Residue of Synthetic Antimicrobial Agent in Eggs by High Performance Liquid Chromatography (고속액체크로마토그라피에 의한 계란중의 합성항균제 잔류량)

  • A simultaneous determination method by HPLC for egg-residues sulfamerazine, sulfamethazine, sulfadimethoxine, furazolidone and zoalene was assesed. The drugs were extracted by dechloromethane, The extract after solvent evaporation, is partitioning in hex ane/water and back-partitioning in dechloromethane and analysis by HPLC. The average recovery rates of the above microbials from the egg spiked standard solution were approximately 81.2%, 87.6%, 92.5%. 86.1% and 79.3% respectively. The limit of detection of sulfamerazine. sulfamethazine and sulfamethoxine were in the levels of 0.2ppb, furazolidone and zoalene 0.5ppb respectively. According to this method 84 commercial eggs were examined. Sulfamethanzine was detected at levels of 0.005-0.008ppm in 3 eggs. Sulfadimethoxine was detected at levels of 0.012-0.019ppm in 4 eggs. No sulfamerazine, furazolidone and zoalene was detected in every samples. The residues of antimicrobial agent were safety level as food generally.

Preparation of high Purity manganese oxide by Pyrolysis of solution extracted from ferromanganese dust in AOD process

  • The high purity manganese oxides were made from the dust, generated in AOD process that produces a medium-low carbon ferromanganese and collected in the bag filter. Manganese oxide content in the dust was about 90%, and its phase was confirmed as Mn₃O₄. In the extraction of manganese, because of remaining amorphous MnO₂, the dust was reduced to MnO by roasting with charcoal. The pulp density of the reduced dust can control pH of the solution more than 4 and then Fe ion is precipitated to a ferric hydroxide. Because a ferric hydroxide co precipitates with Si ion etc, Fe, Si ion was removed f개m the solution. Heating made water to be volatized and nitrates was left in reactor Then nitrates were a liquid state and stirring was possible. Among the nitrates in reactor, only the manganese nitrate which have the lowest pyrolysis temperature pyrolyzed into β-MnO₂powder and NO₂(g) at the temperature less than 200℃. When the pyrolysis of manganese nitrate has been completed about 90%, injection of water stopped the pyrolysis. Nitrates of impurity dissolved and the spherical high purity β-MnO₂powders were obtained by filtering and washing. Mn₂O₃or Mn₃O₄ powder could be manufactured from β-MnO₂powder by controlling the heating temperature. Lastly, a manufactured manganese oxide particle has 99.97% purity.

  • Acne vulgaris, the most common skin disease. can be formed as only a few comedons or severe inflammatory lesions. The pathogenesis of acne involves various factors; excessive androgen, excessive sebum production, abnormal alteration of follicular epithelium, proliferation of Propionibacterium acnes, and inflammation. We investigated acne therapy using oriental herbs described in the Korean traditional medical book(Dong-ui-bo-gam). Oriental herbs(Angelica daurica, Arctium lappa, Coptidis rhizoma, and Glycyrrhiza glabra) were chosen based on their respective property of sebum control, anti-inflammatory activity, and anti-bacterial activity. We examined the effect of acne treatment, in terms of chemotactic inhibition, lipogenesis inhibition, and anti-bacterial activity for P. acnes. 1. Neutrophil chemotaxis assay; P. acnes secrete chemotactic factors and other pro-inflammatory extracellular products. Neutrophil chemotactic activity of P. acnes was measured by 48-well chemotaxis method. Angelica daurica clearly suppressed chemotactic activity of P. acnes. 2. Using sebaceous gland of hamster ear lipogenesis assay; Sebaceous lipogenesis was measured using ear biopsies by incubation or $C^{14}$-acetate in culture media. The $C^{14}$-labeled lipids were extracted and determined by liquid scintilation counting. Coptidis rhizoma markedly inhibited sebum production. 3. Anti-bacterial assay for P. acnes(MIC test); Glycyrrhiza glabra showed anti-bacterial activity. P. acnes did not develop resistance against Glycyrrhiza glabra. Retinoids are effectively to inhibit sebum production and regulate follicular keratinization process, with little anti-inflammatory activity. Angelica daurica suppressed neutrophil chemotaxis, Coptidis rhizoma inhibited sebum production, and Glycyrrhiza glabra showed anti-bacterial activity against P. acnes. A combined formulation of Angelica daurica, Coptidis rhizoma. and Glycyrrhiza glabra is expected to provide effective acne treatment.

Physicochemical Characterization and NMR Assignments of Ginsenosides Rb1, Rb2, Rc, and Rd Isolated from Panax ginseng

  • The fresh ginseng roots were extracted with aqueous methanol, and the obtained extracts were partitioned using ethyl acetate, n-butanol, and water, successively. The repeated silica gel and octadecyl silica gel column chromatogaraphy for n-butanol fraction afforded four diol ginseng saponins, ginsenosides $Rb_1$, $Rb_2$, $R_c$, and Rd. The physicochemical, spectroscopic, and chromatographic characteristics of these ginsenosides were measured and compared with those reported in the literature. Some of the peak assignments in previously published $^1H$- and $^{13}C$-nuclear magnetic resonance (NMR) spectra were inaccurate. This study employed two-dimensional NMR experiments, including $^1H-^1H$ correlation spectroscopy, heteronuclear single quantum correlation, and heteronuclear multiple bond connectivity, to determine exact peak assignments.

Determination of Analytical Method for the Insecticide Clothianidin and its Metabolites in Soil and Surface Water (토양 및 토양수 중 살충제 Clothianidin 및 대사산물 잔류분석법 확립)

  • BACKGROUND: The purpose of this study was to investigate the effects of clothianidin on the soil in terms of clothianidin dissipation and degradation to evaluate its safety in order to provide an analytical foundation for clothianidin and the 5 metabolites related to it. METHODS AND RESULTS: High-performance liquid chromatography(HPLC) was used to separate clothianidin and its metabolites in this study. In soil, after suppressing dissociation-proned ions with weak alkalic $NH_4OH$ and extracting the metabolites with methanol, clothianidin, Methylaminoimidazole(MAI), Methylnitroguanidine(MNG), Thiazolylmethylurea(TZMU) and Thiazolylnitroguanidine(TZNG). Thiazolylmethylguanidine(TMG) were extracted with the addition of neutral $NH_4OAC$ to increasing the intensity of ions. Compounding elements were separated by using Hydrometrix ($ChemElut^{TM}$) and ion-exchanging Solid-phase extraction(SPE) Strong cation-exchanger(SCX) and C18 were used. The recovery rates of clothianidin and 5 metabolites in soil and water ranged from 87.4% to 104.3%. A standard deviation of our analysis for the soil and water samples were less than 5%. CONCLUSION: Well accepted detection limits for clothianidin and 5 metabolites in soil samples based on a dissipation analysis is 0.005 mg/kg and 0.001 mg/L in water samples. The dissipation concentration of this study was decided to be enough to evaluate the dissipation levels of clothianidin and its metabolites.

Studies on Lipid Composition of Egg from Fugu xanthopterus (까치복 알의 지질 조성에 관한 연구)

  • Total lipid were extracted from the eggs of Fugu xanthopterus and were then resolved into lipid subclasses by silica gel column chromatograpy. The fatty acid composition of total lipid, triacylglycerols, phospholipids and wax esters of these oils were analyzed by gas-liquid chromatography. Proximate percentage of total lipid from the sample was 12.63%. The lipid classes of eggs of Fugu xanthopterus fractionated by silica gel column chromatograpy were 59.37% of triacylglycerols, 15.46% of phospholipids, 6.9% of wax esters. The egg lipid obtained from Fugu xanthopterus was enriched with DHA in every class. In particular, the phospholipid contained the highest level of DHA(19.3%) than the other. The sum of polyenoic acids(27.4%). The fatty alcohols were mainly composed of saturated alcohols such as C16:0 alcohol(62.6%), C14:0(7.9%) and C18:0(5.8%), while the level of polyene alcohols was strikingly low compared with that of saturated alcohols.

  • Lipids of Job's tears (Yullmoo) were extracted by the mixture of chloroform: methanol (2:1, v/v) and from these lipids, triglycerides were separated by column chromatography (Sephadex G-25 column) and then confirmed by thin layer chromatography (TLC) and reconfirmed by gas chromatography (GC). The triglycerides were fractionated into 6 groups on the basis of their partition numbers (PN) by reverse phase high performance liquid chromatography (HPLC) on a column Shimpack CLC-ODS using tetrahydrofuran-acetonitril (25:75) mixture as a solvent. Each of these collected fractions gave one to two peaks in the GC chromatograms according to the acyl carbon number (CN) of the triglyceride and fatty acid composition of the triglycerides were also analyzed by GC. From the results, the major triglycerides of Job's tears lipids were estimated to be C18:1 C18:2 C18:2 (OLL, 38.2%), C16:0 C18:2 C18:3 (PLLn, 15.7%), C18:1 C18:1 C18:2 (OOL, 12.6%).

Isolation of Antifungal Lactic Acid Bacteria (LAB) from "Kunu" against Toxigenic Aspergillus flavus

  • The antifungal activity of isolated lactic acid bacteria (LAB) from a locally fermented cereal, "Kunu", was tested against toxigenic Aspergillus flavus. The liquid refreshment, "Kunu", was prepared under hygienic condition using millet, sorghum, and the combination of the two grains. The antifungal potential of isolated LAB against toxigenic A. flavus was carried out using both in vitro and in vivo antifungal assays. The LAB count from prepared "Kunu" ranged from $2.80{\times}10^4CFU/mL$ to $4.10{\times}10^4CFU/mL$ and Lactobacillus plantarum, Lactobacillus delbrueckii, Lactobacillus fermentum, Pediococcus acidilactici, and Leuconostoc mesenteroides were the isolated bacteria. Inhibitory zones exhibited by LAB against toxigenic A. flavus ranged from 5.0 mm to 20.0 mm. The albino mice infected with toxigenic A. flavus showed sluggishness, decrease in body weight, distortion of hair, and presence of blood in their stool, while those treated with LAB after infection were recovered and active like those in control groups. Except for the white blood cell that was increased in the infected mice as $6.73mm^3$, the packed cell volume, hemoglobin, and red blood cell in infected animals were significantly reduced (P<0.05) to 29.28%, 10.06%, and 4.28%, respectively, when compared to the treated mice with LAB and control groups. The antifungal activity of LAB against toxigenic A. flavus can be attributed to the antimicrobial metabolites. These metabolites can be extracted and used as biopreservatives in food products to substitute the use of chemical preservatives that is not appealing to consumers due to several side effects.

Antibacterial Effect of Siegesbeckia pubescens Extract against Fish Pathogenic Streptococcus iniae (희렴(Siegesbeckia pubescens) 추출물의 어병세균 Streptococcus iniae에 대한 항균활성)

  • Antibacterial activity of 80% methanol extract from 13 commercial herb medicines was measured against fish pathogens Streptococcus iniae, causing several diseases in various kind of fish. Siegesbeckia pubescens showed the strongest antibacterial activity against S. iniae. Methanol extract of S. pubescens was further extracted using several organic solvents having different polarity. Extract from n-hexane and ethyl acetate fraction showed strong activity. Minimal inhibitory concentration, MIC of S. pubescens extract was measured and resulted showing 8 μg/mL with n-hexane fraction and 32 μg/mL with ethyl acetate fraction against S. iniae. The growth of S. iniae was fully inhibited by adding 50 μg/mL (final concentration) of n-hexane or ethyl acetate fraction in the liquid media. It is needed that, from these results, purification and isolation of responsible active compound(s) of these activities and further study on the synergy effect using combination with commercial antibiotics against fish pathogenic bacteria.

Study on purification and extraction of nitrate salts from waste scrubbing liquid of de-SOx/de-NOx (탈질/탈황 폐 세정액으로부터 질산염 추출 및 정제 연구)

  • IMO to issue some restricted maritime legislation for reducing the adverse environmental impacts arising from such ship exhaust emissions. According to the IMO policy, every ship entering the Baltic SECAs has to equip the gas cleaning scrubber. The discharged waste solution by gas cleaning scrubber contains many types of salts, which to recover some valuable materials before disposal. This study try to achieve valuable salts including AN and AS throughout a few process such as selective organic solvents salting out, low temperature extraction and thermal evaporation. Amongst them, Thermal evaporation with repetition extraction using inorganic solvent was the most optimum to purify the extracted AN. This valuable salt was evaluated by Elemental analysis and Differential scanning calorimetry.
