Characterization of a Small Cryptic Plasmid from Pseudomonas nitroreducens Strain TX1 (Pseudomonas nitroreducens TX1에 존재하는 작은 플라스미드의 특성 규명)

  • Pseudomonas nitroreducens TX1 was isolated from a rice field drainage in Taiwan. The bacterium is of special interest because of its capability to use a group of nonionic surfactants such as alkylphenol polyethoxylates even at high concentrations as a sole carbon source. In this study, a small cryptic circular plasmid, pTX1, was characterized from P. nitroreducens TX1. It is 2,286 bp in length with a GC content of 63.3% and harbors three open reading frames, $Rep_{pTX1}$ and functionally unidentified ORF1 and ORF2. The predicted $rep_{pTX1}$ gene product is homologous to Rep proteins of plasmids belonging to the pC194/pUB110 family, which is predominantly found in Gram-positive bacteria and is known to replicate by the rolling-circle mechanism. The copy number of pTX1 was estimated to be about 150 in each cell. Based on the genetic fingerprints and comparison with other plasmids, it is concluded that pTX1 replicates by a rolling circle mechanism which is rarely found for Pseudomonas plasmids.

Construction of DNA Profile Data Base of Strawberry Cultivars Using Microsatellite Markers (Microsatellite 마커를 이용한 딸기 품종의 DNA Profile Database 구축)

  • This study was carried out to construct a DNA profile database of 100 strawberry cultivars using microsatellite markers. Two hundred seventy four microsatellite primer pairs were screened with a set of 21 strawberry cultivars with different morphological traits. Twenty five primer pairs were selected because they produced reliable and reproducible fingerprints. These primer pairs were used to develop DNA profiles of 100 strawberry cultivars. Three to thirteen alleles were detected by each marker with an average of 7.50. The average polymorphism information content varied from 0.331 to 841 (average 0.706). Cluster analysis showed that the 100 cultivars were divided into 7 major groups reflecting geographic origin and pedigree information. Moreover, most of the cultivars could be discriminated by marker genotypes. These markers will be useful as a tool for the protection of plant breeders' intellectual property rights in addition to providing the means to intervene seed disputes relating to variety authentication.

Bacterial Leaf Spot of English Ivy Caused by Xanthomonas hortorum pv. hederae (Xanthomonas hortorum pv. hederae에 의한 헤데라 세균성점무의병)

  • English ivy (Hedera helix) in Araliaceae family is an evergreen climbing vine. A severe bacterial disease of English ivy was observed and investigated in January 2005. Initial symptoms on the leaves appeared as small water-soaked lesions on the lower surface. As the spots enlarged, the center became brown to brownish black and greenish-brown water-soaked, irregular margins surrounded the center, The spots developed into large irregular blotches, sometimes 5$\sim$10 mm in diameter, then coalesced. Finally, the water-soaked margins raised, dried out, became corky and broke in the center. A bacterial organism, isolated from the advancing margins of the lesions, was tested for its pathogenicity according to the Koch's postulates and biochemical and physiological tests identified the isolated bacterium as a Xanthomonas. The representative Xanthomonas strains (SL4821 and SL4822) isolated from English ivy were compared with a reference strain X. hortorum pv. hederae for fatty acid profiles, metabolic fingerprints and 16s rDNA sequences, showing that all outcomes were indistinguishable between the representative and reference strains. This is the first report of bacterial leaf spot of English ivy in Korea.

Bacterial Leaf Spot and Dry Rot of Lettuce Caused by Xanthomonas campestris pv. vitians (Xanthomonas campestris pv. vitians에 의한 상추 세균성점무늬병)

  • During 1997 and 1998, a new disease of lettuce (Lactuca sativa) was observed on greenhouse-grown plants in Kwangju and Kwangmyung. Lesions on leaves were irregular, small, pale green to black, water-soaked, and 2 to 5 mm in diameter, Coalescing lesions sometimes caused defoliation of older leaves. Isolations made from diseased leaves on yeast extract dextrose calcium carbonate agar yielded nearly pure cultures of a yellow pigmented bacterium typical of a xanthomonad. Two bacterial strains (SL0246 and SL1352) were purified and used for further tests. Pathogenicity of strains was confirmed on 5-week-old lettuce plants injected with bacterial suspensions containing $10^8$ cfu/ml of phosphate buffered saline. The representative Xanthomonas strains isolated from lettuce were compared with a reference strain X. campestris pv, vitians for fatty acid profiles and metabolic fingerprints using GN2 microplate, showing that all outcomes were indistinguishable between the representative and reference strains. This is the first report of bacterial leaf spot and dry rot of lettuce in Korea.

  • In previous research, results on efficiency versus size and type of Ag particles showed similarity of detection efficiency comparing the particles of flake and spherical type with the gray particle on the market and in the case of nAg (rod, $0.9\;{\mu}m$) particle, relatively good results was given in the various evaluation methods for detection efficiency of latent fingerprint. However, oxidation was occurred when nAg particles laying on nature condition for a month and due to water absorption, detection efficiency was decreased. Therefore, with need to prevent oxidation and water absorption, more research is necessary. In this research, surface modification on nAg particles using silicon oil was conducted in various methods for complementing weakness of oxidation and water absorption. Then detection efficiency of nAg particles and surface modified nAg particles was evaluated by the number of feature points on the surface of non-porous materials (glass, plastic etc.) and degree of particle adhesion with ridges and contrast of detected fingerprint. Improvement of preventing oxidation and water absorbtion was given by surface modification using silicon oil (DC200, 0.5%) on the surface of non-porous materials.

Identification of Heat Stress-related Proteins and Low Molecular Weight HSP Expressed in Stem Tissues of Rice Plants by Proteomic Analysis (프로테옴 분석법에 의한 벼 줄기에서 발현하는 고온 스트레스 관련 단백질 및 저분자량 Heat Shock Protein의 분리 동정)

  • In order to investigate rice stem proteome in response to heat stress, rice plants were subjected to heat treatment at 42$^{\circ}C$ and total soluble proteins were extracted from stem tissues, and were fractionated with 15% PEG (poly ethylene glycol) and separated by two-dimensional polyacrylamide gel electrophoresis (2-DE). After staining of 2-DE gels, 46 of differentially expressed proteins were extracted, digested by trypsin, and subjected to matrix assisted laser desorption/ionization-time of flight mass spectrometry (MALDI-TOF MS) analysis. Proteins were identified through database search by using peptide mass fingerprints. Among them, 10 proteins were successfully identified. Seven proteins were up- and 3 proteins were down-regulated, respectively. These proteins are involved in energy and metabolism, redox homeostasis, and mitochondrial small heat shock proteins. The identification of some novel proteins in the heat stress response provides new insights that can lead to a better understanding of the molecular basis of heat-sensitivity in plants, and also useful to molecular breeding of thermotolerant forage crops.

A Network Analysis of the Research Trends in Fingerprints in Korea (네트워크 분석을 활용한 국내 지문인식연구의 동향분석)

  • Since the 1990s, fingerprint recognition has attracted much attention among scholars. There have been numerous studies on fingerprint recognition. However, most of the academic papers have focused mainly on how to make a technical advance of fingerprint recognition. there has been no significant output in the analysis of the research trends in fingerprint recognition. It's essential part to describe the overall structure of fingerprint recognition to make further studies much more efficient and effective. To this end, the primary purpose of this article is to deliver an overview of the research trends on fingerprint recognition based on network analysis. This study analyzed abstracts of the 122 academic journals ranging from 1990 to 2015. For gathering those data, the author took advantage of an academic searchable data base-RISS. After collecting abstracts, cleaning process was carried out and key words were selected by using Krwords and R; co-occurrence symmetric matrix made up of key words was created by Ktitle; and Netminer was employed to analyze closeness centrality. The result achieved from this work included followings: research trends in fingerprint recognition from 1990 to 2000, 2001 to 2005, 2006 to 2010, and 2011 to 2015.

Korea Coast Guard's Human Biological Materials Storage Project for Identifying Bodies Recovered from the Sea: A Model Suggestion (해양변사자 신원확인을 위한 해양경찰의 인체유래물 보관사업 모델제시)

  • The ocean is a harsh environment and Korea Coast Guard officers often face the threat of death or going missing in the line of duty. In order to promptly identify unknown bodies during mishaps, Korea Coast Guard Research Center has established an identification system using the DNA of its officers. This collected DNA can also be entombed in National Cemeteries when the remains are not recovered. The storage, disposal and quality control of the materials are overseen by the Governing Committee according to the Enforcement Rules for the Human Biological Materials Storage Project for the Identification of Officers Killed or Gone Missing in Action. Approximately 700 bodies are found per year along the Korean coast. An alternative method should be put in place for when identification through fingerprints and teeth are not applicable due to severe decomposition or partial recovery. We believe it would greatly contribute to helping identify the unknown bodies if the storage project could expand its data to include marine industry workers, relevant researchers and those involved in marine leisure activities.

Genetic Diversity of Pleurotus eringii Strains in Korea Based on Morphological Characteristics and PCR Polymorphism (형태적 특성과 PCR다형성 분석에 의한 국내 큰느타리버섯 계통의 유전적 다양성 분석)

  • This study was conducted to investigate genetic characteristics of 25 Pleurotus eringii strains that have been released in Korea based on cultural, morphological features and PCR fingerprints. Strains PER-007 and PER-012 showed distinct cultural characteristics in growth rate, morphological characteristics of mycelial colony and fruiting bodies when compared to those of other strains. Strain PER-007 did not form primordium initiation in sawdust medium and PER-012 also showed different phenotypes on fruiting bodies. Eleven URP primers were used to detect PCR polymophic bands in P. eringii strains. Primers URP1F, URP2R, URP2F, URP4R, URP6R, URP9F and URP17R were selected as useful primers for amplifying PCR polymorphic bands in P. eringii strains. The genetic similarity index was calculated by using PCR polymorphic bands amplified by eight URP primers among the 25 strains. The P. eringii strains were grouped by four distinct clusters on the UPGMA analysis. The genetic similarity values ranged from 100% to 76% were observed in three major groups, suggesting close genetic relatedness of them. Exceptionally, PER-007 and PER-017 were involved in outgroup.

Utilization of [6]-gingerol as an origin discriminant marker influencing melanin inhibitory activity relative to its content in Pinellia ternata (반하(Pinellia ternata)에서의 [6]-gingerol 함량과 멜라닌 저해 활성에 영향을 미치는 원산지 판별 마커로의 활용)

  • Pinellia ternata Breitenbach, the natural medicinal plant of the Araceae family, is a perennial plant originated from the East Asia, but also widely distributed in Europe and North America. Its tuber is used as traditional medicine for treatment of various diseases such as vomiting, inflammation, and traumatic injury. Pharmacological studies revealed that P. ternata possesses anticonvulsant, anti-tumor, insecticidal, and cytotoxic activities. Despite being well-known as the useful medicinal plant, there is no reliable, standardized method for origin discrimination. Ultra performance liquid chromatography-photodiode array detector and quadrupole time of flight-mass spectrometry based metabolite-profiling was applied to explore significant metabolite for origin discrimination between Korean and Chinese P. ternata. One compound was isolated from Korean P. ternata using repeated ODS column chromatography by bioactivity guided fractionation, and determined as [6]-gingerol according to the results of spectroscopic data including nuclear magnetic resonance and MS. This compound was selected as cosmeceutical biomarker by fingerprints, and it was associated to melanin inhibitory effect determining its origin authenticity. Furthermore, the calibration curve of biomarker was prepared using validated method for the comparison of content between Korean and Chinese P. ternata. This is the report to address the selection and successful validation of the discriminant metabolite for confirmation of Korean P. ternata.
