Adaptive Response Induced by Low Dose Ionizing Raditation in Human Cervical Carcinoma Cells

  • Adaptive response induced by low dese .gamma.-ray irradiation in human cervical carcinoma cells was examined. Cells were exposured to low dose of .gamma.-ray irradiation in human cervical carcinoma cells was examined. Cells were exposured to low dose of .gamma.-ray (1-cGy) followed by high doses of r-ray irradiation (0,1,2,3,5,7 and 9Gy for chlnogenic assay or 1.5Gy for micronucleus assay) with various time intervals. Survival fractions of cells in both low dose-irradiated and unirrated groups were analyzed by clonogenic assay. Surviva fractions of low dose-irradiated in cell survival was maximum when low and high dose irradiation time interval was 4 hr. Frequencies of micronuclei which is an indicative of chromosome aberration were also enutained from survival fractions analyzed by clonogenic assay, maximum when low and high dose irradiation time interval was 4hr. Frequencies of micronuclei which is an indicative of chromosome aberration were also enumerated in both low dose-irradiated and unirradiated groups. In consiststent with the result obtained from survival fractions analyzed by clonogenic assay, maximum reduction in frquencies of micronuclei was observed when low dose radiation was given 4 hr prior to high response to subsequent high dose .gamma.-ray irradiation in human cervical carcinomal cells. Our data suggest that one of the possible mechanisms of adaptive response induced by low dose rediation is the increase in repair of DNA double strand breaks in low dose radiation-adapted cells.

Changes in Expression of Connexin Isoforms in the Caudal Epididymis of Adult Sprague-Dawley Rats exposed to Estradiol Benzoate or Flutamide at the Neonatal Age

  • Direct communication between neighboring cells via gap junction in tissue is important for maintenance and regulation of its physiological functions. Each epididymal region has different composition of cell types. It is well recognized that the epididymis is a steroid hormone-responsive tissue. The present study was designed to determine the effect of estradiol benzoate (EB) or flutamide exposured at the early postnatal age on the expression of connexin (Cx) isoforms in the caudal epididymis. The EB or flutamide was subcutaneously administrated to male Spragure Dawley rat at 7 days of age, and expressional changes of Cx isoforms in the adult corpus epididymis were determined by quantitative real-time PCR. The treatment of low-dose EB resulted in decreases of Cx30.3, Cx31.1, Cx37, and Cx45 expression but caused an increase of Cx32 expression. Exposure to high-dose EB led into expressional increases of Cx31, Cx31.1, Cx32, Cx40, and Cx43, even though a decrease of Cx37 expression was found with a high-dose EB treatment. A low-dose flutamide induced increases of Cx31, Cx31.1, Cx32, and Cx43 expression but a decrease of Cx37 expression. Expression of most Cx genes were significantly increased by a high-dose flutamide, while no expressional change of Cx26 and Cx40 was detected by a high-dose flutamide. These results indicate that expression of Cx isoforms in the caudal epididymis is altered by exposure to steroidal compounds at the prepubertal age. It is suggested that a contact with environmental exogenous materials during the early postnatal period would lead to alteration of epididymal functions at the adult.

흉부의 ANALOG-DIGITAL 영상에 대한 비교 평가

  • This paper were studied the evaluation in compare with the conventional and AMBER of analog images, PACS and CR of digital images which were collected every ten sampling chest images with the J.J.Vucicuh chest evaluated chart, and were measured the chest phantom surface dose and the density of several part in chest images. The evaluated numbers were total 22 persons who were 6 persons of the M.D., 6 of the radiotechnological professors and 10 of the radiotechnologists. The obtained results summarized as following : 1. Approaching the optimum standard density of the several part in chest images drew near at the lung round region density in PACS images, the sternum region density in CR and AMBER images, the heart region density in CR AMBER images, the diaphram region density in AMBER and conventional images. 2. The evaluation measured surface dose were appeared orderly lesser dose at the AMBER images (spine 21 mR, lung 2mR, heart 12mR, apex 6mR) than the conventional images(32 mR), CR images(38mR) and PACS images(81mR). 3. The anatomical physical evaluation marks were taken the highest points at CR images(88.3), and orderly PACS images(82), AMBER images(79.2) and conventional images(65.2). 4. It is exposured with lesser surface dose at the analog images, but analog images leaves much room for image quality improvement, and digital images demand for lesser exposure surface dose, although excellent image quality.

Effect of Reducing Scattering Radiation Exposure of Medical Staffs When Additional Shielding is Used in Interventional Radiology (중재적 방사선시술에서 부가 차폐체 사용 시 종사자의 산란선 피폭 감소효과)

  • This article is designed to look into the radiation exposure dose to each body part and the shielding effect for workers using an additional shielding to reduce their radiation exposured by scattering radiation which is generated in a space between the operating table and lead curtain during interventional radiology(IR) procedures. After placing a human phantom on the table of SIEMENS' angiography machine, the following measurements were taken, depending on the presence of an additional shield of lead equivalent of 0.25 mmPb, manufactured for this purpose: dose to gonad, dose to an area where the personal dosimeter is placed, and dose to an area of eye lens is located. An ion chamber(chamber volume 1,800 cc) was utilized to measure scattering radiation. The two imaging tests were carried out as follows: fluoroscopy of the abdomen (66 kV, 100 mA, 60 seconds) and of the head (70 kV, 65 mA, 60 seconds); and digital subtraction angiography(DSA) of the abdomen (67 kV, 264 mA, 20 seconds) and of the head (79 kV, 300 mA, 20 seconds). In all the experiments, the shielding efficiency of the gonad position was the largest at 59.8%. In case an additional shielding was used as protection against scattering radiation that came through the operating table and the lead curtain during an IR, the radiation shielding efficiency was estimated to be up to 59.8%, leading to a conclusion that its presence may effectively reduce the radiation exposure dose of medical staffs.

  • Adaptive response induced by low dose ${\gamma}-ray$ irradiation in human peripheral lymphocytes was examimed. Human lymphocytes were exposured to low dose of ${\gamma}-ray$ (priming dose, 0.01Gy) followed by high dose (challenging dose, 1.5Gy) after various time intervals (4, 7, 20 hours). Frequencies of micronuclei were enumerated in both primed and unprimed groups. Maximum reduction in frequency of micronuclei was observed when challenging dose irradiation was followed by priming dose after 4hr incubation period. When challenging doses were irradiated 7 or 20hr after priming dose, frequencies of micronuclei were reduced slighty. However, these reduction were not statistically significant. In this study, human peripheral lymphocytes were irradiated at Go phase and they showed adaptive response induced by low dose radiation. Since micronucleus assay is relatively simpler and faster than other methods, it may be a good tool for evaluating radiation-induced adaptive responses.

Comparison of the Mercury Levels Between Maternal and Fetal Organs in Pregnant Fisher-344 Rats (염화메틸수은에 폭로된 임신 흰쥐에서 모체와 태자의 장기에 축적된 수은농도의 비교)

  • The purpose of this study was to determine the mercury accumulated at maternal and fetal organs, and compare its levels between maternal and fetal organs on day 20 of gestation, in pregnant Fisher-344 rats which given orally methylmercuric chloride on day 7 of gestation. Pregnant rats were divided four groups by dose: control group, and methylmercuric chloride treatment groups of 10, 20 and 30 mg/kg, respectively. The results obtained are as follows: I The mercury concentrations in maternal organs were the highest in kidney, and followed by blood, spleen, liver and brain. 2. The slopes of regression equation among mercury dose levels in maternal organs were as follows: Kidney 3.62 (r$^2$=0.943), Blood 2.75 (r$^2$=0.941), Spleen 2.49 (r$^2$=0.990), Liver 1.13 (r$^2$= 0.949), Brain 0.33 (r$^2$=0.984). 3. The mercury concentrations in fetal organs and placenta were the highest in liver, and followed by kidney, placenta and brain. 4. The slopes of regression equation among mercury dose levels in fetal organs and placenta were as follows: Liver 1.79 (r$^2$= 0.968), Kidney 0.79 (r$^2$= 0.976), Placenta 0.68 (r$^2$= 0.920), Brain 0.52 (r$^2$= 0.978), All Body 0.58 (r$^2$= 0.941). 5. As to the mercury levels in kidney, dams were 4.8~14.9 times higher than fetus. But as to the mercury levels in liver and brain, fetus were 1.6~2.5 and 1.5~1.9 times higher than dams. In conclusion, the mercury which exposured to pregnant rats can easily pass through the placenta and accumulated in fetus, especially higher in fetal liver and brain.

  • To make a comparative analysis for clinical application of total hip replacement arthroplasty(THRA) using fluoroscopy, we have performed total hip replacement arthroplasty making 30 patients an object with general radiography and fluoroscopy respectively. The results are as follows : 1) Reexamination rate was 80% of patients and mean 3.9 sheets in the general radiography, where as it was 46.7% and 0.37 sheets in the fluoroscopy(p<0.01) 2) Add examination was 2 cases in the general radiography, but fluoroscopy was no add examination. 3) The total film sheet used was mean 10.16 in the general radiography and 6.73 in the fluoroscopy. 4) In the cause of reexmination and add examination, inaccurate position of patient accounted for 72.6% in the general radiography and poor exposure condition accounted for 45.5% in the fluoroscopy. Total hip replacement arthroplasty using the fluoroscopy decreased reexamination and add examination rate, for these reasons, this method was effective abatement of pain, exposured radiation dose, and examination time and so on.

  • Chelidonii Herba (Baekgulchae in Korean: CHE), a commonly used herb in Korea, Japan and China, is widely used in the treatment of stomach cancer, jaundice, gastric ulcer, edema and pain of stomach. In the present study, we demonstrated that CHE induces apoptosis in AGS cells, human stomach adenocarcinoma cell line. One of the most important recent advances in cancer research is the recognition that apoptosis plays a major role in both tumor formation and treatment response, In this study, CHE caused a decrease of viability in AGC cells. When AGS cells were treated with CHE, cells showed dose-dependent manner apoptotic cell death. Increased apoptotic cell death, exposured to CHE, resulted from induction of Bad translocation to mitochondria, cytochrome-c release from mitochondria to cytosol, activation of caspase-3, 8, 9, and PARP cleavage. These results suggest that CHE may be potential therapeutic approach in the clinical management of stomach adenocarcinoma.

Effect of Radix Polygoni Multiflori on Spinal Motor Neurons Damaged by Methylmercuric Chloride (메틸수은으로 손상된 척수신경세포에 대한 하수오의 영향)

  • In order to evaluate the cytotoxicity of methylmercuric chloride(MMC) in cultured spinal motor neurons of neonatal mouse, cell viability was measured by MTT assay in spinal motor neurons treated with 1-30 μM MMC for 48 hours. And also, the protective effect of Radix Polygoni Multiflori(RPM) was examined by cell viability in these cultures. Cell viability was significantly decreased in dose-dependent manner after cultured cells were exposured to 20 μM MMC for 48 hours. Protective effect of RPM on MMC-mediated toxicity was very effective in these cultures. From above the results, it suggests that MMC has toxic effect in cultured mouse spinal motor neurons and herb extract such as RPM is very effective in blocking the neurotoxicity induced by MMC.

  • To establish prediabetes in vitro/ model concerning the etiology of Insulin Dependent Diabetes Mellitus (IDDM) in cellular level we have designed experimental prediabefic model in pancreatic beta cells. RINm5F, HIT-T15 and isolated rat islets were chosen as pancreatic beta cells. Since interleukin-$1{\beta}$-induced beta cell cytotoxicity has been implicated in the autoimmune cytotoxicity of IDDM, we used inteleukin-$1{\beta}$ as diabetogenic agent. For establishment of prediabetic in vitro model, the degree of beta cell deterioration was determined by cell proliferation, insulin release and morphological appearance. Cell proliferation, insulin release and morphology were changed dose-dependently in condition that inteleuldn-$1{\beta}$ was exposured to pancreatic beta cells. The concentration and exposure time of interleukin-$1{\beta}$ to set up prediabetic model in beta cell lines and isolated rat islets were 100${\sim}$1000U/ml, 48hr. And 25${\sim}$100U/ml, 48hr, respectively.
