This study was conducted to test the effect of residual Al on plankton community after dissolved air flotation (DAF) application. Growth rate of phytoplankton after DAF application ($0.37day^{-1}$) was about 2 times lower than that before DAF application ($0.70day^{-1}$). Under the condition of addition of nitrogen and phosphorus without light, growth rate phytoplankton in treatment without residual Al increased in difference with showing the negative growth rate in treatment with residual Al. Under the condition of light without addition of nutrient, growth rate of phytoplankton was no noticeable difference between the before and after DAF application. The relatively high settling rate (0.47 m/day) was observed in treatment after DAF application. Although the abundance of rotifer decreased, the abundance of copepod and cladoceran such as Daphnia galeata, Diaphanosoma sp. and Bosmina longirostris with relative higher grazing was no noticeable difference between the before and after DAF application. In the treatments before and after DAF application with zooplankton, growth rate of phytoplankton was $0.41{\pm}0.08day^{-1}$, $0.20{\pm}0.03day^{-1}$, respectively. This difference was in treatment after DAF application similar with those in treatments before and after DAF application without zooplankton. Those indicate that the filter-feeding effect of zooplankton on phytoplankton community may be not changed by residual Al after the DAF application. These results suggest that residual Al after DAF application be to improve water quality by inhibition of growth rate as well as increasing settling rate of phytoplankton.
Pine(Pinus densiflora Sieb et Zucc.) is one of rhe popular plant drugs which has been used as a medicine in Asia. To investigate the effect of pine needle extract (PNE) on oxygen radicals and their scavenger enzymes in brain membranes of Sprague- Dawley (SD), make SD rats were fed basic diets(control group), and experimental diets (PNE group) with 0.5 and 1.0% of PNE 6 weeks. Mitochondrial hydroxyl radical levels in brain of 0.5%-PNE and 1.0%-PNE groups were significantly inhibited to 30% and 25%, respectively, and microsomal hydrogen peroxide levels in brain of 0.5%-PNE and 1.0%-PNE groups were significantly inhibited to 15% compared with control group. Cytosolic superoxide rdical levels in 1.0%-PNE group were significantly inhibited to 20% compared with control group. Lipid peroxide(LPO) levels in brain mitochondria of 0.5%-PNE and 1.0%-PNE groups were significantly lower(25% and 35%) than that in control group. Mn-superoxide disumtase (SOD) activities in brain of 0.5%-PNE and 1.0%-PNE groups were significantly higher(18% and 12%) than those in control groups, but Cu,Zn-SOD activities in brain of 0.5%-PNE were significantly activated to 15% compared with control group. Glutathione peroxidase(GSHPx) activities in brain of 1.5%-PNE and 1.0% PNE groups were significantly higher(14% and 12%) than those in control group. These results suggest that more beneficial effects such as inhibition of oxygen radicals and lipid peroxide(LPO). and oncreases of scavenger enzymes in brain membranes of SD rats may be effectively modulated by administration of pine needle extract (PNE)
To investigate the effect of pine needle extract (PNE) on membrane fluidirt and neurotransmiter-related enzymes in brain of Spragu-Dawley(SD), male SD rats were fed basic diets (control group), and experimantal diets (PNE group)with 0.5% and 0.1% fo PNE for 6 weeks. pine (pinus tabulaeformis C$_{ARR}$ is one of the popular plant drugs which has used as a medicine in Asia. Cholesterol levels in brain mitochondria of 0.5%-PNE and 0.1%-PNDE groups were significantly decreased in 15% and 25%, respectively, compared with control group, but cholesterol levels in brain microsomes of these PNE groups howed almost no change compared with control group. Lipofuscin accumulations in brain membranes of 0.5%-PNE and 0.1%-PNE groups were sgnificantly inhibited in 18% and 21%, respectively, compared with control group. Brain memberance fluidity was also activated in 50% and 100% by the administration of 0.5%-PNE and 0.1%-PNE. higher acetylcholinesterase(15% and25%) and lower monoamine oxidase B (25% and 15%0 activities were effectively modulated by the administration of 0.5%-PNE and 0.1%-PNDE. These results suggest that more beneficial effects such as inhibition of cholesterol and lipofuscin, increase of membrane fluidity, higher acetylcholinesterase and lower monoamone oxidase activities in brain membranes of SD rats may be effectively modulated by administration of pine needle extract (PNE).
The asymmetric biosynthesis of ethyl (S)-4-chloro-3-hydrobutanoate from ethyl 4-chloro-3-oxobutanoate was investigated by using whole cells of Candida magnoliae JX120-3 without the addition of glucose dehydrogenase or $NADP^+/NADPH$. In a one-phase system, the bioconversion yield was seriously affected on the addition of 12.1 g/L ethyl 4-chloro-3-oxobutanoate. In order to reduce this substrate inhibition, a water/ n-butyl acetate two-phase system was developed, and the bioreduction conditions optimized with regard to the yield and product enantiometric excess value. The optimal conditions were as following: water to n-butyl acetate volume ratio of 1:1, 4.0 g DCW/L active cells, 50 g/L glucose and $35^{\circ}C$. By adopting a dropwise substrate feeding strategy, high concentration of ethyl 4-chloro-3-oxobutanoate (60 g/L) could be asymmetrically reduced to ethyl (S)-4-chloro-3-hydrobutanoate with high yield (93.8%) and high enantiometric excess value (92.7%).
To investigate the effects of low dosed microcystin -LR (MCLR) on the initial stage of subacute hepatotoxicity in vivo, male Sprague-Dawley rats after weanling were divided in four groups. The orally treated group (OG) was administered orally by 1 $\mu\textrm{g}$/kg B.W. at an interval of three days. The free group (FG) has taken the drinking water including 1 $\mu\textrm{g}$/L freely and the control group (CC) was only treated with 0.9% saline solution All groups were treated for a period of 3 weeks. There was a significant correlation in body growth rate between OG and FC and especially, a deterioration of the growth of spleen was observed in the FG after 5 days. The protein levels were also decreased in OG and FG after 9 days. Level of total fat was increased to the 9th day but again decreased up to the initial level. High hemolysis of the isolated erythrocytes occurred only in OG. Activities of ${\gamma}$-G7 of 0G and FG were higher twice-fold than CG, but the values of OG were already higher at the first treatment day. No significant change in aspartate aminotransferase (AST) activity was shown in all groups, but the activity of alanine aminotransferase (ALT) was slightly increased at the beginning state. There were much similarities in the results of OG and FG. except the growth inhibition of spleen in FG. It may be concluded that long -term effects of the low doses of mycrocystins in animals including human being can lead to serious health problems, especially to liver and spleen.
The relation between lipid peroxidation and thrombotic reaction were investigated in streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley male rats weighing 100$\pm$10gm were randomly assigned to normal and STZ-induced diabetic group(DM). Diabetes was experimentally induced by intravenous injection of 55mg/kg of body weight of STZ in citrate buffer(pH 4.3) after 4 weeks feeding of basal diet. Animals were sacrificed at the 6th day of diabetic states. Body weight gains were lower in diabetic group after STZ injection. Serum levels of thiobarbituric acid reacting substances(TBARS) that were markedly increased in DM group compared with of normal group. TBARS levels of HDL and LDL were similar patterns to total TBARA of serum. Activities of platelet phospholipase A2(PLA2) were higher in diabetic group than those of normal group. Activities of platelet cyclooxygenase were 106% in DM group than normal group. Platelet thromboxane A2(TXA2) formation was increased in DM group than normal group. Production of aortic prostacyclin(PGI2) was lower in diabetic group than that of normal group. PGI2/TXA2 ratios were decreased by 55% in DM groups than those of normal group. The present results indicate that STZ-induced diabetic rats are more sensitive to oxidative stess which leads to acceleration of lipid peroxidation and platelet aggregability. In conclusion, accelerating effect of lipid peroxidation and thrombogenesis in diabetic state is regareded to be resulted from enhancement of PLA2 activity and arachidonic acid metabolism, inhibition of antiaggrgating agent and aortic PGI2 formation.
Hypolipidemic effect of Insamsansa-eum (ISE, Ren-Shen-Shan-Zah-Yin in Chinese) consisted of Red ginseng (RG; the steamed root of Panax ginseng C.A. Meyer) and Crataegii fructus (CF, the fruit of Crataegus pinnatifida BGE) is used frequently in China and Korea as a herbal medicine to treat arteriosclerosis, was investigated. Treatment of ISE significantly reduced blood triglyceride (TG) and total cholesterol (TC) levels in Triton WR-1339-induced hyperlipidemic mice and serum TG levels in corn oil-induced hypertriglyceridemic mice. ISE significantly lowered the high blood TG and TC levels as well as body and epididymal mass weights in hyperlipidemic mice induced by long-term feeding of a high-fat diet and increased blood HDL cholesterol level. ISE and its ingredients, RG and CF, inhibited pancreatic lipase and HMG-CoA reductase activities. Of its ingredients, RG reduced the blood TC level and HMG-CoA reductase activity more than CF. CF reduced blood TG level and pancreatic lipase activity more than RG. These finding suggest that the antihyperlipidemic effect of ISE may be due to synergistic inhibition of HMG-CoA reductase and pancreatic lipase by RG and CF, and that ISE may be effective hypolipidemic agents.

The current study examined the effect of quercetin supplements on the lipid-lowering and antioxidant metabolism in ethanol-fed rats. The control $group(E_8)$ received ethanol only diet for 8 wks, whereas the other group$(E_8Q_4)$ received a diet including quercetin supplementation(0.05% wt/wt) for 4 wks while on the ethanol diet for 8wks. The hepatic alcohol dehydrogenase activity was significantly higher in the $E_8Q_4$ group than in the $E_8$ group. Supplementation with quercetin significantly elevated the HDL- cholesterol concentration, the HDL-C/total-C ratio, and lowered the atherogenic index(AI) compared to the $E_8$ group. The hepatic triglyceride and cholesterol contents were significantly lowered by the quercetin supplement compared to those of the control group. The hepatic HMG-CoA reductase and ACAT activities of the $E_8Q_4$ group were significantly lower than those of the $E_8$ group. The overall potential for antioxidant defense was significantly enhanced by the quercetin supplement, as indicated by a decrease in plasma and hepatic TBARS levels. The hepatic GSH-Px and G6PD activities were significantly higher in the $E_8Q_4$ group compared to the $E_8$ group. The current results suggest that dietary quercetin leads to the inhibition of HMG-CoA reductase and ACAT, which in turn lowers cholesterol levels and normalizes antioxidant enzyme activities.
BACKGROUND/OBJECTIVES: Seaweeds have been reported to have various health beneficial effects. In this study, we investigated the potential anti-obesity and anti-inflammatory effects of four types of domestic brown seaweeds in a high-fat diet-induced obese mouse model and bone marrow-derived macrophages (BMDM). MATERIALS/METHODS: Male C57BL/6N mice were fed low-fat diet (LFD), high-fat diet (HFD) or HFD containing Undaria Pinnatifida, HFD containing Laminaria Japonica (LJ), HFD containing Sargassum Fulvellum, or HFD containing Hizikia Fusiforme (HF) for 16 weeks. RESULTS: Brown seaweed supplementation did not affect long-term HFD-associated changes in body weight or adiposity, although mice fed HFD + LJ or HFD + HF gained slightly less body weight compared with those fed HFD at the beginning of feeding. Despite being obese, mice fed HFD + LJ appeared to show improved insulin sensitivity compared to mice fed HFD. Consistently, we observed significantly reduced blood glucose concentrations in mice fed HFD + LJ compared with those of mice fed HFD. Although no significant differences in adipocyte size were detected among the HFD-fed groups, consumption of seaweeds decreased formation of HFD-induced crown-like structures in gonadal adipose tissue as well as plasma inflammatory cytokines. BMDM from mice fed HFDs with seaweeds showed differential regulation of pro-inflammatory cytokines such as IL-$1{\beta}$ and IL-6 compared with BMDM from mice fed HFD by LPS stimulation. CONCLUSION: Although seaweed consumption did not prevent long-term HFD-induced obesity in C57BL/6N mice, it reduced insulin resistance (IR) and circulation of pro-inflammatory cytokines. Therefore, seaweeds may ameliorate systemic inflammation and IR in obesity partially due to inhibition of inflammatory signaling in adipose tissue cells as well as bone marrow-derived immune cells.
열대식물인 인도멀구슬나무(Azadirachta indica)는 병해충과 방제와 의학적인 목적으로 오랫동안 이용하였다. 본고에서는 님제의 활성성분인 아자디라크틴의 해충에 대한 곤충생장조절, 기피, 섭식저해, 산란억제 등 다양한 작용기작과 국내 이용현황에 대하여 고찰하였다. 국내 유통되는 친환경유기농자재 중에서 님제 제품은 총 57종이었다. 그중 효능효과가 표시된 자재는 7종이었고 평균 아자디라크틴 함량은 0.38%로써 세계적으로 유통되는 평균 함량 2.1%와 비교할 때 약 5.5배 낮았다. 국내에서 님제의 방제효과에서 진딧물은 실험에서 다소 변이는 있었으나 대부분 90% 이상의 방제효과를 보였다. 꽃매미 약충에 누적살충률은 73~77%를 보였고, 총채벌레에 대한 방제가는 50~72% 범위로 살포효과가 인정되었다. 노린재와 나방류에 대한 방제효과는 대체적으로 낮았다. 본고는 유통되는 님제품의 이해와 실험자료의 해석에 필요한 주요한 정보를 제공해줄 수 있을 것으로 기대된다.
