Hepatotoxicity in treatment of canine dermatophytosis with ketoconazole (피부사상균 감염개에서 Ketoconazole 경구투여시의 간독성에 관한 연구)

  • The purpose of this study is to compare hepatotoxicity of each treatment for dermatophytosis; one is the administration of the ketoconazole only and the other, ketoconazole with diphenyl-dimeththyl-dicarboxylate. Have chosen the range of 14-24 months of healthy dogs divided by two groups (group 1 and group 2) for the experiment of which test proved positive in dermatophytosis diagnosis and showed normal reaction in terms of physical examination, blood chemistry and especially of liver function. Group 1 was administrated ketoconazole orally at 10 mg/kg/day and of same dose of ketoconazole with diphenyl-dimethyl-dicarboxylate for group 2. After administering, we have tested two groups by blood collecting every one week in order to check the differences of hepatotoxicity state through AST, ALT and r-GTP, the barometers of liver function which lasted for 12 weeks. Moreover, tested Indocyanine Green (ICG), known as susceptible gauge of function of excretion before starting the experiment and tested ICG as well after 12 weeks. The experiment of result the value of group 1 in AST, ALT and r-GTP has been highly rised after administering ketoconazole for 10 weeks meanwhile, of group 2 has shown a steady state troughout the whole experiment. For ICG test, we injected 0.5 mg/kg of ICG into a vein for both groups and tested the retention rate at regular interval of 15, 30, 45 minutes. The results of retention rate in two groups were similar to before the drug administration. However, after 12 weeks the retention rate of group 1 has been delayed, on the other hand, retention rate of group 2 were a steady state. In conclusion, the administration of ketoconazole only for a long period of time induced hepatotoxicity where as, the administration of ketoconazole with diphenyl-dimethyl-dicarboxylate didn't induce hepatotoxicity. Therefore, when doctors prescribes for a dog with dermatophytosis should not administrate ketoconnazole itself but with diphenyl-dimethyl-dicarboxylate and one who has abnormal condition of liver function should not be prescribed ketoconazole treatment. If there is a case needed to prescribe ketoconazole treatment, the regular monitoring should be accompanied by at the same time.

Effects of Cheonggukjang Added Phellinus linteus myceria on Lipid Metalbolism in Adult Female Rats (상황버섯균사체청국장이 고지혈증을 유도한 암쥐의 지질대사에 미치는 영향)

  • This study was performed to investigate the effects of Cheonggukjang (traditional soybean food, CK) and Cheonggukjang added with Phellinus linteus myceria (CKP) on the lipid metabolism, growth, food intake, and food efficiency ratio(FER) in adult female rats. Sprague-Dawley rats were fed a high fat diet (control diet: 0.1% cholesterol, 10% Lard) for 4 weeks. The rats were randomly assigned to each treatment group: control, and two kinds of CK and CKP (powders of CK and CKP as dietary protein sources). After 4 weeks of experimental diet consumption, the body weights and the uterine fat pad weights of the CK and CKP diet groups were more significantly decreased than those of the control diet group. The hepatic cholesterol, triglyceride and total lipid levels were significantly lower in both the CK and CKP diet fed groups than those in the control group. The concentrations in serum triglyceride and LDL-cholesterol were significantly decreased in the CK and CKP diet groups compared to those in the control group. Fecal total lipid, moisture and wet weight excretion in the CK and CKP diet groups were more increased than those in the control group. These results showed that feeding of cheonggukjang and cheonggukjang added with Phellinus linteus myceria both the triglyceride and LDL-cholesterol in serum, as well as the triglyceride and cholesterol in the livers of the rats.

Capsaicin Intake Estimated by Urinary Metabolites as Biomarkers (뇨중 대사체 정량에 의한 capsaicinoid의 생화학적 섭취지표 개발 연구)

  • Reliable data on the exposure of capsaicin, which evokes hot sensation in hot red pepper, are important prerequisites for studying biological functions of capsaicin in human body since its roles are controversial according to animal and epidemiological studies. To get accurate data, the use of urinary biomarkers was considered as a measure of internal exposure of capsaicin. After 2-day-capsaicin depletion five to six women in their 20's were instructed to eat 60 or 80 g/day hot red pepper soybean paste as a capsaicin source with meal which did not contain capsaicinoid, and to collect their urine separately. HPLC conditions were set to detect capsaicinoid and urinary metabolites at the same time. Most of capsaicinoid were excreted in the form of 4-hydroxy-3-methoxybenzoic acid in three subjects at the highest dose. This result suggests the oxidation of the 4-hydroxy3-methoxybenzaldehyde, the hydrolysis product of capsaicin, is the major metabolic pathway in the human body, whereas the reduction of the aldehyde is the major route in rats. However, neither any metabolite nor the parent compound was ever shown in two of the subjects reflecting large individual differences of capsaicin absorption and/or biotransformation.

Behavior of the herbicide metolachlor in carps (제초제 metolachlor의 잉어체내 행적)

  • In order to investigate the behavior of the herbicide metolachlor [2-chloro-6'-ethyl-N-(2-methoxy-1-methylethyl)aceto-O-toluidide] in fish, carps (Cyprinus carpio L.) were exposed to the herbicide at $LC_{10}$(1.93 mg/L) for 4 days. Metolachlor dissolved in water was absorbed rapidly into carps to mark the maximum concentration 6 hours after exposure. The amounts of the $^{14}C$-metolachlor and its metabolites absorbed in gall were much higher than those in the other parts, strongly suggesting that biliary excretion involving enterohepatic recirculation could be an important route for the elimination of metolachlor. The $^{14}C$-radioactivity distributed into aqueous phase fraction in test water and in carp extract was increased in time-dependent manner. Extraction rate of $^{14}C$ absorbed in carp tissues was decreased remarkably up to 6 hours after exposure, suggesting that the possible polar metabolites of metolachlor were transformed into the conjugates to form non-extractable bound residues.

Extracts of Housefly Maggot Reduces Blood Cholesterol in Hypercholesterolemic Rats (고콜레스테롤 랫드에서 파리유충 추출물의 혈액지질 감소기전)

  • The aim of this study was to evaluate the biological mechanism of orally administered ethanolic extract of fly maggot(EM) on hypocholesterolemic rats fed a high-cholesterol diet. Sprague Dawley male rats were divided into four groups (EM dose control=0, 5.0, 7.0, and 9.0 mg/100 g BW) and were treated for 6 weeks. EM groups revealed a significant reduction in serum triglyceride, total cholesterol, and LDL-C when compared with the control group(p<0.05). HMG-CoA reductase activity in EM groups were lower than those of the control group, but total sterol, neutral sterol, and bile acid excretion were increased in EM groups when compared with the control group(p<0.05). To identify the biological mechanism of EM towards the hypocholesterolemic effect, sterol response element binding proteins (SREBPs) and the peroxisome proliferator-activated receptors ($PPAR{\alpha}$ transcription system were determined in rats fed a high-cholesterol diet. It was discovered that EM suppress the expression of SREBP-$1{\alpha}$ and SREBP-2 mRNA in the liver tissues of high-cholesterol diet fed rats, while simultaneously increasing the expression of $PPAR{\alpha}$ mRNA(p<0.05). This finding indicates that EM may have hypocholesterolemic effects in rats fed a high-cholesterol diet, by regulating cholesterol metabolism-related biochemical parameters and SREBP-$1{\alpha}$ SREPB-2 and $PPAR{\alpha}$gene expression.

Cholesterol Contents of Pork Fed Dietary β-Cyclodextrin (BCD를 섭취한 돼지의 부위별 콜레스테롤 함량)

  • This study demonstrates that pork cholesterol levels are reduced in fattening stage swine fed $\beta-cyclodextrin({\beta}-cyclodextrin)$. The study subjects were 120 swine fed their respective chow diets containing 0, 5, 7, or 10% $\beta$-cyclodextrin for 35 consecutive days. Plasma total lipids, triglyceride and total cholesterol of the $\beta$-cyclodextrin treated group were significantly lower than those of the control group (p<0.05). The levels of plasma lipid were significantly decreased by 63.22 mg, 73.98 mg, and 82.12 mg in the fattening swine group fed $\beta$-cyclodextrin at 5%, 7%, and 10%, respectively, compared to those in the control group (p<0.05). When 5, 7, and 10% $\beta$-cyclodextrin was administered to fattening swine, the triacylglyceride levels were decreased by 56.24 mg, 55.48 mg, and 60.02 mg, and total cholesterol concentration was reduced by 25.05 mg, 27.17 mg, and 30.19 mg, respectively, compared to those in the control group (p<0.05). Excretion of total steroid significantly (p<0.05) increased with the increasing amount of $\beta$-cyclodextrin supplementation. The cholesterol levels of swine back fat, belly, loin, and ham were significantly decreased with increasing $\beta$-cyclodextrin supplementation (p<0.05). The pork cholesterol was significantly (p<0.05) reduced by 15.31% in the $\beta$-cyclodextrin treated group, compared to that of the control group. These results suggest that feeding $\beta$-cyclodextrin to fattening swine may produce novel functional pork with low cholesterol levels.

Comparison Between Face Color Change and Its Recognition Difference on Asian: Korean, Indonesian and Vietnamian (아시아인의 얼굴색 변화와 인지도간 상관성 비교 : 한국인, 인도네시아인, 베트남인)

  • Various indicators representing skin characteristics such as skin hydration, sebum excretion rate, lightness, and pH are different depending on environmental and genetic factors. However, they are absolute skin indicators and are different from skin characteristics that a person recognizes. Based on this fact, many recent studies have been mainly conducting researches on perspective changes according to changes of absolute skin. This study was proposed not only to find out differences on skin colors of asian by nations, but also to find out whether there was any difference in skin brightness they perceive depending on actual skin color changing. As many as 410 subjects of three Asia nations were participated in this study, and investigated their responses on skin brightness using questionnaire, which was answered their skin color in three different levels. It was also were analyzed how actual skin brightness were changed depending on their perceived skin color changes of subjects. There was a trend showing that the brightness of the actual skin color was increased when participants felt their skin color got brighter regardless of their nationalities. However, there were some differences in color between perceived color and actual color. In addition, there was a different aspect by nations in changes of skin redness and skin yellowness. In conclusion, it was revealed that factors which help people to perceive their own skin brightness were not based on absolute skin brightness, but on different criteria depending on where they are from.

The Absorption and Metabolism of Fenobucarb and Carbofuran by Susceptible and Carbamate Insecticide-selected Strains of the Brown Planthopper (Nilaparvata lugens Stal) (Fenobucarb 및 Carbofuran의 저항성 벼멸구 체벽 투과량과 체내대사에 관한 연구)

  • Cuticular penetration and detoxication as mechanisms of resistance to the carbamate insecticides in fenobucarb-selected($R_{f}$) and carbofuran-selected($R_{c}$) strains of the brown planthopper (N. lugens Stal) were investigated. Rates of penetration were not significantly different in the susceptible and resistant strains. However, total amount of excretion of the $R_{f}$ and $R_{c}$ strains were much larger than that of the susceptible strain. Fenovucarb and carbofuran were in vivo metabolizd much faster in the $R_{f}$ strain than in the susceptible strain. OSBP(o-sec-butyl phenol) and 3-ketocarbofuran phenol were invitro the major metabolites of fenobucarb and carbofuran in the brown planthopper, respectively. Total amount of the two major metabolites were produced abotu 2 times larger in the $R_{f}$ and $R_{c}$ strains compared to the susceptible strain. OSBP and 3-ketocarbofuran phenol were not so toxic to the brown planthopper ($LD_{50}$ >100 $\mu\textrm{g}$/g hopper). Based on our data, detoxication plays a large role in resistance to fenobucarb and carbofuran in the resistant strain of BPH, although several resistance factors maybe involved.

Systematic Review : Comparative Safety and Efficacy of Mono- and Combination Therapy of Anti-hypertensive Agents Acting on the Renin-Angiotensin System (레닌-안지오텐신계에 작용하는 항고혈압 약제의 단독요법과 병용요법의 안전성 및 유효성에 대한 체계적 문헌고찰)

  • Given that single blockade with angiotensin-converting enzyme inhibitors (ACEIs) or angiotensin receptor blockers (ARBs) can achieve only partial and undurable suppression of the Renin Angiotensin System (RAS), it has been hypothesized that dual blockage would be more beneficial in the management of blood pressure (BP) reduction and prevention of progressive chronic kidney disease (CKD) than either agent alone. Thus, it has been suggested that the combination of an ACEI and an ARB might provide renal benefits to hypertensive patients over and above BP reduction. However, this might also expose patients to additive or synergistic side effects. We attempted to conduct a systematic review to evaluate the benefits and harms of combination therapy in hypertensive patients with or without kidney diseases. MEDLINE and KoreaMed were searched for relevant randomized clinical trials in adult hypertensive patients with or without diabetes (restricted to 1997, limited to trials published in English). Results were summarized using the random-effects model, and between-studies heterogeneity was estimated with $I^2$. A final analysis of ten trials (23,928 patients) revealed that the combination of an ACEI and an ARB reduced blood pressure (SBP/DBP) by 3.95/2.02 mmHg (95% confidence interval [CI], -4.38 to -3.53/-2.33 to -1.71) compared with ACEI monotherapy, and 2.83/2.64 mmHg (95% CI, -3.25 to -2.41/-4.95 to -0.33) compared with ARB monotherapy. Eight trials (391 patients) demonstrated a significant reduction in 24h-proteinuria (weighted mean difference, 0.16 g/day, 95% CI, -0.26-0.05), but they did not translate into an improvement in GFR. Tests for heterogeneity showed no difference in effect among the studies. The combination therapy reduced proteinuria by 30% (95% CI, 23% to 37%) and 39% (95% CI, 31% to 48%) compared with ACEI monotherapy and ARB monotherapy, respectively. However, in patients who had proteinuria more than 0.5 g/day, the combination therapy failed to show significant reduction in urinary protein excretion. The current cumulative evidence suggests that diabetic patients with proteinuria on dual RAS blockade have an increase risk of adverse events such as hyperkalemia, hypotension, and so on, compared with ACEI or ARB alone. It is, therefore, proposed that the combination therapy should not be routinely used for the treatment of hypertension with or without compelling indications.

Analysis of Metabolism and Effective Half-life for Tritium Intake of Radiation Workers at Pressurized Heavy Water Reactor (중수로원전 종사자의 삼중수소 체내섭취에 따른 인체대사모델과 유효반감기 분석)

  • Tritium is the one of the dominant contributors to the internal radiation exposure of workers at pressurized heavy water reactors (PHWRs). This nuclide is likely to release to work places as tritiated water vapor (HTO) from a nuclear reactor and gets relatively easily into the body of workers by inhalation. Inhaled tritium usually reaches the equilibrium of concentration after approximately 2 hours inside the body and then is excreted from the body with a half-life of 10 days. Because tritium inside the body transports with body fluids, a whole body receives radiation exposure. Internal radiation exposure at PHWRs accounts for approximately 20-40% of total radiation exposure; most internal radiation exposure is attributed to tritium. Thus, tritium is an important nuclide to be necessarily monitored for the radiation management safety. In this paper, metabolism for tritium is established using its excretion rate results in urine samples of workers at PHWRs and an effective half-life, a key parameter to estimate the radiation exposure, was derived from these results. As a result, it was found that the effective half-life for workers at Korean nuclear power plants is shorter than that of International Commission on Radiological Protection guides, a half-life of 10 days.
