H. erinaceum의 기능성 소재화 연구의 일환으로, 지금까지 체계적으로 연구된 바 없는 H. erinaceum의 액체배양을 시도하고. 이 버섯의 서로 다른 생물반응기하의 액체배양 특성을 규명하고 대량생산을 검토하였다. 5 L 용량의 jar fermenter와 air-lift fermenter에서의 액체배양의 비교실험을 통해 air-lift fermenter의 사용이 종균 배양에 적합한 것으로 나타났으며, 형태 분석을 통해 적정 크기($2{\sim}4mm$)의 pellet 형태의 유지가 대량 생산의 중요인자인 것을 규명하였다. Air-lift fermenter 하의 배양을 종균으로 하고, 적정 크기의 pellet을 유지하는 500 L까지의 jar fermenter를 사용한 회분 배양의 scale-up 실험결과, 30 및 500 L에서의 교반 및 통기속도는 각각 200 rpm, 1 vvm 및 150 rpm, 1 vvm이었다. 이 조건하에서 배양의 전 기간 중 적정 pellet을 유지하였으며, 최대 균사체량은 약 $13{\sim}14g/L$이었다.
For the efficient production of a new exo-polysaccharide from Ganoderma lucidum ASI 7004, the optimum conditions and methods in submerged cultivation were investigated with an airlift fermenter system. The optimum aeration rate was 2.5 Wm at the initial pH 5.0 and 28$^{\circ}C$. The increase of dissolved oxygen concentration by pure oxygen supply during cultivation did not improved the exo-polysaccaride production and the mycelial growth. The maximum exo-polysaccharide production and the mycelial growth under the optimum culture condition were obtained in media of glucose 60g/L, yeast extract 6g/L, (NH4)2HPO4 1g/L and KH2PO4 0.5g/L. Under these optimum medium and culture conditions, about 7.15g/L of exo-polysaccharide and 13.9g/L of mycelial growth were producted, respectively.
This study was carried out to screen the effective polymeric additives preventing wall growth during mycelial submerged cultivation of Ganoderma. lucidum. Effects of additives on mycelial growth and exo-polysaccharide (EPS) production in flask culture and jar fermenter system under 3 different pH processes were investigated, and changes of mycelial morphology were also examined. From flask culture of G. lucidum with additives of different concentrations, 0.1%(w/v) polyacrylic acid was effective for EPS production. As the polyacrylic acid of 0.1%(w/v) was added in medium, wall growth of G. lucidum mycelium grown in jar fermenter system could be protected. The addition of 0.1%(w/v) polyacrylic acid to medium was also improved the mycelial growth and EPS production in the later of submerged culture G. lucidum and no changes of mycelial morphology were observed.
유기칠 폐기물을 보다 효율적으로 처리하고자, 상 분리 혐기발효와 Chiorella 배양에 의한 생우분의 4 단계-복합처리를 시도하였다. 3배 희석 생우분을 6 일간 산발효시킨 발효액을 4 일의 수리학적 체류시 간으로 1차 에탄발효조에 공급했을 때, 평균 977ml/l C비ture/day의 바이오가스가 생산되었고, 1차 발효 폐액을 역시 4일의 체류시간으로 2차 메탄발효조에 공급했을 때 평균 428ml/l culture/day의 바이오 가스가 생산되었다. 1차 메탄발효의 바이오가스 생 산성은 재래의 단일조식과 비교했을 때 31.4% 증가 되었다. 또한 산발효과정, 1차 및 2차 에탄발효과정 의 유기물 제거율은 각기 71.8%, 42.6% 그리고 2 24.0%였다. 한편, 2차 메탄발효폐액은 Chiarella s sp. PSH3에 의해 10일의 체류시간으로 반연속배양 처리한 결과, 평균 1.8g/l culture/day(2.8$\times$106 cells/ml)의 조체가 생산되었고, 이 과정의 유기물 제거율은 73%였다. 이상의 4단계 처리과정을 통하 여 초발원료인 3배 희석 생우분의 COD가 51, 300ppm에서 최종단계인 Chiarella의 처리에 의해 평균 85ppm으로 감소되어 총유기물 제거율이 99.8 %에 달하였다. 이들의 결과는 유기질 폐기물의 효 율적 처리와 더불어 바이오가스와 균체단백질을 생산하는 복합처리계의 구성이 가능함을 나타낸다.
The optimal temperature, pH and aeration rate for spore production by Bacillus polyfermenticus SCD in 500 ml shake flask and 5-1 jar fermenter were found to be $32^{\circ}C$, 7.0 and 1.0 vvm. respectively. When batch culture processes was performed under optimized culture conditions. viable cells were $3.3{\times}10^{10}$ CFU/ml and spore cells were $3.3{\times}10^{10}$ CFU/ml. Fed-batch culture processes were also examined with regard to higer maximum viable cell and spore production. The highe viable cells and spores were obtained in 5-1 jar fermenter at 72 h cultivation time by strategy in an intermediate feeding mode with 60% glucose solution 150 ml and 5% soybean flour solution 150 ml fed to the fermenter twice, and the productivity of spore cells was significantly increased. Finally. volumetric productivity of spore cells on fed-batch culture indicated $9.9{\times}10^8$ CFU/ml/h, which was approximately 2 times higher than batch culture. Thus, fed-batch culture show a promise as an industrial production method.
노루궁뎅이버섯 균사체(Hericium erinaceum)의 심부발효물로부터 기능성소재를 얻기 위하여 flask 배양에서 적당한 기본 배지를 선정하였고, jar fermenter에서 선정된 mushroom complete medium(MCM)에 수삼추출물을 첨가하여 최적 발효조건을 검토하였다. 기본 액체배지 중 수삼추출물이 0.5% 첨가된 MCM에서 $25^{\circ}C$, pH 5.5의 조건으로 10일 경과 후 5.89 g/L의 가장 높은 균사체 건조균체량(mycelial dry weight, MDW)을 나타내었다. 발효물의 대량생산을 위한 50 L jar fermenter의 액체배양에서도 $25^{\circ}C$, pH 5.5와 교반속도 120 rpm과 0.4 vvm의 통기속도로 최적화 되었으며, 이러한 조건하에서 5일 배양으로 최대 MDW인 4.28 g/L을 얻을 수 있었다. 또한 MCM에 첨가되는 수삼추출물의 양을 jar fermenter에서 검토한 결과, 수삼추출물 5%가 첨가되는 군(HE-GE-5)의 최적 발효조건하에서 4.93 g/L의 최대 건조균체량을 나타내었다. 한편 수삼추출물이 첨가된 노루궁뎅이버섯 균사체의 발효물의 특성을 검토하기 위하여 심부발효물의 조다당획분을 조제한 후 구성분과 면역활성을 살펴본 결과, 최대 MDW를 나타낸 수삼추출물 5% 첨가 심부발효물의 조다당(HE-GE-CP-5)은 주로 중성당(63.2%)과 함께 상당량의 산성당(19.3%)과 소량의 단백질(8.8%)로 구성됨을 알 수 있었으며 마이토젠과 골수세포 증식활성이 균사체만의 심부발효물 조다당보다 증강되고 있음을 알 수 있었다.
Cephalosporium acremonium is a filamentous microorganism producing cephalosporin C. The morphological differentiation of C. acremonium in submerged culture is closely related with the rheological properties of culture broth and production of cephalosporin C. In this study, the rheological and morphological properties of culture broth of C. acremonium were investigated. In the seed broths of shake-flask and fermenter culture, the Herschel-Berkley equation was in excellent agreement with experimental results in the whole range of shear rate. In the seed broths of shake-flask culture, morphological differentiation into arthrospores affected to changes of apparent viscosity. But results in the fermenter culture, morphological factors such as mean hyphal thickness and the number of tips gave more effect on changes of apparent vitacosity than differentiation into arthrospores. Overall, it suggested that the morphological parameters measured by image analysis can be used as a good parameter to indicate the rheological properties of culture broth of C. acremonium M25.
This study was carried out to investigate the effects of agitation, aeration and scale-up on the mycelial growth, exo-polysaccharide (EPS) production, and mycelial morphology in the liquid culture of Ganoderma lucidum. A correlation between roughness and operating variables was also studied to scale-up the liquid culture of G. lucidum in a jar fermenter. When the agitation speed or aeration rate increased, the morphological form was changed from rough pellet to smooth pellet form. Increase of the agitation and aeration reduced the mycelial roughness. On the other hand, in the case of pellet size, it was not affected by aeration. The higher EPS production was obtained at approximately 17% of roughness and mycelial pellet size of 3~5 mm. The morphology at each fermenter was closely correlated with kLa value, and it was found that similarity of morphology would be used as a criteria of scale-up for liquid culture of G. lucidum.
Cellulase production by batch culture using the Trichoderma reesei Rut C30 strain with various concentrations of Solka Floc with 1 % wheat bran was studied in a 2.5 I fermenter. The cellulase activity increased with Solka Floc concentration up to 5%. When 5% Solka Floc and 1% wheat bran were contained in the medium, carboxymethyl cellulose (CMC) and filter paper (FP) activities were 232.4 U/$m\ell$ and 21.25 U/$m\ell$, respectively. The productivity was 143.6 FPU $1^{-l}h^{-1}$ and the yield was 425 FPU/g. The colonial morphology of T. reesei Rut C30 grown on Avicel agar plates and the changes in mycelial morphology of T. reesei Rut C30 with culture time are also presented.

This study investigated the effects of various culture parameters (carbon sources, temperature, initial pH of culture, NaCl concentration, and light) on the growth and canthaxanthin production by Dietzia natronolimnaea HS-1. The results showed that the most effective carbon source for growth and canthaxantin production was glucose, and the best pH and temperature were 7 and $31^{\circ}C$, respectively. In addition, the biomass and canthaxanthin production increased in a medium without NaCl and in the presence of light. Under the optimized conditions, the maximum biomass, total carotenoid, and canthaxanthin production were $6.12{\pm}0.21g/l,\;4.51{\pm}0.20mg/l,\;and\;4.28{\pm}0.15mg/l$, respectively, in an Erlenmeyer flask system, yet increased to 7.25 g/l, 5.48 mg/l, and 5.29 mg/l, respectively, in a batch fermenter system.
