  • The extraction efficiency of orange-yellow pigment from the Gardenia was greatly depended upon the extraction time, extraction temperature, volume of solvent used and fat contents of the Gardenia. From the experimental results, the amounts of extracted pigment (P) was proportional to the $log\;t^{\;1{\cdot}15}$ of extraction time$(t;\;0{\sim}60\;min.)$, the $log\;T^{3{\cdot}73}$ of extraction temperature$(T;\;5{\sim}60^{\circ}C)$, the $log\;S^{3{\cdot}7}$ of volume of solvent$(S;\;5{\sim}50\;ml)$, and the -4X of fat contexts of sample $(X;\;0{\sim}0.\;15)$ at $18^{\circ}C$ for 10 minutes. Finally, the modified empirical equation was derived as follow; $P{\simeq}1.15\;log\;t+3.73\;log\;T+3.7\;log\;S-4X-6.4$ In addition to that, the most optimum conditions of pigment extraction were determined as 30 minutes of operation time, $40^{\circ}C$ of temperature. Deffated Gardenia was more productive than natural Gardenia in the pigment extraction.

  • The composition of four rice protein groups is greatly affected by the extraction conditions. The extraction amounts of albumins and glubulines primarily depended on the temperature rather than the method of extraction. The total amount of glutelins, the major components of rice storage proteins, could be extracted by a successive extraction processes, extraction with 0.5% SDS-0.1M borate buffer(pH 8.3) followed by extraction with 0.5% SDS-0.6% ${\beta}-mercaptoethanol-0.1M$ borate buffer(pH 8.3). The extracted amounts of glutelin with these solvents were 54.1 and 45% respectively. The further purification of SDS soluble glutelins was achieved by Sephadex G-150 gel column chromatography. The molecular weight of the components in four protein groups has been estimated by SDS-polyacrylamide gel electrophoresis with or without ${\beta}-mercaptoethanol.$ The comparison of albumins and globulins by starch gel electrophoresis at pH 3.1 permitted us to identify seven rice varieties. However, at pH 8.95, the specific bands for Japonica type rice varieties were observed.

  • Extraction of DNA from skeletal material is of great importance in the identification of human remains, but is particularly difficult because the high amount of microbial DNA was often co-extracted with human bone DNA. We found that a phenol/chloroform extraction, followed by ultrafiltration, and cleanup by via the $QIAquick^{(R)}$ PCR purification kit yields higher amounts of human genomic DNA compared with extraction by the column affinity $method^{(R)}$ alone. Ultrafiltration extraction of human DNA from ten exhumed bone samples yielded $0.041-1.120ng/{\mu}L$ DNA (mean = $0.498ng/{\mu}L$ DNA), and purification using the column affinity resulted in $0.016-0.064ng/{\mu}L$ DNA (mean = $0.034ng/{\mu}L$ DNA). Although the STR genotyping by the column affinity method was partially successful, all DNA samples by the ultrafiltration method produced full profiles from the multiplex PCR. The efficiency of STR genotyping was in accordance with the amounts of the human DNA extracted.

Microwave Assisted Extraction of Physiologically Active Materials from Agaricus blazei Fruiting Bodies (Agaricus blazei 자실체로부터 마이크로웨이브를 이용한 생리활성물질 추출공정)

  • Physiologically active material ${\beta}$-glucan extracted from mushroom has an antitumor effect. Agaricus blazei murill(A blazel) fruiting bodies were reported to contain large amounts of ${\beta}$-glucan. The purpose of this study was to maximize ${\beta}$-glucn extraction from A. blazei by microwave assisted extraction method. Optimal extraction conditions were obtained under various extraction time and applied power which was examined as 200, 500 and 650 watt. The structure and concentrations of the raffinates were analyzed by nuclear magnetic resonance spectroscopy(NMR) and infrared spectroscopy(IR) It was proved that the extraction yield of microwave assisted extraction method was higher than that of conventional solvent extraction method. It was also observed that microwave assisted extraction method requires less amounts of solvents and extraction time compared to the conventional solvent extraction method.

  • The studied area occupies the southern part of Haenam peninsula located in the southwestern corner of Korea. The stratigraphic sequences of the area are grouped into following three units in ascending order. (1) Late Cretaceous sedimentary rocks of Uhangri Formation which consist of the alternative beds of black shales and tuffaceous sediments, Hwangsan acidic fine grained tuffs and Jindo rhyolite flows. (2) Late Cretaceous biotite granite and acidic porphyries which intruded the Late Cretaceous sedimentary rocks. (3) Tertiary(?) pitchstone. The study purposes to delineate any of geochemical aspects on the deposition of the sequences, the average amounts of organic materials and the effect to the maturation of hydrocarbonization which has been occured by the intrusives. Sixty nine black shale samples were collected from Uhangri Formation in the northern and southern coasts of the peninsula. Organic carbons, total extracted organic matters and hydrocarbons were mainly determined by the Soxhlet extraction method, together with C-H-N elemental analyses. Based on the field and laboratory studies, the following interpretations have been obtained. (1) The paleohydrostatic condition of the sedimentation which took place was relatively calm and stable to delineate a lacustrine environment. (2) The amounts of organic material were more or less constant throughout the period of the deposition of the sediments. (3) The degree of hydrocarbonization of the Uhangri Formation in the northern coast was taken place higher than that in the southern coast due to the differences of thermal effect by the biotite granite which is assumed to be as a heat source in the area. Among the northern coast, some black shales in Uhangri and Mogsam areas which had been under the most moderate temperature environment snow symptomatic oily material, whereas, those in Sinseongri area do not, which were presumably to be intensive thermal alteration by the later acidic porphyries.

Effects of Changes in Composition of Herb Extract Product on Growth of Spoilage Yeast, Zygosaccharomyces sp. (생약복방제의 조성 변화가 부패성 효모 Zygosaccharomyces sp. 의 성장에 미치는 영향)

  • The aim of the present study was to investigate effects of food preservative addition and changes in composition of herb extract product on the growth of spoilage yeast, Zygosaccharomyces sp. Herbs such as Panax ginseng, Cinnamomum cassia, Lycium chinense, Zyzyphus juiuba and Jingiber officinale were altogether put into water and essence was extracted at 80oC, and then the extract was concentrated at 75oC. The herb extract product was made by adding vitamins, amino acids and honey to the concentrated herb extract. The amount of gas produced from the herb extract product was increased as inoculated cell number increased but decreased as Brix concentration increased. Gases were produced in small amount when incubation was made at 4oC but large amounts of gases were produced at 25 or 40oC of incubation. The gas production and growth of Zygosaccharomyces sp. were measured after browning reaction was induced by heating at 85oC for 12 hours. It appeared that heating treatment did not induce any significant change in the gas production and growth of the cell. The effects of addition of various sugar to the herb extract produce were also invesigated. Amounts of gas production were in the order of glucose>sucrose>oligosaccharide>stevioside. The viable cell count was measured as 6.0$\times$107 CFU/g when glucose was added to the herb extract product. The viable cell counts were 5.0$\times$106, 3.0$\times$103, and 3.0$\times$102 CFU/g in sucrose, oligosaccharide and stevioside added herb extract product, respectively. The amount of gas production from the herb extract product was remarkably reduced by addition of such food preservatives as sodium benzoate and DF 100. TLC(thin layer chromatography) chromatogram of the herb extract showed stability of the herb extract in the above treatments.

Essential Oil Components in Herb Teas (Rose and Rosehip) (로즈차와 로즈힙차의 휘발성 향기 성분)

  • The purpose of this study was to characterize the aromas of rose tea and rosehip tea. Aroma compounds were extracted by simultaneous distillation and extraction method using a Likens and Nickerson's extraction apparatus. The concentrated aroma extracts were analyzed and identified by GC and GC-MS. Thirty-eight compounds, including phenylethyl alcohol, citronellol, menthol, menthone, linalool and geraniol, were isolated and identified in rose tea. Thirty-six compounds, including menthol, $\alpha$-anethole, $\alpha$-terpinolene, menthone, linalool and 6-methyl-5-heptene-2-one, were isolated and identified in rosehip tea. Large amounts of phenyl ethyl alcohol and citronellol were found in rose tea, while large amounts of menthol and $\alpha$-anethole were found in rosehip tea.

  • To identify the volatiles of Platycodon roots, the three-year-old and the 24-year-old Platycodon roots cultivated in a local farm were extracted with ethyl ether by SDE method. 54.71mg% and 80.01mg% of volatile components were collected in the three-year-old and in 24-year-old Platycodon roots, respectively. As a result of GC/MS(TIC) analysis, 24 and 28 compounds were identified, respectively; The amounts of volatile components such as hexanol, trans-2-hexenol and nonanal in 24-year-old roots were detected above ten times than those in 3-year-old roots. The Volatiles of 24-year-old roots were fractionated by SCC and identified by GC/MS to be 10 hydrocarbons, 8 alcohols, 10 aldehydes, 5 esters, 6 acids and 1 unknown compound as the major components. The species and total amounts of volatile component in 24-year-old Platycodon roots were considered to be higher than those in 3-year-old Platycodon roots. *Abbreviations 1.   • This investigation was carried out to study the effect of heating treatment on the saponin fractions in ginseng extract and crude saponin. The changes of saponin patterns and amounts were investigated using HPLC and compared with peak area of each fraction. Shape of crude saponin was changed more easy to compare with in ginseng extract. The more extracting temperature risen and treated time longer, the more changes of saponin amounts and patterns were shown, expecially at $100^{\circ}C$ over. All of the saponin fraction except ginsenoside-Rd were relatively unstable in heating treatment. Suitable extracting condition was extracted at $80^{\circ}C$ for 40 hours with $H_2O$.

Application of a Gas Chromatography/Mass Spectrometric Method for the Determination of Butyltin Compounds in Sediment

  • A gas chromatography/mass spectrometric (GC/MS) method has been developed for the determination of trace mono-n-butyltin (MBT), di-n-butyltin (DBT), and tri-n-butyltin (TBT) compounds in sediments. Samples were extracted by 10% acetic acid in methanol containing 0.03% tropolone and were then derivatized for GC/MS analysis. Ethylation by sodium tetraethylborate and phenylation by sodium tetraphenylborate were evaluated as a derivatization reaction of the organotins in sample extract. n-Hexane was added into reaction media in the beginning of the reaction for the continuous extraction of derivatized organotins. Ethylation requires less than 2 hours to get proper derivatization yields for MBT, DBT, and TBT altogether and produces relatively low amounts of side reaction products. Compared to ethylation, phenylation requires much longer time but provides relatively lower yield and produces considerable amounts of side reaction products. Therefore, the ethylation reaction was applied for the analysis of organotin compounds in sediment. An isotope dilution mass spectrometric (IDMS) method based on GC/MS has been applied to the accurate determination of DBT compounds in the sediments. The IDMS results from the analyses of sediment samples showed a reasonable repeatability and a good agreement with the values obtained by IDMS based on liquid chromatography/induced coupled plasma/mass spectrometry.
