Studies on PVA/Chitosan/Fibroin Blend Sponge Sheets: Preparation and Wound Healing Effects in Rats

  • The preparation of PVA/Chitosan/Fibroin (PCF)-blended sponge sheets and wound healing effects of these sheets in rats were investigated. We excised the skin off rat, including the dermis, approximately 2${\times}$2 cm in size. The wound was coveted with PCF-blended spongy sheets. The spongy sheets absorbed the exudate, and gained flexibility and softness. Histopathological inspection of the wound 12 days later showed the increase of a vascular ingrowth and the absence of inflammatory cells. Regeneration of the skin around the wound was faster than that of the control. We also tested wound healing effects of PVA, Chitosan and Fibroini alone of in various combinations. Wound healing was accelerated in the order of PVA/chitosan/Fibroin (PCF)-blended sponge>Chitosan/Fibroin (CF)-blended sponge$\geq$Fibroin (F) sponge>PVA/Chitosan-blended sponge (PC)>Chitosan (C) sponge.

Retinal Blood Vessel Segmentation using Deep Learning (딥러닝 기법을 이용한 망막 혈관 분할)

  • Diabetic retinopathy is a complicated form of diabetes due to circulatory disorder in the peripheral blood vessels of the retina. We segment the microvessel for diagnosing diabetic retinophathy. The conventional methods using filter and features can segment the thick blood vessels, but it has relatively weak for segmenting fine blood vessels. In pre-processing step, noise reduction filter and histogram equalization are applied to suppress the noise and enhance the image contrast. Then, deep learning technique is used for pixel-by-pixel segmentation. The accuracy of conventional methods is between 90% to 94%, while the proposed method has improved as 95% accuracy. There is a problem of segmentation error around the optic disc and exudate due to the network depth. However the accuracy can be improved by modifying the network architecture in the future.

A case report of secondary infection by Vibrio splendidus associated with gas bubble disease in syngnathid fishes (Syngnathus schlegeli and Hippocampus haema)

  • Seahorses, which have been cultivated since the 2000s, are economically very important. Gas bubble disease (GBD) is a significant concern in the cultivation of seahorses; therefore, this study aimed to determine the cause of GBD-induced death in two species of Syngnathidae (Syngnathus schlegeli and Hippocampus haema). Rod-shaped bacteria were observed histopathologically and identified as Vibrio splendidus by conventional and real-time PCR analyses. The lethality of V. splendidus varies depending on the host's immune status, and the disease can be prevented through water quality management or improvement of the breeding environment. In this study, the GBD lesions (gas bubbles) were observed at 12℃, 8.0 mg/L of dissolved oxygen, 30 ppt of salinity, and pH 7.7. In addition, rod-shaped bacteria, infiltration of inflammatory cells, and extensive serous exudate were confirmed in the lesions where gas bubbles were found. PCR analysis was able to detect V. splendidus, possibly a secondary infection of the immunocompromised syngnathid fish. Understanding the risk of immunity control and the correlation between these lesions and causal agents will be of great help to the aquaculture industry and the ornamental fish market.

A Case of Chronic Nummular Dermatitis Improved by Hwangryunagyo-tang (황련아교탕(黃連阿膠湯) 투여로 호전된 만성 동전 모양 습진 1례)

  • Objectives: This study aims to report a case of chronic nummular dermatitis improved by Hwangryunagyo-tang. Methods: The patient in this study was a 15-year-old female, who visited our hospital with symptoms such as redness, erythema, papule, excoriation, scale, exudation, crusting, lichenification, and pruritus of the extremities and trunk. Considering that the skin lesions were coin-shaped and persisted for more than 6 months, the patient's symptoms were diagnosed as chronic nummular eczema. Based on the patient's symptoms and overall condition, Hwangryunagyo-tang was prescribed. Results: Approximately 4 months after administration of Hwangryunagyo-tang, the exudate disappeared, and overall skin symptoms improved, from objective score 33.7 to 6.6, leaving only slight scale, dryness, and pigmentation. As the pruritus of the skin lesion decreased from numeric rating scale 8 to 1, the patient's sleeping condition also improved from numeric rating scale 9 to 0. Conclusion: This study is significant in that the symptoms of chronic nummular dermatitis were effectively improved after administration of Hwangryunagyo-tang.

A Case Report of Contact Dermatitis on the Burn Lesion (화상 부위에 발생한 접촉피부염 치험 1례)

  • Objectives : The purpose of this study is to report the case of contact dermatitis on the burn lesion treated with Korean medicine treatment including Bangpoongtongseongsan gagam, Jaeumganghwatang gagam and pharmacopuncture. Methods : A 28-year-old female patient had complaints of erythema, swelling, pruritus, exudate due to contact dermatitis caused by the band used to treat the left forearm burn lesion from February 10, 2024. The patient visited Dong-Eui Korean medicine university hospital on February 19, 2024, and we treated this patient with herbal medicine, pharmacopuncture, and western medicine. The effects of treatments were evaluated by photo comparision, visual analogue scale(VAS), dermatology life quality index(DLQI) and sleep disorder score. Results : After treatment, the patient's symptoms improved. VAS of pruritus was decreased from 9 to 0, DLQI was decreased 25 to 0 and sleep disorder score was decreased 7 to 0. Conclusions : The results suggest the Korean medicine treatment including herbal medicine and pharmacopuncture could be effective in treating patient with contact dermatitis.

  • Background : Idiopathic pulmonary fibrosis(IPF) is a devastating illness for which there is little effective treatment. The key cytokines currently implicated in the fibrotic process are the transforming growth factor-${\beta}_1$(TGF-${\beta}_1$), tumor necrosis factor-$\alpha$(TNF-$\alpha$), endothelin-1(ET-1) and interferon-$\gamma$(IFN-$\gamma$). The rat model for paraquat-induced pulmonary fibrosis was chosen to investigate the role of ET-1 in this disease. Both ET-1 and TGF-${\beta}_1$ expression in lung lesions were examined using immunohistochemical staining. After $Bosentan^{(R)}$ administration, an orally active ET-$l_A$ and ET-$1_B$ receptor antagonist, the degree of pulmonary fibrosis and ET-1 and TGF-${\beta}_1$ expression were analyzed. Method : Sprague-Dawley rats were divided into three groups, the control group, the fibrosis group, and the fibrosis-$Bosentan^{(R)}$-treated group. The animals were sacrificed periodically at 1, 3, 5, 7, 10, 14 days after administering saline or paraquat. The effects between groups were compared with the results of light microscopy and immunohistochemical staining for ET-1 and TGF-${\beta}_1$. The degree of fibrosis was evaluated by H&E and Masson's trichrome staining, which were graded by a computerized image analyzer. The degree of immunohistochemical staining was categorized by a semi-quantitative analysis method. Results : The lung collagen content had increased in the paraquat instillated animals by day 3, and continued to increase up to day 14. A daily treatment by gavage with $Bosentan^{(R)}$ (100mg/kg) did not prevent the increase in collagen deposition on the lung that was induced by paraquat instillation. There were increased immunohistochemical stains of ET-1 on the exudate, macrophages, vascular endothelial cells and pneumocytes in the paraquat instillated group. Furthermore, TGF-${\beta}_1$ expression was higher on the exudate, macrophages, some inflammatory cells, pneumocytes( type I, and II), vascular endothelium and the respiratory epithelial cells around the fibrotic area. After Bosentan treatment, there were no definite changes in ET-1 and TGF-${\beta}_1$ expression. Conclusion : Fibrosis of the Paraquat instillated group was more advanced when compared with the control group. In addition, there was increased ET-1 and TGF-${\beta}_1$ expression around the fibrotic area. Studies on Constituents of the Higher Fungi of Korea (LIV) Antitumor Components of Favolus alveolarius

  • To find antitumor components from the higher fungi of Korea, the mycelia of Favolus alveolarius (Fr.) Quelet were cultured in a liquid medium. The cultured mycelia were extracted with hot water twice, and a high molecular weight fraction was obtained by adding two volumes of ethanol to the extract. Two grams of Fraction A were obtained by dialyzing it. It was further separated into four fractions by gel filtration with Sepharose CL-4B, and they were designated Fractions B, C, D, and E. The results of the antitumor test showed that Fractions A, B, C, D and E had tumor inhibition ratios of 92.3, 78.5, 59.6, 77.4 and 62.2%, respectively. Anthrone test was carried out to determine the contents of total polysaccharide of the five fractions, and they had 46.3, 27.3, 65.3, 64.6, and 46.1%, respectively. The contents of the total protein of the five fractions were 29.4, 13.9, 14.3, 14.3, and 29.1%, respectively. Monosaccharide subunits of each fraction were analyzed with gas chromatography, and glucose, xylose, mannose, galactose and fucose were identified. Fraction A was examined for immunological effects. It increased the count of hemolytic plaque forming cells 12.8 times to that of the control group, and the population of macrophage in peritoneal cavity 3.2 times to that of the control group.

Inhibitory activities of polysaccharide purified from Phellodendron chinese SCHNEID on melanoma B-16-derived metastatic tumor and hypersensitivity

  • The polysaccharide fractions were isolated and purified from Phellodendron chinese SCHNEID, and antitumor activities using the melanoma B-16-derived metastatic tumors were examined at dosages of 2, 5 and 10 mg/100 g. F-7 and F-8 showed the highest tumor metastatic inhibitory activities (inhibition ratio 60 and 80% in 2 mg/100 g), and in dose of 5 mg/100 g, the inhibitory ratios were 85 and 70%, respectively. Furthermore, 10 mg/100 g of intraperitoneal (i.p.) injection gave 90 and 95% of inhibition. When the effects of polysaccharides on hypersensitivity were examined, the inhibitory activities were not markedly observed in oral administration, indicating that the polysaccharides are directly acting to immune system. Also, the polysaccharides increased the number of circulating blood leukocytes and total peritoneal exudate cells. Although implantation of tumor cells greatly decreased the productivity of antibody (antibody-mediated) and T lymphocyte reactivity (delayed-type) as 6.3 from 9.3 and 5.9 from 7.7, represented by the increase of footpad thickness, respectively. the polysaccharides elevated the reactivity of T lymphocyte in tumor-bearing mice, which were rapidly recovered by discontinuance of sample treatments. Especially, F-2, F-5, F-7 and F-8 remarkably recovered the decreased sensitivity. These results clearly indicated that the i.p. injection is much effective to suppress tumor growth than oral administration.

Isolation and immunohistochemical diagnosis of Haemophilus parasuis from naturally occurring polyserositis in pigs (자연발생한 돼지 다발성 장막염 예로부터 Haemophilus parasuis의 분리와 면역조직화학적 진단)

  • From Jan. 1996 to Oct. 1997, 29 pigs with 40-70 days old showing dyspnea inappetite and polyserositis were collected and carried out necropsy, bacterial culture, histopathology, avidin biotin complex(ABC) stain, fluorescent antibody(FA) test, and electron microscopy. In the study, 4 strains from 3 pigs were isolated from meninges, pleura and synovial fluid and also were identified as Haemophilus parasuis serovar 5. Main histopathological lesions of 29 pigs with polyserositis were frequently composed of fibrinous peritonitis(27), pleurisy(22), interstitial pneumonia(21), fibrinous epicarditis(20), fibrinopurulent meningitis(8) and synovitis(4). By ABC stain, 11/29(37.9%) pigs with polyserositis were confirmed to have H parasuis serovar 5 in the cytoplasm of macrophages and neutrophils in cerebral meninges, epicardium, pleura surface of lung or serosa of spleen. ABC stain(20.8~40.0%) to detect H parasuis serovar 5 in tissues was more sensitive than bacterial culture(10.3%), but less sensitive than FA test(62.5%) using frozen tissues even though the result of 8 cases. By electron microscopy, a bacterium was also detected in the cytoplasm of macrophages in purulent exudate of cerebral meninges. Consequently, we confirmed that H parasuis serovar 5 has been involving to cause pigs with polyserositis and can be detected by FA and ABC stain as reliable diagnostic tools.

Carcass Characteristics and Qualitative Attributes of Pork from Immunocastrated Animals

  • An investigation was carried out to assess the carcass characteristics and meat quality aspects of immunocastrated male pigs of medium genetic potential for lean meat deposition in carcass (53 to 55%). When the crossbred Large White x Landrace pigs (n = 45) were 70 days old, they were distributed in a totally randomized design in three treatments (castrated males, females and immunocastrated males) with three replicates of five animals. The pigs were slaughtered when they were 140 days old. Carcass temperature and pH were recorded twice, at 45 min and 24 h after slaughter. The carcasses were evaluated for hot and cold carcass yield, commercial cuts yield, length and depth, back fat thickness, loin eye area and lean meat percentage. The Longissimus dorsi muscle was extracted for analysis of color ($L^*$, $a^*$, $b^*$), exudate loss, cooking loss and centesimal and sensorial analysis of the meat. There were no significant differences for the evaluated parameters between castrated males, immunocastrated males and females, except for backfat thickness between the 7th and 8th thoracic vertebra and the point P2 (lower for immunocastrated males) and carcass temperature at 45 min post slaughter (higher in immunocastrated males), however, this did not interfer with the rate of pH decrease post mortem and the meat quality. The results from this research did not indicate a benefit of immunocastration on carcass characteristics of pigs of medium genetic potential for lean meat deposition in carcass, when compared to surgical castration.
