Effects of Dietary Cheongkukjang on Liver Superoxide Dismutase Activity of Parrotfish Oplegnathus fasciatus (사료 내 분말 청국장이 돌돔, parrootfish, Oplegnathus fasciatus의 간 내 superoxide dismutase 활성에 미치는 영향)

  • A four-week feeding trial was conducted to investigate the effects of dietary soybean meal (SBM) and powdered Cheongkukjang (CKJ) on non-specific immune responses of parrotfish Oplegnathus fasciatus. Three isonitrogenous (42% crude protein) and isocaloric (17.1 MJ/kg) diets were formulated to replace fish meal by 0, 25% SBM or 25% CKJ (designated as FM, 25SBM and 25CKJ, respectively). Ninety fish (initial body weight 122 g) were randomly allotted into nine 150 L tanks. One of the three experimental diets was fed to triplicate groups of fish for 4 weeks. After the feeding trial, no differences were observed in growth performances and feed utilization among fish groups. Liver superoxide dismutase activity of the fish fed CKJ containing diet was significantly higher than that of the control groups. DPPH radical scavenging and $Fe^{2+}-chelating$ activities of the experimental diets containing SBM or powdered CKJ were significantly higher than that of the control diet. The results of the present study suggest that dietary inclusion of powdered 25CKJ significantly increased liver superoxide dismutase activity and did not affect the growth performances, feed utilization, morphological parameters, as well as hematological values of parrotfish.

  • Ichthyophonus disease had broken out among rock bream in Dec. 1989. Diseased-fish showed markedly stunted growth and darkish coloration, and anatomically the liver with small white nodules, tumefied spleen with its granulous surface, and the markedly tumefied kidney. Microscopic examination of liver, kidney, spleen and gill from 10 rock bream revealed cyst of the fungus Ichthyophonus sp. Rock bream were heavily infected with the highest concentrations of spherical multinucleate bodies in the liver, the spleen, the kidney and the gills. In heavily infected sectors of tissue a common necrotic zone was formed around spore aggregates. Spherical multinucleate hyphal terminal bodies developed thin hyphae longer than 1mm which divided into many branches.

Studies on the Acute Toxicity and Histological Changes in Fish Exposed to Furrural (Furfural 어류급성독성 및 조직병리에 관한 연구)

  • Furfural, an organic solvent, is widely used as synthetic component material in producing chemical products. However, furfural has been reported that it shows strong toxicities to human being showing intense stimulus to skin, eyes, mucous membrane and nerve system. It is also known to cause anemia, liver cirrhosis, kidney failure and genetic toxicity in the human being working in the exposed area. LD$_{50}$ of furfural for peritoneal injected mouse has been known around 20mg/kg, but the acute toxicity on aquatic organisms such as fish, daphnid or algae are not well known, compared to those on rodents. In this experiment, we studied on the fish toxicity of furfural using Japanese Medaka (Orvzias latipes) and Common Carp (Cvprinus carpio). We also observed histological changes in the fish organs. The LC$_{50}$ were 12. Smg/L in Japanese Medaka and 21.8 mg/L in Common Carp, respectively. When Common Carps were exposed to 120mg/L of furfural concentration for 30 minutes, blood congestion in gills and lysis of secondary lamella were shown. Though the muscle of caudal fin was not completely eroded, its epidermic cells were shown to be necrotic in various parts. Tissue atrophy and cell necrosis were also shown in the liver of Common Carps exposed to furfural. From these results, furfural seems to cause histological damages on liver, an internal organ as well as on external organs such as gills and fins eventhough the fish were exposed for a short-term.

The Effort of Dietary Lipids on CVD Risk Factors in Ovariectomized Rats

  • This experiment was performed to investigate the effect of different dietary lipids on the risk factors of coronary vascular disease(CVD) in ovariectomized rats. female rats of Sprague-Dawley stram were divided into sham-operated(sham) and ovariectomized(ovx) groups and then each group was divided into a beef fallow group, a soy bean oil group and a fish oil group. After 16 weeks of feeding on experimental diets, animals were sacrificed and blood, liver, kidney and perirenal fat pad were obtained. Food intake and weight gain of fish oil group were significantly lower than other dietary lipid groups. food intake and weight gain tended to be higher in ovx groups than in sham groups. The weight Index(g/100g body weight) of liver and kidney was higher in the fish oil group than the other groups and weight index was lower in ovx groups compared to sham groups. The weight of the perirenal fat pad was the highest in the beef tallow group and the lowest in the fish oil group. The fish oil group showed the lowest total cholesterol(TC) and triglyceride (TG) levels in serum. Serum TG levels were lower in all ovx groups than in sham groups, but serum TC levels were not influenced by ovariectomy. fatty acid composition of serum reflects the recent dietary Intake of fat. Linoleic acid content was tile highest in soy bean oil group and eicosapentaenoic acid(EPA) and docosahexaenoic acid(DHA) contents were the highest in fish oil group. fatty acid composition of adipose tissue, especially EPA and DHA contents in perirenal fat pad, was highest in the fish oil group. Saturated fatty acid(SFA) and monounsaturated fatty acid(MUFA) in serum and adipose tissue did not reflect fatty acid intake. The activities of glucose-6-phosphate dehydrogenase, a lipogenic enzyme, in the blood of the beef tallow and soybean oil groups showed the tendency to be high and that of the fish oil group to be low in ovx. Carnitine acetyltransferase, a lipolytic enzyme, showed the highest activity in the liver of the fish oil group and was least active in the soy bean oil group.

  • The purpose of this experiment is to estabilish the sensitive and specific ELISA (enzyme linked immunosorbent assay) system for the detection of fish metallothionein (MT). Silver carp were injected with CA of 1-8mg/kg body wt. 4 times during 10 days. Silver carp was very tolerant species to CA. Cd induced MT in liver was seperated and purified by gel filtration chromatography and ion exchange chromatography and identified by spectrophotometry, native gel electrophoresis and western blot analysis. The rabbit antiserum was produced by immunizing rabbit with lyophilized MT, and the competitive ELISA system was estabilished for the detection of fish MT. In the present ELISA system, the detection limit was about 33 ng/ml. When this ELISA system was employed to determine the MT level in the supernatant sample of fish liver homogenate, the reaction curve showed a good parallel corelationship with the calibration curve over a certain dilution range. The results indicate that the competitive ELISA can be a useful tool for the detection of fish MT in the toxicological study and the evaluation of water pollution.

Effect of Cholesterol on Hepatic Phospholipid Metabilism in Rats Fed a Diet Containing Fish Oil and Beef Tallow

  • The influence of dietary cholesterol on phospolipid metabolism in rat liver microsmes was studied in rats fed a diet containing fish oil(FO) or beef tallow (BT). The hepatic phospholipid content decreased wherease gepatic triglyceride and cholesterol increased significantly in both groups after cholestered supplementation. Plasma concentrations of phospholipid and traiglyceride increased with cholesterol supplement in both groups while cholesterol decreased only moderately in the FO group. Dietary cholesterol affected microsomal phosphiolpids in liver ; the proportation of phosphatidylcholine decreased in the FO group, an d it also slightly decreased in the BT group at the expense of phosphatidylethanolamine. The activity of CTP : phospocholine cytidylytransferase , the rate-limiting enzyme of phosphatidylcholine synthesis, increased inhepatic mocrosomes whreas it decreased in hepatic cytosol of both groups by cholesterol supplementation. In conclusion, these indicated that the dietary cholesterol profoundly influences phospholipid metabolism in the rat liver.

An outbreak of Motile Aeromonas Septicemia in cultured Nile tilapia, Oreochromis niloticus with reference to hematological, biochemical and histopathological alterations

  • The current investigation dealing with the causative agent of mass mortalities in cultured Oreochromis niloticus. The diseased fish showed external hemorrhage, unilateral and bilateral eye opacity, ended by blindness and fish death. The postmortem lesions revealed congested friable kidney and spleen, and liver has yellow nodules. Obtained isolates were identified as Aeromonas hydrophila (the causative agent of Motile Aeromonas Septicemia) and found to be highly pathogenic as they contained hemolysin virulence gene causing mortality reached to 100 and 70% in intraperitoneal and intramuscular infection. The prevalence of MAS was 80% among the surveyed O. niloticus. Blood and serum were collected from naturally diseased, intraperitoneal and intramuscular injected O. niloticus for hematological and biochemical examination. Similarly, gills, musculature, kidney, liver and spleen were collected for histopathological evaluation, and micropathomorphological analysis of spleen was done. Macrocytic hypochromic anemia was recorded in the intraperitoneal infection. Serum protein, albumin and globulin were decrease only in naturally diseased fish. Leucocytosis with heterophilia and lymphocytosis were observed in naturally diseased and intraperitoneal infected fish. There were severe degenerative changes and hemorrhagic necrosis in the examined tissues which were more obvious in intraperitoneal than intramuscular infection. Activation and proliferation of melanocytes macrophages centers with severe hemosiderosis were recorded in spleen of naturally diseased and experimentally infected fish.

Toxicity of the Puffer fish, Takifugu pardalis (Jolbok) and Takifugu niphobles (Bokseom) from Coastal Area of Korea (한국 연안산 졸복(Takifugu pardalis)과 복섬(Takifugu niphobles)의 독성)

  • The toxicity of two species of puffer fish, Takifugu pardalis and T. niphobles, collected from the coastal regions of Korea was determined using a mouse bioassay. In T. pardalis collected at Tongyeong, the proportion of toxic specimens containing ${\geq}10MU/g$ exceeded 90% for the skin, fins, liver, intestine, ovary, and gallbladder, 11.1% for the testis, and 6.9% for the muscle. In each of the organs, the highest toxin levels were several tens (14-39) of mouse units (MU) per gram in the muscle, testis, and eyeball, but thousands (1,444-5,755) of MU per gram in the skin, liver, intestine, ovary, and gallbladder. The organs of T. pardalis exhibited remarkable variation in toxicity. In T. niphobles, the proportion of toxic specimens exceeded 90% for the ovary and skin, 60-80% for the fins, liver, intestine, and gallbladder, and 4.5% for the muscle; no toxicity was detected in the testis or eyeball using the mouse bioassay. The highest toxin levels were thousands (2,291-7,777) of MU per gram in the liver, intestine, ovary, and gallbladder, hundreds(146-328) of MU per gram in the skin and fins, and 18 MU/g in the muscle. Takifugu niphobles toxicity also exhibited remarkable regional variation. The toxicity in the edible muscle of T. pardalis and T. niphobles was at acceptable levels for human consumption, while the toxicity of the skin of both species of puffer fish was very high, so that care must be taken when used for human consumption.

Induction of oxidative stress in Clarias gariepinus from Eleyele River in Nigeria

  • This study evaluated some markers of oxidative stress in the organs of African Catfish, Clarias gariepinus from Eleyele River in Oyo State, Nigeria. Clarias gariepinus (250 g-400 g) were collected from Eleyele River (a suspected polluted River) and Clarias gariepinus from a clean fish farm (Durantee fisheries) were used as the control. Levels of Malondialdehyde (index of lipid peroxidation), Glutathione (GSH) and activities of antioxidant enzymes- Superoxide dismutase (SOD), Catalase and Glutathione-S-Transferase (GST) were evaluated in the liver, kidney and gills of the fish. From the results, there were significant (p<0.001) increases in malondialdehyde and GSH levels in the liver, kidney and gills of Clarias gariepinus from Eleyele River compared with control. The activity of GST increased significantly (p<0.05; p<0.001) in the liver and kidney of fish from Eleyele River compared with control. There was a significant decrease (p<0.05; p<0.001) in SOD activity in all the organs of Clarias gariepinus from Eleyele River compared with conrol and also a significant (p<0.001) decrease in catalase activity in the gills and kidney of the fish but catalase activity increased in the liver. Increase in lipid peroxidation and alterations in antioxidant status in Clarias gariepinus from Eleyele River show that the fish were under oxidative stress. These suggest that the River is polluted probably as a result of various wastes frequently discharged into the River. This could pose serious health risks to consumers of water and aquatic organisms from the River.

Effects of Fish Oil and Some Seed Oils on Fatty Acid Compositions of Liver and Brain Tissue in Rats (어유 및 식물 종자유의 급이가 흰쥐가 간장, 뇌조직의 지방산 조성에 미치는 영향)

  • This study was designed to observe the effects of the fish oil and some seed oils on the improvement of the fatty acid compositions of liver and brain tissue in rats. In order to induce the hypertriglyceridemia in the rats of the Sprague-Dawley, 12% coconut oil and 3% each of olive oil, lard, fish oil, perilla oil, corn oil, red pepper seed oil and evening primrose oil were administered to the rats for 4 weeks. In the fatty acid composition of liver lipid, n-3 PUFA contents were most in the fish oil and perilla oil groups of phospholipid fraction, and n-6 PUFA contents were most in the corn oil, red pepper seed oil and evening primrose oil groups of triglyceride fractions. Fatty acid composition of liver lipid fractions were influenced from the fatty acid composition of the test lipids. In the fatty acid composition of brain phospholipid, n-3 PUFA contents (8.8~17.2%) were most in the fish oil group, and n-6 PUFA (34.6~38.2%), though it contains high percentage, showed little difference between groups.
