에 대한 젓갈 분리균의 효과 연구)

  • Jeotgal (salted seafood) has been one of major fermented foods in Korea for long time. Although there are many studies about Jeotgal in various aspects of food, its immunological importance on hosts has not been elucidated yet. In this study, we investigated if several bacteria isolated from Jeotgal may modulate the function of dendritic cells (DCs), powerful antigen-presenting cells equipped with special immunological capabilities. 4 Jeotgal bacteria were selected as representatives and used for experiments. To treat viable DCs, those bacteria were killed at $60^{\circ}C$ for 30 min. The viability of DCs treated with Jeotgal bacteria was verified and two isolates significantly induced high production of interleukin-12, a representative cell-mediated cytokine of DCs. Surface activation and maturation markers (MHC class II, CD40, CD86) of DCs were analyzed by flow cytometer. In addition, the treated DCs showed significantly high lymphocyte stimulatory capability compared to control DCs based on allogeneic mixed lymphocyte reactions. These observations suggest that Jeotgal isolates can function as immunostimulating bacteria in hosts, like Lactobacillus. Taken together, these experimental evidences may broaden the use of Jeotgal isolates in immunological fields in addition to as a fermented food.

Effect of silver nanoparticles on the performance of riverbank filtration: Column study (강변여과에서의 은나노입자의 영향 : 실험실규모 컬럼 실험)

  • Soil column experiments were evaluated effects of silver nanoparticles (i.e., 0, 2.5, 5, and 10 mg/L) on the microbial viability which is strongly associated with the degradation of organic matter, pharmaceutically active compounds(PhACs) and biological oxidation of nitrogenous compounds during river bank filtration. The addition of silver nanoparticles resulted in almost no change in the aqueous matrix. However, the intact cell concentration decreased with addition of silver nanoparticles from 2.5 to 10 mg/L, which accounted for 76% to 82% reduction compared to that of control (silver nanoparticles free surface water). The decrease in adenosine triphosphate was more pronounced; thus, the number and active cells in aqueous phase were concurrently decreased with added silver nanoparticles. Based on the florescence excitation-emission matrix and liquid chromatograph - organic carbon detection analyses, it shows that the removal of protein-like substances was relatively higher than that of humic-like substances, and polysaccharide was substantially reduced. But the extent of those substances removed during soil passage was decreased with the increasing concentration of silver nanoparticles. The attenuation of ionic PhACs ranged from 55% to 80%, depending on the concentration of silver nanoparticles. The attenuation of neutral PhACs ranged between 72% and 77%, which was relatively lower than that observed for the ionic PhACs. The microbial viability was affected by silver nanoparticles, which also resulted in inhibition of nitrifiers.

Runoff Characteristics and Non-point Source Pollution Loads from Cheongyang-Hongseong Road (청양-홍성간 도로에서의 강우 시 비점오염 유출특성 및 오염부하량 분석)

  • Nowadays, the importance of non-point source pollution treatment is being emphasized. Especially, the easy runoff characteristic of highly concentrated pollutants in the roads makes the circumstance more complicated due to impermeability of roads. When the pollutants flow into steam it could make water quality in stream worse and it also causes a bad influence in the aquatic ecosystem because the effluents of rainfall-runoff may contain indecomposable materials like oil and heavy metals. Therefore, we tried to figure out the property of non-point source pollution when it is raining and carried out an assessment for the property of runoff for non-point source pollution and EMC (Event Mean Concentrations) of the essential pollutants during this study. As the result of the study, the EMC was BOD 5.2~21.7 mg/L, COD 7.5~35.4 mg/L, TSS 71.5~466.1 mg/L, T-N 0.682~1.789 mg/L and T-P 0.174~0.378 mg/L, respectively. The decreasing rate of non-point pollutant in Chungyang-Hongsung road indicates the maximum decrease of 80% until 5 mm of rainfall based on SS concentration; by the rainy time within 20~30 minutes, the decreasing rate of SS concentration was shown as 88.0~97.6%. Therefore it was concluded that it seems to be possibly control non-point pollutants if we install equipments to treat non-point pollutants with holding capacity of 30 min. It is supposed that the result of this study could be used for non-point pollutants treatment of roads in Chungyang-Hongsung area. We also want to systematically study and consistently prepare the efficient management of runoff from non-point source pollution and pollutant loading because the characteristics of non-point source pollution runoff changes depending on different characteristics and situations of roads and rainfall.

A Study on the Impulse Waves Discharged from the Exit of the Convergent and Divergent Pipes (축소관과 확대관 출구로부터 방출되는 펄스파에 관한 연구)

  • The present study is to investigate the propagation characteristics of the impulse waves discharged from the exit of the convergent and divergent pipes. An experiment is carried out using a shock tube with an open end and is compared to the computation of the axisymmetric, compressible, unsteady Euler equations, which are solved by the second-order total variation diminishing (TVD) scheme. For the computational work, several initial compression waves are assumed inside the pipe so that those are the same to the experimental ones of the shock tube. The results show that the peak pressures of the impulse waves discharged from the exit of convergent and divergent pipes decrease with an increase in the wavelength of the initial compression wave. All of the impulse waves have a strong directivity toward the pipe axis, regardless of the exit type of the pipe employed. The impulse waves discharged from the divergent pipe are stronger than those from the straight pipe, while the impulse waves of the convergent pipe are weaker than those from the straight pipe. It is found that the convergent pipe can play a role of a passive control to reduce the peak pressure of the impulse wave. The present computations represent the experimented impulse waves with a good accuracy.

  • The experimental study on the reduction of $In_2O_3$ was performed by using thermogravimetric analyzer. The reduction of $In_2O_3$ was occurred at above $300^{\circ}C$. The reduction rates were rapidly increased with the reaction temperature, whilehardly affectedby the flow rate of hydrogen gas. It was found that the unreacted core model could be applied for the analysis of the reduction data and the rate control step was the chemical reaction of $In_2O_3$ with hydrogen on the surface of unreacted $In_2O_3$. The apparent activation energy for this reaction was 20kcal/g-mol $H_2$ and the rate equation of $In_2O_3$ reduction with hydrogen could be expressed in the following equation. ${\frac{dX}{dt}}=1.6{\times}10^5e^{-20000/RT}(1-X)^{2/3}$

Activation of Caspase-3 and -7 on Porcine Bone Marrow Derived Mesenchymal Stem Cells (pBM-MSCs) Cryopreserved with Dimethyl Sulfoxide (DMSO) (동결 보호제(DMSO) 농도에 따른 돼지 중간엽 줄기세포의 Caspase 3과 7 발현)

  • Adult stem cell transplantation has been increased every year, because of the lack of organ donors for regenerative medicine. Therefore, development of reliable and safety cryopreservation and bio-baking method for stem cell therapy is urgently needed. The present study investigated safety of dimethyl sulfoxide (DMSO) such as common cryoprotectant on porcine bone marrow derived mesenchymal stem cells (pBM-MSCs) by evaluating the activation of Caspase-3 and -7, apoptosis related important signal pathway. pBM-MSCs used for the present study were isolated density gradient method by Ficoll-Paque Plus and cultured in A-DMEM supplemented 10% FBS at $38.5^{\circ}C$ in 5% $CO_2$ incubator. pBM-MSCs were cryopreserved in A-DMEM supplemented either with 5%, 10% or 20% DMSO by cooling rate at $-1^{\circ}C$/min in a Kryo 360 (planner 300, Middlesex, UK) and kept into $LN_2$. Survival rate of cells after thawing did not differ between 5% and 10% DMSO but was lowest in 20% DMSO by 0.4% trypan blue exclusion. Activation of Caspase-3 and -7 by Vybrant FAM Caspase-3 and -7 Assay Assay Kit (Molecular probes, Inc.OR, USA) was analyzed with a flow cytometer. Both of cryopreserved and control groups (fresh pBM-MSCs) were observed after the activation of Caspase-3 and -7. The activation did not differ between 5% and 10% DMSO, but was observed highest in 20% DMSO. Therefore 5% DMSO can be possibly used for cell cryopreservation instead of 10% DMSO.

The Effect of the Combined Operation of Sprinkler and Vent Systems on the Smoke Control in a Horizontal Corridor (스프링클러와 배연설비의 통합작동이 수평통로의 연기제어에 미치는 영향)

  • Fire scenarios in a space $20.0m\;\times\;4.0m$ floor and 3.0m high were simulated by using computational fluid dynamics program (FDS 4.0.3) to investigate the effect of the combined operation of both sprinkler and vent systems, which are installed for cooling and blocking smoke which propagates beneath the ceiling of a horizontal corridor, on the temperature and smoke density of it. It was shown that the combined operation both sprinkler and vent systems was more effective than each operation for cooling and blocking smoke, the number of operating sprinklers was two because of corner effect of wall, and over-installed sprinklers deteriorated the effect of cooling and blocking smoke. This study showed that the case of two sprinklers and vent flow rate $3.0m^3/s$ in fire scenario was the most effective for cooling and blocking smoke. It was confirmed that the smoke downdrag occurs in operating sprinkler system, and the more smoke droplets produced by increasing fire size, the greater smoke downdrag occurred.

A Study for Pressure Difference and Critical Velocity by Pressurization of Elevator Shaft at High Rise Apartment (고층 공동주택의 승강로가압을 이용한 차압 및 방연풍속에 관한 연구)

  • It is not recommended that elevator use for egress at (super) high rise buildings because elevator shaft main roles to spread of fire smoke. But in North America used to protect this area by elevator shaft pressurization. These tests are performed at high rise apartment to verify that elevator shaft pressurization can protect to spread of fire smoke or not. and verify to used for egress at fire. Pressurization at elevator shaft make pressure difference of 50 Pa all floor at 150 CMM because this method have low friction loss from air flow. Also when dwelling door and elevator door are opened that critical velocity is performed to protect of back-layering from fire room for escape routs by 180 CMM. Therefore through out these pressurization tests by elevator shaft are estimated to have less overpressure because supply air difference are low between to satisfy critical velocity at one door opened and maintain to pressure difference all doors closed. Finally we verified that disable or residual people can use elevator for egress at fire by elevator shaft pressurization.

Antioxidant Effect of some Chelating Agents on Soybean Oil (식용대두유에 대한 Chelating agent의 항산화 효과)

  • Some chelating agents are evaluated as an antioxidant for the autoxidation of soybean oil. Soybean oil is autoxidized under a mild condition (the flow rate of 67ml $O_{2}/min$ and $50^{\circ}C$). The antioxidant effect is measured by active oxygen method, and the spectral change of autoxidized soybean oil examined. The antioxidant effect of chelating agents is increased in order of diphenic acid, naphthoquinone, pyromellitic acid, quinolinic acid and naphthalic acid, and particularly the effect is low in diphenic acid and naphthoquinone. It is found that the effect is more clearly demonstrated by NMR rather than IR and UV and that the effect is dependent on the functional group and geometric molecular structure of chelating agents.

Antioxidant Effect of some Phenolics on Soybean Oil (식용대두유에 대한 Phenolics의 항산화 효과)

  • Some phenolics were examined as an antioxidant for the autoxidation of soybean oil. Soybean oil was autoxidized under a mild condition (the flow rate of 67ml $O_{2}/min$ and $50^{\circ}C$). The antioxidant effect was estimated by active oxygen method. Phenolics show distinctive antioxidant effect, and the effect is prominent when cupper or iron was added. Phenolics showed a tendency to increase antioxidant effect with an increase of the number of hydroxyl group, and the increasing order was ferulic acid, quinalizarin, sesamol, alizarin, fisetin and purpurogallin. However, the effect was remarkably low in ferulic acid, alizarin and quinalizarin. It was found that the antioxidant effect was dependent on the functional group and geometric molecular structure of phenolics.
