Effects of Dietary Lipids and Dexamethasone on Mucosal Hyperplasia after 70% Jejunoileal Resection in Rats (식이지방과 Dexamethasone 주입이 소장절제후 소장 점막의 증식에 미치는 영향)

  • Dietary mehaden oil enhances mucosal hyperplasia that normally occurs after massive small bowel resection. In contrast, dexamethasone and aspirin inhibit the adaptation response. In order to gain insight on the mechanism of these effects, male Sprague-Dawley rats weighing approximately 150gram were randomly divided into two groups and fed diet containing either 15% safflower oil or 14% menhaden oil and 1% safflower oil. Ten days later they were subjected to 70% jejunoilear resection. Immediately after surgery each group was further divided into two groups and receive either vehicle or 125ug/kg/day dexamethasone subcutaneously. All animals were sacrificed seven days after the surgery, and the remaining intestine was removed and divided at the anastomotic site. Dexamethasone, which decreased gut hyperplasia in both dietary groups, decreased both serum IGF-I levels and ileral PGE2 synthesis. Menhaden oil enhanced gut hyperplasia, but did not increase IGF-I or IGF-II levels in serum. PGE2 synthesis was lower in the ileum of menhaden oil-fed rats compared to that of safflower oil-fed rats. The effects of menhaden oil on adaptation did not apper to be mediated either through IGFs or PGE2 synthesis. Other factors could have played a role in enhancing adaptation following menhaden oil feeding.

  • The success of continuous aluminum powder combustion with steam plasma is different from hydrocarbon ignition source. Ignition characteristics of aluminum powder with high temperature thermal plasma is studied with oxidizer-free environment. Experiment with argon plasma has same temperature conditions at 4500 K and particle feeding condition for previous combustion test with steam plasma and swirl combustor. The temperature of the plasma was measured using Optical Emission Spectroscopy method. Ignition characteristics were analyzed by SEM image and EDS. Aluminum powder with plasma has rapid evaporation mechanism contrast to hydrocarbon ignition source. It enhances to aluminum powder effective ignition characteristics.

Mechanical Properties and Microstructures in WC-12%Co/Low Carbon Steel Metal Matrix Composites(MMC) Welding Overlay

  • Metal matrix composites(MMC) consist of metal matrix into which is distributed a second solid phase. The normal intension is to develop a material with superior mechanical properties (for example increased toughness, stiffness and wear resistance) compared to those inherent in the matrix component. In this study, WC-12%Co/low carbon steel MMC overlays have been prepared by Gas Metal Arc Welding(GMAW) according to feeding rate of WC-12%Co grit. The macro and microstructures were examined using optical microscopy (OM) and scanning electron microscopy(SEM) each other. The characteristics of hardness and wear resistance have been investigated. WC-12%Co/low carbon steel MMC overlays which have been taken good beads without porosity and cracks were manufactured by method of GMAW. Matrix of overlayed surface was seen as fish bone and faceted dendrite structures. It was known that structures were iron tungsten carbides, Fe$_{6}$W$_{6}$C which have been occurred by melting of WC-12%Co grits. After MMC had been tested by block-roll wear test it was known that WC-12%Co/low carbon steel MMC has a excellent wear resistance by exiting Fe6w6c and WC-12%Co grit. The consequence was that region of overlay with Fe$_{6}$W$_{6}$C phase has been showed a model of adhesive wear, but region of overlay with WC-12%Co grit was restrained as a result of mechanism that wear of WC-12%Co grit is not adhesive but fracture.racture.

  • The obese population has been increasing worldwide and obesity has become one of the socioeconomic problems. Obesity raises more concerns as more studies regarding its direct and indirect relativity to several diseases such as type II diabetes, hypertension, etc. are published. Since leptin, an important signal in the chronic control of food intake and energy expenditure, was discovered in 1994, there has been a great accumulation of knowledge on fighting obesity by facilitating pharmacological and nutritional strategies on the molecular level of the body weight control system. In particular, evidences are accumulating that particular food components affect our physiological function and gene expressions which are associated with body weight control. In this study, we review the four mechanisms for weight control and antiobesity functional agents such as HCA, L-carnitine, CLA, chitosan, calcium supplements capsaicin contained in red pepper, and oriental herbal mixture. We also describe about the efficacy and working mechanism of these functional agents on the basis of antiobesity mechanisms.

Cholera Toxin Disrupts Oral Tolerance via NF-κB-mediated Downregulation of Indoleamine 2,3-dioxygenase Expression

  • Cholera toxin (CT) is an ADP-ribosylating bacterial exotoxin that has been used as an adjuvant in animal studies of oral immunization. The mechanisms of mucosal immunogenicity and adjuvanticity of CT remain to be established. In this study, we investigated the role of indoleamine 2,3-dioxygenase (IDO), which participates in the induction of immune tolerance, in CT-mediated breakdown of oral tolerance. When IDO-deficient ($IDO^{-/-}$) mice and their littermates were given oral ovalbumin, significant changes in antibody responses, footpad swelling and $CD4^+$ T cell proliferation were not observed in $IDO^{-/-}$ mice. Feeding of CT decreased IDO expression in mesenteric lymph nodes (MLN) and Peyer's patch (PP). CT-induced downregulation of IDO expression was reversed by inhibitors of nuclear factor-kappa B (NF-${\kappa}B$), pyrrolidine dithiocarbamate and p50 small interfering RNA. IDO expression was downregulated by the NF-${\kappa}B$ inducers lipopolysaccharide and tumor necrosis factor-${\alpha}$. CT dampened IDO activity and mRNA expression in dendritic cells from MLN and PP. These data indicate that CT disrupts oral tolerance by activating NF-${\kappa}B$, which in turn downregulates IDO expression. This study betters the understanding of the molecular mechanism underlying CT-mediated abrogation of oral tolerance.

Multiple-Case Studies of Hand-on Breast Massage Techniques used by Breastfeeding Experts (산후 유방 마사지 손기술에 대한 다중사례분석)

  • Purpose: The aim of this study was to understand the hand-on breast massage techniques used by well-known experts in breastfeeding clinics. Methods: A qualitative multiple-case design was applied that involved a feasibility test. Four experts sampling qualitative data collected by observing participants and in individual interviews were analyzed by content analysis, linking data to the propositions, and cross-case pattern matching. This study explored differences within and between cases, and the possibilities of replicating findings across cases. Thirty-nine postpartum women participated voluntarily in the feasibility test, which investigated the usability of four massage techniques. Results: The four techniques showed considerable similarities in terms of the application of stimulation to the breast base and increased flexibility of the wired flexible body, which was the core mechanism underlying the techniques. The breast management strategies were consistent with existing practice guidelines with the exception of using cold cabbage to control engorgement pain. There was insufficient scientific evidence for supporting the massage techniques used by the experts. All of the techniques showed 100% education completeness, but application rates were higher for self-control-oriented techniques. Conclusion: The massage techniques applied by experts in breastfeeding were based on hypotheses and self-control techniques are feasible to apply in practice.

  • Recently, improving the energy efficiency of a pneumatic system and reducing the consumption of compressed air were a concern of scholars at domestic and abroad. The using fields of a pneumatic system are widely used in factory automation of manufacturing line, chemical factories with explosiveness danger and petroleum industries etc. In particular, pneumatic cylinder is applied to feeding work of workpiece. jig tools and press mechanism, reciprocation and rotary motion with rack and pinion. In this study, the experimental apparatus consisted to pneumatic cylinder, dual supply pressure regulator and solenoid valve. The dual supply pressure regulator connected to outlet port of solenoid valve. The supply pressure($4.5kg_f/cm^2$) of compressed air goes into the rodless chamber 1 to drive the piston rod forward which is named working stage. The supply pressure ($2kg_f/cm^2$) of compressed air goes into the rod chamber 2 to drive the piston rod backward which is named no-working stage. Accordingly, the research results of this study can be obtained to Energy-Saving Effects of the compressed air about 35%.

Analysis of Plasma Effects on Seed Germination and Plant Growth

  • Plasma technology has been widely used for decontamination, differentiation, and disease treatment. Recently, studies show that plasma has effects on increasing seed germination and plant growth. In spite of increasing number of studies about plasma effects, the interaction between plasma and plants has been rarely informed. In this study, we have analyzed the effects of nonthermal atmospheric pressure plasma on seed germination and growth of coriander (Coriandum sativum), a medicinal plant. We used to Ar, air, and N2 plasma on seed as feeding gases. Plasma was discharged at 0.62 kV, 200 mA, 9.2 W. Seed germination was increased over time when treated with N2 based DBD plasma for exposure times of 30 seconds and 1 minute, everyday. After 7 days, about 80~100% of seeds were germinated in the treatment with N2 based DBD plasma, compared to control (about 40%, only gas treated seeds). In order to elucidate the mechanism of increased germination, we have analyzed characteristics of changes in plant hormones and seed surface structure by SEM.

  • In order to cut the round bale easily, a roughage cut-feeder for the round bale was developed in this study. This study consists of two parts. One is development of a cutting mechanism and a performance test reported in the previous paper. This is the second part of the study. For the study, recent trend of traditional cutting system was analyzed. Also, cost of the prototype cut-feeder was analyzed and determine the break-even point of farm size was determined by comparing with traditional method using a traditional cutter. A prototype cut-feeder was investigated with the fact that working performance, fiber length and shape of roughage and feeding quality was good. Operating cost of the prototype cut-feeder decreased rapidly with an increase of farm size. Break-even point in terms of farm size was 36 heads for beef and 28 heads for dairy. Also, costs estimated were 118,000 won/head year and 148,000 won/head year, respectively.

Induction of Apoptosis with Kigelia africana fruits in HCT116 Human Colon Cancer Cells via MAPKs Signaling Pathway

  • Kigelia africana (Lam.) Benth. (Bignoniaceae) is a flowering plants in South, Central and West Africa and commonly known as the sausage tree (Eng.); worsboom (Afr.); umVunguta, umFongothi (Zulu); Modukguhlu (North Sotho); Muvevha (Venda). The dried, powdered fruits are used as dressing for wounds and ulcers, haemorrhoids, rheumatism, purgative, skin-firming, lactation in breast-feeding mothers. The aim of this study is to investigate the cytotoxic and apoptotic potentials of 70% ethanolic extracts of Kigelia africana fruits in HCT116 human colon cancer cells. Treatment of Kigelia africana fruits with various concentrations resulted in a sequence of characteristic of apoptosis, including loss of cell viability and morphological changes. Flow cytometry analysis showed Kigelia africana fruits increased the sub-G1 phase (apoptosis) population. Apoptosis confirmed by annexin V-fluorescein isothiocyanate and propidium iodide double staining in HCT116 human colon cancer cell lines. Moreover, analysis of the mechanism indicated that Kigelia africana fruits showed an increased Bax and Bcl-2 expressions in a dose-dependent manner, resulting in activation of hallmarks of apoptotic events, caspase-3, caspase-9 and cleaved poly-ADP-ribose polymerase. This is the first report to demonstrate the cytotoxicity of Kigelia africana fruits on HCT116 human colon cancer cells.
