벼 키다리병 저항성 검정을 위한 기내 유묘 검정법 개발 (Development of In Vitro Seedling Screening Method for Selection of Resistant Rice Against Bakanae Disease)

  • 벼 키다리병은 매우 중요한 벼의 병 가운데 하나로 아시아 지역에서 넓게 발생하고 있다. 포장에서의 저항성 검정체계가 개발되어 있지만, 그 검정 결과가 환경조건에 따라 다르게 나타나는 문제점을 가지고 있다. 본 연구에서는 정확하고 빠른 키다리병 품종 저항성 검정을 위하여 기내에서 검정하는 시스템을 개발하였다. 병원균을 접종한 '남평벼', '주남벼', '추청벼', '삼천벼', '오대벼', '화신벼' 등 6개 품종의 종자를 MS배지가 10 ml 씩 담겨 있는 직경 30 mm, 길이 450 mm 시험관에 벼 종자 3립씩 파종 후 $25^{\circ}C$$28^{\circ}C$ 항온기에 치상하고 하루 12시간 광조건으로 배양하였다. 키다리병 증상은 벼가 도장한 후에 2-3주 경과시점 부터 잎 끝이 마르기 시작하여 고사하게 된다. $10^5$ 포자/ml 농도와 $28^{\circ}C$ 조건에서 '남평벼'는 파종후 28일째부터 잎 끝이 마르는 증상을 보이기 시작하여 35일째에 완전히 고사를 하는 반면 다른 품종들은 19-23일째에 모두 고사하였다. 기내유묘검정법을 이용하여 72개 품종에 대해 저항성검정을 한 결과 '남평벼'와 '인월벼'를 저항성으로 '화동벼', '석정벼', '삼광벼', '삼평벼', '농호벼', '흑진주벼', '조안벼'를 중도저항성으로 선발하였다. 기내유묘검정법을 이용할 경우 파종 후 35일 이내에 키다리병 품종저항성 검정을 완료할 수 있었다.

  • 포장에서 참다래 저장병균들에 의한 감염을 최소화할 수 있는 화학적 방제체계를 확립하기 위하여, 국내에서 참다래 저장병 약제로 등록된 베노밀 수화제와 지오판 수화제 등 2가지 약제를 대체할 수 있는 새로운 약제를 선발하기 위하여 본 연구를 실시하였다. 시험에 사용한 8가지 약제 중에서 후루실라졸 수화제, 이프로 수화제 및 터부코나졸 수화제는 모두 실내 시험을 통하여 국내에서 참다래 주요 저장병원균으로 밝혀진 Botryosphaeria dothidea, Diaporthe actinideae 및 Botrytis cinerea에 대해 탁월한 균사생장 저지효과를 나타내었을 뿐만 아니라 포장 시험에서도 베노밀 수화제와 지오판 수화제와 비등하거나 우수한 방제 효과를 나타내었으므로 장차 참다래 저장병 장제약제로 사용할 수 있을 것으로 기대된다.

개선된 RFID 기술을 이용한 u-헬스케어 서비스 인증 프로토콜 (Improved u-Healthcare Service Authentication Protocol based on RFID Technology)

  • 최근 RFID 기술은 u-헬스케어 서비스와 접목되어 의료서비스 분야에서 주목 받고 있는 추세이다. u-헬스케어 서비스는 개인 의료 정보를 다루는 분야로서 단순한 건강 검진 및 치료의 수준을 넘어 생명과도 밀접한 관계가 있다. 만약 개인 의료 정보가 불법적으로 노출되거나 악용될 경우 프라이버시 침해 뿐만아니라 생명까지도 위협받을 수 있으므로 보안성을 고려한 u-헬스케어 서비스 인증이 필수적으로 요구된다. 2012년에 Jeong 등은 RFID 기술을 이용하여 초기화 과정과 환자 인증 과정을 구분한 J-L 환자 인증 프로토콜을 제안하였다. Jeong등은 제안한 프로토콜에서 재사용 공격, 스푸핑 공격, 정보노출방지 및 불추적성에 대해 안전하다고 주장하였지만 보안성과 연산 효율성 문제를 발생시킨다. 따라서 본 논문에서는 Jeong 등이 제안한 프로토콜의 보안성과 연산 효율성 문제를 증명하고, 안전성과 효율성을 강화한 RFID 기술의 기반으로 하는 실용적인 u-헬스케어 서비스 인증 프로토콜을 제안한다.

FUNDAMENTAL UNDERSTANDING OF CRACKING AND BULGING IN COKE DRUMS

  • Cracking and bulging in welded and internally lined pressure vessels that work in thermal-mechanical cycling service have been well known problems in the petrochemical, power and nuclear industries. However, published literature and industry surveys show that similar problems have been occurring during the last 50 years. A better understanding of the causes of cracking and bulging causes is needed to improve the reliability of these pressure vessels. This study attempts to add information required for increasing the knowledge and fundamental understanding required. Typical examples of this problem are the coke drums in the delayed coking units refinery process. This case was selected for experimental work, field study and results comparison. Delayed coking units are among the refinery units that have higher economical yields. To shut down these units represents a high negative economical impact in refinery operations. Also, the maintenance costs associated with repairs are commonly very high. Cracking and bulging occurrences in the coke drums, most often at the weld areas, characterize the history of the operation of delayed coking units. To design and operate more robust coke drums with fewer problems, an improved metallurgical understanding of the cracking and bulging mechanisms is required. A methodology that is based field experience revision and metallurgical analyses for the screening of the most important variables, and subsequent finite element analyses to verify hypotheses and to rank the variables according to their impact on the coke drum lives has been developed. This indicated approach provides useful information for increasing coke drum reliability. The results of this work not only order the most important variables according to their impact in the life of the vessels, but also permit estimation of the life spans of coke drums. In conclusion, the current work shows that coke drums may fail as a combination of thermal fatigue and other degradation mechanisms such as: corrosion at high and low temperatures, detrimental metallurgical transformations and plastic deformation. It was also found that FEA is a very valuable tool for understanding cracking and bulging mechanisms in these services and for ranking the design, fabrication, operation and maintenance variables that affect coke drum reliability.

일부 농업인에서의 농약 중독증상과 농약 사용정보에 대한 예비조사 (A Pilot Study for Pesticide Poisoning Symptoms and Information on Pesticide Use among Farmers)

  • This study aimed to determine the feasibility of survey questionnaires for pesticide poisoning. We conducted a pilot study to develop a questionnaire for evaluating pesticide poisoning symptoms and to examine the validity of self-reported information for individual pesticides used. In December 2010 we interviewed 20 farmers with self-reported pesticide poisoning histories residing in Gyeonggi Province. For validity of pesticide information, the purchased pesticide lists from the agricultural cooperative federation were compared with individual self-reported pesticides used. Neurological symptoms were the most common among clinical symptoms related to pesticide poisoning, followed by gastrointestinal, dermal and eye symptoms. Insecticides were the main causative pesticide class for poisoning. Twenty one major symptoms were selected for screening for acute pesticide poisoning in a field study based on the pilot results and literature reviews. Substantial under-reporting was found in data on self-reported individual pesticides used (9.2%), as well as low accuracy (36%) compared with their actual purchase lists. In this pilot study, we suggest the selected symptom lists may apply to a field survey of pesticide poisoning. However, the self-reported information on individual pesticides may not be valid and alternative methods need to be developed.

미꾸라지에 성장호르몬 유전자 이식을 위한 최적 조건 개발 (Factors Affecting the Efficiency of Introducing Growth Hormone Gene into Mud Loach : Gene Transfer via Electroporation)

  • 미꾸라지 (Misgurnus mizolepis) 정자에 electroporation 처리를 통해 난과 인공수정시킨 후 수정율, 부화율, 초기 생존율 및 유전자 이식 효율을 조사하였다. pRSV/luc을 reporter유전자로 이용하여$0\~1$ 625 V/cm의 field strength와 $0\~1,000$ ${\mu}F$의 capacitance 범위의 electroporation을 시도한 결과, 실험군간에 수정율, 부화율, 초기 생존율에서 유의한 차이는 나타나지 않았다. 유전자 이식 효율을 PCR 분석으로 조사한 결과, $0\~20\%$로 나타났으며 elertroporation조건에 따라 큰 차이는 관찰할 수 없었다. 그러나 pMT/hGH의 경우, 대조군 및 처리군 모두에서 homology sequence에 의한 nonspecific amplication이 관찰됨으로서 PCR을 이용한 transgene의 검색이 불가능한 것으로 나타났다

더덕에 발생하는 당근뿌리혹선충의 증식억제 식물 탐색 (Screening of Medicinal Plants to Suppress Population of Meloidogyne hapla in Codonopsis lanceolata Trautv)

  • 비름과 쇠무릎등 총 36과 81속 90종의 약용식물을 대상으로 당근뿌리혹선충의 감염 여부를 조사한 결과, 쇠무릎 등 70종의 식물에서 당근뿌리혹선충 감염을 확인하지 못했고, 이들 70종 중 42종의 식물을 당근뿌리혹선충에 의한 더덕의 피해가 많았던 포장에 심고, 감염여부를 조사한 결과, 쇠무릎 등 26종은 당근뿌리혹선충에 거의 감염되지 않았고, 지치 등 16종은 감염되었음을 확인하였다. 또한 42종의 식물중 30종의 식물을 Pot 재배를 통하여 당근뿌리혹선충 접종밀도별 뿌리혹수와 토양 내 유충밀도를 조사한 결과 패랭이, 원추천인국, 기린초, 아주까리, 지모, 해변아욱, 애기똥풀, 흑참깨, 짚신나물, 큰뱀무, 오이풀, 현삼 등 12종은 당근뿌리혹선충에 거의 감염되지 않았음을 확인하였다. 반면에 더덕은 당근뿌리혹선충 접종밀도가 높을수록 뿌리혹수가 많아지고, 토양 내 유충밀도 역시 높아졌으며, 이에 따라 더덕의 생육은 접종밀도가 높을수록 나빠지는 경향이었다.

Isolation and Characterization of Oligotrophic Bacteria Possessing Induced Systemic Disease Resistance against Plant Pathogens

  • Biocontrol microbes have mainly been screened among large collections of microorganisms $via.$ nutrient-rich $in$ $vitro$ assays to identify novel and effective isolates. However, thus far, isolates from only a few genera, mainly spore-forming bacilli, have been commercially developed. In order to isolate field-effective biocontrol microbes, we screened for more than 200 oligotrophic bacterial strains, isolated from rhizospheres of various soil samples in Korea, which induced systemic resistance against the soft-rot disease caused by $Pectobacterium$ $carotovorum$ SCC1; we subsequently conducted in $planta$ bioassay screening. Two oligotrophic bacterial strains were selected for induced systemic disease resistance against the $Tobacco$ $Mosaic$ $Virus$ and the gray mold disease caused by $Botrytis$ $cinerea$. The oligotrophic bacterial strains were identified as $Pseudomonas$ $manteilii$ B001 and $Bacillus$ $cereus$ C003 by biochemical analysis and the phylogenetic analysis of the 16S rRNA sequence. These bacterial strains did not exhibit any antifungal activities against plant pathogenic fungi but evidenced several other beneficial biocontrol traits, including phosphate solubilization and gelatin utilization. Collectively, our results indicate that the isolated oligotrophic bacterial strains possessing induced systemic disease resistance could provide useful tools as effective biopesticides and might be successfully used as cost-effective and preventive biocontrol agents in the field.

  • Determining of Strategic Business Units in the organization is a major critical process for improving the organizational performance. On increasing the demands for extension in function of health center, many health centers are trying to provide the various services. But most health center determined the kind and level of service without scientific considerations. The purposes of this study are to develope the model for determination of strategic business units in health center and to test the availability of implementation for it. Our model is rooted from the McKinsey matrix analysis of Product Portfolio Analysis which used widely in marketing field. We modified the evaluation criteria of the McKinsey matrix analysis for health care field appropriately. Our evaluation criteria are categorized into two concepts; contribution of service, availability of service. At first, in terms of contribution of service, market size, market growth rate, needs and demands of regional people, existences of alternative services in that region, correspondence with health policy. The other component, availability of service are included the availability of manpower, financial availability, the level of knowhow on service, acceptance of health care manpower. In the result of analysis, we could conclude that antismoking and antialcoholics education programs, health screening program are important strategically in that aspects of contributions and availability of services. Also, vaccination program is important in that aspect of service availability and diet and exercise program, health library are meaningful in the aspect of service contribution. Therefore, we think that efforts to investigate the evaluation criteria for priority setting or determination of service area in health center are useful challenges.

Identification of Novel Source of Resistance and Differential Response of Allium Genotypes to Purple Blotch Pathogen, Alternaria porri (Ellis) Ciferri

  • Purple blotch, caused by Alternaria porri (Ellis) Cifferi, is a serious disease incurring heavy yield losses in the bulb and seed crop of onion and garlic worldwide. There is an immediate need for identification of effective resistance sources for use in host resistance breeding. A total of 43 Allium genotypes were screened for purple blotch resistance under field conditions. Allium cepa accession 'CBT-Ac77' and cultivar 'Arka Kalyan' were observed to be highly resistant. In vitro inoculation of a selected set of genotypes with A. porri, revealed that 7 days after inoculation was suitable to observe the disease severity. In vitro screening of 43 genotypes for resistance to A. porri revealed two resistant lines. An additional 14 genotypes showed consistent moderate resistance in the field as well as in vitro evaluations. Among the related Allium species, A. schoenoprasum and A. roylei showed the least disease index and can be used for interspecific hybridization with cultivated onion. Differential reaction analysis of three A. porri isolates (Apo-Chiplima, Apn-Nasik, Apg-Guntur) in 43 genotypes revealed significant variation among the evaluated Allium species (P = 0.001). All together, the present study suggest that, the newly identified resistance sources can be used as potential donors for ongoing purple blotch resistance breeding program in India.
